Objective To establish osteosarcoma vaccine by the LM9 osteosarcoma derived from C3h mice hybridized with activated B lymphocytes and study its biological behavior and the antitumor efficacy. Methods The LM9 osteosarcoma derived from C3h mice was fused with LPS activated B lymphocytes by using 50%PEG. The fused cells was selected by HAT medium and cultured in vitro, and the biocharacter and efficacy of the fusion vaccine were investigated. Results In contrast to LM8, the fused cells grew significantly slowly in vitro. All the mice under the protection of fused vaccine survived without tumor (8/8), while all the mice in the control group succumbed to the tumor with no survival (8/8), 75%of the mice inoculated subcutaneously with cell fusion vaccine survived without tumor burden after implantation of LM8 cells subcutaneously. The mice in the control group developed tumors and died within 45 days without any exception. Conclusion It is possible that the LM9 osteosarcoma biology characteristics change after fused with LPS activated B lymphocytes. Cell fusion osteosarcoma vaccine could produce prophylactic and therapeutic effects in mice.

Objective To investigate the effect of tourniquet compression on axonal transport in sciatic nerve of rats. Methods Twenty-four 12-week old male SD rats weighing 250-300 g were randomly divided into 4groups according to the duration of tourniquet compression(n=6 each):1,2,4 and 12 h.The tourniquet was applied to the middle 1/3 of thigh. In each animal whether the left or right thigh was compressed was determined by a flip of coin. The tourniquet was released for 10 min after every hour of compression.A 3-cm segment of sciatic nerve was removed at the end of tourniquet compression(1.5 cm proximal and 1.5 cm distal to the site of compression).Immuno-histochemistry was used to measure the expression of insulin-like growth factor-1(IGF-1)in the sciatic nerve. The ratio of average optic density of the compressed sciatic nerve to that of control was used to estimate the degree of IGF-1 accumulation. The regression equation of the interaction between the duration of compression and accumulation of IGF-1 was analyzed. Results There was significant accumulation of IGF-1 in the sciatic nerve proximal to the compressed site. The accumulation increased with the duration of compression. There was no significant accumulation of IGF-1 in the sciatic nerve distal to the compressed site. The regression equation of the interaction between the duration of compression(X)and accumulation of IGF-1(Y)was Y=0.422X+0.887.Conclusion Tourniquet compression of sciatic nerve can inhibit axonal transport. The accumulation increases with the duration of compression.

Recent Advances in Ophthalmology 2000;20(5):353-355
Objective To study the value of comprehensive examination of visual electrophysiology in prognosis of visual function after cataract operation. Methods 315 patients with cataract(408 eyes) underwent preoperative routine examination with pattern reversal visual evoked potential(PVEP), red and blue flicker electroretinogram(ERG) and oscillatory potential (OPs). Normal control group included 162 cases (266 eyes). All of the ocular fundi were examined postoperatively and corrected visual acuities were obtained 3 months after operation. Results Postoperative corrected visions ≥0.5(Ⅰ) and <0.5(Ⅱ) in case group compared with that in control group, amplitudes of PVEP P100-wave decreased significantly and PVEP P100-wave latency times prolonged significantly in group Ⅰ and Ⅱ. There was much more prominent changes in P100-wave of group Ⅱ due to other factors influencing visual function. There wasn't significant difference of the amplitudes and the latency times of a-wave and b-wave of Red-Blue ERG in group Ⅰ and the control group preoperatively. The amplitudes of a-wave and b-wave of Red ERG decreased, the latency times of a-wave and b-wave of Red ERG prolonged, the amplitudes of a-wave and b-wave of Blue ERG decreased but latency times didn' t prolong in group Ⅱ. The value of OPs decreased in groupⅡ compared with that of the control group, which was related to the retinopathy due to blood circulation obstruction. Conclusion Combined examination of visual electrophysiology can estimate visual function thoroughly. The prognosis will be good if various examinations are normal before operation, but will not be good if PVEP P100 latency time prolong, the amplitudes of a-wave and b-wave of Red ERG decrease and the latency time prolong, the amplitudes of a-wave and b-wave of Blue ERG decrease and the value of OPs decrease.

Objective: To further understand the role of folic acid supplements rivaling MTHFR gene silencing in pathogenesis of NCLP, RNA interference (RNAi) was applied to knock down MTHFR in mouse embryonic palatal mesenchymal (EPM) cells. Methods: MTHFR ShRNA expression vector were transfected into the primary cultured EPM cells. MTT was used to observe cell proliferation after MTHFR gene silencing. FCM was used to observe cell cycle after MTHFR gene silencing. Results: The results showed the cells proliferation had an inequality amelioration after using folic acid supplements in MEPM cells with MTHFR gene silencing. Using folic acid supplements rivaled the effect of MTHFR gene silencing had a dose-dependent manner. Using 20 μg/ml folic acid supplements could improve the cell proliferation to achieve normal level of cell proliferation. Conclusion: MTHFR gene is an important candidate gene of NCL/P. Using folic acid supplements could prevent teratogenic MTHFR gene silencing for embryonic palate development.
