2.The Effects of Human Endostatin on the Growth of Ovarian Cancer Cells A2780 in vitro and in vivo and the Possible Mechanism

Objective To investigate the effects of human endostatin(hES)on ovarian cancer cells A2780 growth in vitro and in vivo and the possible mechanism.Methods The coding region for hES was amplified by PCR from human liver tissue and then subcloned into pcDNA3.1 vector.The growth rate of A2780 cells transfected with hES was observed.A2780 cells(1× 10~7/ml)were inoculated subcutaneously into 20 nude mice,and the mice were randomly divided into two groups:the hES-trans-fected group(group A,n=10)and the control group(group B,n=10).One month later,the size of subcutaneous tumor was measured,and the tumor inhibition rate was calculated.The specimens were stained with HE for the histological analysis.Cell apoptosis in ovarian neoplasm transplantation tissues was determined by flow cytometry.The expression of hES and Bcl-2 was detected by Western blot.Results The growth rate of the hES-transfected ovarian cancer cells A2780 was significantly decreased.In group A,growth of tumor in nude mice was significantly slowed as compared with group B(P<0.01),with the tumor inhibition rate being 74% in group A.The tumor necrosis was increased and the basophilia stain of nucleus decreased significantly in group A as compared with group B.The expression of hES was increased significantly,but that of Bcl-2 was decreased significantly in group A.Flow cytometry revealed that hES transfection could significantly incyease the apoptosis rate of tumor cells(P<0.05).Conclusion Transfection of hES could suppress the growth of ovarian cancer cells A2780 in vitro and in vivo,which may be related tO the promotion of apoptosis.
3.Clinical features and mutation analysis of LRRK2 gene in patients with autosomal dominant familial Parkinson's disease

Objective To investigate the clinical features and LRRK2 gene mutation in patients with autosomal dominant familial Parkinson's disease (PD). Methods The clinical features of 16 autosomal dominant familial PD probands were analyzed in terms of age at onset, onset symptoms, UPDRS scores, response to the levodopa treatment and drug-induced dyskinesia. The LRRK2 gene exons 5,13,31,32,35,37,41 and 48 of 16 probands were sequenced after polymerase chain reaction. The novel mutation was further screened in 24D sporadic PD patients and 214 controls using PCR-RFLP for the genotypo frequency analysis. Results Clinically, most of 16 probands had late-onset age. Resting tremor (9patients, 56. 25%,t=0.558,P=0.679)and bradykinesia (9 patients,56.25%,t=0.369,P=0.454)were common onset symptoms followed by rigidity(6 patients,37.50%,t=1.324,P=0.735)and postural instability(5 patients,31.25%,t=2.369,P=0.956).Majority of them had good response to levedopa treatment and rare occurrence of drug-induced dyskinesia. Among the 16 autosomal dominant familial PD probands,6 variants were identified:c.457 T>C(Leu153Leu),c.1432 G>T(Asp478Tyr),c.5457 T>C(Gly1819Gly),c.7153 G>A(Gly2385Arg),IVS31+28 T>G and IVS37+162 T>C. The c.1432G>T(Asp478Tyr)variant was a novel mutation and it was not detected in 240 sporadic PD patients and 214 controls. The reported mutations associated with the PD, such as Arg1441 Cys/Gly/His, Arg1514Gln, Tyr1699Cys, Ile2012Thr, Gly2019Ser and Ile2020Thr,were not found in our study. Conclusions The autosomal dominant familial PD patients present with classical symptoms of PD and bear the LRRK2 variantsAsp478Tyr and Gly2385Arg.

The new pattern mainly deals with the cultivation of the students' creativity by reforming and exploring the plan,the content,the method of experimental teaching and the ways of checking the students' work, adding general and designing experiment,and working out the PPT of the experiments.The result shows the new experimental teaching pattern contributes to the cultivation of the students'abilities of performing experiment, the ways of thinking, creativity and comprehensive analysis. It's better than the traditional experimental teaching pattern.A new medical microbiology experimental teaching systerm which is suit to the students majoring in anaesthesia has been established.
5.Influence of β-amyloid peptide on expressions of synaptophysin, dynamin Ⅰ and adaptor protein 180 in human neuroblastoma SH-SYSY cells

Objective To study the effect of β-amyloid peptide (Aβ1-42) on expressions of synaptophysin (Syn), dynamin Ⅰ (Dyn Ⅰ) and adaptor protein 180 (AP180) in human neuroblastoma SH-SY5Y cells. Methods Human SH-SY5Y neuroblastoma cells were treated with 0.01, 0.1, 1, 2 and 5 μmol/L Aβ1-42, and control cells were given no treatment. MTT [3-(4,5-dimethylthiazol- 2- yl) - 2, 5-diphenyltetrazolium bromide]reduction of the cells was measured by spectrophotometry. The protein levels of Syn, Dyn Ⅰ and AP180 in SH-SY5Y cells treated with 0.5 and 1 μmol/L Aβ1-42 were surveyed by Western blotting. The mRNA levels of Syn, Dyn Ⅰ and AP180 were detected by Real-time PCR in SH-SY5Y cells treated with 1 μmol/L Aβ1-42. Results SH-SY5Y cells showed obviously decreased reduction rates of MTT after exposure to Aβ1-42(0.1 μmol/L) as compared with the controls (P<0.05), and dose-dependent negative correlation was noted in these SH-SY5Y cells. The protein level of Dyn Ⅰ in cells treated with 0.5 μmol/L Aβ1-42 was significantly decreased as compared with that in controls (P<0.05). The protein and mRNA levels of Syn and Dyn Ⅰ in cells treated with 1 μ mol/L Aβ1-42 Were obviously decreased as compared with those in controls (P<0.05), but the levels of AP180 were not changed. Conclusion Aβ1-42 reduces the levels of Syn and Dyn Ⅰ in SH-SY5Y cells, which might be a mechanism in eonnection with cognitive deficit of AD.
6.Effect of quercetin on neural stem cell proliferation in the subventricular zone of rats after focal cerebral ischemia-reperfusion injury.

OBJECTIVETo study the effect of quercetin on the proliferation of neural stem cells in the subventricular zone (SVZ) of rats after focal cerebral ischemia.
METHODSAn adult rat model of middle cerebral artery occlusion (MCAO) model was established by placement of an intraluminal filament at the origin of the MCA. Quercetin was administered intraperitoneally in the rats at a dose of 50 mg/kg every 3 days starting at 6 h after MCAO, and BrdU (50 mg/kg daily) was also injected intraperitoneally starting at 4 h after MCAO. BrdU-positive cells in the SVZ were counted at 7, 14 and 21 days after MCAO.
RESULTSCompared with the sham-operated group, the rats in the ischemic model group showed significantly increased BrdU-positive cells in the ipsilateral SVZ 7 days after MCAO, reaching the peak level on day 14 and beginning to decrease on day 21 (P<0.05). The number of ipsilateral BrdU-positive cells in quercetin group was significantly greater than that in the model group on days 7, 14 and 21 (P<0.05), and maintained the high level on day 21.
CONCLUSIONQuercetin can maintain a high level of neural stem cell proliferation in the SVZ after focal cerebral ischemia in adult rats.

7.Analysis of varieties and standards of Leguminosae plants used in Tibetan medicine.

In this paper, the domestic varieties and quality standard of Leguminosae medicinal plants used in Tibetan medicine were analyzed. The results showed that there were 36 genera and 142 species (including varieties), as well as 64 medicinal materials varieties of Leguminosae plants were recorded in relevant literatures. In relevant Tibetan standards and literatures, there are great differences in varieties, sources, used parts, and efficacy of medicinal plants. Among them, about 38.0% (including 54 species) of the endemic plants, about 25.4% (including 36 species) of the original plants have medicinal standard legal records, except 9 kinds of traditional Chinese medicine general quality standard more fairly completed, the most varieties have only description about characters, identification, etc. Therefore it is necessary to reinforce study for the herbal textual, resources and the use present situation, chemical components and biological activity, quality standard, medicinal terms specification, to promote establishment of quality standard system for variety-terminologies-sources of Tibetan medicinal plants.

8.Analysis of varieties and standards of Scrophulariaceae plants used in Tibetan medicine.

In this paper, the popular domestic varieties and quality standard of Scrophulariaceae plants used in Tibetan medicine were analyzed. The results showed that there were 11 genera and 99 species (including varieties), as well as 28 medicinal materials varieties of Scrophulariaceae plants were recorded in the relevant literatures. In relevant Tibetan standards arid literatures, there are great differences in varieties, sources, parts, and efficacies of medicinal plant. Among them, about 41.4% (including 41 species) of endemic plants, about 15.2% (including 15 species) of the original plants have medicinal standard legal records, except the medicinal materials of Scrophalaria ningpoensis, Lagotis brevituba, Picrorhiza scrophulariiflora, Veronica eriogyne general, most varieties have not completed quality standard. Consequently it is necessary to reinforce the herbal textual, resources and the use present situation investigation, the effects of the species resources material foundation and biological activity, quality standard, specification the medical terms of the plants, and promote Tibetan medicinal vareties-terminologies-sources such as the criterion and quality standard system for enriching the varieties of Tibetan medicinal materials and Chinese medicinal resources.

9.Traightened on Chinese endemic seed plant species of medicine plants used in Tibetan medicine.

This paper is in order to discussion with the composition and characteristics of Tibetan medicine plant resources, and promote the reasonable protection and utilization of the resources of Tibetan materia medica. Statistical analysis of species, distributions, and others of Chinese endemic seed plant from Tibetan medicine plants and usually used in the clinic of Tibetan medicine. The results showed that there are 523 species (25%) of Chinese endemic seed plant, belonging to 65 families and 162 genera, in about 2 000 varieties of Tibetan medicine plants recorded in relevant literatures. There are 180 Chinese endemic seed plant species (28%) belonging to 42 families and 72 genera from 625 medicine plants usually used in the clinic of Tibetan medicine. Specifically, the most of these Chinese endemic seed plant species are characteristic crude drug used in Tibetan medicine, and mainly or only distributed in Qinghai-Tibet Plateau. And a few species of them were intersected with traditional Chinese medicines (TCM) and other ethnic medicines. In addition, about 10% are listed in China Species Red List. The Qinghai-Tibet Plateau is the most abundant areas of Areal-types of the Chinese endemic seed plant. This is the biological and ecological reason formation the characteristics of Tibetan medicine plant resources. Therefore, strengthen the research of Chinese endemic seed plants used in Tibetan medicine is great significance for the reasonable protection and utilization of Tibetan medicine plant resources.

10.Aging performances for resisting low-temperature of three dental Yttria-stabilized zirconia ceramic core materials.

BACKGROUNDThe low-temperature resistance aging performance of Yttria-stabilized tetragonal zirconia polycrystal (Y-TZP) is the key effective factor that influences the long-term success rate of prosthesis. The objective of this study was to test and compare the aging performances for resisting low temperature of Lava Frame, Cercon Smart, and Upcera Yttria-stabilized zirconia core materials, via analyzing the micro and the crystal phases of the materials, and measure the three-point bending strength and the fracture toughness.
METHODSThe three zirconia green bodies were prepared as 60 test samples for three-point bending strength and as 60 test samples for fracture toughness. The test samples for three-point bending strength and fracture toughness were assigned to five groups and were treated respectively for 0, 5, 10, 15, and 20 hours to observe the micro and the crystal phases of the test samples. Then the three-point bending strength and fracture toughness were tested by X-ray diffraction (XRD).
RESULTSThe m phase content of Lava Frame was raised from 7.70% to 13.01%; the m phase content of Cercon Smart was raised from 4.95% to 8.53%; and Lava Frame is raised from 10.84% to 35.18%. The three-point bending strengths of the three zirconia core materials were higher than 1100 MPa and the fracture toughness was higher than 3 MPa·m(1/2). The three-point bending strength and the fracture toughness of Upcra zirconia decreased the most, followed by Lava Frame, and then by Cercon Smart.
CONCLUSIONThe aging resistance sequences of the three zirconia core materials are, from strong to weak, Cercon Smart, Lava Frame, and Upcera.
