The molecular characterization of self-antigens expressed by human malignancies that are capable of elicitation of anti-tumor immune responses in patients has been an active field in hematology, oncology, and tumor immunology. More than 2000 tumor antigens have been identified. These significant progresses have led to the renaissance of tumor immunology and studies on novel anti-tumor immunotherapies in lymphomas, other hematologic malignancies and tumors. However, despite of the progress in the identification of these self-tumor antigens, current antigen-specific immunotherapies for tumors are far less satisfactory than that expected, which reflects the urgent need to improve our understanding on the basic principles underlying the selection of these self-tumor antigens. In order to develop more effective antigen-specific anti-tumor immunotherapies and to monitor the responses to these immunotherapies in patients with lymphomas and other malignancies, many additional questions need to be addressed. In this brief review, the progress in the identification of tumor antigens in lymphomas and other malignancies was outlined and the new principles of self-tumor antigens and their significance for future immunotherapies to these malignancies were summarized.

3.Effect of Ca2+-activated Chloride Channel Density on Gating Properties of Anoctamin 1

Objective To investigate the effect of channel density on the gating properties of Anoctamin 1(Ano1,TMEM16A)Ca2+?activated chlo?ride channel. Methods Ano1 expression plasmids were transiently transfected into HEK293 cells. High density and low density of Ano1 was ob?tained after expressing the protein for 24 h and 6 h,respectively. Electrophysiological recordings were performed in the whole?cell patch clamp con?figuration. The activation kinetics of current traces was fitted by exponentials. Results The current density was significantly higher in cells express?ing Ano1 for 24 h than those expressing Ano1 for 6 h(P<0.05). The activation of Ano1 current in cells with low density was well fit by a single expo?nential withτslow of 292.71±38.11 ms. The activation of Ano1 current in cells with high density was well fit by two exponentials withτfast of 47.78±4.58 ms andτslow of 385.74±71.44 ms. ANO1 current in cells with high density has a rapid active component(τfast)more than low density. There was no significantly different of theτslow between cells with high density and low density of Ano1(P>0.05). Conclusion Our findings suggested that chan?nel density regulates the gating of Ano1. High channel density promotes activation of Ano1.

5.Application experience of transumbilical abdominal wall within the three-channel single-incision laparoscopic combined cholecystectomy and appendectomy

Objective To explore the clinical valuation and feasibility of transumbilical abdominal wall within the three-channel single-incision laparoscopic combined cholecystectomy and appendectomy.Methods The clinical data of 40 patients who underwent transumbilical abdominal wall within the three-channel single-incision laparoscopic combined cholecystectomy and appendectomy from January 2011 to March 2013 were retrospectively analyzed.Results Forty patients were successfully completed surgery,no transfer laparotomy and death cases,surgery time was 60-80 min,average 70 min,without postoperative incision infection,wound bleeding,bile duct injury,appendix stump fistula and abdominal residual abscess,neither postoperatively used analgesics,1 month after without incision scar,cosmetic effect was remarkable.Half a year's follow-up,the excellent operation evaluation.Conclusions The performer should have accomplish laparoscopic operation skills,and master the indications and the principle.The method of transumbilical abdominal wall within the three-channel single-incision laparoscopic combined cholecystectomy and appedectomy is safe,feasible and valid.The postoperative wound is more minimally invasive and aesthetic effect is obvious.
6.The effects of diammine silver fluoride solution on demineralized dentine adhesion ability

Objective:To investigate the effect of silver diamine fluoride(SDF)on the bonding strength between dentine and glass ion-omer cement(GIC).Methods:1 2 extracted sound molars were prepared into dintine samples and distributed into sound dentine group and demineralized dentine group.According to the treatment methods,the samples in each group were respectively divided into 3 sub-groups:A(control group),B[coated with 38% Ag(NH3 )F2 ]and C(SDF treatment with additional lighting-curing)(n =20).Then a hand-mixed conventional glass ionomer cement Fuji IX was placed on the dentine surface.After 24 h,micro tensile bond strength test and scanning electron microscope (SEM)analysis were conducted.Results:The bonding strength of demineralized dentine was higher than that of sound dentine(P <0.01 ).SDF with additional lighting-curing treated dentine showed a higer bonding strength value than only SDF treated dentin(P <0.01 ).Conclusion:SDF may improve the bonding between dentine and GIC.
7.Inhibition of the vasculogenic mimicry formation by targeting fibroblast growth factor receptor in glioma

Objective To explore the effect of fibroblast growth factor receptor(FGFR)inhibitor BGJ398 on the vasculogenic mimicry(VM)formation of glioma cells. Methods The phosphor-FGFR(pFGFR)was de-tected by Western blot,the expressions of MMP2 and MMP14 were detected by Western blot and immunocytochem-istry;the VM formation of U87MG and U251MG was tested by tube formation assay;subcutaneously implanted tu-mor model in nude mouse was established and tumor sections were CD34/PAS double-stained to detect the forma-tion of VM in vivo. Results Western blot showed that pFGFR in the experimental groups decreased significantly (P < 0.05);western blot and immunohistochemical staining showed that the expression of MMP2 and MMP14 in the experimental groups decreased significantly compared to the control group. In the tube formation assay ,the tube formation of U87MG and U251MG cells were restrained. In the subcutaneously implanted tumor model ,the VM number of the experimental group(13.85 ± 3.96)was significantly lower than that in the control group(26.40 ± 5.06,P < 0.05). Conclusions In vivo and in vitro experiments confirmed that BGJ398 can inhibit the activa-tion of FGFR,and inhibit the VM formation of glioma cells. These indicate FGFR signaling pathway is involved in the formation of VM.
8.Effect of sulforaphane on human small cell lung cancer NCI-H446 cells proliferation, invasion and the activity of MMP-9

Objective To investigate the effects of sulforaphane (SFN) on proliferation and invasion of human small cell lung cancer NCI-H446 and the activity of matrix metalloproteinase (MMP) -9.Methods NCI-H446 cells were cultured with 0,25,50,100 μmol/L SFN for 24 ~ 72 h,then MTT assay was employed to detect cell proliferation.Chamber invasion assay was used to study the cell invasion,and gelatin zymography assay was implied in MMP-9 enzyme activity.Results After treatment of 25,50,100μmol/L SFN,the growth of NCI-H446 cells were inhibited.When cells were incubated with 25,50,100μmol/L of SFN for 72h,the inhibition ratio was ( 11.1 ± 2.26 ) %,( 25.2 ± 3.24 ) % and ( 44.6 ±4.2) %,respectively,the difference was statistically significant compared with the solvent control group ( t =10.685,8.417,5.264,P <0.05 ).Chamber invasion assay showed that NCI-H446 cell invasion could be reduced.25,50,100 μmol/L of SFN could decrease the trans-membrane cells to (48.6 ± 1.84)%,(35.4 ± 2.22) % and (27.8 ± 1.36) %,and it was statistically significant compared with the solvent control group ( t =6.341,5.562,4.925,P <0.05 ),respectively.In addition,MMP-9 activity was significantly inhibited by SFN.25,50,100 μmol/L of SFN could decrease the gray value of MMP-9 to 764 ±18.4,685 ± 14.74 and 638 ± 21.54 ( control group 822 ± 12.53,t =4.971,7.582,11.235,respectively,P <0.05).Conclusions SFN can inhibit NCI-H446 cells growth,invasion and the activity ofMMP-9.

10.Output strategies for publication of international papers on clinical trials of Chinese medicine.

Scientific research output of clinical trials in Chinese medicine (CM) is insufficient, but international papers hold an important scientific position in China. Based on the current situation, we analyzed the present publication situation of international medical papers in our country, and the feasibility and urgency of publishing international papers on clinical trials of CM. Finally, we proposed to use the PDCA (plan-do-check-action) cycle method to improve the quality control and management of CM clinical trials. Moreover, by combining our experience in relevant scientific research launched at our department, we expounded strategies for improving the quantity and quality of international papers in CM.
