3. Human tumor-related protein hyrdC: Prokaryotic expression, antibody preparation and its preliminary detection in gastric cancer

Objective: To prepare pure prokaryotically expressed protein of hyrdC and the monoclonal antibody against hyrdC, and to detect the hyrdC protein in gastric cancer with the prepared monoclonal antibody. Methods: The target DNA sequence of hyrdC was obtained by RT-PCR and subsequently inserted into prokaryotic expression vector pET32a. The TRX-hyrdC fusion protein was purified and was used to immunize 6-week-old BALB/c female mice to prepare monoclonal antibodies. The supernants of hybridoma cells were screened by enzyme-linked immunoabsorbent assay(ELISA) and identified by Western blot. The prepared monoclonal antibody was used to detect the expression of hyrdC protein in gastric cancer and adjacent normal tissues by immunohistochemical method (SP). Results: The sequence of byrdC ORF was conformed to the reported in Gen-Bank. Two lines of hybrids secreting monoclonal antibodies against hyrdC were established, both belonging to IgG2a subtype. Western blot showed that the prepared monoclonal antibodies specifically combined with human hyrdC protein. Immunohisto-chemistry (SP) results showed that the expression of hyrdC protein in gastric cancer was significantly higher than that in normal gastric mucous membrane (P<0.001). Conclusion: The monoclonal antibody against hyrdC has been successfully obtained. There are significant differences in the expression of hyrdC between the gastric tumor tissues and normal tissues.

5.Tensegrity of Cell Structure

Tensegrity structure is comprised of compression-resis tant elements and a set of continuous tensile elements that are interconnected w ith each other. The stability of such system depends on maintenance of tensional integrity inside the structure, or what has come to be termed “tensegrity”. A ccording to studies on biology, cell structures are assembled on the basis of te nsegrity mechanism. Tensegrity of cytoskeleton can affect cell shape and functio n. Furthermore, some basic rules of mechanochemical transduction in cells can be well explained using tensegrity theory.
7.Correlations of plasma levels of brain natriuretic peptide and Meprin-α with the severity of coronary-artery stenosis

Objective To explore the relationship of the severity of coronary-artery stenosis with plasma levels of brain natriuretic peptide (BNP) and Meprin-α.Methods Totally 237 patients in our hospital were divided into control group (CON group), stable angina group (SA group) and acute coronary syndrome group (ACS group), according to coronary artery angiography.Patients with acute coronary syndrome were divided into 3 subgroups: unstable angina (UA) group, non-ST segment elevation myocardial infarction (NSTEMI) group and ST segment elevation myocardial infarction (STEMI) group.Patients with coronary artery disease (CAD) were divided into 3 subgroups: low-score, medium-score and high-score groups, according to coronary angiography and Syntax score.BNP and Meprin-α levels were determined in patients with coronary artery disease, and the degree of coronary artery stenosis was evaluated.The differences in above indexes were analyzed and compared among the three groups.Results Plasma levels of BNP and Meprin-α were higher in ACS group than in CON group [(233.16± 78.22)ng/L vs.(33.48 ± 13.71)ng/L, (26.89 ± 6.45) nmol/L vs.(12.83±0.66)nmol/L, both P<0.05].Compared with UA group, plasma levels of BNP and Meprin-α were increased in NSTEMI and STEMI groups (all P<0.05).Compared with the control group, plasma levels of BNP and Meprin-α in the Syntax scores-divided subgroups were increased (all P< 0.05).The plasma levels of BNP and Meprin-α in CAD patients were significantly increased along with the increase of Syntax Score.Spearman correlation analysis showed that low density lipoprotein cholesterol, glucose, BNP and Meprin-α levels had positive correlations with the occurrence of coronary heart disease, while high density lipoprotein level was negatively correlated with the occurrence of coronary heart disease (all P<0.05).Conclusions BNP and Meprin-α levels in peripheral blood are significantly elevated in patients with coronary heart disease, and they are correlated with Syntax score.The risk of ACS is increased along with the increased BNP and Meprinα levels.
8.Value of MRS in the differential diagnosis of corpus callosum lesions

Objective To explore the value of multivoxel 1H-MRS in the differential diagnosis of corpus callosum lesions.Methods 17 patients with corpus callosum lesions confirmed by clinical or pathological method underwent MRS examination.These patients included 2 cases of lymphoma,4 cases of Marchiafava-Bignami disease,3 cases of glioma, 7 cases of infarction,and 1 case of multiple sclerosis.MRS features of lesions were analyzed.Results 2 cases with corpus callosum lymphoma revealed significantly increased Cho peak, decreased Cr and NAA peak,towering Lip peak.Among 4 cases with Marchiafava-Bignami diseases, 2 cases revealed increased Cho/Cr and decreased NAA/Cr,1 case showed normal,1 case revealed inverted Lac peak.For 3 cases with corpus callosum glioma,NAA peak decreased or disappeared,and Cho peak increased in varying degrees.7 cases with corpus callosum infarction revealed significant inverted Lac peak, and NAA peak decreased in varying degrees.1 case of multiple sclerosis revealed increased Cho peak,decreased NAA peak,and inverted Lac peak during acute period with enhancement,while these features returned normal during inactive period.Conclusion Multivoxel 1H-MRS plays an important role in the differential diagnosis of corpus callosum lesions.
9.Effects of burn serum on the membrane of intestinal epithelial cell in rats

AIM: To explore the mechanism and significance of the intestinal epithelial cellular membrane damage following burn serum. METHODS: The intestinal epithelial cell(IEC-6) were cultured. The changes of total membranous phospholipid contents fluidity of the IEC membrane were dynamically examined with fluorescence polarization technique and HPCE. RESULTS: In the early stage after stimulation by 20% burn serum, the membranous fluidity obviously decreased. The total phospholipid contents decreased, the content of PLA 2 markedly increased. CONCLUSION: The serial changes in IEC after burned could result in the damages of IEC membrane structure, the integrity of cell membrane and function.
10.Impact of High Pulmonary Blood Flow on Content and Metabolism of Collagen in Rats

Objective To explore impact of high pulmonary blood flow on the content and metabolism of collagen in rats.Methods Thirty-two male SD rats were randomly divided into shunt group and control group.Rats in shunt group were subjected to an abdominal aorta-inferior vena cava shunt to create an animal model of high pulmonary blood flow.In control group,rats experienced the same expe-rimental processes except the shunting procedure.After 4 and 11 weeks of experiment,these changes of pulmonaryartery collagen Ⅰ,collagen Ⅲ,matrix metalloproteinase(MMP-13)and tissue inhibitor of metalloproteinase(TIMP-1) protein expression of rat were investigated by immunohistochemistry.Results After 4 weeks and 11 weeks of shunt,the collagen Ⅰ,collagen Ⅲ,MMP-13 and TIMP-1 of pulmonary artery in rats of shunt group increased significantly compared with those of control group,respectively(all P
