1.Genome-Wide Genetic Study on Central Neurocytoma by Comparative Genomic Hybridization

Objective To detect the genome-wide genetic alterations in central neurocytoma,and to study the pathogensis of central neurocytoma. Methods Comparative genomic hybridization(CGH) analysis was performed in 10 central neurocytomas. Results Chromosomal imbalances were demonstrated in 6 cases.Overrepresentation of genetic material was detected in 4 cases on Chromosome 2p and 10q,and 3 cases on Chromosome 18q. Conclusion(Genetic abnormalities) on Chromosome 2p,10q and 18q may be associated with the pathogenesis of central neurocytoma.
2.Nuclear calcium oscillation in cultured neonatal rat myocytes

AIM: To determine whether nuclear Ca 2+ is independently regulated from the cytosolic Ca 2+ and nuclear Ca 2+ oscillation induced by many modulating factors in cultured rat neonatal myocytes and its mechanism. METHODS: Rat neonatal cardiac myocytes were cultured, and fluo-4/AM was loaded as calcium probe. The changes of cytosolic and nuclear Ca 2+ were observed by confocal laser microscopy. RESULTS: Calcium fluorescent intensity oscillated slightly in myocardiocytes and the average intensity was much higher in the nucleus than that in the cytosole. Ca 2+ oscillation in nucleus and cytosole induced by norepinephrine, isoproperenol, ATP were completely blocked by Ca 2+ -ATPase antagonist thapsigargin (10 -6 mol/L),L-type Ca 2+ channel blocker verapermil (500 ?mol/L) and KCl (20 mmol/L). Ca 2+ -ATPase antagonist thapsigargin completely blocked the propagation of Ca 2+ waves and simutaneouly induced a temporary Ca 2+ increase followed by a magnificient drop and loss of response to norepinephrine. CONCLUSIONS: The results suggested that generation and maintenance of calcium oscillation both in cytosole and nucleus depended on extracellular Ca 2+ influx, membrane potential, Ca 2+ release and uptake of cytosolic and nuclear calcium stores. The difference between cytosolic calcium and nuclear calcium indicated that calcium regulating system relatively independent of cytosole may exist in nucleus.
3.Breast reconstruction with the bipedical transverse rectus abdominis

Objective To study the operational method and the clinical application of breast reconstruction with the bipedical transverse rectus abdominis. Methods 5 cases of breast reconstruction were performed with bipedical transverse rectus abdominis post mastectomy, and 3 cases of nipple-areola reconstruction. Immediate reconstruction was 3 cases, and delayed reconstruction was 2 cases. Results 4 cases of the breast reconstruction were survival, the morphologies were natural, and touch feeling was normal. Only one case skin flap was partial necrosis, nearly 3cm?2cm. Conclutions The breast reconstruction with bipedical transverse rectus abdominis is a good choice for breast cancer patients with caesarotomy scar.
4.The inhibitory effect of ozanimod on the growth and biofilm formation of Staphylococcus aureus

Abstract: Objective To screening new compounds that can inhibit the growth and biofilm formation of Staphylococcus aureus. Methods Compounds that can inhibit the growth of Staphylococcus aureus were screened from the FDA approved drug library by 96 well plates. The absorbance value of 600 nm wavelength (OD600) was measured by Microplate Reader to detect the growth of Staphylococcus aureus planktonic cells in the culture supernatant. The minimum inhibitory concentration (MIC) of ozanimod against Staphylococcus aureus clinical isolates were detected by micro broth dilution method. The inhibitory effect of sub-inhibitory concentrations of ozanimod on the biofilm formation of Staphylococcus aureus was detected by crystal violet staining. Results This study found that ozanimod could significantly inhibit the growth of Staphylococcus aureus SA113 (screening reference strain), and the MIC was 25.00 μmol/L. The MIC of ozanimod against 119 clinical isolates of Staphylococcus aureus [65 isolates of methicillin sensitive (MSSA) and 54 isolates of methicillin resistant (MRSA)] was 12.50 or 25.00 μmol/L. The MIC50 and MIC90 of ozanimod against the 119 Staphylococcus aureus isolates all were 25.00 μmol/L. This study found that 6.25, 12.50, 25.00 μmol/L of ozanimod could significantly inhibit the biofilm formation of 2 MSSA and 2 MRSA. The sub-MIC concentration of ozanimod (12.50 μmol/L) could significantly inhibit the biofilm formation of 14 MSSA and 11 MRSA, but had no inhibitory effect on the growth of planktonic cells of these Staphylococcus aureus isolates. Conclusion Ozanimod can inhibit the growth of Staphylococcus aureus, including MRSA, and has good antibacterial activity. The sub-MIC concentration of ozanimod could significantly inhibit the biofilm formation of Staphylococcus aureus.
5.Effect of air humidity on traditional Chinese medicine extract of spray drying process and prediction of its powder stability.

In order to solve the adhesion and the softening problems of traditional Chinese medicine extract during spray drying, a new method of adding dehumidified air into spray drying process was proposed, and the storage stability conditions of extract powder could be predicted. Kouyanqing extract was taken as model drug to investigate on the wet air (RH = 70%) and dry air conditions of spray drying. Under the dry air condition, the influence of the spray drying result with different air compression ratio and the spray-dried powder properties (extract powder recovery rate, adhesion percentage, water content, angle of repose, compression ratio, particle size and distribution) with 100, 110, 120, 130, 140 °C inlet temperature were studied. The hygroscopic investigation and Tg value with different moisture content of ideal powder were determined. The water activity-equilibrium moisture content (aw-EMC) and the equilibrium moisture content-Tg (EMC-Tg) relationships were fitted by GAB equation and Gordon-Taylor model respectively, and the state diagram of kouyanqing powder was obtained to guide the rational storage conditions. The study found that in the condition of dry air, the extract powder water content decreased with the increase of air compression ratio and the spray drying effect with air compression ratio of 100% was the best performance; in the condition of wet air, the extract powder with high water content and low yield, and the value were 4.26% and 16.73 °C, while, in the dry air condition the values were 2.43% and 24.86 °C with the same other instru- ment parameters. From the analysis of kouyanqing powder state diagram, in order to keep the stability, the critical water content of 3.42% and the critical water content of 0.188. As the water decreased Tg value of extract powder is the major problem of causing adhesion and softening during spray drying, it is meaningful to aid dehumidified air during the process.

6.Determination of equilibrium solubility and n-octanol/water partition coefficient of pulchinenosiden D by HPLC.

OBJECTIVETo determine the equilibrium solubility of pulchinenosiden D in different solvents and its n-octanol/water partition coefficients.
METHODCombining shaking flask method and high performance liquid chromatography (HPLC) to detect the n-octanol/water partition coefficients of pulchinenosiden D, the equilibrium solubility of pulchinenosiden D in six organic solvents and different pH buffer solution were determined by HPLC analysis.
RESULTn-Octanol/water partition coefficients of pulchinenosiden D in different pH were greater than zero, the equilibrium solubility of pulchinenosiden D was increased with increase the pH of the buffer solution. The maximum equilibrium solubility of pulchinenosiden D was 255.89 g x L(-1) in methanol, and minimum equilibrium solubility of pulchinenosiden D was 0.20 g x L(-1) in acetonitrile.
CONCLUSIONUnder gastrointestinal physiological conditions, pulchinenosiden D exists in molecular state and it has good absorption but poor water-solubility, so increasing the dissolution rate of pulchinenosiden D may enhance its bioavailability.

7.Rapid production of tissue-engineered bone based on two kinds of adult stem cells in bone fusion

BACKGROUND:Despite tissue repair with a kind of stem cels has achieved great outcomes, but the combination of two or more kinds of stem cels to construct tissue-engineered bone is stil rare.
OBJECTIVE:To evaluate the efficacy of tissue-engineered bone constructed based on combining adipose-derived stem cels with bone marrow mesenchymal stem cels and alogenic bone in rabbit posterior intertransverse lumbar interbody fusion models.
METHODS:Seventy-five New Zealand rabbits were randomly divided into five groups. L5,6 interbody fusion models were made in each group. Then, these five groups were respectively treated as folows (1) group A: bone marrow mesenchymal stem cels/alograft iliac bone with adipose-derived stem cels; (2) group B: bone marrow mesenchymal stem cels/alograft iliac bone with normal saline; (3) group C: alograft iliac bone with adipose-derived stem cels; (4) group D: alograft iliac bone with normal saline; group E: autologous iliac bone alone. Each group was detected by PET/CT to contrast the standard uptake value at 1, 3, 5 weeks postoperatively.
RESULTS AND CONCLUSION: PET/CT images showed varying degrees of bone fusion and bone metabolism enhancement in the five groups. With time, the standard uptake value was increased in each group, and except the group D, the standard uptake value was higher in al the groups at 3, 5 weeks than at 1 week (P < 0.05). There was no difference in the standard uptake value of al the five groups at weeks 3 and 5 after operation (P > 0.05). The standard uptake value of the group A was better than that in the group B and group E (P < 0.05). Taken together, the combination of adipose-derived stem cels and bone marrow mesenchymal stem cels for rapid production of tissue-engineered bone has good osteogenesis and vascularization.
8.Detecting Hepatitis B Virus Deoxyribonucleic Acid Gene in Breast Milk by Nested Polymerase Chain Reaction

Objective With the method of detecting hepatitis B virus deoxyribonucleic acid(HBV-DNA) gene in breast milk,in ordered to diagnose whether there is HBV in breast milk in those mothers whose HBsAg are positive.Methods Collected breast milk from 187 mothers whose HBsAg were positive,and complified HBV-N gene in breast milk by Nested-Polymerase chain reaction (Nested-PCR).Results Nested-PCR could detect HBV-DNA in breast milk,and the positive rate was 3.2%.Conclusion The method of detecting HBV-N gene in breast milk by Nested-PCR can detect HBV-DNA in breast milk,and it can be an laboratory evidence for whether breast feeding or not.
9.Clinicopathology of mixed epithelial and stromal tumor of kidney

Objective To investigate the pathological features and differential diagnosis of mixed epithelial and stromal tumor of the kidney(MESTK). Methods Three patients with MESTK were studied by light microscopy and immunohistochemistry,and related literatures were reviewed. Results In the three patients,two were females and one was male,with the mean age of 20 years old.Examined grossly,the tumors were well circumscribed and typically composed of multiple cysts and solid areas.Microscopically,the tumors were composed of epithelial and spindle cells,both of the which were well differentiated with no atypia and mitosis of the nuclei.The immunohistochemical staining showed positive for the cytokeratin in the epithelial cells,and Vimentin,SMA,actin,PgR or ER and WT-1 in the spindle cells.No tumor recurrence and metastasis was found in all the three patients by 25 to 29 months of follow up. ConclusionMESTK is an uncommon mixed renal neoplasm with a favorable prognosis.
10.Expression of Recombinant Snake Venom Cystatin in Yeast Pichia pastoris and Its Effects on B16F1 Melanoma Invasion in vitro

To investigate the biological role of snake venom cystatin(sv-cystatin) in tumor progression, the cDNA of sv-cystatin amplified by PCR from pUC18-cystatin plasmid was cloned into methanol-inducible expression vector pPICZ?A. The linearized recombinant plasmid pPICZ?A-cystatin was transfered into Pichia pastoris, strain GS115 by electrophoration. Transfermants with phenotype Mut+ selected were identified by PCR analysis and induced in 1.0% methanol. The reombinant sv-cystatin protein was examined by SDS-PAGE, Western blot analysis. The molecular mass of expression product was about 14 kDa and approximately 16 mg/L of recombinant sv-cystatin was produced from one of GS115-cystatin transformants. The chromatography purified protein could reduce the activity of papain.
