Transforming growth factor-?_1 (TGF-?_) mRNA or protein expression in renal cortex of diabetic rats was assessed by real-time quantitative RT-PCR with SYBR Green,immunohistochemistry or Western blot.After melatonin treatment,the expressions of TGF-?_1 mRNA and protein were decreased,suggesting that beneficial effect of melatonin may result from its antioxidative property and inhibiting TGF-?_1 expressions.

ObjectiveTo investigate current iodine nutritional status of different groups of people in Harbin city, and to provide the basis for development of salt iodization standard and scientific iodine supplementation.MethodsThree urban districts and three surrounding counties were chosen in Harbin,2011.In each chosen urban district and county,one district office (township) was selected,and one residents committee (village) was chosen in each of the district office(township),and 30 households were selected by systematic sampling.Iodized salt,water iodine and iodine intake per capita were investigated.In each of the residents committee (village),20 adults aged 18 - 45,30 pregnant women or lactating women,and 100 school children aged 8 - 10 were selected.Urine samples were collected and urinary iodine level were tested.Salt iodine was determined by direct titration,water and urinary iodine by arsenic cerium catalytic spectrophotometry.Iodine uptake and iodine nutritional status of different populations in Harbin urban and rural areas were compared.ResultsThe edible rate of qunlified iodized salt were 93.3％(84/90) and 96.3％(156/162) in Harbin urban and rural residents,respectively,which were all greater than 90％,and the highest value of salt iodine were 38.3,46.0 mg/kg,respectively,in urban and rural areas,which all did not exceed the upper limit(50 mg/kg) of qualified iodized salt,but there were some samples of salt iodine content below the national standard(20 mg/kg).Water iodine value in urban and rural areas,even the highest value(9.40,8.40 μg/L),was failed to meet the national standard 10 μg/L; salt eaten by rural people perperson a day(8.33 g) was significantly higher than that of the urban people(7.03 g,Z=- 2.750,P ＜ 0.01); in addition to rural children aged 8 - 10,whose urinary iodine value(228.6 μg/L) was higher,the values in urban and rural adults ( 111.3,195.6 μg/L),pregnant women ( 193.0,172.9 μg/L),lactating women ( 128.4,173.7 μg/L)and urban children ( 186.8 μg/L ) were all in appropriate level.The urinary iodine medians ( 195.6,228.6 μg/L )of adults and children in rural were significantly higher than that of urban adults and children(111.3,186.8 μg/L,Z =- 2.294,- 5.434,P ＜ 0.05 or ＜ 0.01,respectively).Population composition of iodine deficiency in both urban and rural adults,lactating and pregnant women[46.7％(28/60),21.6％(13/60) ; 21.1％(19/90),21.3％ (18/89) ; 27.8％ ( 25/90 ),42.2％ (38/90) ] were significantly higher than that of the population composition with iodine excess[4.6％(4/60),5.0％(3/60) ; 16.7％(15/90),16.9％(15/89) ; 4.4％(4/90),0.0％(0/90)],but proportion of iodine excessive population in rural children [26.3％(79/300)] was significantly higher than proportion of iodine deficiency[5.6％(17/300)].ConclusionsThe natural environment of Harbin city is still in the iodinedeficient state.In addition to children in rural areas,the iodine intake and iodine nutrition level is basically appropriate; the risk of disease caused by iodine deficiency in adults,lactating and pregnant women is higher than by iodine excess,but the situation of children in rural is on the opposite.Therefore,we should strengthen the monitoring of different populations,and supplement iodine scientifically based on their need.

Objective To establish a simple and sensitive method for the determination of serum copper by spectrophotometry.Methods Nitro-PAPS was used as a coloring agent for serum copper in the presence of surfactants Tween-80 and Triton X-100 and the formed complex was measured by spectrophotometry.Results The maximum absorption wavelength of the complex was 570 nm and the molar absorption coefficient was 7.95?10~4 L/(mol?cm).The lineafity of the method was up to 63.0 ?mol/L and the recoveries ranged from 98.6% to 103.1%.The within-run and between-run CVs were 2.1%-3.3% and 2.7%-3.8%.The method(Y)was compared with an AAS method(X)and a correlation of Y=1.01X -0.27(r=0.998 2)was obtained.A reference interval(x~-?2s)determined with this method on 68 individuals was 9.7-24.1 ?moL/L.Conclusions A simple and sensitive method for serum copper has been established.It may used for the analysis of serum copper in clinical laboratories.

Objective To investigate the causes and therapeutic methods of compartment syndrome following radial artery cannulation in the left forearm.Methods The data(including puncture pin type,puncture frequency, onset time and INR) and therapeutic methods(including using manicol,cold compress,fasciotomy,dactylolysis and dermatoplasty) of 6 patients were retrospectively analyzed,which had compartment syndrome after radial artery can- nulation in the left forearm.Results The type of puncture pin and puncture frequency attributed to this disease to some extent.After therapy,4 patients recovered completely.2 patients underwent fasciotomy,and Volkmann syn- drome happened in 1 patient,dactylolysis happened in the other because of necrosis in the left thumb and finger.Con- clusion Compartment syndrome may be avoided by correct selection of puncture pin type and no repeated punctur- ing,and early diagnosis and therapy are important for fine prognosis.

Objective As brucellosis epidemic is constantly rising in Harbin in recent years,we analyze the epidemic and surveillance results at municipal surveillance spots of brucellosis from 2008 to 2012,to find out the epidemic characteristics of Harbin brucellosis,and to provide scientific evidences for development of countermeasures.Methods Brucellosis surveillance data which were collected from the National Diseases Surveillance Information Management System from 2008 to 2010,the epidemic characteristics of Harbin brucellosis (including regional,time and population distribution) and the municipal surveillance results in Harbin (including epidemiological investigation and serological surveillance) were analyzed.Results A total of 1348 cases of brucellosis were reported in Harbin from 2008 to 2012,and the incidence of brucellosis increased from 1.79 per 100 000 to 4.62 per 100 000; in addition to Pingfang District (2008-2011) and Fangzheng County(2009 and 2011),all eighteen districts (counties or cities) in Harbin had reported epidemic in the past five years,the highest incidence was in Yilan (382 cases),and the incidence was significantly increased in Shuangcheng(111 cases) and Binxian(73 cases) in 2012; the time of onset focused on March to July,which accounted for 62.54％(843/1348) of annual incidence; age distribution,mainly young adults aged 30 to 60(71.59％,965/1348); gender distribution predominantly male (75.45％,1017/1348) ; occupational distribution farmers(84.87％,1144/1348),and the number of infected students and house workers were also high in the past five years(4.23％,57/1348 and 2.52％,34/1348).In the municipal surveillance spots from 2008 to 2012,13 010 cases were surveyed,of which 2695 cases received serological test,the positive rate was 3.93％ (106/2695),and the positive rate was increasing year by year.Conclusions The incidence of Harbin brucellosis is increasing year by year,onset season mainly in the spring and summer,young men with a high incidence,and farmers are main professional people.Brucellosis has also occurred among students,domestic workers,which should be vigilant.Therefore,we should further strengthen surveillance,timely analyze and report the epidemic,and provide a scientific basis to ensure each measure of eliminating the sources of infection put in place,vigorously develop health education,strengthen inter-departments cooperation,and control the spread of epidemic.

Background Diabetic complication is associated with lipid peroxidation.Aldehyde dehydrogenases (ALDH) catalyze the irreversible oxidation of a variety of biological aldehydes,including lipid-derived aldehydes (LDAs),and thus protect organs and tissues from toxic LDAs.Understanding the activity of ALDH in different ocular tissues in diabetic subjects is very important for prevention and treatment of diabetic ocular complications.Objective This research aimed to investigate the activity and expression of ALDH in different ocular tissues in diabetic rats and to explore the mechanism of ALDH in diabetes-induced eye disease.Methods Twenty-eight healthy SPF male Sprague-Dawley(SD) rats weighted 170-180 g were randomly divided into the normal control group and diabetic group.The diabetic animal model was established by intraperitonial injection of 4％ streptozotocin at 65 mg/kg.Isometric citric acid buffer was injected in the rats of the normal control group.The rats were sacrificed in each group 2 and 4 months after the establishment of the diabetic models,and eyeballs were obtained for the preparation of corneal,lens and retinal homogenates.ALDH activity was detected using a multifunctional microplate reader SpectraMax M5,and ALDH content was measured by ELISA at the wavelength of 450 nm with the SpectraMax M5 ELISA reader.Results The blood glucose level in diabetic rats was significantly elevated at various time points compared with the normal control group(P=0.000),and body weights were evidently lower in the diabetic group than in the normal group (P =0.000).The activities of ALDH (A340) in corneal,lens and retinal tissues in the diabetic group were increased in comparison with the normal control group (F =396.601,P=0.000),and showed an enhancement with the lapsing of time (F =53.139,P =0.000).In addition,the highest level of ALDH was found in the cornea and the lowest level in the lens(F =6973.000,P=0.000).The expression level of ALDH in the corneal,lens and retinal homogenates was significantly higher in the diabetic group compared with the normal control group (F=312.985,P =0.000) and showed a considerable increase over the course (F =19.203,P=0.000).The highest expression level was seen in the cornea and the lowest was in the lens,with a significant difference among these three kinds of tissues (F =3243.000,P =0.000).Conclusions ALDH can protect ocular tissue from the damage of lipid peroxidation.Thess results suggest that ALDH plays a role in preventing diabetes-related ocular complications.

ObjectiveTo determine the utility of the simple body weight support training apparatus used to improve ambulatory function in patients with hemiplegia or paraplegia.Methods38 cases with hemiplegia or paraplegia were randomly divided into control(C,n=10),regular body weight support treadmill training group(R,n=14),simple body weight support training group(S,n=14).All the patients received comprehensive rehabilitation for 6 months.Function Independence Measured(FIM),10 meters ambulatory velocity,Fugl-Meyer Assessment(FMA),Barthel Index(BI) were used to evaluate the effect of training.ResultsThe FIM scores and ambulatory velocity of the patients in group R and S improved significantly compared with that of group C.There was no difference of FIM scores and ambulatory velocity between group R and S.ConclusionThe effect of simple body weight support training on improving the ambulatory function is similar to regular body weight support treadmill.

Objective To explore the relationship between coping style and family cohesion and adaptation among postoperative patients with breast cancer undergoing chemotherapy.Methods A cross-section survey was conducted among 300 postoperative patients with breast cancer undergoing chemotherapy,and Medical Coping Modes Scale and Family Cohesion and Adaptation Scale were used for evaluating.Results The average score of confronce in breast cancer patients was significandy higher than that in domestic norm [ ( 18.53 ± 1.44) vs ( 19.39 ± 3.04 ) ],and the average scores of resignation and avoidance were significantly lower than that in domestic norm [ ( 14.67 ± 1.10) vs ( 8.47 ± 3.58 ),( 16.38 ± 1.63 ) vs ( 14.45 ± 2.97 ) ;t =4.66,29.36,10.54,respectively;P ＜0.01 ].The mean scores of ideal and unsatisfied family cohesion of patients were significantly higher than that in domestic norm,and the mean scores of ideal and unsatisfied family adaptation were significantly lower than that in domestic norm( P ＜ 0.01 ).A significant correlation was found and family cohesion,unsatisfied family cohesion,real family adaptation,and unsatisfied family adaptation were correlated with the coping style( r =-0.14 -0.25,P ＜0.05 ).Conclusions The coping style of postoperative patients with breast cancer undergoing chemotherapy is correlated with their family cohesion and adaptation,so it is important to take action in the intervention to patients' family and.improve the coping style for patients.

Objective To establish a quantitative RT-PCR method with self-quenched fluorogenic probe for detection of bcr/abl mRNA in patients with chronic myeloid leukemia for providing a useful tool for diagnosis of CML,evaluation of therapeutic effect and monitoring of minimal residual disease(MRD). Methods bcr/abl gene from cultured K562 cells was amplified by conventional RT-PCR.The standard quantitative plasmid was constructed by A-T clone method.The self-quenched fluorogenic quantitative RT- PCR method(FQ-RT-PCR)for determination of bcr/abl mRNA was established successfully using the ABI PRISM 7000 PCR Detector.The linear range,sensitivity,stability,and repetitiveness of the method were determined.The marrow samples from 25 CML patients and 3 ALL patients were assessed.Results The sensitivity of the FQ-RT-PCR was 10 copies/?l recombined plasmid,and bcr/abl mRNA can be detected from 1 K562 cell in 10~5 normal cells.The linear range was 10~2-10~9 copies/?l recombined plasmid.The coefficient variation(CV)value was 2.1% in intra-assay and 6.1% in inter-assay.The median ber/abl mRNA expression level was 4.50?10~4 copies/?g RNA [(0.45-89.00)?10~4],5.45?10~4 copies/?g RNA [(2.95-19.30)?10~4 ],13.00?10~4 copies/?g RNA [(4.10-89.00)?10~4] and 2.35?10~4 copies/?g RNA [(0.45-5.12)?10~4] in 25 CML patients,11 patients in the incipient chronic phase,6 patients in blastic crisis,8 patients in chronic period after treatment,respectively.The bcr/abl mRNA level in blastic crisis was significantly higher than that in chronic phase(q= 3.41,P
