1. HPLC-ELSD determination of Fructopyrano-(l → 4)-glucopyranose in Radix Isatidis

OBJECTIVE: To establish an HPLC-ELSD method to determine a new compound named Fructopyrano-(1 → 4)-glucopyranose in Radix Isatidis. METHODS: The HPLC method was conducted on an Agilent XDB-C18 column (4.6 mm × 150 mm, 5 μm), with methanol-water (5:95) as the mobile phase. The drift tube temperature was 115°C and flow fate of carrier gas was 3 L · min-1. RESULTS: The calibration curve was linear for FG in the range of 8.40 - 21.00 ng (r = 0.9990). The recovery was 97.27% - 102.72%. CONCLUSION: This method is accurate, reliable, rapid, sensitive, reproducible, and can be used for the determination of FG in Radix Isatidis. 2.Effect of the regulation of adiponectin on the expression of nitric oxide synthase in the retinas of diabetic rats

Background Diabetic retinopathy (DR) is a common complication of diabetic mellitus,but its pathogenesis is still unclear.Adiponectin may restrain inflammatory reaction and reduce adhesion of vascular endothelial cell to influence diabetic microangiopathy.Relation between adiponectin and nitric oxide synthase (NOS)is less reported.Objective This study was to observe the effect of adiponectin on the expression of NOS in the retinas of diabetic rats.Methods Forty 8-10 weeks Sprague-Dawley (SD) female rats were collected.The rats were randomized into the normal control group (10 rats),adiponectin group (15 rats) and diabetic model control group (15 rats) using the random number table method.Tetraoxypyrimidine was intraperitoneally injected to establish the diabetic model in the rats of the adiponectin group and diabetic model control group,and 10 μg/kg of adiponectin was then injected into the rats of the adiponectin group.Western blot was used to detect the expression of the adiponectin protein in the rat retinas,and the expression of NOS in rat retina was located by immunochemistry.The use of the animals complied with the Regulations for the Administration of Affairs Concerning Experimental Animals by State Science and Technology Commission.Results The relative amount of adiponectin (adiponectin/β-actin)in the retinas was 0.85 ± 0.21,0.79 ± 0.17 and 0.42 ± 0.08,respectively,in the normal control group,adiponectin group and diabetic model control group,with significant differences among the 3 groups (F =4.236,P =0.000).The adiponectin/β-actin ratio in the retinas of the diabetic model control group was significantly declined in comparison with the normal control group and adiponectin group (q =6.615,P =0.000 ; q =6.026,P =0.000).The NOS levels (A value) in the retinas were 0.244 ± 0.035,0.262 ± 0.032 and 0.367 ± 0.066,respectively,in the normal control group,adiponectin group and diabetic model control group,showing a significant difference among them (F =3.752,P =0.001).The A value of NOS in the diabetic model control group was significantly increased in comparison with the normal control group and adiponectin group (q =3.488,P =0.002 ; q =3.079,P =0.005).NOS expression was localized to the inner nuclear layer and retinal ganglion cell layer.Conclusions Adiponectin reduces NO content in diabetic rat retinas by regulating NOS expression.
3.Application of complexity and wavelet packet transformation to motion recognition

Complexity algorithm and wavelet package transform are discussed in this paper. To improve the classification ability of complexity algorithm in motion pattern recognition, it's used together with wavelet package transform. This method shows a better performance than a single complexity algorithm.
4.Investigation of physical growth in 115 infants with eczema

Objective To explore the physical growth in infants with eczema who treated with regular use of glucocorticoid.Methods A total of 115 full-term infants newly diagnosed with eczema were included in this study and treated with topical glucocorticoids regularly.The severity of eczema was assessed and anthropometric indices were measured regularly.Results Both weight for age (WAZ) and length for age (LAZ) at birth and 6-month-old were higher than those of World Health Organization (WHO) child growth standards.However,the LAZ at diagnosis and 3 months after treatment were lower than that of WHO child growth standards.The weight,length,and the growth rate from birth to the time of diagnosis were all lower than those of WHO child growth standards.While the growth rate after treatment for 3 to 6 months was higher than that of WHO child growth standards.The increases of WAZ and LAZ were significantly different among three periods:at birth to diagnose,at diagnosis to 3 months after treatment,and 3 months to 6 months after treatment (P<0.001).Conclusion In the early stage of eczema,the growth rate was decreased when the symptoms were not controlled.However,after treatment,the growth rate was increased.It is suggested that treatment of regular topical glucocorticoids has no adverse effects on growth of eczema infants,and also is beneficial to the catch-up growth of infants.

7.Recent advances in drug development targeting bile acids transporters and related disease

Bile acids (BAs) are a major component of bile salt, which plays a vital role in the metabolism of lipids in humans. Ninety-five percent of bile acids are recycled by the enterohepatic circulation (EHC), and therefore EHC is essential for bile acid homeostasis. There are four transporters that mediate the transmembrane transport of bile acids, each of which plays an important role in the enterohepatic circulation. Gene defects in bile acid transporters can lead to disorders of the enterohepatic circulation, ultimately leading to clinical phenotypes such as metabolic diseases and even death. Bile transporter expression is altered in patients with various metabolic disease states, suggesting that disruption of bile acid transporters may be a pivotal pathological mechanism for the development of metabolism diseases. Thus, many drugs targeting bile acid transporters are being developed. We provide a concise overview of the progress of bile acid transporters research, discuss the relationship between different bile acid transporters and disease development, and summarize the current progress in drug development targeting bile acid transporters.
9.Meta-analysis of Domestic Sparfloxacin for Acute Bacterial Infections with Oral Administration

OBJECTIVE:To assess the clinical efficacy and safety of domestic sparfloxacin for acute bacterial infections.METHODS:Domestic literatures about sparfloxacin for acute bacterial infections were retrieved by computer and their quality was evaluated to extract data(1993~2009).RevMan 4.2.2 software was used for Meta-analysis.RESULTS:A total of 10 RCT were enrolled.The comparisons of 2 groups were homogeneous in terms of clinical cure rate,clinical response rate,bacterial clearance rate and safety.There was statistical significance in comparison of combined effect variable between 2 groups in respect of cure rate,response rate and bacterial clearance rate(P0.05).CONCLUSION:The currently available evidence shows that clinical efficacy of domestic sparfloxacin for acute bacterial infections is better and incidence of ADR was lower.
10.Targeting of p38 mitogen-activated protein kinases to early growth response gene 1 (EGR-1) in the human paclitaxel-resistance ovarian carcinoma cells.

To investigate the relationship between the expression of early growth response gene 1 (EGR-1) and p38MAPK pathway in the paclitaxel resistance of ovarian carcinoma cells, the effect of p38MAPK inhibitor SB203580 on cell apoptosis was examined by using Hoechst 33258 staining. The intracellular Rh123 (Rhodamine 123) accumulation was detected by the flow cytometry (FCM). The 50% inhibition concentration (IC50) of paclitaxel for A2780/Taxol cells was determined by MTT method. Electrophoretic motility shift assay (EMSA) was employed to examine the EGR-1DNA binding activity. MDR1 and EGR-1 mRNA were assessed by RT-PCR. The expressed of p-gp, phosphorylated p53 and p38 were detected by Western blotting. SB203580 could remarkably promote the apoptosis of A2780/Taxol cells, and the cell apoptosis was in a time-dependent manner. Cellular Rh123 accumulation was increased, and the IC50 of paclitaxel for A2780/Taxol cells was decreased significantly. A2780/Taxol cell line after SB203580 treatment was shown to have a significantly higher level of EGR-1 DNA binding activity. SB203580 down-regulated the activity of p38MAPK pathway, but up-regulated EGR-1 expression. SB203580 significantly increased the level of cellular phosphorylated p53 protein, but decreased the p-gp protein level and MDR1 mRNA level in A2780/Taxol cells. There existed a close relationship between p38MAPK pathway and the paclitaxel resistance of ovarian carcinoma cells. The expression of EGR-1 mediated by p38MAPK pathway plays a critical role in paclitaxel resistance of ovarian carcinoma cells.
