2.Effect of propofol on hypoxic pulmonary vasoconstriction in rats

Objective To investigate the effect of propofol on hypoxic pulmonary vasoconstriction(HPV)and assess the underlying mechanism in rats.Methods Twenty male SD rats weighing 300-400 g were.anesthetizedwith intraperitoneal phenobarbital.Heart and lungs were removed after thoracotomy.Pulmonary.arterial rings 4 mmin length and 1.0-1.4 mm in diameter were prepared and suspended in Earl solution maintained at 37℃ with a pHof 7.40 and aerated with 20% O_2-5% CO_2-25% N_2.The rings were stimulated with phenylephrine(PE)10~(-6)mol?L~(-1) with different preloads(300,500,700,900,1 100 mg).The isometric tension of the arterial rings wasmeasured.The optimal preload was determined to be 900 mg which allowed best contractility.Hypoxia was inducedby aerating the solution with 95% N_2-5% CO_2 and the flow rate was adjusted to maintain PO_2 of the solution at40-20 mm Hg and pH at 7.40.HPV of the rings were recorded.Then propofol was added to achieve a finalconcentration of 1,3,10,30,100 ?mol?L~(-1) and HPV was again induced and the changes in HPV wererecorded.In addition the effects of propofol(10,30 ?mol?L~(-1))on vasoconstriction produced by KCI and PE werealso measured.Results The lower doses of propofol(10 ?mol?L~(-1))significantly inhibited HPV(P

5.Changes of the Soleus Muscle Mass and Expression of Myosin Heavy Chain Isoforms mRNA after Acute Spinal Cord Transection

ObjectiveTo investigate the rule of changes of the soleus mass and expression of myosin heavy chain (MHC) isoforms mRNA.Methods40 female Wistar rats were divided randomly into the control group and three spinal cord transection (ST) groups, ST7, ST15, and ST30 with 10 animals in each group. Rats in ST groups were subjected to a complete ST between T8 and T10 levels. The right soleus was dissected and weighed at 7, 15, 30 days after ST, and the expression of MHC mRNA isoforms was measured by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR).ResultsThe Absolute and relative soleus masses in three ST groups were lower significantly than those of control group (P<0.05). The soleus mass in ST15 and ST30 groups were lower than that of ST7 group (P<0.05). The soleus of control group predominantly expressed MHC-I and some MHC-IIa, whereas the soleus began to express MHC-IIx and MHC-IIb after ST, except for MHC-I and MHC-IIa. ST induced consistently down-regulation of MHC-I mRNA and up-regulation of MHC-IIx and MHC-IIa at three time points after ST. The level of MHC-IIb mRNA expression was very low at three time points after ST.ConclusionST can influence the soleus mass at early stage after ST. ST induces a shift toward a faster muscle phenotype from slow to fast MHC isoform. MHC demonstrates plasticity in response to decrease neuromuscular activation.

9.Application of expertise-based randomized controlled trials in acupuncture studies.

Conventional randomized controlled trial is considered as the gold standard of clinical trials in evalua ting pharmacologic effectiveness. Whereas, it still has some faults in the non-pharmacy interventions, such as surgery, acupuncture, massage, etc. Expertise-based randomized controlled trial makes up for some deficiencies of conventional randomized controlled trial, which has made better developments in the field of surgery, however, there is no any relevant retrieval literature found in the acupuncture field. It is feasible and practical of the expertise-based randomized controlled trial to be applied and promoted in the acupuncture-moxibustion clinical research for a further degree. Expertise-based randomized controlled trials might become the trend of acupuncture-moxibustion clinical stu dies in the future.

10.Single nucleotide polymorphism of FCGR2A gene in Han Chinese children with Kawasaki disease.

OBJECTIVETo study the association of FCGR2A gene single nucleotide polymorphism (SNP) rs1801274 with Kawasaki disease (KD) susceptibility and the efficacy of intravenous immunoglobulin (IVIG) therapy in Han Chinese children.
METHODSThirty-five KD children and 25 age-and gender-matched healthy children (control group) were enrolled in the study. Polymerase chain reaction (PCR) and gene sequence analysis were applied to detect SNP of FCGR2A gene rs1801274. These KD patients were classified into two subgroups based on the presence of coronary artery lesion (CAL) following IVIG therapy: CAL (n=13) and non-CAL (n=22).
RESULTSFCGR2A gene SNP rs1801274 was detected in all subjects, including three genotypes (AA, AG and GG). For FCGR2A gene SNP rs1801274, there were significant differences in the genotype and allele frequencies between the KD and control groups (P<0.05), and significant differences in the genotype and allele frequencies were also found between the CAL and non-CAL subgroups (P<0.05). A allele and AA genotype were linked to an increased risk of KD susceptibility (A allele: OR=3.39, 95%CI:1.53-7.50; AA genotype: (OR=4.93, 95%CI:1.61-15.1). Both AG (OR=5.43, 95%CI:1.06-27.8) and G allele (OR=4.88, 95%CI:1.44-16.5) were linked to an increased risk of CAL in KD children.
CONCLUSIONSPolymorphism of the FCGR2A gene SNP rs1801274 is one of the important factors probably influencing susceptibility to KD and efficacy of IVIG therapy on KD in Han Chinese children.
