3.Construction of Streptococcus mutans comE Mutant Strain Using In-frame Deletion System

The intraspecies quorum sensing system of Streptococcus mutans is involved with com genes family.ComE is a kind of response regulator and act as a promoter to the quorum sensing genes.S.mutans comE mutant strain IFD140?comE was constructed using the inframe-deletion system via twice homologous recombination.In current genetic studies of S.mutans,insertion duplication and allelic exchange mutagenesis techniques routinely create polar effects to downstream genes.In-frame deletions are essentially free of these polar effects because the mutation does not introduce any genetic markers.By PCR,sequencing and RT-PCR,it was confirmed that IFD140?comE has only 717bp deleted within the comE gene and the transcription of the downstream gene comD was not interfered.The research of the morphological characteristics indicated that IFD140?comE formed clumps and cells accumulated at the bottom of the glass tubes and light-microscopic observations displayed that the mutant strain formed significantly longer chains compared to those formed by the wild strain.The successfully construction of IFD140?comE lay a foundation for further quorum sensing research.
4.Blind Sample Assessment of Sodium Saccharin in Drinks

Objective: To evaluate the testing level of the relevant items in food inspection agency laboratory objectively, analyze the existing problems and help them enhance the ability of detection.Methods: The blind sample assessment of sodium saccharin in drinks was organized and performed.The test samples at high and low concentrations were prepared and studied by the tests of homogeneity and stability.Those met the requirements of blind sample assessment were randomly distributed to 193 laboratories and the returned data were analyzed statistically.Results: Totally 182 valid data were collected.Among the reported results, those from 85 laboratories were satisfied in the low concentration group with the satisfaction rate of 87.6% , those from 12 laboratories were not satisfied, and no results were suspicious;as for the high concentration group, the above data was 67(78.8%), 12 and 6, respectively.The overall satisfaction rate was 83.5%.The results, especially the dissatisfied and suspicious data, were analyzed by the original records of each participating laboratory, the analysis focused on 8 aspects including detection method, instrument brand, control product variety, pretreatment method, recovery rate, column selection and the other influencing factors, and suggestions were given to every laboratory to improve its own situation.Conclusion: The statistical data reflect that our food testing laboratory has strong detection ability in the determination of saccharin sodium in beverages, and can provide powerful technical support for the regulatory authorities.
5.Effect of flavopiridol on the proliferation, invasiveness and apoptosis of human prostatic cancer cell line LNCaP

Objective To investigate the effect of flavopiridol on the proliferation,invasiveness and apoptosis of human prostate cancer cell line LNCaP,and to explore the possibility of its application in clinical treatment.Methods MTT assay was used to detect cell proliferation,cell invasion in vitro was detected by Transwell assay,and flow cytometer was used to observe apoptosis.Results Flavopiridol inhibited the growth of LNCaP cells in a concentration-dependent and time-dependent way (P < 0.05),and reduced the ability of invasion capacity.After treated by 10 nmol/L flavopiridol for 24 h,the apoptosis rate was increased significantly to (7.5±0.9) % compared with the control group [(5.3±0.5) %] (P < 0.05).Conclusion Flavopiridol can inhibit proliferation of LNCaP cells and induce apoptosis,which may be applicable for the treatment of prostate cancer.
6.Atypical adenomatous hyperplasia of the lung:correlation of radiographic and pathologic findings

Objective To study radiographic features of atypical adenomatous hyperplasia(AAH) of lung and correlate them with their pathologic findings,so as to improve the imaging diagnose.Methods The imaging features of 8 patients with pathology-proved AAH were reviewed.All patients were women with age ranging from 35 to 74 years.All cases had chest radiography and HRCT.The radiographic findings were studied retrospectively and correlated with those of pat hology.Results On chest X-ray,the foci in five patients presented as nodule-like area of a bit high attenuation,while the chest radiograph in the other three patients were negative.On HRCT,all 8 patients presented as non-solid nodules.All lesions were round or oval shaped with well-defined margin(n=4)or poor-defined margin(n=3).The greatest dimension of the lesions ranged from 5.0 mm to 20.0 mm.Their CT value ranged from-362.7 HU to-485.6 HU,the mean CT value was(-423.0?47.0)HU.Air bronchograms and/or bubbles were seen in 5 lesions.Coarse spiculation and pleural tag was not seen in any lesion.Before operation,all the nodules did not change in size over a follow-up period from one month to six months.Pathologic findings showed atypical epithelial cell proliferation along thickened alveolar septa without alveolar collapse.Conclusion AAH should be considered in cases with the following features:(1)nodules are found in lung cancer screening or incidentally detected;(2)the diameter of the nodule is usually less than 10 mm;(3)the lesion presented as non-solid nodule on HRCT,air bronchogram and/or bubble sign can be seen;(4)no coarse spiculation and plural tag was seen.
7.Studies on chemical constituents of Zhuang medicine Excoecaria venenata and their cytotoxic activity.

Fourteen compounds were isolated from 95% ethanol extract by silica gel, MCI, and ODS column chromatography. These compounds were respectively identified as quercetin (1), kaempferol (2), (+)-catechin (3), fraxin (4), protocatechuic acid (5), gallic acid (6), methyl gallate (7), ethyl gallate (8), apocynol A (9), baccatin (10), cerevisterol (11), ellagic acid (12), 3, 3',4'-tri-0-methylellagic acid(13) and N-benzoyl-L-phenylalaninyl-N-benzoyl-L-phenylalaninate(14) by analyzing their spectral data and comparing with the previously reported literatures. Except for gallic acid (6), all other compounds were isolated from this plant for the first time. Compounds 1, 2 and 6 showed moderate anti-proliferation activities on tumor cells.

8.Quality of life among patients receiving chemotherapy in the outpatient departments of a general hospital and its correlation with pain.

OBJECTIVETo analyze the relationship of quality of life and pain in patients receiving chemotherapy in the outpatient departments in a general hospital.
METHODA questionnaire-based survey was conducted to collect data on pain status, sleep disorder,vomiting,constipation as well as the social function status.
RESULTSAmong the 428 patient-times of survey, 12.2% of the patients had moderate to severe pain,of which 78.8% was consistent pain. The occurrence of pain was closely related to the metastasis of cancer (P = 0.000). The intensity and location of pain were independently correlated to the origin of cancer (P = 0.035, P = 0.000). Nearly a quarter of patients (24.5%) agreed that talking with his/her family was helpful to relieve pain, while 17.3% of patients had ambulatory restriction or changes in social function. The changes in social function were closely related with sleeping disorder, constipation, fatigue, and intensity of pain (all ,0.05). Furthermore, sleeping disorder was related to pain intensity (P=0.000), while constipation had no relationship with analgesic medicines (P = 0.798).
CONCLUSIONSA significantly large proportion of patients who are receiving chemotherapies in the outpatient departments of general hospital are suffering from moderate to severe pain. Their decreased social functions correlate with the cancer-related somatic symptoms.
