2.STUDY OF TT VIRUS IN EXPERIMENTALLY INFECTED RHESUS MONKEY

To study tissue distribution of TTV in experimentally infected Rhesus monkey and if the TT virus is hepatotropic. Total DNA was extracted from tissues of 5 experimentally infected Rhesus monkeys. TTV was detected by PCR,and dot hybridization was done with virus double DNA strand probe or single antisense strand probe. The double strand probe was hybridized with DNA of liver, bone marrow, and spleen, stomach, small intestine, colon. and sera. In the above tissues , the virus was also positive as shown by PCR. The single strand antisense probe was only hybridized with DNA of liver, bone marrow,and small intestine of all 5 monkeys, but not with that of other tissues. It suggested that TTV could infect many tissues of Rhesus monkey. Only the liver ,bone marrow, and small intestine presented the virus positive single strand, which might be a replicative intermediate of the virus. It suggests that TT virus replicates in liver , bone marrow, and small intestine,and it might be hepatotropic.
3.Percutaneous Laser Hypothermia in the Treatment of Lumbar Discogenic Pain

Objective To evaluate the efficacy of percutaneous laser hypothermia in the treatment of lumbar discogenic pain. Methods From June 2006 to March 2008,35 patients with lumbar discogenic pain were treated by low-frequency laser hypothermia in our hospital with 1 second impulse time,1 second interval time,3.5 w power,and 30 minutes duration. The effectiveness of the therapy was evaluated afterwards. Results Among the cases,15 patients were treated by one operation,20 patients received a second therapy,and 6 underwent the therapy for totally 3 time; the treatment was effective in 32 cases (excellent in 7,good in 24,and fair in 1). The therapy is ineffective in 2 cases. One case was converted to open surgery for unsatisfying outcomes of the laser hypothermia. The total effective rate was 91.4%(32/35),and fineness rate was 88.6%(31/35). Conclusions Percutaneous laser hypothermia is an effective treatment for lumbar discogenic pain. The indications for the procedure should be selected strictly.
4.Natural infection of 4-Lyme disease Borrelia burgdorferi in Diebu and Huajian areas of Gansu province

Objective In order to offer scientific evidence for prevention and therapy of Lyme disease, we had investigated the natural infection of lyme disease Borrelia burgdorferi(Bb) of 4 ticks in Diebu and Huajian areas of Gansu province. MethodsEpidemiology detection of natural infection of lyme disease Bb was accomplished for four dominant tick ( Dermacetor silvarum , Dermacentor nuttalli , Haemaphysalis Japonica and Ixodes Crenulatus ) at Diebu, Diebu forest zone(Qin mountain), north of Min Mountains of Huajian region and Huajian forest zone, north of Qilian Mountains of Sunan region, Gansu, from March to June, 2010. The methods of dispersion of clamp every 10 m which was put in the morning and retrieved at night were used to capture rodent animal and gnawer retrorse hair inspection insect methods were used to collect parasitic tick. Flagging methods were used to collect free tick.Four kinds of live adult ticks were dissected after cleaning and disinfection. Intestinal contents were smeared, lyme disease Bb was observed under dark-field microscope. Etiological agent was cultivated and separated. Separated spirochete was confirmed by spirochete monoclonal and polyclonal antibody. Results The ticks collected were classified as 2 families, 8 genera, 36 species, i.e. Ixodidae 6 genera, 33 species and Argasidae 2 genera, 3 species. The method of dark-field microscope was used to detect the lyme disease spirochete in 201 ticks intestines after dissection of Dermacentor silvarum, Dermacentor nuttalli, Haemaphysalis Japonica and Ixodes Crenulatus, the lyme disease Bb-positive were 25 with positive rate of 12.44%(25/201). The lyme disease Bb was cultivated and separated from 12 ticks with the positive rate of 18.46%(12/65) after inoculation and cultivation of 3 species, 65 ticks of Dermacentor silvarum, Dermacentor nuttalli and Haemaphysalis Japonica. ConclusionsDermacentor silvarum,Dermacentor nuttalli and Haemaphysalis Japonica have different degrees of natural infection of lyme disease Bb.

10.Study on the therapeutic effects of hyperbaric oxygen on human umbilical cord Wharton jelly mesenchyme cells on myocardial infarction rats

Objective To explore whether hyperbaric oxygen could enhance the effects of the transplanted stem cell on treating myocardial infarction rats.Methods A total of 70 Sprague-Dawley (SD) Rats was randomly divided into four groups,sham operated,acute myocardial infarction (AMI),mesenchymal stem cells (MSCs) (Stem Cell Transplanting),and hyperbaric oxygen (HBO) + MSCs group,respectively.Myocardial infarction rat model was established,hyperbaric oxygen treatment was carried out every day at the first to 30 day after AMI.Human umbilical cord Wharton jelly mesenchyme cell was transplanted at the third day after AMI.The densities of polymorphonuclear neutrophils and macrophages in the infracted border zones and in the infarcted area of heart were determined with hematoxylin and eosin (HE) staining.The cardiac function was assessed with M-mode transthoracic echocardiography,in vivo.Results (1)Compared to MSCs group,the stem cell proliferation and survival rates were increased significantly at the forth week (276.6 ±73.8 versus 20.5 ± 12.3,P <0.01).(2)Compared to the AMI and MSCs groups,the infracted area and the number of inflammatory cell infiltration were significantly reduced in HBO alone and MSCs + HBO groups.(3) Compared to the AMI group,the cardiac function was significantly improved at the different degree in the other three groups;among these groups,cardiac function was significantly improved in the MSCs + HBO group (P <0.01),except for the volume of end-dilation (P >0.05).Conclusions Hyperbaric oxygenation could significantly promote the proliferation and survival rate of stem cells,and improve the cardiac function.Thus further enhance the effect of the transplanted stem cell on treating the infarcted heart.
