1.Effects of haloperidol on the expression of HSP70 induced by ketamine in the rat hippocampus

Objective This experiment was designed to determine the effect of haloperidol on HSP70 induced by ketamine, and to explore the possibility of using haloperidol to prevent or treat the brain injury caused by ketamine.Methods 48 rats were divided into 8 groups, In group 1-3 different doses of haloperidol (1.0,5.0,10.0 mg/kg)were given 1h before the administration of ketamine. There were two control groups. In control group 1 normal saline was given twice with 1h interval between the two injections. In control group 2 ketamine 80.0 mg/kg was given 1h after the administration of normal saline.The volume of each injection was 3ml and intraabdominal injection was used as the route of administration.HSP70 mono-clone antibody immunocytochemistry was used to detect HSP70 expression in rat hippocampus, and MIAS-2000 photography analytic software to analyze HSP70 expression in hippocampus of the rat brain.Results Ketamine induced HSP70 expression in the rat brain. Pretreatment with haloperidol inhibited HSP70 expression induced by ketamine, and the inhibition was dose-dependent, but haloperidol given after the administration of ketamine could not decrease HSP70 expression.Conclusions Ketamine injures neurons of rat hippocampus and induces the expression of HSP70 and haloperidol pretreatment can prevent neuronal injury caused by ketamine, but haloperidol can not antagonize the injury caused by ketamine.
2.Effect of ketamine-midazolam on myocardial HSP 70 expression in septic shock in rat

Objective To evaluate the myocardial protection afforded by ketamine midazolam in its septic shock and its possible mechanism by studying the impact of ketamine midazolam on myocardial HSP 70 expression in LPS induced septic shock in rats Methods 112 healthy adults SD rats of both sexes, weighing 180 205g were randomly assigned to one of following groups:(Ⅰ)control group received normal saline ip (group NS); (Ⅱ) endotoxin group received LPS 20mg?kg -1 ip(group E); (Ⅲ) endotoxin midazolam group received midazolam 0 5mg?kg -1 20 min before LPS (group EM); (Ⅳ) endotoxin ketamine group received ketmine 80mg?kg -1 20 min before LPS (group EK); endotoxin ketamine midazolam group received ketmine 80mg?kg 1 and midazolam 0 5mg?kg -1 20 min before LPS (group EKM) One hour later half of the initial dose of ketamine and/or midazolam was given to reinforce their effect Heart was harvested 2h after LPS or when the rats died of LPS induced septic shock(pupil did not respond to light) for determination of HSP 70 expression using monoclone antibody immunocytochemistry The survival time of every rat was recorded Results The expression HSP 70 was higher in group E than that in group NS and was lower than that in group EK and EKM The survival rate of group EK and EKM was higher than that of group E and EM (P0 05) Conclusions Ketamine can enhance myocardial HSP 70 expression in septic shock This may be one of the mechanisms of its myocardial protection Joint use of ketamine midazolam dose not affect the myocardial protective effect of ketamine
3.Preliminary comparative proteomics study of cervical carcinoma tissues with different sensitivity to concurrent chemoradiotherapy

Objective To investigate the proteomics differences between the high-sensitivity(HS) and the low-sensitivity(LS)groups of cervical carcinoma treated by concurrent chemoradiotherapy,and to confirm the sensitivity associated proteins in intermediate stage and advanced cervical carcinoma.Methods Fresh carcinoma tissues were collected from 10 untreated cervical carcinoma patients.According to the response to concurrent chemoradiotherapy,the tissues were classified into HS group and LS group.In the first part of our experiment,protein separation was performed using two-dimensional gel electrophoresis(2-DE)with Amersham 18 am linear pH 3-10 immobilized pH gradient(IPG)strips.The images of the gels were analyzed by PD-quest 7.0 software to find the differentially expressed protein-spots in each group.Then the differentially expressed protein-spots were incised from the gels and digested by trypsin.The peptide mass flngerprintings(PMF)was acquired by matrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).The proteins were identified by data searched in the Mascot-database.Two differentially expressed proteins were assayed by western blot and immunohistochemical methods.Results Most of the gels were clear and successfully analyzed by PD-quest 7.0 software.Most of the protein-spots concentrated on the area of 20-100 KDa(Mw)and pH4-8.The average number of the protein-spots was 781±74 in HS group and 766±52 in LS group.The match rate was 87.6%between the two groups.Eight proteins highly in HS group but lowly expressed in LS group included hemoglobin subunit beta,caspase-14 precursor,calmodulin-like,S100-A9 protein(MRP-14),galectin-7,HSKERC4,keratin 19 and actin.Ten proteins highly in LS group but lowly expression in HS group included anti HBs antibody light-chain Fab,laminB1,WARS protein,flavin reductase,glutamate dehydrogenase 1,nuclear matrix protein 238,retinal dehydrogenase 1,AFl 65172,subunit of replicative DNA polymerase and HSP70.The higher expression of HSP70 in LS group and galectin7 in HS groups were further confirmed by western blot and immunohistochemical method.Conclusions The 2-DE gels images are successfully acquired from high-sensitivity group and low-sensitivity group of intermediate stage and advanced cervical carcinoma tissues treated by concurrent chemoradiotherapy.Some differentially expressed proteins between the two groups can be further confirmed by western blot and immunohistochemical method.
4.The clinical diagnosis, treatment and prognostic analysis of renal secondary tumor

Objective To investigate the clinical features,diagnosis,treatment and prognosis of the renal secondary tumor.Methods From January 2000 to January 2014,the data from 31 patients,including 23 male patients and 8 female patients,with renal secondary tumors were analyzed retrospectively.Their mean age was 56 years old (ranging from 38 to 75 years old).The 31 renal secondary tumors rooted in lung cancer(n =14),lymphoma(n =5),colorectal cancer and gastric cancer(n =3),breast cancer(n =2),esophageal cancer(n =1),thyroid cancer (n =1),cervical cancer (n =1) and bladder cancer (n =1),respectively.There were 22 patients (71.0%) of renal metastasis accompany with other organs or lymph node metastasis.9 cases (29%) suffered with independent renal metastasis and 21 cases (67.7%) suffered with unilateral renal metastasis.5 cases(16.1%) were diagnosed as primary tumor with the renal metastasis at the same time,and the remaining 26 cases were found renal metastasis within 9 to 72 months after primary tumor (mean 30 months).There were only 5 patients (16.1%) with symptom.Ultrasound showed low echo range in 20 cases (65.6%) or uneven echo in 11 cases (34.4%).CT showed equal density (77.4%) in 24 cases or slightly low density shadow (22.6%) in 7 cases,most of which were endogenous,mild enhancement.10 cases (32.3%) were bilateral renal metastasis,unilateral renal multiple metastases was found in 6 cases (19.4%),and single metastasis was noticed in 15 cases (48.4%).The average diameter of the renal metastasis was 2.7 cm (ranging from 0.9 to 6.8 cm).Except 4 cases gave up the treatment,the remaining 27 cases were accepted comprehensive therapy about the primary tumor.the 9 patients with renal metastasis only were treated with chemotherapy or targeted therapy for the advanced primary tumor.Among the 9 patients,6 cases were undergone NSS or radical nephrectomy (RN) treatment.Results In 9 cases with only renal metastasis,6 cases,treated by surgery,recovered well.Postoperative pathological and immunohistochemical results confirmed the renal metastasis.Up to January 2015,the follow-up duration ranged from 2 months to 60 months [mean (22.6 ± 18.4) months].The survival time ranged from 1 month to 51 months [mean (13.2 ± 13.2) months].Among 22 cases with multiple metastasis,4 cases gave up treatment,whose average survival time was (2.0 ± 1.4) months.However,the average survival time in remaining 18 cases was (11.1 ± 4.7) months (P < 0.05).In 9 cases with independent renal metastasis,the average survival time in 6 cases,accepted the procedure,was (26.2 ± 18.6) months.While,the average survival time in remaining 3 non-surgical cases,was (10.3 ± 4.0) months (P < 0.05).Conclusions Renal secondary tumor was rare in clinic.Most cases have isolated lesion.Renal secondary tumor was advanced manifestation of the primary tumor,which could prolong the survival time according to the comprehensive treatment for the primary tumor.Surgical resection of the lesion before the comprehensive treatment could be chosen in the independent renal metastasis.
5.The value of mucin 7 in the detection of bladder cancer

Objective To investigate the diagnosis value of mucin 7 in plasma and urine in the detection of bladder cancer.Methods Expression of mucin 7 was detected quantitatively in plasma and urine from 23 patients with bladder cancer and 23 patients as control group by enzyme-linked immunosorbent assay(ELISA) from November 2012 to September 2013.There were 15 were male and 8 were female in the 23 bladder cancer patients,aged from 41 to 95 years,with an average of 64 years.Of the 23 bladder cancer pathology,12 with high grade,11 with low grade,11 with invasive disease and 12 with noninvasive disease.There were 19 males and 4 females in the 23 patients in control group,aged from 28 to 85 years,with an average of 61 years.Results The amount of mucin 7 in plasma was significantly higher in bladder cancer group than that in control group [(7.43±4.54) ng/ml versus (4.55±.1.98) ng/ml,P=0.017].While there was no significant difference in urine between the two groups [(12.44±7.1 1) ng/ml versus (11.96±8.41)ng/ml,P=0.840].There was no significant differences in the amount of mucin 7 had in different grades and stages of bladder cancer (P>0.05).Conclusions Expression of mucin 7 in plasma of patients with bladder cancer is significantly higher than that in control group.Mucin 7 expression has no significant correlation with the grading and staging of bladder cancer.Detecting mucin 7 expression quantitatively with ELISA for diagnosis of bladder cancer is a method with certain value.

9.Study on seed quality test and quality standard of Pesudostellaria heterophylla.

Referring to the rules for agricultural seed testing (GB /T 3543-1995) issued by China, the test of sampling, seed purity, weight per 1 000 seeds, seed moisture, seed viability and germination rate had been studied for screening seed quality test methods of Pesudostellaria heterophylla. The seed quality from different collection areas was measured. The results showed that at least 6.5 g seeds should be sampled and passed through 10-mesh sieve for purity analysis. The weight of 1 000 seeds was determined by using the 500-seed method. The phenotypic observation and size measurement were used for authenticity testing. The seed moisture was determined under the higher temperature (130 ± 2) degrees C for 5 hours. The seeds were dipped into 0.2% TTC sustaining 1 hour at 40 degrees C, then the viability could be determined. The break dormancy seeds were cultured on sand at 10 degrees C. K cluster analysis was applied for the data analysis, the seed quality from different collection areas grading of P. Heterophylla was described as three grades. The seed quality of each grade should reach following requirements: for first grade seeds, germination rate ≥ 86%, 1 000-grain weight ≥ 2.59 g, purity ≥ 87%, moisture ≤ 13.1%; for second grade seeds, germination rate ≥ 70%, 1 000-grain weight ≥ 2.40 g, purity ≥ 77%, moisture ≤ 14.3%; for third grade seeds, germination rate ≥ 41%, 1 000-grain weight ≥ 2.29 g, purity ≥ 76%, moisture ≤ 15.8%. The seed testing methods for quality items of P. heterophylla had been initially established, as well as the primary P. heterophylla seed quality classification standard.
