1.In vitro biological characteristics and immune regulation defects of bone marrow-derived mesenchymal stem cells from patients with aplastic anemia

BACKGROUND: Previous research has indicated that bone marrow-derived mesenchymal stem cells had immunological regulation and hematopoiesis role. There were significant differences in immunological regulation and hemopoietic function between patients with aplastic anemia and normal cases. OBJECTIVE: To summarize the in vitro biological characteristics and its immune regulation defects of bone marrow-derived mesenchymal stem cells from patients with aplastic anemia.METHODS: A computer-based online search was conducted in PUMMED (1987-2009) and VIP database (1989-2009) with the key words of "mesenchymal stem cell, bone marrow" in both Chinese and English. There were 55 articles in total. Articles which were related to biological characteristics and its immune regulation defects of bone marrow-derived mesenchymal stem cells were included; however, duplicated articles were excluded. Finally, 32 articles were included, containing 3 reviews in English, 23 original articles in English, and 6 original articles in Chinese. RESULTS AND CONCLUSION: Bone marrow-derived mesenchymal stem cells had the capacities of high proliferation, self-renewal and multilineage differentiation; in addition they had the roles of supporting hematopoiesis, promoting implantation and hematopoietic reconstitution in vivo. Bone marrow-derived mesenchymal stem cells still had the effects of negative immune regulation and reducing the incidence of graft-versus-host disease (GVHD). Aplastic anemia correlated with hematopoietic stem cells in the pathogenesis of intrinsic defects in the proliferation or differentiation of hematopoietic microenvironment and immune system abnormalities and other related disorders. Bone marrow-derived mesenchymal stem cells of patients with aplastic anemia played an important role in the pathogenesis of this disease.

Objective] To investigate and compare with western medicine on the advantages of traditional Chinese medicine in the treatment of subacute thyroiditis. [Method] Select subacute thyroiditis clear diagnosis, single disease cases in clinical work.For patient treated with pure Chinese medicine internal and external treatment. Observation, comparison and analysis of the degree of symptom relief, the size of the side effect, the prognosis of patients and the satisfaction of patients with medical treatment.Discussion on Chinese medicine in the treatment of subacute thyroiditis the high compliance of advantages, the advantage of fewer side effects,therapeutic advantage and the advantage of improving the prognosis and reducing the sequela of disease. [Result] Through clinical observation and comparison, traditional Chinese medicine to western medicine in the treatment of subacute thyroiditis, good treatment compliance, curative effect are exact, side effects rarely, high patient satisfaction,good prognosis show obvious advantage. [Conclusion] Chinese medicine treatment of subacute thyroiditis, compared with the western medicine has obvious advantage for early clinical intervention, controling an illness as soon as possible, improving prognosis, reducing hypothyroidism incidence are of great significance.
3.Investigation into physical activity and eating behaviours of obese children aged 1~6 years

Objective To analyze the risk factors related to obesity of preschool children. Methods The physical activity and eating behavior of 120 obese children aged 1~6 years were investigated with questionnaire by case control 1:1 matched. Results Obesity group spent significantly longer time to watch TV than control group, but the time for physical activity, feeding pattern during the first 4 months, the time of initial supplementary foods and the sleeping duration were not different between both groups. Conditioned logistic regression of multi-variables showed that good appetite, fast eating, overeating of carbohydrate and meat , and snacks before breakfast were the risk factors of child obesity. Conclusion The bad eating behaviors and the lack of physical activity were related to obesity in preschool children.
4.Elimination Effect of Radix Pulsatillae Liquid Extracts on R Plasmid in Pseudomonas Aeruginosa

Objective To explore the elimination effect of radix Pulsatillae extracts on R plasmid of pseudomonas aeruginosa.Methods R plasmids of pseudomonas aeruginosa were isolated from the sputum of elderly patients suffering from respiratory infection,and the elimination test of radix Pulsatillae extracts on these plasmids was performed in vitro and in vivo.Results Being treated with the extracts for 24,48 and 72 h in vitro,the elimination rate of R plasmids was 0,1.8 %and 3.4 %respectively,while in SDS(sodium dode canesulphonate)control group the elimination rate was 0,0.8 %and 1.6 %respectively.Being treated with the extracts for 24,48 and 72 h in vivo,the elimination rate was 0,2 %and 12.8 %respectively,and 0 in the control group.Conclusions Both in vitro and in vivo,radix Pulsatillae extracts have an elimination effect on R plasmid of pseudomonas aeruginosa,and the effect in vivo is stronger than that in vitro.
5.The feasibility of the anterior reverse transpedicular screw (ARTPS) fixation at upper thoracic spine

Objectives: To verify the feasibility and safety of the anterior transpedicular screw(ATPS) fixation of the upper thoracic spine (T1-T4) through the radiological anatomy study on the cadaveric specimens. Methods: The upper thoracic spine thin-section CT data of 40 cases were collected from the radiology de-partment′s database(20 males and 20 females, aged from 18 to 68 years, the mean age was 39.7 years). The data of OPW(outer pedicle width), OPH(outer pedicle height), PAL(pedicle axis length), TPA(transverse section angle), SPA(sagittal section angle), DTIP(distance of transverse intersection point) and DSIP(distance of sagittal intersection point) of each pedicle were measured on the transverse and sagittal sections through the axis of each pedicle. The data were recorded and statistically analyzed. 10 upper thoracic spine(C7-T6) specimens of adults(5 males and 5 females, with unknown ages), with no damage to their appearance, the costovertebral joints and paravertebral soft tissue were completely retained. Then simulate surgical operations were done on the cadaveric specimens based on the obtained data. Screws were implanted anteriorly by free hand. After that, the specimens accepted X-ray fluoroscopy and CT scan. At last, the screws were removed, the speci-mens were sawed along the transaction and sagittal section of the screw channel. Then the success rate of the screw placement was evaluated according to Rao′s worn out classification standard of pedicle screws. Results: From T1 to T4, the OPW decreased from 8.14mm to 3.47mm; the OPH increased from 6.89mm to 10.29mm; the TPA decreased from 32.96° to 11.64°; the DTIP increased from 1.80mm to 5.50mm; the SPA increased from 104.95° to 115.74°; the DSIP increased from 5.95 to 8.76mm; the PAL changed irregularly, from 32.95 to 35.96mm. The pedicle diameters of T3 and T4 were too small to implant ATPS, but the ARTPS can be implanted successfully. The diameter of ATPS was about 4.0mm; the length of ATPS was about 35mm. The diameter of ARTPS was about 5.0mm; the length of ARTPS was about 35mm. 80 pedicle screws were implanted anteriorly, according to Rao′s worn out classification standard of pedicle screws, the fine rate was 90%. The internal walls of 7 pedicles were broken by screws of less than 2mm and no compression to the spinal cord. The internal walls of 5 pedicles were broken of 2 to 4mm, 1 at T1, 1 at T3 and 3 at T4, with varying degrees of spinal cord compression. The internal walls of 2 pedicles were broken of greater than 4mm, 1 at T2 and 1 at T4, with serious spinal cord compression. The external wall of 1 pedicle was broken at T2. Conclusions: The ATPS techniques at T1, T2 and the ARTPS techniques at T3, T4 are feasible, but the safety and clinical practice and further research is needed.
6.The survival analysis of chronic severe hepatitis B with antivirus treatment

Objective To analyse the effect of antivirus treatment on the survival time of chronic severe hepatitis B retrospectively.Methods one hundred and twenty-one patients with chronic severe hepatitis B and positive HBV DNA were divided into 3 groups:42 patients in entecavir(ETV)group,34 patients in lamivudine(LVD)group and 45 patients in control group.The primary data were recorded and followed.The survival curve was described using Kaolain-Maier method.The comparison of survival rate among groups was done using Logrank test.Results The baseline characteristics were well balanced among 3 groups.When the follow-up ended,the mean survival time of ETV group,LVD group and control group was(49.4±5.8)weeks,(51.6±6.7)weeks,(32.8±5.7)weeks;the total survival rate was 0.567,0.557,0.318 respectively,there was a statistical difference between ETV group and control grouP(χ2=5.742,P<0.05),or between LVD group and control group (χ2=5.472,P<0.05).There was not statistical difference between ETV group and LVD group in the week of 2,4,8,12,24,48.Conclusion ETV and LVD can improve the survival rate of chronic severe heoatitis B,and ETV iS not different from LVD.
7.Instant effect of moxibustion at different points on electrogastrogram of patients with functional dyspepsia

Objective:To observe different effects of moxibustion at Zusanli (ST 36) and Yanglingquan (GB 34) on electrogastrogram (EGG) of patients with functional dyspepsia (FD). Methods:A total of 65 cases were randomized into a Zusanli (ST 36) group (n=33) and a Yanglingquan (GB 34) group (n=32) by random serial number, to receive moxibustion therapy to the corresponding point respectively. The 8-lead EGG was used before and after treatment to collect EGG parameter of the gastric body, antrum of stomach, lesser curvature and greater curvature, including the dominant frequency, slow wave percentage, average frequency, response area and motility index. Results:After treatment, the dominant frequency and slow wave percentage were improved in nearly the whole stomach area in Zusanli (ST 36) group, showing statistical significances (bothP<0.05); the slow wave percentage and average frequency were improved in stomach body and antrum of stomach in Yanglingquan (GB 34) group, showing statistical significances (bothP<0.05); moxibustion at Zusanli (ST 36) had a significantly higher influence on EGG than Yanglingquan (GB 34) , in which the lifting of slow wave percentage was significantly higher than Yanglingquan (GB 34) group (P<0.05). Conclusion: Moxibustion at Zusanli (ST 36) and Yanglingquan (GB 34) has the effect of adjusting abnormal EGG pattern in FD patients. Compared with Yanglingquan (GB 34), Zusanli (ST 36) has a superior effect in improving EGG in stomach area.
8.The effects of hyperbaric oxygen in reducing brain damage from hypoxic ischemia in neonatal rats

Objective To investigate the protective effects of hyperbaric oxygen(HBO)against brain dam- age from hypoxic ischemia(HIBD)in neonatal rats.Methods One hundred and seventeen 7-day-old Sprague- Dawley rats were randomly divided into 4 groups:a control group(n=32),a hypoxic ischemia brain damage group (HIBD group,n=30),a hyperbaric air group(HBA group,n=27),and a hyperbaric oxygen group(HBO group, n=28).The HIBD model was established by permanent occlusion of the left common carotid artery followed by expo- sure to a mixture of 8% oxygen/92% nitrogen for 2 h(at 37℃).HBO therapy was administered to the HBO group after the hypoxia exposure once a day for 7 d,as was HBA therapy to the HBA group.Apoptotic cells in the cortex and hippocampus(A_(CH)cells)were measured using TUNEL at 9 d after birth,and the ratios of left and right cerebral hemisphere weight(R_(L/R))and rate of weight gain(GRW)were recorded 14 d after birth.A radial arm maze acquisi- tion test(RAMAT)was administered at 30 to 35 days.Lastly,the neuron density in the CA_1 subfield of the rats' hip- pocampi(ND_(CAI)was measured with Nissl staining.Results R_(L/R)and GRW in the HIBD group were significantly lower than in the control group(P<0.01),while R_(L/R)was increased in the HBO and HBA groups,especially in the HBO group(P<0.01),although there was no significant difference in GRW between the groups.Compared with the control group,A_(CH)cells were increased and ND_(CAI)was decreased in the HIBD group(P<0.01),while A_(CH)cells were decreased and ND_(CAI)was elevated in the HBO group in comparison with the HIBD group(P<0.01).There was no change in A_(CH)cells or ND_(CAI)in the HBA group.The RAMAT results for the HIBD group,including the time to find the arms baited with water,average times of working errors and reference memory errors,were significantly high- er than those of the control group,while these values for the HBO group were obviously lower than for the HIBD group,and there was no change for the HBA group(P>0.05).Conclusion HBO therapy might increase the re- covery of learning and memory function by attenuating HIBD in neonatal rats.
9.Neurotoxicity of quinolinic acid to spiral ganglion cells in rats.

Our study investigated the neurotoxicity of quinolinic acid (QA) to spiral ganglion cells (SGCs), observed the protective effects of N-methyl-D-aspartate (NMDA) receptor antagonist MK-801 and magnesium ions on the QA-induced injury to SGCs, and analyzed the role of QA in otitis media with effusion (OME)-induced sensorineural hearing loss (SNHL). After culture in vitro for 72 h, SGCs were exposed to different media and divided into 4 groups: the blank control group, the QA injury group, the MK-801 treatment group, and the MgCl(2) protection group. The apoptosis rate of SGCs was analyzed by Annexin V and PI double staining under the fluorescence microscopy 24 h later. SGCs were cultured in vitro for 72 h and divided into four groups: the low concentration QA group, the high concentration QA group, the MK-801 group, the MgCl(2) group. The transient changes of intracellular calcium concentration were observed by the laser scanning confocal microscopy. Apoptosis rate in QA injury group was higher than that in blank control group and MgCl(2) protection group (both P<0.05), but there was no significant difference between MK-801 treatment group and blank control group (P>0.05). In high concentration QA group, there was an obvious increase of the intracellular calcium concentration in SGCs, which didn't present in low concentration QA group. In MgCl(2) group, the peak values of the intracellular calcium concentration in SGCs were reduced and the duration was shortened, but the intracellular calcium concentration in SGCs had no significant change in MK-801 group. It was concluded that QA could injure SGCs by excessively activating NMDA receptors on the cell membrane, which might be the mechanism by which OME induced SNHL, while Mg(2+) could protect the SCGs from the neurotoxicity of QA.
10.The clinical value of MSCTA in diagnosing lower extremity arterial disease of diabetic foot ulcers

Objective To study the value of MSCT angiography(MSCTA)in diagnosing lower extremities arterial disease (LEAD) of diabetic foot ulcers and analyze the possible influential factors on the onset of diabetic foot ulcers.Methods 80 diabetic patients for MSCTA examination were divided into two groups by whether combined with foot ulcers.The tibial artery calcification score (TACS) and peripheral arterial occlusion index (PAOI) were calculated and the possible risk factors of diabetic foot ulcer were analyzed.Results There were 30 cases suffered with foot ulcer and 18 cases occurred lower extremity arterial occlusion (LEAO), the average PAOI was 4.40(3.60, 5.75).Non-foot ulcer cases were 50, and there were 8 cases suffered with LEAO, the average PAOI was 2.05(1.43, 3.10).Compared to patients without foot ulcer, patients with foot ulcer were older, had a lower BMI, and were more likely to have a history of tobacco use.They usually had higher TACS and severer peripheral arterial occlusive disease (PAOD).Stepwise regression model showed that foot ulcers had significant correlation with TACS and PAOI.Partial correlation analysis showed TACS was independent from PAOI in impacting diabetic foot ulcer.Conclusion MSCTA has important clinical value in diagnosing and valuing diabetic foot ulcer LEAD.Age, smoking history,BMI,TACS and PAOI affect the incidence of diabetic foot ulcers, and TACS and PAOI are the independent risk factors for diabetic foot ulcers.
