OBJECTIVE:
To investigate the mechanism of induction of ferroptosis by brazilin in breast cancer cells.
METHODS:
Breast cancer 4T1 cells were divided into 6 groups: control, brazilin 1/2 half maximal inhibitory concentration (IC50), IC50, 2×IC50, erastin (10 µg/mL) and capecitabine (10 µg/mL) groups. The effect of brazilin on the proliferation of 4T1 cells was detected by cell counting kit-8 assay, and the treatment dose of brazilin was screened. The effect of brazilin on the mitochondrial morphology of 4T1 cells, and the mitochondrial damage was evaluated under electron microscopy. The levels of Fe2+, reactive oxygen species (ROS), malondialdehyde (MDA), glutathione (GSH) and glutathione peroxidase 4 (GPX4) were estimated using various detection kits. The invasion and migration abilities of 4T1 cells were detected by scratch assay and transwell assay. The expressions levels of tumor protein p53, solute carrier family 7 member 11 (SLC7A11), GPX4 and acyl-CoA synthetase long-chain family member 4 (ACSL4) proteins were quantified by Western blot assay.
RESULTS:
Compared to the control group, the 10 (1/2 IC50), 20 (IC50) and 40 (2×IC50) µg/mL brazilin, erastin, and capecitabine groups showed a significant decrease in the cell survival rate, invasion and migration abilities, GSH, SLC7A11 and GPX4 protein expression levels, and mitochondrial volume and ridge (P<0.05), and a significant increase in the mitochondria membrane density, Fe2+, ROS and MDA levels, and p53 and ACSL4 protein expression levels (P<0.05).
CONCLUSIONS
Brazilin actuated ferroptosis in breast cancer cells, and the underlying mechanism is mainly associated with the p53/SLC7A11/GPX4 signaling pathway.

Objective:To investigate the current situation of standardized patient programs and standardized patient educators in China, and to analyze the existing problems in the implementation. Methods:Questionnaire survey was used in this study. The questionnaire was made by Delphi method, and distributed via E-mails or through the internet to medical colleges, affiliated hospitals and general hospitals across the country. Microsoft Office Excel 2016 was used for data reduction and analysis and cartography. Results:A total of 94 medical colleges and hospitals of 27 provinces and municipalities participated in the survey, of which 43.62% had carried out standardized patient programs. The primary factor affecting the program implementation was the lack of standardized patient educators. The existing trainers were mainly clinicians or nurses, and the main way of training the educators was to send them to other universities for learning. A majority of respondents (94.68%) think it is necessary to establish and formulate a unified national certification system for standardized patient educators. Conclusion:The development of standardized patient program is unbalanced in China, and the lack of standardized patient educators has become the primary factor restricting the development. In order to make standardized patients a greater role in medical education, we should encourage different types of personnel to join in the standardized patient training team, to clarify the responsibilities of standardized patient educators, to standardize the process of trainers training, and to establish the certification system of standardized patient educators.

Aim To study the protective effect of ZN-RF2 on OGD/R-induced injury and the autophagy-related mechanism in PC12 cells. Methods PC12 cells were cultured in vitro and divided into normal group and OGD/R group. qRT-PCR and Western blot were used to measure the mRNA and protein expressions of ZNRF2. To explore the effect of ZNRF2 on OGD/R-induced injury in PC12 cells, cells were grouped into normal group, OGD/R group, LV-ZNRF2 group, LV-NC group, siR-ZNRF2 group and siNC group. Cell viability was detected by MTT assay, cell apoptosis was measured by flow cytometry and the expressions of autophagy-related proteins LC3II, p62, Beclin-l were accessed by Western blot. Results Compared with normal group, the cell viability decreased in OGD/R group, the cell apoptosis increased markedly, and the expressions of ZNRF2 mRNA and protein were downregulated significantly. Simultaneously, the proteins expressions of LC3II and Beclin-1 increased, and the expression of p62 protein decreased in OGD/R group. Compared with OGD/R group, the cell viability was enhanced, the cell apoptosis and autophagy were decreased in LV-ZNRF2 group. In contrast, the cell viability decreased and the cell apoptosis and autophagy were aggravated after transfecting siR-ZNRF2. Conclusions ZNRF2 protects PCI2 cells from the injury caused by OGD/R and its mechanism may be related to the inhibition of autophagy.

Based on the story of Chinese idiom, (a magical and skilled form of craftsmanship) as the breakthrough point, this paper discusses the both (cook) and the experienced acupuncture practitioner have the same high skills and explores the potential relationship between mind-regulation in treatment with acupuncture and flow theory. It is believed that the skills of ancient acupuncture practitioner in mind-regulation with acupuncture is not only a kind of "Tao" mode, but also a state of "flow". By the discussion on mind-regulation and flow theory, modern people may have more clear recognition on the mind regulation in treatment with acupuncture so as to better determine the therapeutic methods of acupuncture for mind-regulation.

Chinese Journal of Epidemiology 2020;41(10):1686-1691
Objective:To explore the influencing factors on perinatal mortality of pregnant women with HIV infection to reduce the mother-to-child transmission in Sichuan province. Methods:In this study, 4 786 perinatal infants of the HIV-infected pregnant women were included. Related data on perinatal epidemiology was reported by all the 183 medical and health care institutions where the HIV prevention of mother-to-child transmission program was initiated in 2005-2016. Univariate χ2 test and multivariate logistic regression methods were used to analyze the perinatal mortality outcomes and influencing factors. Results:The overall perinatal mortality rate was 25.7‰ (123/4 786) among HIV-infected pregnant women, with annual downwarding trend (trend χ2=32.220, P=0.000). Perinatal mortality rate appeared the highest ( χ2=4.130, P=0.042), with more fetal deaths and stillbirths and less early neonatal death within 7 days in Liangshan county ( χ2=29.626, P=0.000). Results from the multivariate logistic regression analysis showed that fewer pregnant numbers would contribute to the, lower perinatal mortality rate (1-2 pregnancies OR=0.417, 95 %CI: 0.184-0.943; 3-4 pregnancies OR=0.447, 95 %CI: 0.223-0.895). Perinatal deaths were more likely to be prevented if LPV/r protease inhibitor-based triple antiviral therapy was provided ( OR=0.530, 95 %CI: 0.285- 0.986) or delivery was taken place in the hospital (hospital of municipal-level and above OR=0.222, 95 %CI:0.098-0.499; county-level hospital OR=0.282, 95 %CI: 0.166-0.480; township-level hospital OR=0.134, 95 %CI: 0.031-0.586) among HIV-infected pregnant women. However, premature delivery or neonatal asphyxia would increase the risk of perinatal mortality (premature delivery OR=8.285, 95 %CI: 5.073-13.533; neonatal asphyxia OR=9.624, 95 %CI: 4.625-20.028). Conclusions:The perinatal mortality rate of HIV-infected pregnant women appeared significantly higher than that in the province or the whole country. Strategies involving LPV/r-based triple antiviral therapy, promotion of hospital delivery, reducing the incidence rates of premature deliveries and neonatal asphyxia, should be strengthened.
