Schizophrenia is one of the most severe psychiatric disorders.Although the etiology is unknown, schizophrenia appears to be a polygenic disorder associated with environmental factors. There are evidences that intracellular signaling plays an important role in the pathogenesis of schizophrenia.Canonical Wnt pathway, which is closely related to the development and function maintenance of central nervous system, have drawn wide attention.Evidences for potential disturbances of Wnt signaling pathways in schizophrenia have accumulated, which are in favour of the hypothesis that Wnt signaling alterations may be important in the pathogenesis of schizophrenia. The research advances in the relationship between canonical Wnt signaling pathway and schizophrenia are reviewed in this paper.

BACKGROUND:How to promote wound healing is always the research focus of the surgical physicians in the clinic. Recombinant human epidermal growth factor (rhEGF) can effectively promote wound healing. However, as a biological agent, it is easy to be decomposed under normal temperature.
OBJECTIVE:Toexplorethe mechanisms underlying the promotion of wound healing in the back of rabbits by bletila carrying exogenous rhEGF.
METHODS:Model rabbitswith ful-thickness skin defects in the back were treated with bletila carrying exogenous rhEGF (combined treatment group), bletila, rhEGF, or saline (control group).
RESULTS AND CONCLUSION:The time of wound healing was the shortest (P< 0.05) and the wound healing rate was the highest (P< 0.05) in the combined treatmentgroup. On postoperative days 3 and 10, newly formed granulation tissue, capilaries, and colagenous fibersshowed by hematoxylin-eosin staining staining and Masson staining and the strongest immunoreactivity of vascular endothelial growth factor determined by immunohistochemical staining were found in the combined treatment group (P< 0.05). These findings confirm that bletila carrying exogenous rhEGF promotes wound healing by accelerating the forming of granulation tissue, new-born capilaries, and colagenous fibers, and the effects are superior to either of them alone.

Chinese Pharmacological Bulletin 2010;26(2):182-185
Aim To provide practical method for screening human carbonic anhydrase Ⅱ(hCA Ⅱ) inhibitors in drug discovery. Methods hCA Ⅱ protein was obtained from induced BL21(DE3) E.coli containing plasmid pET-28b-hCA Ⅱ.The hCA Ⅱ activity was detected under pH 7.6 and 25℃ by its esterase activity which could decompose PNPA to increase the photoabsorption at 348 nm. After the assay conditions were finally selected, 35 new compounds were tested. Results A practical method for screening hCA Ⅱ inhibitors was successfully constituted by using recombinant hCA Ⅱ protein expressed in E.coli as the source of hCA Ⅱ enzyme.10 new compounds had better inhibitory effect and 9 new compounds had the same inhibitory effect on hCA Ⅱ compared with acetazolamide. Conclusions The hCA II inhibitor screening technique constituted in this work possesses advantages of being reliable, rapid, and practical. 19 new compounds are worth further research for developing high efficiency and low side effect drugs used for high-altitude illness.

Objective To investigate the effect of MDR1 and MDR3 gene silence by shRNA of human breast carcinoma cell line MCF-7/Adr,and explore the role of MDR1 and MDR3 in adriamycin-resistance of breast carcinoma cells. Methods shRNA plasmid vector specifically targeting MDR1 and MDR3 gene was transfected into cells. The control group was transfected with empty vector. The concentration of adriamycin was detected by the flow cytometry (FCM). Cell apoptosis was analysed by FITC-Annexin-V/PI double staining. Cell viability and the IC50 of adriamycin on MCF-7/Adr cells were determined by MTT method. MDR1 and MDR3 mRNA were assessed by RT-PCR. P-gp expression was detectedby immunochemistry. Results After treatment with ABCB1 and ABCB4 shRNA plasmid vector, the apoptosis of MCF-7/Adr cells was (30.21±1.65)%and (22.07±2.17)% respectively. Compared with untransfecedgroup and empty vector transfection group the difference was significant(P<0.01). MDR1 and MDR3 shRNAcould increase cellular adriamycin accumulation of MCF-7/Adr cells. MCF-7/Adr cells viability and the IC50were significantly decreased after transfection. Compared with untransfeced group and empty vector transfectiongroup, the mRNA level of MDR1 and MDR3 in MCF-7/Adr cells were decreased by (89.5±0.8)%and(85.1±1.2)%, the reduction of MDR1 and MDR3 mRNA was in a time-dependent manner. Immunochemistry proved that the expression of p-gp was significantly inhibited. Compared with untransfeced group and empty vector transfection group the difference was significant (P<0.05). Conclusion The shRNA can effectively and specifically silence the expression of MDR1 and MDR3 gene, reverse the adriamycin-resistance mediated by P-gp in MCF-7/Adr cells. The reversal effect of adriamycin-resistance by shRNA of MDR1 is more effective than that of MDR3.
