2.Expression of Fas/FasL in Children with Aplastic Anemia and Its Clinical Significance

Objective To investigate the expression of Fas/FasL in children with aplastic anemia(AA) and its clinical significance.To explore the pathogenesis of AA.Methods CD34+ cell counts and the expression of Fas/FasL in 12 children with severe AA(SAA),18 with chronic AA(CAA)and 10 normal children were detected by flow cytometry combining monocolon antibody.Results The expression of Fas in children with AA were significantly higher than that in control group(P0.05).Conclusions Fas/FasL systeam take part in inducing CD34+ cell apoptosis.This may contribute to understanding the decrease number of stem cells and bone marrow failure.
3.Ultra-Structural Features of Interstitial Cells of Cajal in Fetal Enteron

Objective To study the ultra-structural features of interstitial cajal cells (ICC)in fetal enteron and then classify them.Methods Ultra-structural features of ICC in fetal enteron with spontaneous abortion or premature birth were detected under JEM-2000EX transmission electron microscope for the cause of fetal death, including two fetus specimens of a gestation 18 months and 28 months stained with lead nitrate and uranium acetate.Results ICC had a big oval karyon and a little of cytoplasm. ICC formed spindle or stellate cells with 2 to 5 long cell processes . From the esophagus to the terminal ileum ICC mostly had the same ultra-structural features, but with mitochondria and a well-developed endoplasmic reticulum and Golgi complex in the whole colon and the rectum , more than those of esophagus and small intestine. In the whole colon and rectum ICC had similar ultra-structural features. ICC also possessed an abundance of mitochondria and a well-developed endoplasmic reticulum and Golgi complex. ICC also possessed Caveloae lipid droplet with more electron dense and heterochromatin. Two types of ICC were identified under JEM-2000EX transmission electron microscope. One type was present from the esophagus to the terminal ileum and the other type was observed in the colon and rectum. The first type ICC in circular muscle layer was bipolar cells which extended to tapering processes in opposite directions. These processes rarely branched, and their appearance was similar to smooth muscle cells.Most of them ran parallel to the circular muscles . The second type of ICC in the myenteric plexus and longitudinal muscle layer was mostly multipolar and rarely bipolar cells with long processes. They showed an irregular appearance characterized by numerous short spike-like branches. Processes of multipolar cells extended in every direction and connected with each other. ICC nerve cells and smooth muscle cells were connected with gap-like junction, which was the main connection mode .Conclusions The ultra-structural features of ICC in fetal enteron varied with the diffe-rent locus and different tissue sheets in the enteron. The mitochondria and a well-developed endoplasmic reticulum and Golgi complex of the whole colon and rectum are more and more developed than that of esophagus and small intestine. Ultra-structural features of ICC will develop further with the gestational age. The gap- like junction among ICC nerve cell and smooth muscle cell are highly important for ICC to educe function.
4.Screening and isolation of fibrinolytic active compound from marine microorganism

Objective To isolate bioactive compound of enhancing fibrinolysis from secondary metabolites of marine microorganism.Methods The separation of microorganism from seawater samples,screening of producing fibrinolytic compound's strain,selection of the active strain's optimum fermentation medium and refining of active compound were done by the method of selective cultivation,measuring of compound's fibrinolytic activity and semipreparative HPLC,respectively.Results Nine hundred and thirty-six single strains from 31 samples were collected 100 meters off the coast,and cultures of the fungus(FG216) contained enhancing fibrinolytic compound.Compounds from modified Czapek medium as the fermentation medium of FG216 showed significant fibrinolytic effect.Finally,active fraction were isolated and refined from cultures of FG216.Conclusion In this paper,active compound of enhancing fibrinolysis were gained from secondary metabolites of isolated single microorganism from seawater.
6.HPLC simultaneous determination of contents of 5 saponin constituents in Ophiopogonis Radix.

This research is to establish an HPLC method for the simultaneous determination of ophiopogonin D, ophiopogonin D', ophiopogonin C, deacetylophiopojaponin A and ophiogenin-3-O-α-L-rhamnosyl-(1-->2)-β-D-glucoside in Ophiopogonis Radix. HPLC-ELSD analysis was performed on a Kromasil 100-5 C₁₈ column (4.6 mm x 250 mm, 5 µm), with the mobile phase of acetonitrile (A) -water (B) in gradient elution mode (0-45 min, 35%-55% A), at a flow rate of 1 mL · min⁻¹. The column temperature was 35 °C and the drift tube temperature was 100 °C in a gas flow rate of 3.0 L · min⁻¹. The result showed that baseline of all the 5 constituents was well separated, and every constituent had wide linearity range and good linear relation (r > 0.999). The recovery rate was between 95.75% and 103.1%. The new established method for simultaneous determination of saponin constituents in Ophiopogonis Radix was sensitive and has good, repeatability. It could be applied to quality evaluation of Ophiopogonis Radix.

7.Relationship between Apoptosis of Bone Marrow Cells and Tumor Necrosis Factor-?,Fas/FasL Expression in Children with Aplastic Anemia

Objective To investigate the relationship between apoptosis of bone marrow cells and tumor necrosis factor-?(TNF-?),Fas/FasL expression in children with aplastic anemia.Methods The concentration of TNF-? in supernatant of cultured bone marrow mononuclear cell(BMMC) was measured by enzyme linked immunosorbent assay.The expression of Fas/FasL,apoptosis ratio of bone marrow CD34+ cell were evaluated by flow cytometry.Results The concentration of TNF-? in supernatant of cultured BMMC in AA group[SAA (27.9?8.20) ng/L;CAA (23.6?6.78) ng/L] increased significantly compared with control group(t=3.432 P0.05).Apoptosis ratio of CD34+ cell in SAA group[(24.6?9.56)%] and CAA group[(20.9?7.32)%] significantly increased compared with control group(t=3.492,3.458 Pa0.05).The concentration of TNF-? was positively correlated with the expression of CD34+ Fas+ in all types AA children(r=0.542 P0.05).Conclusions TNF-? and Fas/FasL system can be involved in inducing CD34+ cell apoptosis.This may contribute to understanding the decreased number of stem cells and bone marrow failure.

9.Experimental study on the theraputic effect of crescent Euphorbia on lewis lung cancer in mice.

OBJECTIVETo observe the inhibitory effects of crescent euphorbia on tumor growth, immunoregulation, the side effect on the mice blood system, cell cycle and to investigate its effect on apoptosis of tumor cells.
METHODCrescent euphorbia was administered gastrally to C57BL/6J mouse implanted with Lewis lung cancer for 12 days. Inhibition on tumor growth, immunoregulation,the side effect on the mice blood system and cell cycle were observed and its effect on apoptosis of lung cancer cells were investigated.
RESULT(1) Inhibitory rates of crescent euphorbia at doses of 7.5, 15, 30 g x kg(-1) and 60 g x kg(-1) were 0.61%, 16.93%, 32.81% and 58.26% respectively, significant differenel with those of controls at doses of 30 g x kg(-1) and 60 g x kg(-1)(P < 0.05). (2) Although no effect on thymus index in normal mouse at all dose of crescent euphorbia was found, spleen index in tumor-bear mice was reduced significantly at dose of 60 g x kg(-1)(P < 0.05). (3) No side effect on the miceblood system was found. (4) The percentage of S phase of cell cycle was increased in the group of crescent euphorbia at dose of 30 g x kg(-1), 60 g x kg(-1) and the apoptotic rate was (16.43 +/- 18.69)% and (24.37 +/- 15.48)% by flow eytometry (P < 0.05). The apoptotic index (AI)of these two groups were (4.00 +/- 7.50)% and (5.93 +/- 5.96)% by TUNEL, however apoptosis was found in the group at doses of 60 g kg(-1) (P < 0.01).
CONCLUSIONCrescent euphorbia can inhibit the growth of Lewis lung cancer in mice, no side effection for the mice blood system and the effect might be associated with induction of apoptosis on cancer cells.

10.A randomized controlled trial: acclimatization training on the prevention of motion sickness in hot-humid environment.

OBJECTIVEIncidence and severity of motion sickness (MS) in hot-humid environment are extremely high. We tried to know the effect of two-stage training for reducing incidence and severity of ms.
METHODSSixty male subjects were divided into experimental group and control group randomly. Subjects in experimental group received: (2) adaptation training including sitting, walking and running in hot lab. After adaptation confirmation based on subjective feeling, rectal temperature, heart rate, blood Pressure, sweat rates and sweat salt concentration, we tested both groups by Coriolis acceleration revolving chair test and recorded Graybiel's score and grading of severity to evaluate whether adaptation training was useful; (2) Anti-dizzy training 3m later of deacclimatization contained revolving chair training for 10 times. Then we did the same test as mentioned above to evaluate effect of anti-dizzy training. RESULST: Graybiel' s score and grading of severity had no difference between two groups through acclimatization training (P > 0.05). While they had difference through anti-dizzy training (P < 0.01).
CONCLUSIONAdaptation training seems useless for reducing incidence and severity of MS in hot-humid environment, but anti-dizzy training is useful.
