2.Recombinant Adenovirus Infected HaCat Cells Express Human Kerotinocyte Growth Factor (hKGF)

Human kerotinocyte growth factor (hKGF) gene amplified by PCR was inserted into the shuttle vector pAdTrack-CMV to get the recombinant plasmid pAdTrack-CMV-hKGF, which was linearized with Pme I and transferred into Escherichia coli BJ5183 containing the adenoviral bone plasmid pAdEasy-1 to obtain the recombinant adenoviral plasmid pAdEasy-hKGF. The recombinant adenoviral plasmid was then transfected into HEK-293 cell lines via Lipofectamine 2000 to package and amplify the recombinant adenovirus containing hKGF gene detected by PCR. The recombinant adenovirus produced could effectively infect HaCat cells. The result of Western blotting showed that HaCat cells infected with the recombinant adenovirus expressed and secreted hKGF protein.
3.Comparison of cold blood and cold crystalloid cardioplegia on myocardial protection effects in valve replacement surgery

Objective To compare the myocardial protection effects of cold blood cardioplegia and cold crystalloid cardioplegia in patients undergoing valve replacement surgery.Methods One hundred adult patients with cardiac function Ⅱ~Ⅲ class undergoing primary elective valve replacement surgery due to rheumatic heart disease were retrospectively analyzed.For patients with cardiac arrest,cold blood cardioplegia was applied to 50 cases(Group B),and cold crystalloid cardioplegia to the others(group C).The process of heart resuscitation,the postoperative need of dopamine,the blood pressure and heart rate,the postoperative level of serum myocardial enzyme,the intensive care unit(ICU) stay after operation were collected and statistically analyzed.All the cardioplegic solution was perfused in a similar manner and topical cooling was employed simultaneously.Results There were less cases in group B than in group C who needed isoprenaline for cardiac resuscitation after clamp removal(P0.05).Conclusion The cold blood cardioplegia and cold crystalloid cardioplegia have similar myocardial protection effects on rheumatic heart disease patients with cardiac function Ⅱ~Ⅲ class undergoing valve replacement surgery.
4.Changes in electrophysiological and morphological properties of neuron in the ventral partition of medial geniculate body during the postnatal development of rats.

We investigated the electrophysiological and morphological characteristics of the neurons in the ventral partition of medial geniculate body (MGBv) in the rat during postnatal development (postnatal day 3-30, P3-30) with whole-cell patch clamp recording and intracellular labeling technique. The results are as follows: (1) There was an increase in the negativity of the resting membrane potential, a decrease in the input resistance and the time constant of the MGBv neurons of rats during the postnatal development; the amplitude, threshold and duration of the action potentials were also different on different postnatal days; (2) During the postnatal development, the changes in voltage-dependent ion channels dramatically influenced the waveform of action potential in the MGBv neurons. Application of 4-AP, a K(+)-channel blocker, reduced the amplitude and prolonged the durations of the action potentials in P6 neurons, and inactivated the neurons at the end of the pulse in P16 rats. Application of CdCl2 to block the voltage-dependent Ca(2+)-channel decreased the amplitude and increased the duration of the action potential in P16 rats; (3) Biocytin staining revealed that immature MGBv cell had smaller somata and short, simple dendritic aborization. During postnatal development, the matured neurons had oval somata and 3-4 primary dendrities that radiated irregularly and projected outside the section; spines also appeared on the dendrites of the mature neurons. These results reveal that the morphologic and electrophysiological properties are not matured during early postnatal development, and that the electrophysiological and morphological features of the developing MGBv neurons are interrelated.

5.Clinicopathological analysis of 34 cases of primary small intestine lymphoma

Objective To analyse the clinicopathological features of primary small intestine lymphoma(PSIL), and explore the relationship between clinical stage,histological findings,therapeutic modality and prognosis. Methods The clinical data of 34 cases of PSIL were collected,the pathohistological features and results of immunohistochemical examinations were obtained,and the follow-up findings were adopted for comprehensive analysis. Results Among these 34 cases of PSIL,abdominal pain or discomfort,gastrointestinal bleeding and abdominal mass were the predominant symptoms.PSIL mainly involved ileum,especially the bottom of ileum and ileocecal area.Among the 26 patients with follow-up for more than one year,the 1-year survival rate was significantly higher in patients without tumor perforation than those with tumor perforation(76.2% vs 20.0%)(P
6.Clinical analysis of vitrectomy in treatment of 48 eyes with metallic foreign bodies

AIM:To investigate the effect of vitrectomy in treatment of metal intraocular foreign bodies and the factors affecting visual prognosis.
METHODS:Fourty seven cases ( 48 eyes ) with foreign bodies from January 2010 to June 2013 in our hospital underwent vitrectomy combined with intraocular foreign body removal were retrospectively analyzed. The changes of visual acuity, the size of foreign body, the complications of preoperative, and the relationship between treatment time and visual acuity were recorded.RESULTS: Totally 46 eyes were successfully removed the complete foreign body, 2 eyes of orbital foreign bodies had not been removed. After 6mo of followed-up, the best corrected visual acuity were all improved in varying degrees (P<0. 05), intraocular foreign body size combined with retinal detachment was the risk factor for visual impact (P<0. 05).
CONCLUSION:Vitrectomy has obvious advantages in the treatment of metal intraocular foreign bodies. Timely and appropriate vitrectomy is very important to the recovery of visual function.
7.Effects of iodine excess on mitochondrial superoxide production and mitochondrial membrane potential in rat thyroid cell line cells

Objective To investigate the effects of iodine excess on mitochondrial superoxide production and mitoehondrial membrane potential(△ψ)changes in Fisher rat thyroid cell line(FRTL)cells.Methods FRTL cells were treated with 10-4mol/L potassium iodine(KI),10 U/L thyrotropin(TSH),10-4 mol/L KI+10 U/L TSH respectively for 24 h.Effects on cell proliferation were assayed by methyl thiazolyl tetrazolium(MTT)colorimetric method.Changes of mitochondrial superoxide production and △ψ were measured by live cell imaging and spectrofluorometer using MitoSOX and rhodamine 123(rh123)respectively.Results Absorbance(A)in the KI group (0.794±0.144)showed a significant decline compared to the control group(1.000 ±0.183,P<0.05),whereas a significant elevation was observed in the TSH group(1.215±0.156,P<0.05).No significant differences was found between the KI+TSH group(1.025±0.254)and the control group(P>0.05),but the former was marked higher than the KI group(P<0.05).Compared to the control group(9.74±3.24).MitoSOX mean fluorescence intensity (MFI)in the KI and KI+TSH groups(18.16±6.57,13.33±2.92)were significantly increased(all P<0.05),which was a significant decline in the TSH group(6.64±2.15,P<0.05).MitoSOX MFI in the KI+TSH group was lower than the KI group(P<0.05).Rh123 MFI in the KI and KI+TSH groups(210 593±31 328,295 525±34 243)showed significant decline than the control group(407 824±37 198,all P<0.05).Compared with the KI group.the KI+TSH group pronouncedly attenuated the reduction of Rh 123 MFI(P<0.05).No significant differences of Rh 123 MFI were found between the TSH group(411 187 ± 72 852) and the control group(P > 0.05). Conclusion Iodine excess (10-4 mol/L KI) may lead to peroxide damage on the mitochondria of FRTL cells, and cell proliferation is inhibited. Combining treatment with 10 U/L TSH may attenuate mitochondrial peroxide damage and inhibition of cell proliferation caused by iodine excess.
8.Proteomic study of the effect of recombinant hFGF-10 adenovirus on HaCat cells.

This study is to investigate the effect of recombinant hFGF-10 adenovirus on the proteome of HaCat cells, and to speculate further the possible mechanism of the effect of hFGF-10 on HaCat cells via differentially expressed proteins identified. Two-dimensional gel electrophoresis (2-DE) combined with tandem time-of-flight mass spectrometry was applied to identify the differentially expressed protein spots on the 2-DE maps of the whole-cell proteins from Ad infected and rAd-hFGF-10 infected HaCat cells. The mRNA and protein levels of the differentially expressed proteins were confirmed with semi-quantitative RT-PCR and Western blotting. The results showed that the 2-DE maps with high resolution were obtained, and four selected differentially expressed proteins involved in cell apoptosis, cytoskeleton regulation and protein degradation were identified with MALDI-TOF/TOF. The mRNA and protein levels of one of the differentially expressed proteins, VDAC2, were up-regulated in HaCat cells infected with the recombinant hFGF-10 adenovirus. The differentially expressed protein, VDAC2, may be related to the bioactivities of hFGF-10.

9.Study on critical indicators for nursing practice environment

Objective To identify the critical indicators of nursing practice environment in China, as a reference for related studies. Methods Descriptive systematic review was used to retrieve literature of nursing practice environment, collect all potential indicators and design questionnaires based on those indicators. In the end, 39 potential indicators were identified, for a questionnaire survey of nurses from cooperating medical institutions in 30 provinces in China, with the data subject to statistical analysis in multiple linear regression model. Results Questionnaire items of higher mean score were usually found with a smaller 95% confidence interval, while coefficient of variation tended to be lower. Seventeen indicators were recommended as key indicators for nursing practice environment in China, including proper working hours and intensity, opportunities for nurses to participate in hospital management, good working relationship with doctors within a department,nursing managers′support for nurses′right decisions. The remaining components turned out to be insignificant in the analysis. Conclusions The influencing factors for the nursing practice environment in China are different from other countries. In this consideration, nursing managers should further explore and focus on such special indicators, and proactively improve the quality of nursing practice environment for better nursing service.
10.Somatic symptoms and dynamic changes after treatment in elderly inpatients with depression:A 1-year follow-up study

Objective:To explore the characteristics of somatic symptoms and their dynamic changes after treatment in elderly inpatients with depression,so as to give supported data in differential diagnosis and management of somatic symptoms in the elderly patients.Methods:Totally 69 consecutively hospitalized depression patients(≥60 years)were enrolled in the study,and 62 of them completed the 12-month follow-up.The diagnoses were made according to International Statistical Classification of Diseases and Related Health Problem-10th revision(ICD-10) criteria.The Hamilton Depression Scale(HAMD),Hamilton Anxiety Scale(HAMA)and Somatic Symptom Self-rat-ing Scale-Somatic factor(SSS-S)were used to assess the severity of depression,anxiety and somatic symptoms, which were evaluated at baseline,2 weeks,4 weeks and the end of 12-monthr espectively.The Minimum Mental State Examination(MMSE)were used to evaluate cognitive function,and Global Assessment Function(GAF)were used to evaluate social function at the end of 12-month follow-up.Result:Somatic symptoms presented in the all patients at the stages of depressive episode.The most common ones were gastrointestinal symptoms(91%),dizzi-ness or headache(66%),cardiovascular symptoms(59%).SSS-S scores at 2 weeks and 4 weeks of antidepressant treatment were significantly improved compared with those of baseline,[(13.6 ±3.2),(12.4 ±2.8)vs.(17.1 ± 4.8),analyzed by repeated measures of ANOVA,P <0.05].The changes of SSS-S scores were positive associated to the changes of anxiety symptoms and the effect of physical diseases,analyzed by hierarchical linear model analy-sis.At the stages of rehabilitation,residual somatic symptoms were positive associated to the degree of anxiety symptoms(OR =2.132,P <0.05),and unrelated to the level of HAMD,MMSE and GAF scores analyzed by logis-tic regression analysis.Conclusion:Somatic symptoms are various in elderly inpatients with depression during the a-cute phase,which are improved greatly as effective antidepressant treatment,and still have residual somatic symp-toms at the stage of rehabilitation.The effect of physical diseases and anxiety symptoms have influence on somatic symptoms.
