1.A method of assessing the thickness of myocardial basement membranes

Aim To observe the thickness changes of cardiomyocyte and microvascular basement membranes(BM)and the expression of collagen Ⅳ in Gk rats of different months and STZ-induced diabetic rats,and to explore the method that operated easily and could assess the thickness of myocardial BM.Methods GK rats of the fourth,fifth,sixth,seventh months,Streptozotocin induced diabetes rats of fifth months and normal Wister rats were respectively divided into different groups with ev eight rats in each group.The myocardial section was stained by methenamine silver and immunohistochemistry was performed for collagen Ⅳ.Left ventricular endocardium was selected as measuring view.Through computer image analysis,the ratio of BM positive staining density(BMPSD)and positive staining density of collagen Ⅳ(CPSD)were calculated separately in all myocardium.Take the ratio to assess the thickness of myocardial BM and the expression of collagen Ⅳ.Results The glycemia,BMPSD and CPSD of the two diabetes rats were higher than those of the normal rats,having highly remarkable difference(P0.05).Conclusion By methenamine silver staining for myocardial section,calculating BMPSD through computer image analysis,can be an index to assess the thickness of myocardial BM.

3.Non-coding RNAs and erectile dysfunction: An update.

Erectile dysfunction (ED) is a disease associated with a variety of factors such as age, psychological factors, physical conditions, and lifestyle, and it severely affects the patients quality of life. In the past, some non-coding RNAs (ncRNAs) transcribed by genes but not translated into the protein were regarded as the "waste" in the process of gene expression. But in the recent years, ncRNAs have been found to play a crucial role in regulating gene expression and its products, which may affect penile erectile function. This review focuses on the recent progress in the studies of the relationship between ncRNAs and erectile dysfunction.

4.In vitro and in vivo models of osteosarcoma for research of enhanced green fluorescent protein expression

Background and purpose: Mouse osteosarcoma model was widely used in osteogenic malignant tumor research, and it was helpful for studying the invasion and metastasis of the tumor cells when it was well marked in vivo. The purpose of this study was to establish mouse sarcoma cell lines (S180) that were infected with enhanced green fluorescent protein(EGFP). Methods: EGFP-S180 expressing strong EGFP fluorescence was acquired by electroblot, and supplemented with G418 (800 mg/mL), c-Myc was detected by laser scanning confocal microscopy. Meanwhile, the cancer-bearing model was established subcutaneously within the abdominal cavity. Results: EGFP-S 180 cells were cloned. There was no significantly difference between c-Myc expressions in S180 cells and those in EGFP-S180 cells (P>0.05), and between the cancer-bearing time subcutaneously and the time within abdominal cavity (P>0.05). Conclusion: According to in vitro and in vivo assay, it showed that EGFP-expressing S180 cells could be used for studying further the tumor biological behavior with fluorescence technology.
5.Exploration of establishment of medical information technology curriculum system

Principles of constructing medical information technology curriculum system were pro-posed revolving round the core problem of how to combine needs of developing life science with culturing talents who can independently apply information technology knowledge in studying and researching. De-tailed implementing measures for constructing medical information technology curriculum system were summarized after repeated practical teaching. Longitudinal classification based on discipline and trans-verse classification based on teaching level were combined to construct the whole standard course. Finally, courses were appropriately distributed according to detailed culturing objectives for students in medical universities and colleges,thus a suitable medical information technology curriculum system in medical universities and colleges was formed.
6.Reform of training model for medical information professionals

After the need of medical information professionals was analyzed, the reform of training model for medi-cal information professionals in Department of Information Management and Information System, Hubei Medical and Pharmaceutical College, was described in aspects of the position of its training target, construction of its teaching team, optimization of its course teaching system structure, updating of its teaching methods, and improvement of its practical ability training system in order to provide certain beneficial experiences and reference for the training of medical information professionals in higher education institutions.
7.Preventative and Curative Effect of Tongxinluo on Microvascular Inflammatory Injury and Vasospasm in Cardiac Muscle of Streptozotocin Diabetic Rats

Objective To observe the preventative and curative effect of Tongxinluo on microvascular inflammatory injury and vasospasm in the cardiac muscle of streptozotocin (STZ) diabetic rats. Methods STZ rats were divided into STZ group, insulin group, Tongxinluo group, with the other 8 Wistar rats as normal control. The normal group and STZ group were intragastric administered with NS, insulin group was subcutaneous injected with insulin, and Tongxinluo group was intragastric administered with Tongxinluo suspension for 8 weeks. The sections of cardiac muscle was stained in HE staining and NF-?B immunohistochemical staining. The level of adhesion molecule-1 (sICAM-1), tumor necrosis factor (TNF-?), endothelin-1 (ET-1), angiotensin-Ⅱ (AT-Ⅱ) in myocardial homogenate from intercellular, and level of AT-Ⅱ, neuropeptide Y (NPY) in plasma were determined, with sICAM-1 by enzyme-linked immunosorbent assay and the others by radioimmunoassay. The sections stained by NF-?B immunohistochemical staining were image analyzed using computer. The extent of its expression was evaluated by the mean of the density ratio of the positive side. Results Compared with normal rats, the content of sICAM-1, TNF-?, ET-1, AT-Ⅱ and the expression of NF-?B in the myocardial homogenate of STZ rats was increased (P0.05). 8.Postpartum MRI appearance of placental implantation abnormality

Objective: To explore postpartum MRI appearance and diagnostic value of MRI for placental implantation abnormality (PIA). Methods: Postpartum MRI findings of 11 PIA patients (mean age:28?4.5 years; range 23-25) were analyzed retrospectively. All patients underwent hysterectomy because their placentae were not discharged spontaneously, the postpartum hemorrhage added up to 800-4 500 ml and PIA was diagnosed by MRI 2-6 d after delivery. Results:Compared with the signal of the outer layer of the myometrium on the same scanning sequence, the placental signals appeared isointense or slightly hyperintense and the dividing line between the placenta and uterine wall was indistinct on T_1WI. On T_2WI the placental signal intensity was higher than that of the uterine wall. Placenta accreta showed that the placenta was confluent with the uterine wall and the junctional zone continued and the thickness of junctional zone was normal or slightly thinning. Placenta increta showed that the placental tissue invaded the myometrium in the shape of “triangle”, “nodule” or “mushroom”, and the junctional zone became thinner locally or discontinued. On Gadolinium-enhanced T_1WI, the intensity of placental enhancement was higher than that of uterine wall and the placental tissue was confluent with the uterine wall. The surgico-pathologic examination found 7 cases of placenta accrete and 4 cases of placenta increta. MRI diagnosis showed 8 cases of placenta accreta and 3 cases of placenta increta. There was one misdiagnosis by MRI. Conclusion: MRI is an ideal tool for postpartum diagnosis of PIA and can accurately evaluate placental configuration, the extent of placental accreta and the location and deepness of placental invasion.
10.Forefoot plantar pressure changes of the first tarsometatarsal joint fracture-dislocation fixation by different internal fixations.

OBJECTIVETo measure the changes of plantar pressure of the first tarsometatarsal joint fracture and dislocation by three different implants to provide experimental reference in selecting implants.
METHODSEight fresh foot specimens were made into the models of the first tarsometatarsal joint fracture and dislocation, which were fixed with 3.5 mm cortical screw, 1/4 tubular plate and compressive staple in turn. After the loading of 600 N, the changes of the plantar pressure in forefoot were measured by the method of the F-scan plantar pressure system.
RESULTSAfter first tarsometatarsal joint fracture and dislocation, the peak pressure under the first metatarsal head would decrease, while the pressure under the second metatarsal head would increased,whose differences were statistically significant (P<0.05). When the first tarsometatarsal joint was fixed with screw or plate respectively; the peak pressure under the two metatarsal heads would tend to be normal. However,the staple fixation showed the statistical significant difference compared with normal state, although the peak pressure under the first and second metatarsal heads were recovered in some extent(P<0.05).
CONCLUSIONAfter the first tarsometatarsal joint fracture-dislocation, the plantar pressure might be compensated partly by the adjacent metatarsal heads according to the regulation of the load transfer mechanism. While the first tarsometatarsal joint fracture-dislocation was fixated by screw or plate, the plantar pressure of the forefoot would return to the normal state. However,if the joint was fixated by the staple, it would still be difficult to return the plantar pressure to be normal.
