3.Research on the construction of the 5 +3 integrated clinical medical personnel training objective system

Objective To construct the objective system of 5+3 integrated clinical medicine personnel training mode.Methods For the attitude,knowledge and ability clinical medical students should have,who received 5+3 integrated training,we adopted literature research,expert interviews intending to make a preliminary questionnaire items,and invited experts and graduates to modify the expressions of the items a number of times,and eventually formed the final questionnaire that met the study objective.We asked 500 teachers in basic medicine,clinical medicine,and medical humanities and so on,to evaluate the importance of the questionnaire.406 valid questionnaires were recovered and the effective recovery rate was 81.2%.Epidata 3.1 and SPSS 19.0 were used for survey data summary statistics,exploratory factor analysis and factor weighting method were used to get the goal of talent training base,and on these basis target system was constructed.Results In 5+3 integrated medical education model,clinical medical personnel training target system included knowledge,attitude and ability of the part,a total of 9 target groups.Each group contained elements in the training target system for different weights,among which attitude included a common factor,proportion of 27.51%;knowledge consisted of three factors,the proportion of 29.34%;and the ability contained five common factors,accounting for highest proportion of 43.15%.Conclusion 5+3 integrated clinical medical talents training target system established in this study highlights the ability requirements,emphasizes the comprehensive quality training,which accords with the principles of talent training goal and the requirements of 5+3 medical education model,has certain guiding significance for the curriculum reform,and can be used for reference in the development of talents training program for medical colleges and universities.
4.Studies on Shedding Virus and the Formation of Aerosol of H_9N_2 Subtype AIV

In order to study the regularity of shedding virus from infected SPF chickens and the formation of aerosol of H9N2 subtype AIV, SPF chickens were bred in a positive and negative pressure isolator. Aerosol samples were collected by AGI-30 (All Glass Impinger-30) extractor, and simultaneously trachea and cloaca samples were collected by tracheal swabs and cloacal swabs in different periods after challenged with vi- ruses. The above-mentioned samples were detected by HI, Dot-ELISA and RT-PCR methods. The results in- dicated that aerosols were isolated from the 4 days to the 43 days after inoculation. It was proved that H9N2 subtype AIV could copy themselves in respiratory passage and cloaca, and then could formation of aerosols. AIV H9N2 subtype could be isolated from cloacal and tracheal swabs 3 days after inoculation and lasted for 45 days, viruses were detected from all infected SPF chickens on 7 days.
5.Effect of Pingtang recipe containing drug-serum on INS-1 pancreatic beta cells lipoapoptosis.

OBJECTIVETo observe the effect and mechanism of Pingtang Recipe containing drug-serum (DS-PTR) in improving INS-1 beta pancreatic cells lipoapoptosis.
METHODSExperimental INS-1 beta cells were divided into 5 groups (6 pools for each group), namely, the blank control group treated with rat's serum (C), the other 4 model groups induced into lipoapoptosis by palmitic acid and treated respectively by rat's serum (M), high, middle and low dose DS-PTR (DSh, DSm and DSI). Cell apoptosis was detected by TUNEL staining; Caspase-3 activity of cells was measured by chemiluminescence method; intracellular production of reactive oxygen species (ROS) was detected by DCHF-DA incorporation, and expressions of uncoupling protein-2 (UCP-2) was determined by RT-PCR.
RESULTSINS-1 beta cell apoptosis in Group M was significantly higher than that in Group C (P < 0.01), while that showed a decreased trend in the three DS-PTR treated groups. Caspase-3 activity was enhanced in Group M, it decreased significantly in Group DSm (P < 0.05). The over-produced ROS in cells after modeling was inhibited in Groups DSm and DSI (P < 0.05), meantime, expression of UCP-2 excited by PA (2.244 +/- 0.421) was reduced significantly in Group DSI and Group DSm to 1.286 +/- 0.373 (P < 0.01) and 1.627 +/- 0.348 (P < 0.05) respectively.
CONCLUSIONDS-PTR shows a protective effect on INS-1 beta pancrentic cells against lipoapoptosis, which is possibly play its mechanism through regulating ROS and UCP-2.

6.Influence of humic acid fertilizer on biomass accumulation and quality of Angelica sinensis.

OBJECTIVETo study the influence of humic acid fertilizer on plant growth, assimilation base, dried biomass accumulation, yield, quality and disease infection of Angelica sinensis.
METHODThree kinds of humic acid fertilizer and an amino acid liquid fertilizer were tested in randomized groups at 1 level with 3 times repeat.
RESULTT1 promoted plant and root growth effectively, increased dried biomass accumulation and fresh root average weight remarkably, the yield was increased, the content of ethanol extract was increased by 11.31%. T3 promoted plant and root growth quickly, enlarged leaves area and increased dried plant weight, but effect lasted shortly, the content of ethanol extract was increased by 5.23%. T4 increased more leaves in late growth period, enlarged leaves area, the yield was increased, the content of ethanol extract was increased by 3.09%. T2 increased fresh root average weight remarkably, the yield was increased.
CONCLUSIONHumic acid fertilizer and amino acid liquid fertilizer could effectively promote plant growth, enlarge leaves area, promote dried biomass accumulation and transformation to root and increase yield and content of ethanol extract effectively.

7.Isolation and Characterization of a Temperature-sensitive p-Nitrophenol Degradation Related Genes Mutant Strain

A temperature-sensitive mutant strain was isolated after transposon mutagenesis with Tn5 and named MT54.PCR was carried out with primers designed according to the sequence of transposon,the PCR products showed that the MT54 carried transposon in the genome. The mutant grew well at 30℃in minimal medium(MM)containing p-nitrophenol(PNP)as sole carbon source,while it cannot grow at 37℃in the same medium,NO_2.detection results also proved that.Comparing the degradation rate of PNP and hydroquinone of MT54 and DLL-E4 at different temperature,it was speculated that the mutant site locate in the PNP degradation related genes.
10.Relationship between angiotensin converting enzyme gene polymorphism and essential hypertension in children.

OBJECTIVETo study the relationship between insertion/deletion (I/D) polymorphism of 287 bp in the 16th intron of angiotensin converting enzyme (ACE) and essential hypertension in children.
METHODSI/D polymorphism of 287 bp in the 16th intron of ACE was detected using PCR in 105 children with essential hypertension and 105 normal children as control group.
RESULTSThere was an I/D polymorphism in the 16th intron of ACE in the hypertension and the control groups: type II, type ID and type DD. The genotype frequencies of type DD, type ID and type II in the hypertension group were 30.5%, 47.6% and 21.9%, respectively. The genotype frequencies of type DD, type ID and type II in the control group were 14.3%, 46.7% and 39.1%, respectively. There were significant differences in the genotype frequencies of types DD and II between the two groups (P<0.01). The allele frequency of type D (54.3% vs 37.6%) was significantly higher in the hypertension group; in contrast, the allele frequency of type I (45.7% vs 62.4%) was significantly lower than in the control group (P<0.01).
CONCLUSIONSPolymorphism of type II, type ID and type DD exits in ACE. The deletion of 287 bp in the 16th intron of ACE might be associated with the occurrence of essential hypertension in children.
