4.Morphologic Localization and Purified Cultivation of Sinoatrial Nodes Isolated from Newborn Rabbits

Objective To localize the sinoatrial node (SAN) of the newborn rabbits in vivo and cut it for purifying cultivation and study the morophologic characters of primary cultured pacemaker cells of SAN under light microscope and transmissional electron microscope. Methods Hearts of the newborn rabbits were embedded in paraffin for HE-staining and observed the location, form of SAN under optical microscope; SAN cells isolated from neonatal rabbits cultured and purified with the method of differential attachment and BrdU-treatment.Results SAN localized in the anterior wall of the superior vena cava and the posterior-lateral atrial wall.There was about 0.32 mm between its lowest point and sulcus terminalis. Three distinctly different types of cells were observed among the cultured cells of SAN: spindle, araneiform and polygon. The spindle cells covered the greatest proportion of the cultured cells of SAN (59.6%?7.3%). The frequency of spontaneous contraction of spindle cells was the highest among the constrcting cells (145 ?9)time/min. The results of ultrastructure observation showed that myofibrils and other organelles in spindle cells were poorly organized and significantly decreased in number compared with araneiform cells. There was no significant difference between araneiform cells isolated from SAN and from atrial muscle.Conclusion Among the cultured cells from neonatal rabbits SAN, the spindle cells are the pacemaker cells of SAN.
5.Effect of the cryopreserved Schwann cells on the peripheral nerve regeneration.

AIMTo compare the effects of primary culture Schwann cells and cryopreserved Schwann cells on the injured peripheral nerve regeneration.
METHODSThe primary culture Schwann cells and cryopreserved Schwann cells were transplanted into the silicone tubes that bridged the resected sciatic nerves. At different time after transplantation, HRP was injected into the sciatic nerve trunks at the distal end of the silicone tubes, and then the HRP labeled neurons in dorsal root ganglions and anterior horns of spinal cord were counted. The complex action potential of regenerated nerve fibers was measured. Myelination on regenerated nerve fibers was investigated under electronic microscope.
RESULTSAt different time after transplantation, there were not significant differences that the HRP labeled neurons in dorsal root ganglions and anterior horns of spinal cord, the complex action potential of regenerated nerve fibers and myelination on regenerated nerve fibers (P > 0.05), between primary culture and cryopreserved Schwann cells.
CONCLUSIONThe cryopreserved Schwann cells still play the same important role in the regeneration of injured peripheral nerves as the primary culture Schwann cells do.

6.Effect of Glomus versiforme and Trichoderma harzianum on growth and quality of Salvia miltiorrhiza.

The present study aimed to investigate the effect of Glomus versiforme and Trichodema harzianum on the growth and quality of Salvia miltiorrhiza continuous cropping under field conditions. The field plot experiment was conducted, these active components in the plant were analyzed by HPLC, the root diseases incidence rate of S. miltiorrhiza determined by observation and counting, and relative parameters were measured. The data was statistically processed. The result showed that inoculation of G. versiforme and combined inoculation of G. versiforme with T. harzianum significantly decreased the root diseases incidence rate of S. miltiorrhiza, and combined inoculation of G. versiforme with T. harzianum was better than other treatments. All treatments improved accumulation of active ingredients in root. Inoculation of G. versiforme and combined inoculation of G. versiforme with T. harzianum significantly increased the content of salvianolic acid B and cryptotanshinone of root (P < 0.05), Inoculation of G. versiforme, T. harzianum and combined inoculation of G. versiforme with T. harzianum significantly enhanced the content of tanshinone I and tanshinone II(A) of the root (P < 0.05). It may conclude that inoculation of G. versiforme and combined inoculation of G. versiforme with T. harzianum can effectively reduce the root diseases incidence of continuous cropping S. miltiorrhiza, and improve the quality of S. miltiorrhiza.

7.Effects of Bushen Wenyang Huayu Recipe on Expressions of HIF-1α, PHD2, and VHL in Endometriosis Rats with Shen Yang Deficiency Blood Stasis Syndrome.

OBJECTIVETo observe the effect of Bushen Wenyang Huayu Recipe (BWHR) on hypoxia inducible factor-1α (HIF-1α), proline hydroxylase2 (PHD2), von Hippel Lindau disease (VHL) suppressor gene expressions in endometriosis (EM) rats with Shen yang deficiency blood stasis syndrome (SYDBSS), and to explore the pathogenesis of EM and the mechanism of BWHR for treating EM.
METHODSTotally 50 SD rats were randomly divided into five groups, i.e., the blank control group, the sham-operation group, the model group, the Chinese medicine (CM) group, and the Western medicine (WM) group, 10 in each group. Rats in the blank control group and the sham-operation group were fed routinely. Rats in the rest 3 groups received 30-day "extended refrigerator freezing and ice water immersion" and combined with " autotransplantation" to establish EM rat model with SYDBSS. One Milliliter BWHR at 3.33 g/mL was administered to rats in the CM group by gastrogavage. Gestrinone at the daily dose of 0. 5 mg/kg was administered to rats in the WM group by gastrogavage. Equal volume of normal saline was administered to rats in the model group, the blank control group, and the sham-operation group. The size and morphology of ectopic foci in rats were observed after 4 weeks of medication. Expressions of serum CA125, plasma cyclic adenosine monophosphate (cAMP), and plasma cyclic guanosine monophosphate (cGMP) were detected by radioimmunoassay. Morphological changes of eutopic endometrium and ectopic tissue were observed under the optical microscope by HE staining. Protein expressions and contents of HIF-lα, PHD2, and VHL were detected by immunohistochemical SABC method and Western blot. mRNA expressions of HIF-1α, PHD2, and VHL were detected by RT-PCR.
RESULTSThe ectopic foci grew significantly in the model group. Their volumes were obviously contracted after treated by CM and WM. Compared with the blank control group and the sham-operation group, serum CA125 and plasma cGMP obviously increased, cAMP obviously decreased (P < 0.05); expressions and contents of HIF-1α mRNA and protein all decreased (P < 0.05); mRNA and protein expressions and contents of PHD2 and VHL all decreased in the model group (P < 0.05). Compared with model group, levels of CA125 and cGMP obviously decreased; cAMP levels obviously increased, expressions and contents of HIF-1α mRNA and protein all increased, mRNA and protein expressions and contents of PHD2 and VHL all increased in the WM group and the CM group (P < 0.05). Compared with the CM group, PHD2 protein contents were higher in the WM group (P < 0.05). HIF-1α was negatively correlated with PHD2 (r = -0.799, P = 0.00). HIF-1α was negatively correlated with VHL (r = -0. 625, P = 0.003).
CONCLUSIONSBWHR could effectively treat EM. Its mechanism might be associated with reducing contents of HIF-1α, serum CA125, and plasma cGMP, and up-regulating expressions of PHD2, VHL, and cAMP.

8.Histologic study of local infiltration of spinal bone giant cell tumor and chordoma.

OBJECTIVETo study the local infiltration length of spinal bone giant cell tumor and chordoma in various para-tumorous tissues.
METHODSSurgical specimens from 7 patients with bone giant cell tumor and 10 patients with chordoma were divided into 6 groups: cortical bone group (CBG, para-tumorous tissue is bone cortex with periosteum), cartilage group (CG, para-tumorous tissues contain intervertebral disc, cartilage endplate and facet joint cartilage), trabecular bone group (TBG), paraspinal muscle tissue group (MTG), adipose tissue group (ATG) and scar tissue group (STG). Macroscopically margin (MSM) in the fresh specimens, histological margin (HLM) in the H&E sections and molecular margin (MCM) in the immunohistochemical staining sections were measured respectively. Three types of the margins of each group and MSM-HLMs, and MSM-MCMs among all groups were compared.
RESULTSAnalysis of Variance comparing MSM, HLM and MCM within each group showed that there were significant differences in all groups (P < 0.05) except for CBG and CG. MSMs were significantly longer than HLMs and MCMs in TBG and MTG (P < 0.05), and MSMs were significantly longer than MCMs in ATG and STG (P < 0.05). MSM-HLMs and MSM-MCMs of TBG and ATG were significantly larger than those of CG and CBG. MSM-HLMs and MSM-MCMs of MTG and STG were significantly larger than those of CG; in addition, MSM-MCM of STG was significantly larger than CBG's. The maximum of tumor infiltration length in CBG, MTG, ATG and STG were 11.68 mm, 13.08 mm, 8.64 mm and 9.98 mm respectively.
CONCLUSIONSEach vertebra can be seen as a compartment, so total spondylectomy is an optimal procedure when bone giant cell tumor and chordoma localized in the compartment. To achieve a wide resection, a 1.3 cm wide margin in para-tumorous cancellous bone, a 1.5 cm wide margin in para-tumorous muscle and 1.0 cm wide margin in adipose tissues are necessary. First-time resection should be more aggressive and avoid tumor cell contamination. The surgical extent of salvage revision should contain para-tumorous scar tissue with the margin no less than 1.0 cm.

Background Stromal cell-derived factor-1_?(SDF-1_?)has been demonstrated to be essential for stern cell mobilization/homing.Recent evidence indicates that SDF-1_? has been expressed in injured carotid arter- ies.Besides,high SDF-1_? plasma levels are clinically associated with stable coronary artery disease.Objective To investigate whether SDF 1 involves in mobilization of endothelial progenitor cells(EPC)and reendothelialization after vascular injury.Methods SDF-1_? was detected by RT-PCR and Western blot in carotid arteries of mice at different time points after wire-induced injury.SDF-1_? determination in peripheral blood samples and BM was per- formed by SDF-1_? enzyme-linked immunosorbent assay(ELISA)kit.EPC in peripheral blood collected at different time points after vascular injury were quantified by flow cytornetry.In subgroup,blocking SDF-1 rnonoclonal anti- body was injected,peripheral blood EPC were quantified after vascular injury and reendothelialization of injured ar- teries was determined 14 days later.Results Expression of SDF-1_? was evident at day 1,and peaked at day 3 after arterial injury.A rise in plasmatic concentration of SDF-1_? and a significant reduction of SDF-1_? in bone marrow concentration was noticed at all time points following injury.The amount of circulating EPC was increased shortly after induction of vascular injury and persisted up to 7 days(P
10.Effects of shengmai for injection on cardiogenic shock.

OBJECTIVETo study the protective effects and the possible mechanism of shengmai for injection(SM) against the experimental acute cardiogenic shock.
METHODThe experimental acute cardiogenic shock model was established by ligating the anterior descending cornonary in dogs. The effects of SM on cardiogenic shock were investigated by measuring the hemodynamics parameter, the activity of LDH, CK, SOD and the contents of MDA in blood serum.
RESULTIn the dogs treated with SM, the mean arterial pressure (MAP), heart rate (HR), left ventricular pressure (LVP), the maximum of its first derivative (+/- dp/dtmax), the cardiac output (CO) and the cardiac index (CI) were increased significantly. The left ventricular end diastolic pressure (LVEDP) and the peripheral vascular resistance (TPVR) were decreased significantly, the myocardial infarct size was redused observely. In addition, the activity of LDH, CK and the contents of MDA in serum were decreased significantly, however the activity of SOD was increased observely.
CONCLUSIONThe results indicate that SM has the protective effects on cardiogenic shock.
