1.Enriching and Purifying Achyranthes Bidentata Saponins with Macroporous Adsorptive Resins

OBJECTIVE:To enrich and purify Achyranthes Bidentata Saponins with macroporous adsorptive resins.MET_HODS:The macroporous adsorptive resin columns on the Achyranthes Bidentata extractives were eluted respectively with water and different concentrations of ethanol,and the elution rate and purity coefficient of Achyranthes Bidentata Saponins were detected.RESULTS:The elution ratio of the portion of Achyranthes Bidentata Saponins that enriched in 50% alcohol eluant was above 58.42%.CONCLUSION:It is feasible to enrich and purify Achyranthes Bidentata Saponins with macroporous adsorptive resins.
2.Study on Separation and Purification of Total Saponins from Radix Clematidis with Macroporous Absorptive Resin

Objective To establish a method for the separation and purification of total saponins from Radix Clematidis by column chromatography of macroporous absorptive resin. Methods Radix clematidis was extracted with 70 % ethanol and then the extraction was purified with D101 macroporous absorptive resin. After eluting with distilled water and 30 % , 50 % and 70 % ethanol, absorption capability and elution parameters of macroporous absorptive resin were measured by the indexes of elution rate and refinement rate of total saponins. Results Total saponins of Radix Clematidis were enriched in the 50 % ethanol eluted solution fraction. The elution rate of macroporous absorptive resin was 71.26 % and absorption capability was 53.36 mg/g . Conclusion It is feasible to use macroporous absorptive resin for the isolation and purification of total saponins from Radix Clematidis. The optimum elution condition is: removing water- soluble impurity by water, eluting total saponins with 50 % ethanol.
3.Experimental Studies on the Anti-inflammatory and Immunity Effects of Huatan Jiangqi Capsule

Objective To study the anti-inflammatory and immunity effects of Huatan Jiangqi capsule. Method The anti-inflammatory and immunity effects were investigated by ear swell model of mice induced by dimethylbenzene,granulation tissue hyperplasia model of rats,viscera coefficient and swimming time of mice. Results Huatan Jiangqi capsule could reduce ear swell degree of mice and granulation tissue hyperplasia of rats. It could also increase viscera coefficient and prolong swimming time of mice. Conclusion Huatan Jiangqi capsule had significant anti-inflammatory and immunity effects.
4.The mechanism of Pseudostellaria Polysaccharides decreasing leptin level on Diabetic Rats

Objective To investigate potential mechanism of decreasiy leptin level of Pseudostellaria Polysaccharides on experimental diabetic rats. MethodsThe diabetic rats were induced by the abdominal cavity injection of alloxen(ALX) 200 mg· kg-1. The levels of leption and TNF-α were determined. Results Pseudostellaria Polysaccharides (0.6 g·kg-1 , 1.2 g·kg-1 ) could increase Leptin level and could decrease TNF-α level of DM rats(P<0.05 or P<0.01). Conclusion Pseudostellaria Polysaccharides can play an threpedtic rale in the experiment aldialetic rats through multi-pathway the experimental diabetic rats.
5.Research on antithrombotic effect of total saponins of Astragalus

Objective: To research antithrombotic effect and mechanism of total saponins of Astragalus (TSA).Methods: Effects of TSA on artery thrombosis anud platelet aggregation were studied, at dosage of 50,100,200mg/kg. And level of TXA 2,PGI 2 in plasma, content of NO in serum of rats were determined.Results: TSA could restrain artery thrombosis and platelet aggregation of rats, and increase PGI 2 and NO.Conclusion: There was apparently antithrombotic effect of TSA.
6.Research progresses of plant polysaccharides on cell adhesion.

Cell adhesion mediated by cell adhesion molecules (CAMs) constitutes essential life phenomenon. In inflammation, immunity, infection, thrombosis, tumor metastasis and wound healing, cell adhesion comes into being the basic physiological and pathological process. Intervening with cell adhesion has been the important therapeutic and prophylactic strategies for diseases. Accumulated evidence has indicated that plant polysaccharides especially those exacted from Chinese traditional and herbal drugs displayed various pharmacological effects such as anti-inflammation, anti-cancer, anti-infection, immunomodulation, cardiovascular protective effects and so on. In this paper, the research progress of plant polysaccharides on cell adhesion is reviewed.
7.Effects of p27mt gene transfection mediated by replication deficient adenovirus on the proliferation and apoptosis of human hepatocellular carcinoma cells

Objective To assess the effects of p27mt gene transfection on the proliferation and apoptosis of human hepatocellular carcinoma cell (HCC) lines SMMC-7721. Methods A replication deficient adenovirus vector encoding p27mt (Ad-p27mt) was used and p27mt cDNA was transfected into human SMMC-7721 cell lines in vitro. The synthesis of DNA in SMMC-7721 cells was determined by using 3H-thymidine incorporation; the cell apoptosis was determined by flow cytometry, TUNEL method and DNA fragmentation analysis. Results The virus titer was 7.95?1012 cfu/ml, the transduction efficiency was 100 % when multiplicity of infection ≥50, FCM analysis revealed a sub-G1 cell peak in Ad-p27mt transduced hepatocellular carcinoma cell lines. 8.The effect of Connexin43 downregulation on biological functions of HUVEC.

Connexin43 has been shown to play a pivotal role in wound healing process. Wound repair is enhanced by acute downregulation of connexin43, by increasing proliferation and migration of keratinocyte and fibroblast. Angiogenesis is also a central feature of wound repair, but little is known about the effects of connexin43 modulation on functions of endothelial cells. We used connexin43 specific small interference RNA (siRNA) to reduce the expression of connexin43 in human umbilical vein endothelial cell (HUVEC), and investigated the effects of connexin43 downregulation on intercellular communication, viability, proliferation, migration and angiogenic activity of HUVEC. Treatment of siRNA markedly reduced the expression of connexin43 by -80% in HUVEC (P < 0.05), and decreased the intercellular communication by -65% (P < 0.05). The viability, proliferation, migration and angiogenic activity of HUVEC decreased significantly (P < 0.05), compared with that of the normal cells. The results suggest that temporally downregulation of connexin43 expression at early stage of wound to inhibit the abnormal angiogenesis characterized with leaky and inflamed blood vessels, maybe a prerequisite for coordinated normal healing process.
9.Effects of Tougu Xiaotong Capsules on the expression of Rac1 and Cdc42 in chondrocytes

BACKGROUND:Tougu Xiaotong Capsule has pretty good clinical therapeutic effect on osteoarthritis of early and middle periods. However, the mechanism of Tougu Xiaotong Capsule is not ful y clarified. The RhoA GTPases can regulate chondrocyte apoptosis and hypertrophy.
OBJECTIVE:To observe the Tougu Xiaotong Capsule on the expression of Rac1and Cdc42 in tumor necrosis factor-α-induced in vitro cultured rat articular chondrocytes, and to explore its mechanism of action for combating osteoarthritis.
METHODS:Knee cartilage of the 4-week-old Sprague-Dawley rats was used to stably establish in vitro culture system of chondrocytes. Passage 3 chondrocytes were identified by toluidine blue staining. Chondrocyte apoptosis was successful y induced by 20μg/L tumor necrosis factor-αand then Tougu Xiaotong Capsule at different dosage (500, 100, 20 mg/L) was given after 24-hour incubation. MTT assay was used to detect cellsurvival, flow cytrometry to measure mitochondrial membrane potential, and western blot assay to determine the protein expression of Rac1, Cdc42, Bax and Bcl-2.
RESULTS AND CONCLUSION:Tougu Xiaotong Capsule could reduce tumor necrosis factor-α-induced apoptosis of chondrocytes to improve the survival rate of the cells, and at the same time, could down-regulate the protein expression of Rac1, Cdc42 and Bax and increase the protein expression of Bcl-2 significantly (P<0.05). Tougu Xiaotong Capsule possibly plays a therapeutic efficacy on osteoarthritis by reducing promote apoptosis Rac1, Cdc42 and Bax expression and increasing apoptosis inhibiting gene Bcl-2 expression, thereby to inhibit apoptosis of chondrocytes.
10.Identification of adenovirus epidemic strains and analysis of hexon gene characteristics in Anqing area from 2013 to 2015

Objective To investigate the characteristics of human adenovirus ( HAdv) epidemic strains and the variations of hexon protein and amino acid residues in acute respiratory infections in Anqing. Methods A total of 596 throat swab samples of children with acute respiratory infections were collected from influenza surveillance sites between 2013 and 2015 and detected with real-time fluorescent PCR adeno-virus nucleic acid test kit. Hep cells were used to separate viruses from positive samples. PCR amplification of hexon gene and sequencing analysis were conducted. Homologous alignment and phylogenetic tree analysis were performed between the obtained sequences and those published in GenBank. Results HAdv-positive samples accounted for 11. 4% (68). Thirty-four viruses were successfully isolated, including nine HAdv-3 (26. 5% ), 12 HAdv-7 (35. 3% ), 12 HAdv-14 (35. 3% ) and one HAdv-55 (2. 9% ). The 9 strains of HAdv-3 had a close genetic relationship with KX384958, sharing a homology of 99. 8%-100% . Three muta-tions in main amino acid residues were found in them as compared with reference strains. The 12 strains of HAdv-7 were genetically related to KX897164 and KU361344 with a homology ranging from 99. 8% to 100% and had seven major amino acid residue mutations in comparison to reference strains. The 12 strains of HAdv-14 were highly similar to JF420883 with a homology of 99. 6%-99. 9% , and possessed three major variations in amino acid residues in comparison to reference strains. The HAdv-55 strain was closely related to KP279748 and KX289874, showing a homology of 100% in both nucleotide and amino acid sequences. HAdv-7 strains had the greatest variations, followed by HAdv-14 strains. Conclusion From 2013 to 2015, the epidemic adenovirus strains causing acute respiratory infections in Anqing area were mainly HAdv-3, HAdv-7 and HAdv-14 with a small number of HAdv-55. The hexon genes of HAdv-55 strains were stable and no variation occurred. However, HAdv-3, HAdv-7 and HAdv-14 strains all had some variations in nu-cleotides and amino acids. Amino acid variations in the antigenic determinants of HVR1, HVR2 and HVR7 regions were detected.
