1.Validation Research And Regulation Exploration Of High Fat-introduced Hyperlipidemia Model In Rat

Objective To investigate the feasibility of high fat-introduced hyperlipeidemia model in male SD rat and study the time rule of molding .Methods 30 Male adult rats of SD Strain bred in the animal house of the institute were divided into 3 groups after 1 week adaptation , group 1:control group, normal diet;group 2:model 1 group, high fat high cholesterol diet;group 3:model 2 group, high fat high cholesterol diet .The period of experiment was 8 weeks.Food and water intake were measured everyday and body weight were measured every four days .Blood were collected by orbital venous at the end of fourth ,sixth,eighth week to test their serum lipid level .At the end of experiment ,animals were killed to collect liver and aorta tissue for HE stain .Results Compared with control group ,the food intake of model 1 was higher and model 2 was significant lower , water intake of model 2 was significant lower , the ratio of liver/weight of two model groups were significant heavier ,and weight of model groups were higher .High fat diet significantly increased TC levels of model groups at the end of fourth ,sixth week.The level of LDL-c in model 1 group were higher and the HDL-c were lower compared with control group .HE stain showed the livers of control group were regular ,arrangements of the liver cells were trim, dyeing present uniformity .The two model groups showed a large range of hepatocyte fatty change ,a few liver blood sinus were in congestion and infiltrated with inflammatory cells .Aorta HE stain showed no significant change among 3 groups.Conclusions The method of high fat-introduced hyperlipeidemia model in male SD rat is feasible and the model turned out to present hypercholesterolemia with severe fatty liver .On the other hand,levers of serum lipid increased within an increase—inter-adjustment—increase state .In the process of modeling ,how to overcome the symptom of anorexia and the state of cholesterol inter-adjustment in animals is the key to successfully establish hyperlipeidemia model .
2.Effects of Multiple Cleaning and Disinfection Interventions on Infectious Diseases in Children: A Group Randomized Trial in China.

OBJECTIVETo assess the effectiveness of multiple cleaning and disinfection interventions in the homes and kindergartens, in reducing gastrointestinal and respiratory illnesses of children.
METHODSFrom October 2010 to September 2011, we performed a prospective, controlled study in China. 408 children under 5 years old were recruited and group randomized into intervention and control groups. Families and kindergartens in the intervention group were provided with antibacterial products for hand hygiene and surface cleaning or disinfection for one year. Each child's illness symptoms and sick leave were recorded every day.
RESULTSA total of 393 children completed the study, with similar baseline demographics in each of the 2 groups. Except for abdominal pain, the odds of symptoms (fever, cough and expectoration, runny nose and nasal congestion, diarrhea), illness (acute respiratory illness and gastrointestinal illness), and sick leave per person each month were significantly reduced by interventions. The rates of fever, diarrhea, acute respiratory illness, gastrointestinal illness and sick leave per person per year were significantly decreased as well.
CONCLUSIONNot only the acute respiratory and gastrointestinal illness but the sick leave rate in children were significantly reduced by multiple interventions.

3.Comparative observation on acupuncture and western medicine for treatment of minimal brain dysfunction.

OBJECTIVETo compare the therapeutic effects of acupuncture and western medicine on minimal brain dysfunction (MBD) and to search for a clinically effective therapy for MBD.
METHODSSixty-eight cases were randomly divided into an acupuncture group and a western medicine group, 34 cases in each group. The acupuncture group were treated by acupuncture at Dazhui (GV 14) and Shenque (CV 8), and the western medicine group by taking Haloperidol orally. One month constituted one course. After treatment, the total effective rate and scores of Connell's scale for diagnosis and behavior of MBD were compared between the two groups.
RESULTSThe total effective rate and the score after treatment were 97.1% and 10 +/- 0.37 in the acupuncture group and 82.4% and 15 +/- 0.93 in the western medicine group, with a very significant difference between the two groups (P < 0.01, P < 0.000 5), the acupuncture group being better than the western medicine group. Follow-up survey for 2-10 months showed the effects of the acupuncture group still were kept.
CONCLUSIONAcupuncture at Dazhui (GV 14) and Shenque (CV 8) can effectively cure MBD.

4. The mediating role of psychological capital between the sense of occupational mission and job engagement in preschool teachers

OBJECTIVE: To explore the mediating role of psychological capital in the sense of occupational mission and work engagement in preschool teachers.METHODS: A method of stratified cluster sampling was used to select 464 preschool teachers from 16 kindergartens in a city of Hubei Province. Occupational Mission Scale, Psychological Capital Questionnaire and Job Involvement Questionnaire were used to investigate their sense of occupational mission, psychological capital and job engagement. RESULTS: The total score of occupational mission was(48.6±6.9); the total score of psychological capital was(87.1±11.8); and the total score of work engagement was(79.8±11.4). Occupational mission was positively correlated with job engagement [correlation coefficient(r) was 0.607, P<0.01].The self-efficacy, hope, tenacity and optimism were positively correlated with job engagement(r were 0.526, 0.544, 0.573, 0.415, respectively, P<0.01). The mediating effect of psychological capital on occupational mission and work engagement was 0.15, accounting for 26.8% of the total effect. CONCLUSION: The sense of occupational mission can directly or indirectly promote job engagement. The psychological capital plays a mediating role between occupational mission and job engagement.
5.Genetic analysis of the CACNA1F gene in a family affected with incomplete form Schubert-Bornschein type congenital stationary night blindness

Objective:To determine the pathogenic gene mutation in a family with incomplete congenital quiescent night blindness (CSNB) of Schubert-Bornschein type.Methods:A retrospective clinical study. In February 2021, one patient and his parents and elder brother from a Han Chinese incomplete CSNB of Schubert-Bornschein type family diagnosed by clinical and genetic examination at Henan Provincial People's Hospital were included in the study. The patient’s medical history, family history were inquired; best corrected visual acuity (BCVA), color vision, fundus color photography, full-field electroretinogram (ERG), and frequency domain optical coherence tomography (OCT) were examined in detail. Five ml of the subject’s peripheral venous blood was collected and the whole genome DNA was extracted. The genomic DNA of the subject was library constructed, and all-exon probes were polymerized for capture. The suspected pathogenic mutation site was verified by Sanger, and the pathogenicity of the gene mutation site was determined by parallel bioinformatics analysis.Results:The BCVA of both eyes of the proband (Ⅱ2) was 0.4; the color vision test could not recognize the red color. Fundus examination showed no obvious abnormalities. The retina thickness in the macular area of both eyes was slightly thinned. ERG examination of the whole field showed that the amplitude of ERG b wave was significantly reduced under the stimulation of binocular dark adaptation 3.0 and showed a negative waveform. The mother of the proband (Ⅰ2) had normal BCVA, color vision, fundus color photography, and frequency domain OCT examination. The full-field ERG examination showed that the amplitude of each eye reaction was slightly reduced, and the amplitude of the dark adaptation shock potential was significantly reduced. Genetic testing showed that the proband (Ⅱ2) had a c.1761dupC hemizygous mutation in exon 14 of the voltage-dependent calcium channel α1F subunit gene ( CACNA1F gene). The results of protein sequence homology analysis showed that the site was highly conserved in multiple species; the results of bioinformatics analysis showed that the CACNA1F gene c.1761dupC (pY588fs) subsequently had a frameshift mutation and became a stop at position 10. Codons appear translational termination in the conserved regions of the protein. According to the standards and guidelines of the American College of Medical Genetics and Genomics, the mutation was judged to be a possible pathogenic variant. The mother of the proband (Ⅰ2) was a carrier of this site mutation. The clinical and genetic test results of the father and elder brother of the proband were not abnormal. Conclusion:CACNA1F gene c.1761dupC is the pathogenic mutation site of the Schubert-Bornschein type incomplete CSNB family.
