1.Study on safety of argon plasma coagulation with submucosal injection for colon polyps

Objective To investigate the safety of argon plasma coagulation (APC) with submucosal injection for colon polyps.Methods A total of 30 sets of fresh sigmoid colon from healthy pigs were assigned to control group to receive direct APC,and to treatment group to receive APC with submucosal injection,respectively.After same time and energy of APC,each specimen was sampled for pathological evaluation and the damage extent was determined as mucosa,submucosa (superior 1/3,middle1/3,inferior 1/3),and muscularis propria.Ten cases of colon sessile polyps with diameter of about 1-2 cm,elevated within 3mm by endoscopic ultrasonography(EUS).Direct APC or APC with submucosal injection were performed,respectively,and the difference was observed by EUS.Results In 30 cases of control group,the injury depth of 5 cases reached to the muscularis propria and of 25 cases to submucosa layer,among which 4 cases showed damage to superior submucosa 12 middle submucosa and 9 inferior submucosa.In 30 cases of treatment group,there was no damage to muscularis propria.The injury depth of 26 cases reached submucosa layer,among which 22 cases showed damage to superior submucosa,4 middle submucosa.The results showed significant difference between two groups (P <0.01).In patients who underwent APC with submucosal injection,EUS showed effective isolation of mucosa and muscularis propria.It confined the injury to the mucosal layer.Without treatment of submucosal injection in advance,it seemed easier to damage the submucosa and muscularis propria.Conclusion Submucosal injection can protect the colon injured by APC,and may reduce the risk of colon perforation by APC for colon polyps.
2.Analysis of human papillomavirus 16 subtypes E2 gene and long control region sequence in Chengdu

Objective To explore the relevance between sequence variation of human papillomavirus (HPV)16 subtypes E2 gene or long control region (LCR) and cervical lesions.Methods Fifty specimens from HPV16 infected people in Chengdu were collected,including cervical exfoliated cells from 38 HPV carriers,papilloma tissues from 8 cases of genital warts,2 with cervical intraepithelial neoplasia (CIN) Ⅱ and 2 with CIN Ⅲ in this study.Polymerase chain reaction was used to amplify E2 gene and LCR,then an evolutionary tree was constructed.Results In all the 50 specimens,there were 12 mutation sites in E2 gene,among which,C→A existed in one specimen of genital warts,and ≥2 mutation sites existed in all the other 48 specimens.There were 28 mutation sites of LCR sequence of all the specimens.Ten specimens were chosen to construct evolutionary tree and were sequenced.The data showed that 8 specimens were Asian variants,E2 gene mutation existed in all the specimens while the LCR 7867 G→A only existed in the four CIN.Conclusion LCR 7867G→A is a correlative mutation site of cervical lesions in Chengdu.
3.Roles of 99mTc-MDP and SPECT/CT fusion imaging in the diagnosis and treatment decisions of bone metastasis of malignant tumors

Objective To evaluate the role of (99)Tc(m)-MDP SPECT/CT bone fusion imaging in diagnosis and treatment strategy establishment of patients with bone metastatic malignancy.Methods Retrospective study was carried out on 66 patients (55 patients with primary malignant tumors,11 patients with primary benign bone disease) chosen from 117 patients who had undergone whole body bone scintigraphy and SPECT/CT fusion imaging examination.Comparison was carried out on diagnostic efficacy for bone metastases and changes of treatment between SPECT/CT fusion imaging and other anatomical imaging (CT and/or MR).Cases excluded are extensive bone metastases and no final diagnosis of patients.Results For diagnosis of patient with bone metastasis and bone metastasis lesion,the sensitivity of SPECT/CT fusion imaging was (90.62%,93.88%),specificity (79.41%,89.47%) and accuracy (84.84%,90.97%),while sensitivity of simple anatomical imaging was (59.38%,51.02%),specificity (94.11%,94.73%) and accuracy (77.27%,79.86%),with a significant difference (P < 0.05).SPECT/CT fusion imaging changed 30.31% (20/66) the patient's treatment plan,while 16.67% (11/66) for simple anatomical imaging,with a significant difference (P < 0.05).Conclusions SPECT/CT fusion imaging could increase the accuracy of diagnosis of bone metastases and have an important role in establishing the treatment strategy.
4.Clinical effect of retinal photocoagulation combined with drug on retinopathy of diabetic retinopathy and its effect on VEGF and IGF-1 level

Objective To explore the effect and application value of retinal photocoagulant combined with compound anisodine in the treatment of diabetic retinopathy , and its effect on vascular endothelial growth factor (VEGF) and insulin-like growth factor-1(IGF-1) level.Methods 116 patients with diabetic retinopathy were selected ,and they were randomly divided into observation group and control group according to the digital table ,58 cases in each group .The control group was given retinal photocoagulation treatment , and the observation group was given retinal photocoagulation combined with compound anisodine treatment .The therapeutic effect was compared between the two groups.Results The total effective rate of the observation group was 93.10%,which was significantly higher than 77.59% of the control group, and the difference between the two groups was statistically significant (χ2 =5.582,P<0.05).The edema absorption time,bleeding absorption time,exudate absorption time of the observation group were (4.19 ±1.24) d,(2.19 ±1.01) d,(9.48 ±2.13) d,respectively,which of the control group were (5.83 ±2.85)d,(3.25 ±1.88)d,(13.54 ±3.88)d,respectively,and the differences between the two groups were statistically significant (t=4.018,3.782,6.985,all P<0.05).After treatment,the VEGF and IGF-1 levels in the observation group were (85.12 ±4.64) ng/L,(90.17 ±4.95)μg/L,respectively,which in the control group were (100.48 ±9.26)ng/L,(132.69 ±10.04)μg/L,respectively,the differences between the two groups were statistically significant (t=11.294,28.928,all P<0.05).Conclusion Retinal photocoagulation surgery combined with compound anisodine in the treatment of diabetic retinopathy can improve clinical curative effect ,promote the patients'visual acui-ty,adjust the concentration of VEGF ,the effect is remarkable ,it is worthy of popularizing in clinical application .
5.Clinical study of Dihuang-Yinzi Decoction combined with donepezil hydrochloride tablets in the treatment of mild to moderate Alzheimer's disease

Objective:To evaluate the efficacy of Dihuang-Yinzi Decoction combined with donepezil hydrochloride in the treatment of mild to moderate Alzheimer's disease (AD). Methods:114 patients with mild to moderate AD in the First Hospital of Yulin in Suide District from February 2017 to February 2019 were divided into two groups according to the method of random number table, with 57 cases in each group. The control group took donepezil hydrochloride tablets orally, and the study group took Dihuang-Yinzi Decoction based on the control group. Both groups were treated continuously for 3 months. The activities of daily living scale (ADL), Mini-Mental State Examination (MMSE) and the Alzheimer’s disease assessment scale-cognitive section (ADAS-cog) were used to evaluate the life ability and cognitive function of patients. The concentrations of Notch 1 signaling protein, a disintegrin and metalloproteinase 10 (ADAM10) and β-amyloid precursor protein endonuclease 1 (BACE1) in serum were detected by Western blot. The levels of acrolein were detected by ELISA, the adverse reactions during the treatment were recorded, and the clinical efficacy was evaluated. Results:The total effective rate was 84.2% (48/57) in the study group and 64.9% (37/57) in the control group. The difference between the two groups was statistically significant ( χ2=5.596, P=0.018). After the treatment, the ADL and ADAS-cog scores of the study group were lower than those of the control group ( t values were 5.939, 6.124, respectively, all Ps<0.01), and MMSE scores were higher than that of the control group ( t=6.087, P<0.01). The levels of Notch1 signaling protein (3.43 ± 0.58 vs. 1.31 ± 0.47, t=21.440) and ADAM10 (1.86 ± 0.23 vs. 1.12 ± 0.25, t=16.446) in the study group were higher than those of the control group ( P<0.01), and the levels of BACE1 (0.62 ± 0.15 vs. 0.98 ± 0.17, t=11.988) and acrolein (2.19 ± 0.39 nmol/mg vs. 4.76 ± 0.54 nmol/mg, t=12.354) were lower than those of the control group ( P<0.01). The adverse reaction rate was 14.7% (11/57) in the study group and 14.0% (8/57) in the control group, and there was no significant difference between the two groups ( χ2=0.568, P=0.451). Conclusions:Dihuang-Yinzi Decoction combined with donepezil hydrochloride tablets can effectively improve the cognitive function and life ability of patients with mild to moderate AD, with clinical efficacy.
6.Age-related changes in prevalence and symptom characteristics in kidney deficiency syndrome with varied health status:a cross-sectional observational study

Objective:This study aimed to reveal the age-related changes in prevalence and symptom characteristics in kidney deficiency syndrome (KDS) with varied health status.Methods:This cross-sectional observational study was conducted in6 hospitals.Investigators queried participants aged 20-79 about their health,including symptoms if any,and completed questionnaires to collect participants responses.Prevalence,severity,and frequency of KDS and other relevant parameters were observed and recorded.Comparative analysis of count-able variables,including prevalence,was performed by frequency analysis and x2 test and ex-pressed as a composite ratio.Comparative analysis of quantitative scores of the severity and frequency of symptoms was performed by the rank-sum test.Regression analysis of the corre-lation of KDS with potential contributing factors was performed by non-conditional binary lo-gistic stepwise regression of numerical variables.Results:Prevalence of KDS in healthy and unhealthy participants correlated with increasing age (P < 0.05).In those with chronic disease,KDS prevalence was comparatively high,but the trend was not obvious.KDS prevalence in participants age 20-39 showed an increasing trend with deteriorating health (P < 0.05).Compared to healthy individuals,KDS prevalence in subhealthy persons and those with chronic disease showed an increasing trend (P < 0.05)in the 40-59 and 60-79 age groups,whereas there was no difference between subhealthy per-sons and those with chronic disease age 40-59 and 60-79.Symptom severity scores of KDS showed an increasing trend with increasing age and deteriorating health status (P < 0.05).Higher symptom frequency scores were also positively correlated with increasing age(P < 0.05),but health status deterioration was not significantly correlated (P > 0.05).Age,health status,lower back pain,shin soreness or heel pain,tinnitus or deafness,hair loss or loose teeth,incomplete bladder emptying or incontinence,and sexual dysfunction or infer-tility were potential factors contributing to KDS (P < 0.05),but age was the only independent variable for which OR >1.Moreover,the distribution of typical KDS-related symptoms showed dramatic regularities.Conclusion:Prevalence and symptom characteristics of KDS were found to increase consis-tently with increasing age and deteriorating health status.Kidney deficiency may be an impor-tant mechanism of aging in the subhealthy and chronic disease states.
7.Association of single nucleotide polymorphisms in H2BFWT with male infertility in southwest China.

OBJECTIVETo assess the association of single nucleotide polymorphisms (SNPs) in exons of H2BFWT gene with male infertility in southwest China.
METHODSThree hundred and twelve infertile men and 211 fertile men were recruited. PCR was employed to amplify the target fragments of H2BFWT, and PCR products were sequenced. Prevalence of SNPs in the two groups was analyzed by statistical method.
RESULTSThe detected SNPs have mainly distributed in the first exon of the H2BFWT gene. The ratios of 368G/A (rs553509) and -9C/T (rs7885967) were significantly higher in infertile group than fertile group. Additionally, a context-dependent effect was observed between 368G/A and -9C/T which the allele 368G combined with allele -9T will considerably increase the risk of male infertility.
CONCLUSIONThe present study has revealed that the SNPs in H2BFWT are associated with male infertility, and may increase the susceptibility of male infertility in southwest China.

8.Rapid detection of SOX2 gene by primed in situ labeling.

OBJECTIVETo rapidly detect SOX2 gene using primed in situ labeling (PRINS).
METHODSHuman peripheral blood samples were cultured using an optimized method. Sequence of the SOX2 gene was amplified in situ with biotin-labeled specific primers and processed with a tyramide signal amplification (TSA) biotin system. Subsequently, fluorescence-stained signal was detected by streptavidin-Texas red. For the control group, MCF-10F cells were transfected with Lentivirus hSox2.
RESULTSBy VideoTesT-FISH software analysis, the long arm of chromosome 3 in the experimental group showed a specific red fluorescence signal, whilst the control samples showed no specific signals for SOX2. Transfected MCF-10F cells showed various efficiency of SOX2 gene integration.
CONCLUSIONPRINS utilizes a highly sensitive in situ PCR technique combined with fluorescence labeled oligodeoxynucleotides can synthesize probes in situ, thus greatly reducing the cost of probe and time for detection. It can facilitate identification and classification of induced pluripotent stem cells, and has many potential applications in this prospect.

9.Design of a storage device for lymph node specimens of gynecological malignant tumors and its application

Objective To design a storage device for lymph node specimens of gynecological malignant tumors and to evaluate its application effect.Methods The specimen depository is composed of 3 parts,including storage frame,partition plate and marking plate.391 patients with gynecological malignant tumor surgeries requiring collection of retroperitoneal lymph nodes were selected in the operating room of a tertiary A hospital in Zhejiang Province from January to December 2022 in accordance with the operation schedule.They were randomly divided into an experimental group(n=196)and a control group(n=195).Patients in the experimental group used a newly developed storage device,while the control group was with traditional device.The incidence of additional verification specimens by doctors,the average time for disposing of lymph node specimens,and the job satisfaction survey of doctors and nurses were compared.Results Within a period of 12 months after utilizing this easily recognizable specimen storage device,the additional verification rate in the experimental group was 10.62%compared with the rate of 72.48%in the control group.The average time of disposing lymph node specimens was reduced from(8.3±0.9)to(5.2±0.3)minutes.Doctors and nurses in the experimental group reported a higher level of satisfaction for the specimen storage device(P<0.05).Conclusion Utilizing the newly designed specimen storage device can help medical staff to alleviate their workload,enhance their work efficiency,raise their job satisfaction,and ensure the accuracy of pathological results.
10.Toll-like receptor 3 (TLR3) regulation mechanisms and roles in antiviral innate immune responses.

Toll-like receptor 3 (TLR3) is a member of the TLR family, mediating the transcriptional induction of type I interferons (IFNs), proinflammatory cytokines, and chemokines, thereby collectively establishing an antiviral host response. Studies have shown that unlike other TLR family members, TLR3 is the only RNA sensor that is utterly dependent on the Toll-interleukin-1 receptor (TIR)-domain-containing adaptor-inducing IFN-β (TRIF). However, the details of how the TLR3-TRIF signaling pathway works in an antiviral response and how it is regulated are unclear. In this review, we focus on recent advances in understanding the antiviral mechanism of the TRIF pathway and describe the essential characteristics of TLR3 and its antiviral effects. Advancing our understanding of TLR3 may contribute to disease diagnosis and could foster the development of novel treatments for viral diseases.
