1.Analysis on illuminated dose level of operators' eyes in imaging quality control test of positron emission tomography

Objective To investigate the eye lens dose to the operators who tended to test the quality control of positron emission tomography (PET).Methods Before encapsulation and in preparation of point source,line source 1 and line source 2,the two operators were worn with lens thermoluminescence dosimeter each at the left of the left eye,the front of the left eye,between the right eye and the left eye,the left of the right eye,and the front of the right eye.Measurement and analysis were made of the radiation doses to eye lens received by the operators in order to calculate their maximum annual doses.Results The maximum lens dose was 2 439.80 μSv for the test of 5 PETs.There appears to be the same trend in the eye lens doses for the first and second operators.(x2 =15.629-16.155,P < 0.05).The first operator have received higher eye lens dose higher than the second(Z =2.611,P < 0.05).Conclusions The dose to the eye lens for a single PET test is relatively low.
2.Quality of life and mental health of pre-post household service workers who held a high school graduation certificate or above and had been engaged in different work

Objective To survey the quality of life and mental health status of pre-post household service workers who held a high school graduation certificate or above and had been engaged in different work.Methods A total of 254 household service workers completed Symptom Checklist 90 (SCL-90) and World Health Organization Quality of Life-Brief (WHOQL-BREF).Statistical analyses of variance and correlation were carried out to evaluate mental health status and quality of life of the participants.Results Abnormal score of SCL-90 was found in 8% of household service workers who had graduated from high school.The psychological score of household service workers who had worked as workers(60.59± 2.18)was lower than those who had worked as service workers (66.23 ± 12.13) or peasants (65.40 ± 12.24) (P <0.05).Almost all factors of SCL-90 were negatively correlated with WHOQL-BREF (correlation coefficient -0.140 to-0.376).Conclusions Our data suggest that mental health status can be well in pre-post household service workers who held a high school graduation certificate or above.Special training should be given to the household service workers in accordance with their working experience.
3.Regulatory role of long-chain non-coding RNA HIF1A-AS1 on hypoxia-induced the autophagy of pancreatic cancer PANC1 cells

Objective To observe the regulatory role of long non-coding RNA HIF1A-AS1 on the autophagy of pancreatic cancer PANC1 cells induced by hypoxia.Methods The pancreatic cancer PANC1 cells were cultured in a three-gas incubator filled with hypoxic gas mixture (94% N2,5% CO2,1% O2) for 3, 6, 12, 24, 36 and 48 h.HIF1A-AS1 overexpression and low expression PANC1 cells were obtained by the infection of recombinant adenovirus carrying HIF1A-AS1 and the transfection of HIF1A-AS1 targeting siRNA by liposome, and routinely cultured PANC1 cells served as control.The expression of HIF1A-AS1 of PANC1 cells was detected by real-time quantitative PCR after being cultured in hypoxia-induced condition for 24 h.The apoptosis rate was detected by flow cytometry.The autophagy related proteins Beclin 1 were detected by western blot.Results The expression of HIF1A-AS1 in hypoxic cells was increased as the hypoxic time increased since 6 h and peaked at 36 h, which was significantly higher than that in control group (P<0.01).HIF1A-AS1 relative expression in HIF1A-AS1 overexpression and low expression PANC1 cells was 4.49±0.53 and 0.49±0.07, which were normalized to that of control group with the relative expression of 1.Control group had lower HIF1A-AS1 expression than HIF1A-AS1 overexpression PANC1 cells but higher HIF1A-AS1 in HIF1A-AS1 low expression PANC1 cells, and the differences were statistically significant (P<0.01).The cell apoptosis rate of control, HIF1A-AS1 overexpression and low expression PANC1 cells was (8.27±1.28)%, (6.56±1.49)% and (19.9±2.34)% after 24 h hypoxic culture.Control group had higher HIF1A-AS1 expression than HIF1A-AS1 overexpression PANC1 cells but lower HIF1A-AS1 in HIF1A-AS1 low expression PANC1 cells, and the differences were statistically significant (P<0.01).The expression of Beclin 1 protein was protein 1.05±0.11, 1.29±0.19 and 0.38±0.18, respectively.Control group had lower Beclin 1 expression than HIF1A-AS1 overexpression PANC1 cells but higher Beclin 1 in HIF1A-AS1 low expression PANC1 cells, and the differences were statistically significant (P<0.01).Conclusions HIF1A-AS1 can promote autophagy of pancreatic cancer PANC1 cells induced by hypoxia and participate in the pathogenesis and metastasis of pancreatic cancer.

Objective To investigate the role and potential mechanism of high mobility group box-1 protein (HMGB1) on improving the chemosensitivity of gemcitabine-resistant pancreatic cancer PANC1 ceils.Methods Gemcitabine-resistant pancreatic cancer PANC1 (PANC1-GR) cell line was established by using increased gradient concentration of gemcitabine.The si-HMGB1-PANC1 and si-HMGB1-PANC1-GR cells were established by the transfection with HMGB1 siRNA using liposome.The 50% inhibitory concentration (IC50) and Resistance index (RI) of gemcitabine in 4 PANC1 cell lines with or without HMGB1 siRNA transfection were determined and calculated by CCK-8 assay.Western blot assay was used to detect the protein expression of HMGB1 in PANC1 and PANC1-GR cells and the expression of autophagy marker protein Beclin1 in the 4 PANC1 cell lines.Flow cytometry assay was used to evaluate the apoptosis rate of 4 pancreatic caner cell lines.Results The gemcitabine-resistant pancreatic cancer cell line PANC1-GR was successfully established,which could grow stably and passage in media with 100 μmol/L gemcitabine.The IC50 of gemcitabine in PANC1,PANC1-GR,si-HMGB1-PANC1,and si-HMGB1-PANC1-GR cells lines were (4.7 ±0.4) μmoL/L,(166.5 ± 13.6) μmol/L,(3.2 ± 0.3) μmol/L,and (52.4 ± 8.4) μmol/L,respectively.The IC50 in PANC1-GR wassignificantly higher than that in PANC1,while the IC50 in the transfected cells was significantly lower than that in untransfected cells,and the differences were both statistically significant (bothP < 0.01).The RI value of gemcitabine in transfected and untransfected PANC1-GR cells was 35.4 and 16.4.The relative protein levels of HMGB1 in PANC1 and PANC1-GR were 0.17 ± 0.08 and 0.38 ± 0.11.The expression of HMGB1 in PANC1-GR was obviously higher than that in PANC1,and the difference was statistically significant (P<0.01).The relative protein levels of Beclin1 in PANC1,PANC1-GR,si-HMGB1-PANC1 and siHMGB1-PANC1-GR cells were 2.68 ± 0.23,3.28 ± 0.15,0.68 ± 0.23 and 0.78 ± 0.11,which in two transfected cells was greatly lower than those in untransfected cells.The apoptosis level was (34.58± 3.14)%,(79.56±3.58)%,(19.41± 1.53)%,and (34.57±2.94)%.The apoptosis level in the 2 transfected cell lines were significantly higher than those in the 2 untransfected cell lines,and the differences were both statistically significant (P < 0.01).Conclusions The inhibition of HMGB1 could improve the chemosensitivity of gemcitabine in pancreatic cancer PANC1 cells,which might be mediated by the activation of autophagy.

Objective:
To observe the predictive value of serial platelet function testing (PFT) on outcome in patients undergoing complex percutaneous coronary intervention (PCI).
Methods:
Six hundred and two consecutive patients undergoing complex PCI in Anzhen hospital were enrolled during October 2011 to June 2012.Adenosine diphosphate(ADP)-induced platelet aggregation was measured by light transmission aggregometry on the first, sixth and twelfth month after PCI and the mean value was calculated.The cut-off value of high on-treatment platelet reactivity (HTPR) was defined as 40%.The primary endpoint was major adverse cardiovascular and cerebral event (MACCE). Clinical outcomes were analyzed by the Kaplan-Meier method and differences were compared using the log-rank test.Multivariate analyses by Cox proportion hazards regression were applied to identify variables independently associated with the adverse outcomes.

Objective:To explore regulatory effects of short-chain fatty acids (SCFA) on hypoxia-induced oxidative stress and activation of pancreatic stellate cells (PSCs) .Methods:PSCs were cultured in normoxia or hypoxia conditions to establish normoxia or hypoxia group. PSCs were pre-treated with SCFA working solution (10 mmol/L sodium acetate, 0.5 mmol/L sodium propionate and 0.5 mmol/L sodium butyrate), and then cultured in hypoxia conditions to establish the hypoxia-SCFA group. PSCs pre-treated by normal saline was set as the hypoxia-control group. The relative growth viability of the cells was detected by the CCK-8 assay. Relative levels of reactive oxygen species (ROS) were detected by DCFH-DA fluorescence probe method. The mitochondrial membrane potential was detected by JC-1 fluorescence probe. Protein expression of cyclin-associated marker cyclin A and cyclin D, hypoxic marker HIF1α, activation marker α-SMA, and antioxidant marker NRF2 and HO-1 was detected by western blotting.Results:The relative viability of PSCs in hypoxia group was significantly higher than that in normoxia group at 48 h (1.23±0.05 vs 0.99±0.04), but the relative viability of hypoxia-SCFA group was significantly lower than that of the hypoxic-control group at both 36 h and 48 h (0.69±0.01 vs 0.86±0.03, 0.86±0.02 vs 1.25±0.05). The relative level of ROS was significantly higher in hypoxia group than normoxia group (1.74±0.11 vs 1.00±0.10). The relative level of ROS was significantly lower in the hypoxia-SCFA group than the hypoxia-control group (1.39±0.14 vs 1.66±0.11). The fluorescence signals of JC-1 polymer in hypoxia group were significantly higher than those in normoxia group (1.36±0.05 vs 1.00±0.11), whereas the fluorescence signals of JC-1 polymer were significantly lower in hypoxia-SCFA group than in hypoxia-control group (1.11±0.03 vs 1.32±0.06). The expression of cyclin A, cyclin D, HIF1α, α-SMA, NRF2, and HO-1 was significantly higher in hypoxia group than those in normoxia group (1.19±0.01 vs 0.63±0.02, 0.93±0.02 vs 0.83±0.03, 1.18±0.07 vs 0.41±0.02, 1.19±0.14 vs 0.66±0.04, 1.22±0.11 vs 0.61±0.04, 1.28±0.12 vs 0.68±0.02), but the expression of cyclin A, cyclin D, α-SMA, NRF2, and HO-1 in Hypoxia-SCFA group was significantly lower than those in hypoxia-control group (0.79±0.04 vs 1.15±0.03, 0.88±0.01 vs 0.95±0.03, 0.87±0.01 vs 1.18±0.05, 0.84±0.01 vs 1.22±0.04, and 0.92±0.02 vs 1.27±0.06). All these differences were statistically significant (all P values <0.05) . Conclusions:SCFA significantly improves the oxidative stress state of PSCs under hypoxic conditions, maintains the stability of mitochondrial membrane potential, and inhibites hypoxia-induced activation of PSCs.
8.Application of fuzzy combination of TOPSIS method and rank sum ratio method to comprehensive evaluation of radiodiagnosis and radiotherapy services

Objective:To explore a comprehensive evaluation method for radiodiagnosis and radiotherapy services, with 16 cities in Shandong province as a case study.Methods:The survey data from 3 475 radiodiagnosis and radiotherapy facilities in the province in 2023 were analyzed using a weighted fuzzy combined approach of the TOPSIS method and rank sum ratio method. Seven key indicators were selected, including the number of radiodiagnosis and radiotherapy facilities per million population, the number of radiological workers per million population, the number of items of radiodiagnosis and radiotherapy equipment per million population, the number of X-ray diagnoses per thousand population, the number of interventions per thousand population, the number of radiotherapies per thousand population, and the number of nuclear medicine diagnoses and therapies per thousand population, to provide a comprehensive assessment of the quality of radiodiagnosis and radiotherapy services in the 16 cities of Shandong province in 2023. The study also explored the association between the quality of radiodiagnosis and radiotherapy services in each city and its per capita GDP.Results:The findings indicated an unbalanced development of radiodiagnosis and radiotherapy services in Shandong province, with Jinan, Dongying, and Qingdao ranking the top three cities, and Heze, Linyi, and Dezhou ranking the bottom three. Furthermore, the number of radiological workers per million population, the number of X-ray diagnoses per thousand population, the number of interventions per thousand population, and the outcomes of the weighted fuzzy combination method, TOPSIS method, and RSR were shown to be positively correlated with the per capita GDP in each city ( r=0.597– 0.750, P < 0.05). Conclusions:The combined application of the TOPSIS method and rank sum ratio method offers an impartial evaluation of radiodiagnosis and radiotherapy services across various cities in Shandong province. This method is considered appropriate for the comprehensive assessment of radiodiagnosis and radiotherapy services and holds practical significance for broader implementation.
9.Study on estimation methods of absorbed dose to eye lens of infants from CT head and neck scanning based on one-year old anthropomorphic phantom

Objective To study the methods to estimate absorbed dose to eye lens of infants from CT scanning using different protocols and try to find a practical quick way for estimating absorbed dose to eye lens.Methods By scanning one-year old anthropomorphic phantom using 7 kinds of different protocols,all TLDs were measured for final estimation of absorbed dose to eye lens using two different dose conversion methods.Meanwhile,linear regression equation was established between absorbed dose to eye lens and CTDI.Results Absorbed doses to eye lens of infants from children CT scanning using 7 kinds of different protocols were (9.96±0.69) mGy in head axis,(7.01±0.42) mGy in head helical,(12.60± 0.97) mGy in sinus,(12.97±0.42) mGy in inner ear,(0.63±0.03) mGy in neck soft tissue,(8.89± 0.44) mGy in cervical vertebra,and (0.34± 0.01) mGy in chest,respectively.There were statistically significant difference in doses among different groups (F =846.826,P < 0.05).For different scanning locations,there was linear relation between absorbed dose to eye lens and CTDI(r=0.986-0.999,P< 0.05).Conclusions Absorbed dose to eye lens of infants from children CT scaning with single dose may not be above threshold dose.In addition,absorbed dose to eye lens can be estimated quickly by linear regression equation between absorbed dose to eye lens and CTDI.
10.Short-term outcomes of dual kidney transplantations in adults: a report of 7 cases

Objective:To explore the short-term outcomes of dual kidney transplantation and summarize its safety and feasibility.Methods:From September 2018 to September 2019, a total of 7 dual kidney transplantations were performed. And retrospective analysis was performed for baseline profiles, clinical data and postoperative complications.Results:The mean age was (62.7±8.5) years for donors and (43.9±9.3) years for recipients. The Remuzzi score of 6 paired kidneys ranged from 4 to 6 points. During follow-ups, the survival rate of 7 dual kidney transplantation grafts and recipients was 100%. The median follow-up period was 16 months. Renal function of 6 recipients normalized within 1 week and delayed graft function (DGF) occurred in one case. All of them underwent unilateral kidney transplantation with an average operative duration of (5.6±1.4) hours. There was no onset of operative complications. One case of rejection was not confirmed by biopsy. Among three patients of lung infections, there was one case of severe pneumonia. In 3 cases, lateral plasma flow of transplanted kidney exceeded that of medial plasma flow.Conclusions:Dual kidney transplantation in adults is both safe and feasible so as to expand the availability of donated kidney.
