1.Expression and clinical significance of epidermal growth factor receptor and vascular endothelial growth factor in non-small cell lung cancer
Chinese Journal of Primary Medicine and Pharmacy 2012;19(11):1634-1636
Objective To observe the expression of epidermal growth factor receptor(EGFR) and vascular endothelial growth factor(VEGF) in non-small cell lung cancer(NSCLC) and the reletionship with clinic-pathologic parameters. Methods Immunohistochemistry of tissue sections was used to study the expression of EGFR and VEGF in 124 NSCLC tissues and 15 normal lung tissues adjacent to cancer tissues. Results The positive rates of EGFR and VEGF in 124 NSCLC samples were 52.4%,66.1%,while no expressed in the 15 normal lung tissues adjacent to cancer tissues. The positive rate of VEGF with stage Ⅲ was higher than that with stage Ⅰ - Ⅱ ( 80.4% vs 56.2%,P <0.05).The positive rate of VEGF with lymph node metastasis was higher than that without lymph node metastasis (78,2% vs 53.3%,P < 0.05 ).The expression of VEGF and EGFR had no relationship with clinic pathological factors including histological type of lung cancer, sex, age.EGFR and VEGF expression was statistically correlated with each other( r =0.93,P <0.05 ).Conclusion EGFR and VEGF were overexpressed in NSCLC tissues. The expression of VEGF was correlated with TNM stages and lymph node metastasis. The expression of EGFR was correlated with VEGF expression in NSCLC tissues, which indicates they might act synergistically in angiogenesis of NSCLC.

Objective To discuss the relationship between self-control and aggressive structure of middle school students. Methods Using Self-control Questionnaire,Aggressive Questionnaire and the Implicit Association Test(IAT)to test 298 middle school students. Results The effects of students'thought control, emotion control and behavior control related not remarkable on implicit aggression(r=-0.11,r=-0.07,r=-0.03,P>0.05),but remarkable on the explicit aggression(β=-0.232,β=-0.188,β=-0.231,P<0.05).The moderating effect of implicit aggression to the relationship between self-control and explicit aggression is weakly(△R2=0.007,P>0.05).Conclusion Self-control has remarkable inhibitory action to the explicit aggression, and the inhibitory action will not be influenced by implicit aggression.

The aimed of this study was to prepare stabilized thiomers to overcome the poor stability character of traditional thiomers. Poly(acrylic acid)-cysteine (PAA-Cys) was synthesized by conjugating cysteine with poly(acrylic acid) and poly(acrylic acid)-cysteine-6-mercaptonicotinic acid (PAA-Cys-6MNA, stabilized thiomers) was synthesized by grafting a protecting group 6-mercaptonicotinic acid (6MNA) with PAA-Cys. The free thiol of PAA-Cys was determined by Ellmann's reagent method and the ratio of 6MNA coupled was determined by glutathione reduction method. The study of permeation enhancement and stabilized function was conducted by using Franz diffusion cell method, with fluorescein isothiocyanate dextran (FD4) used as model drug. The influence of polymers on tight junctions of Caco-2 cell monolayer was detected with laser scanning confocal fluorescence microscope. The results indicated that both PAA-Cys and PAA-Cys-6MNA could promote the permeation of FD4 across excised rat intestine, and the permeation function of PAA-Cys-6MNA was not influence by the pH of the storage environment and the oxidation of air after the protecting group 6MNA was grafted. The distribution of tight junction protein of Caco-2 cell monolayer F-actin was influenced after incubation with PAA-Cys and PAA-Cys-6MNA. In conclusion, stabilized thiomers (PAA-Cys-6MNA) maintained the permeation function compared with the traditional thiomers (PAA-Cys) and its stability was improved. The mechanism of the permeation enhancement function of the polymers might be related to their influence on tight junction relating proteins of cells.

Objective To determine the role of Bmi-1 in the submandibular gland (SMG) of mice. Methods SMG of 4-week wild-type(WT) and Bmi-1 null(Bmi-1-/-) mice was analyzed on the weight, salivary flow rate,hematoxylin-eosin staining morphological differences and the changes in proliferation and aging by histology, immunohistochemistry and Western blotting. Results Compared with WT mice, the average static salivary flow rate [WT:(0.21 ±0.02) μg/min,Bmi-1-/-:(0.10 ±0.02) μg/min] (P =0.001) and the submandibular gland weight [WT:(1.89 ± 0.15) μg],Bmi-1-/-:[(1.34 ± 0.07) μg] (P =0.003) of the male Bmi-1-/-mice were significantly decreased, the number of gland duct increased, and the granular convoluted duct showed reduced diameter and branches.More senescence-associated β-galactosidase positive cells existed in SMG of Bmi-1-/-mice (WT:0.00,Bmi-1-/-:0.18 ± 0.02),and Ki-67 immunopositive cells decreased in SMG of Bmi-1-/-mice(WT:0.40 ~ 0.47,Bmi-1-/-:0.18 ~ 0.20) (P =0.000).The expression of p16 (WT:1.00 ± 0.12,Bmi-1-/-:0.00 ±0.00) (P =0.003) and p19(WT:0.97 ±0.09,Bmi-1-/-:5.09 ±0.21) (P =0.004) were up-regulated dramatically in SMG of the Bmi-1-/-mice.Conclusions Bmi-1 gene deficiency causes abnormal function of SMG by inducing senescence phenotype of SMG.
