1.Effects of joint synovial fluid on tendon-bone healing of the reconstructed ligaments

[Objective]To explore the effects of joint synovial fluid on tendon-bone healing of reconstructed ligament. [Methods]Forty female New Zealand rabbits,aged 6 months,weight 2.5~3.5 kg,were divided into 4 groups randomly.Transfer of semitendinosus tendon to the bone tunnels was performed to make bone-tendon healing model,on the right knee the bone tunnel was not communicated to the joint cavity,in contrast,the bone tunnel on the left knee was connected with synovial fluid.Animals in one group were killed at 8 weeks after operation,bone-tendon healing samples were harvested for biomechanical test.Animals in other three groups were killed at 2,4 and 8 weeks after operation respectively to harvest the samples for histological observation.HE and Masson staining were conducted to examinant histological change.[Results]At 2 weeks after operation,there was no significant difference between non joint synovial fluid group and joint synovial fluid group in bone-tendon interface healing,which were filled with granulation tissue.At 4 weeks postoperatively,interface healing in non joint synovial fluid group was obvious better than that in joint synovial fluid group.Fibroblast cells and cartilage cells increased obviously,and collagen fibers synthesized.At 8 weeks after operation,tendon-bone interface in joint synovial fluid group was filled with connective tissue,the collagen fibers arranged disorderly.In contrast,more compact collagen fibers were seen in non joint synovial fluid group.Interface cell accounts in non joint synovial fluid group were superior to that in joint synovial fluid group at 2,4,8 weeks after operation (P
2.The effect observation of carvedilol and metoprolol on MMP-9 changes and left ventricular remodeling in patients with hypertension heart disease

Objective To study the curative effect of MMP-9 changes on left ventricular hypertrophy patients with hypertension heart disease by carvedilol and metoprolol.Methods 168 cases were divided into two groups.The control group(84 cases)applied metoprolol.The observation group applied carvedilol.The changes of IVST,LVPWT, LVMI and LVGI,expression of MMP-9 were observed in two groups after treatment.Results The values of IVST, LVPWT,LVMI and LVGI,expression of MMP-9 were decreased after treatment.But the decreased values of IVST [(1.72 ±0.35 )mm vs (0.36 ±0.11 )mm,t =5.75,P <0.05 ],LVPWT[(0.66 ±0.25 )mm vs (0.24 ± 0.18)mm,t =4.32,P <0.05],LVMI[(16.23 ±3.35)g/m2 vs (7.44 ±2.24)g/m2 ,t =9.86,P <0.05 ],LVGI [(0.52 ±0.12)g/m2 vs (0.40 ±0.11)g/m2 ,t =4.24,P <0.05],MMP-9[(76.23 ±20.87)vs(42.55 ±10.87),t =6.54,P <0.05]were higher in the observation group than in the control group.Conclusion The treatment of carvedilol is satisfied,can improve ventricular remodeling,decreased the expression of MMP-9,and is worthy of being recommended in left ventricular hypertrophy patients with hypertension heart disease.
3.Bacteriostatic Effect of Volatile Oil from Artemisia Argyi in Vitro and in Vivo

Objective To study the bacteriostatic effect of the volatile oil from Artemisia argyi. Methods Minimal inhibitory concentration (MIC) was measured with double-fold didution method. Protective effect of the volatile oil from Artemisia argyi were determined by experimental treatment of infected mice in vivo. Results The MIC90 of 3 kind of pathogenic bacteria were 0.78~25 ?L/mL and protective rate for infected mice were 40%~50%. Conclusion The volatile oil from Artemisia argyi has better inhibiting effect on Staply lococcus aureus, Escherichiacoli and Pseudomona than Erythromycin. It has better protective effect for infected mice.
4.EVALUATION ON THE EXTERNAL QUALITY ASSESSMENT OF NEWBORN SCREENING FOR INHERITED METABOLIC DISEASE LABORATORY

Objective To evaluate our results of external quality assessment (EQA) program of the newborn screening laboratory for 6 years, summarize the problems in the screening measurement and improve the quality of measurement.Methods The phenylalanine (Phe) and thyrotropin (TSH) values in dried blood spot specimens on filter paper distributed by national center were measured by fluorometric method and time -resolved fluorescence im-munoassay ( DELFIA ) respectively.The results sent by national center were analyzed .Results In the 90 specimens of 18 batches, the bias range of Phe and TSH measurement was wide (-62.50%~66.67%and-66.67%~54.31%respectively) and their average values were 8.62%and 10.18% respectively;85 quantitative results of Phe and 84 quantitative results of TSH measurement were in the quality control , accounting for 94.44%and 93.33%respective-ly.The qualitative analysis of 90 Phe and TSH specimens all conformed to the expected results , accounting for 100%.The result of every batch was up to 80%.Conclusion Our neonatal screening laboratory is in control and it is useful to evaluate the measurement competence , find out the problems in the Phe and TSH measurement and re-solve them in time through participated in the national EQA in neonatal screening laboratory , so as to improve the quality of measurement and ensure the accuracy and reliability of results .
5.Improved conjunctival transplantation for corneal ulcer

Objective To evaluate the therapeutic effect of pedical conjunctival flap transposition on refractory corneal ulcer. Methods The data of 56 eyes of 56 patients with refractory corneal ulcer who underwent pedical conjuctival transposition in a tertiary eye unit in China from June 2005 to June 2009 were retrospectively analyzed. There were 36 infective corneal ulcers, 9 keratoplasty graft ulcers, 4 traumatic ulcers, and 7 chemical burn ulcers. All patients were treated with focal debridement and pedical conjuctival flap transposition. Results Forty-seven out of 56 (83.9%) were cured after one surgery, and 5 (8.9%) had conjunctival flap retraction, among which 3 underwent the second surgery and 2 had the third surgery to completely heal. Four patients (7.2%) turned to lamellar keratoplasty because of fungal corneal ulcer relapse. Ulcer in 52 patients healed and the globe preserved. They had different levels of vision improvement. Conclusion Pedical conjunctival flap transposition is effective for refractory corneal ulcer.
6.FORM AND STRUCTURE OF LIGAMENTUM TRIANGULARE SINISTRUM HEPATIS AND ITS FIBROUS APPENDIX

The left triangular ligament of liver and its fibrous appendix(free border) was studied in 42 Chinese adult male cadavers. 37 fibrous appendices were further studied histologically.In order to prove the formation of the fibrous appendix 30 left triangular ligaments of new born cadavers were also studied. The average length of the left triangular ligament of adult cadavers was 13.91cm.,and that of their fibrous appendices was 6.86cm. Almost all of the left triangular ligaments were attached to the superior surface of the left lobe of liver,consequ- ently, the left suprahepatic space could be properly subdivided into anterior and posterior left suprahepatic spaces by the ligament. The forms of the fibrous appendices were classified into 5 types:triangular (23.8%), narrow band(14.3%), triangular-narrow band(50.0%),membranous (7.1%), and bifurcate type(4.8%).The left triangular ligaments of the 30 new born cadavers were devoid of fibrous appendix. Histological observations of the 37 fibrous appendices showed:aberrant bile ducts were found in 89% of the cases, rudimentory liver-cell cord 65%,nerves 78%, and blood vessels 100%.Their size and number diminished gradually from the pro- Kimal (hepatic) end of the fibrous appendix to the distalend. The authors agreed with Toldt and Zuckandl that the fibrous appendix might be the result of the retrogression of the left lobe and suggested that the occurrence of the fibrous appendix was after birth. The clinical importance of the fibrous appen- dix was discussed.
7.Effects of Pericarpium aurantiin water extract on sleeping and spontaneous activity of Kunming mice

BACKGROUND: Effects of Pericarpium aurantiin water extract on sleeping and spontaneous activity in Kunming mice has been investigated in three experiments, that is, the experiments of sleeping time due to supraliminal and subliminal dosage of sodium pentobarbital, and shaking cage for energy transducer, respectively.OBJECTIVE: To observe the influence of Pericarpium aurantiin water extract on sleeping and spontaneous activity of Kunming mice.DESIGN:Randomized controlled trial.SETTING: The Staff Rooms of Pathology, Physiology and Pharmacology in Gannan Medical College.MATERIALS: The experiment was conducted in the Central Laboratory of Science Research, Gannan Medical College from January through February 2005. Ninety adult Kunming mice, grade of clearness, were enrolled and randomly divided into three groups with 30 mice for each experiment:10 in control group, 10 in large-dosage group and 10 in small-dosage group.METHODS: ① In energy transforming trial, spontaneous activity of mice was recorded, small wave with wave amplitude < 5 mm was considered as the waveform of gentle activity in mice, such as fur licking and tickling; middle wave with wave amplitude of 5-10 mm as the waveform of moderate activity, such as walking, standing, climbing up and down;large wave with wave amplitude > 10 mm as the waveform of intensive activity, such as running, jumping up and down. Mice in the small-dosage group, large-dosage group and control group were respectively given intraperitoneal injection of 5 mg/g, 10 mg/g Pericarpium aurantiin water extract and equal volume of normal saline, and placed into the cage after 15 minutes. Then, 3 minutes later, waveforms of mice within 15 s was recorded. ② In the experiments on duration of sleep in mice due to supraliminal or subliminal dosage sodium pentobarbital, 0.02 mL/g or 0.01 mL/g sodium pentobarbital (2.5 g/L) were administrated intraperitoneally into mice in the small-dosage, large-dosage and control group 15 minutes after intraperitoneal injection of 5 mg/g, 10 mg/g Pericarpium aurantiin water extract and equal volume of normal saline were given to the three groups, respectively. Subsequently, comparisons of the number of sleeping mice, time to falling asleep, and sleeping time were carried out among the three groups.MAIN OUTCOME MEASURFS: ① Times of spontaneous activity, ②Sleeping time due to supraliminal dosage of sodium pentobarbital, ③ Duration of sleep due to subliminal dosage of sodium pentobarbital.RESULTS: All the mice were involved in the result analysis without loss.① There were fewer middle and large waves in the small-dosage and largedosage groups as compared with that in the control group (90.5±14.7,58.3±12.5, 45.6±10.1, t=2.341-3.215; P <0.05 or P <0.01), indicating that Pericarpium aurantiin water extract can evidently inhibit spontaneous activities in mice. ② In comparison with the control group, the time to falling asleep and the duration of sleep due to supraliminal dosage of sodium pentobarbital were shortened and prolonged significantly, respectively,in the small-dosage and large-dosage groups. ③ In comparison with the control group, the time to falling asleep and duration of sleep due to subliminal dosage of sodium pentobarbital were shortened and prolonged significantly, respectively, in the small-dosage and large-dosage groups.CONCLUSION: Pericarpium aurantiin water extract can prolong the duration of sleep and inhibit the spontaneous activities of Kunming mice,which is in cooperation with sodium pentobarbital in central inhibition.
8.Inhibitory effects of xanthotoxol on neutrophil infiltration and brain edema induced by focal cerebral ischemia-reperfusion injury in rats

Aim To investigate the inhibitory effects of xanthotoxol(XT) on neutrophil infiltration and brain edema induced by focal cerebral ischemia-reperfusion injury in rats.Methods Focal cerebral ischemia-reperfusion model in rat was induced by transient occlusion of the middle cerebral artery for 2 hours and followed by 24 hours of reperfusion.XT(2.5,5 and 10 mg?kg-1,ip)was administered at 1 hour and 12 hours after the onset of ischemia,respectively.After 24 hours of reperfusion,the influence of XT on neurological deficit score,brain edema and infarct size were evaluated;the activity of Na+,K+-ATPase,Ca2+-ATPase and myeloperoxidasse(MPO) in the ischemic hemisphere cortex of the middle cerebral artery area was assayed by spectrophotometry;the expression of intercellular adhesion molecule-1(ICAM-1) and E-selectin was measured with immunohistochemistry.Results XT significantly reduced the neurological deficit score,brain edema and infarct size,enhanced activity of Na+,K+-ATPase and Ca2+-ATPas,suppressed the injury-induced upregulation of MPO activity and cell adhesion molecules(ICAM-1 and E-selectin) expression in the brain tissue.Conclusion XT attenuates brain damage following focal cerebral ischemia-reperfusion in rats and its mechanism may partly be due to the inhibition of inflammation and brain edema induced by ischemia-reperfusion.
9.Diagnostic value of cardiopulmonary exercise test for coronary heart disease

Cardiopulmonary exercise test and electrocardiogram exercise test were performed in 68 patients with suspected coronary artery disease,the diagnostic value of two tests was compared with coronary angiography results as the gold standard.The results show that the sensitivity,specificity,positive predictive value and negative predictive value of electrocardiogram exercise test for coronary heart disease were 51.28%,68.97%,68.97% and 51.28% respectively; those for ratio of O2 pulse peak in cardiopulmonary exercise test were 51.28%,75.86%,74.07% and 53.66% respectively.It suggests that the results of cardiopulmonary exercise test may have the same value as electrocardiogram exercise test in diagnosis of coronary artery disease.
10.Diagnostic value of serum ischemia modified albumin in coronary artery disease

Objective To assess value of serum level of ischemia modified albumin (IMA) in diagnosis for myocardial ischemia of coronary artery disease (CAD). Methods Seventy-two patients with clinically suspected myocardial ischemia of CAD admitted to The First People's Hospital of Hangzhou during November 2009 to May 2010 ready for undergoing coronary angiography, the gold standard for diagnosis of CAD, were randomly selected for the study. The patients were divided into CAD and non-CAD groups based on their coronary angiography. Serum level of IMA was determined with cobalt-albumin binding ( ACB) assay before coronary angiography, which served as diagnostic standard for CAD. Logistic regression analysis method was used to evaluate varied levels of IMA with area under the receiver operating characteristic curve (AUCROC) in diagnosis for myocardial ischemia of CAD. Results Mean level of IMA was (97 ±24) U/ml and (81 ±15) U/ml for CAD group (n =51) and non-CAD group (n =21), respectively. Sensitivity and specificity of a cut-off value of IMA 83.69 U/ml in diagnosis for myocardial ischemia of CAD was 80 percent and 57 percent, respectively, with a predictive value of a positive test 82 percent and that of a negative test 55 percent, respectively, from AUCROC. Logistic regression analysis demonstrated that both hypertension (P=0. 022, 6 = 1.421, OR=4. 141) and level of IMA (P=0.003, b= 1.780, OR=5.928) were independent predictors for CAD. Conclusions Sensitivity, specificity and predictive value of a positive test of the level of IMA are relatively high in diagnosis for myocardial ischemia of CAD, which is an independent predictor of it.
