1.Study of Quality Control Method for Compound Weilining Nose Drops

OBJECTIVE:To establish the quality control method for compound Weilining nose drops.METHODS:First order derivation spectrophotometry was used to determine the content of Weilining in nose drops at wavelength224.7nm.RESULTS:A linearity of concentration(C)and amplitude(D)of Weilining solution was found in range of5~20?g/ml with a regression equation of C=—1.3438+520.78D;(r=0.9998),The average recovery rate was100.35%(n=9),RSD=0.87%.CONCLUSION:The Weilining nose drops is simple in preparation and stable in quality,and this method can be used for quality control of the preparation.
2.Odontogenesis-related gene expression during in vitro culture of tooth germ cells

BACKGROUND:Some studies have indicated that different genes in tooth germ tissue play a role at different time, contributing to tooth germ development.
OBJECTIVE:To observe the expressions of dentin matrix protein 1, enamel protein, col agen I and homeobox gene 1 at different stage of in vitro culture of tooth germ cells.
METHODS:RNA from tooth germ cells was extracted at days 1, 3, 6 of in vitro culture. After reverse transcription, real-time quantitative PCR detection was adopted to measure relative expression of dentin matrix protein 1, enamel protein, col agen I and homeobox gene 1 mRNA.
RESULTS AND CONCLUSION:Dentin matrix protein 1, enamel protein, and col agen I mRNA expressions increased with culture time, and reached the peak at day 3 (P<0.05), whereas homeobox gene 1 mRNA decreased with culture time (P<0.05).
3.Evaluation of right ventricular systolic function in patients with primary pulmonary hypertension using real-time three -dimensional echocardiography(RT -3DE)and ultrasonic two -dimensional speckle tracking imaging technology

Objective To evaluate the right ventricular(RV)systolic function in patients with primary pulmonary hypertension(PPH)using real -time three -dimensional echocardiography(RT -3DE)and ultrasonic two-dimensional speckle tracking imaging(STI).Methods Patients with PPH were divided into three groups according to pulmonary arterial systolic pressure(PASP),consisted of mild group,moderate group and severe group,with 20 cases in each group.20 healthy cases were included as the control group.The global RV dysfunction,including RV end -diastolic volume(EDV),end -systolic volume(ESV),stroke volume(SV),ejection fraction(EF),tricuspid annular plane systolic excursion(TAPSE),longitudinal peak systolic strain(SL),longitudinal peak systolic strain rate(SRL), longitudinal peak systolic velocity(VL)in RV free wall and interventricular septum for basal,mid and apical segment were detected by using RT -3DE and STI.Results Global RV ESV increased in all groups(t =3.10,7.53,7.72,all P <0.01),but The RV EDV in the moderate and severe group was higher than that of normal and mild groups(t =4.02,5.82,3.04,5.15,all P <0.01),the RT SV and EF in all case groups was fewer than that of normal(t =-9.33 ~-2.71,all P <0.05).Linear correlation analysis showed negative correlation between EF and ESV,EDV (r =-0.776,-0.638,all P <0.01).The level of RV free wall peak systolic displacement of tricuspid annulus, ventricular septal annulus peak systolic displacement,tricuspid annulus peak systolic displacement midpoint and the right longitudinal ventricular shortening fraction were significantly lower than those of the control group(all P <0.01). Linear correlation analysis showed positive correlation between EF and TAPSE(r =0.761,0.593,0.745,0.773,all P <0.01).The VL and SL in RV free wall and interventricular septum for basal,mid and apical segment were fewer than the control group(P <0.05 or P <0.01),but SRL of RV for all segments showed no significantly different in all groups(P >0.05).Conclusion RT -3DE and STI technology can be useful in assessing RV function every period.
4.Performance comparison of 3D printing sheep vertebral bone meal/polyvinyl alcohol scaffold, nano-hydroxyapatite/polyvinyl alcohol scaffold and sheep vertebral bone meal/polyvinyl alcohol nonporous bone plate

BACKGROUND:With the promotion of 3D printing technology, 3D printing scaffolds for bone tissue engineering have become the new ideas for jaw bone repair. OBJECTIVE:To compare the physical and biological properties of sheep vertebral bone meal/polyvinyl alcohol (PVA) scaffold, nano-hydroxyapatite (nHA)/PVA scaffold, and sheep vertebral bone meal/PVA nonporous bone plate. METHODS:3D printing technology was used to print sheep vertebral bone meal/PVA scaffold, nHA/PVA scaffold, and sheep vertebral bone meal/PVA nonporous bone plate. Porosity, morphology, water absorption rate and mechanical properties of different scaffolds were detected. Three kinds of scaffolds were al used to culture bone marrow mesenchymal stem cel s, and cel proliferation ability was detected using cel counting kit-8 at 1, 4, 7 days of culture. RESULTS AND CONCLUSION:Under scanning electron microscope, the sheep vertebral bone meal/PVA scaffold and nHA/PVA scaffold exhibited regular and interconnected pores with good continuity and clear network structure;the sheep vertebral bone meal/PVA nonporous bone plate had no obvious pores;however, it had dense and evenly distributed micropores with different sizes on its surface. The porosity of nHA/PVA scaffold was lower than that of the sheep vertebral bone meal/PVA scaffold (P<0.05). The water absorption rate was highest for the nHA/PVA scaffold fol owed by the sheep vertebral bone meal/PVA scaffold and the sheep vertebral bone meal/PVA nonporous bone plate (P<0.05). In contrast, the scaffold toughness was highest for the sheep vertebral bone meal/PVA nonporous bone plate, fol owed by the sheep vertebral bone meal/PVA scaffold and nHA/PVA scaffold. In addition, the cel proliferation activity of cel s cultured on the sheep vertebral bone meal/PVA scaffold was significantly higher than that cultured on the other two kinds of scaffolds. Taken together, the 3D printing sheep vertebral bone/PVA scaffold has good physical and chemical performance.
5.The expression and the clinical guiding significance of HIF-1α and Hp in Mongolian and Han two nationalities

Objective To detect the expressions of two protein factors hypoxia-inducible factor (HIF)-1α and Helicobacter pylori (Hp) in gastric cancer in Mongolian and Han two nationalities respectively,and to explore the relationship of HIF-1α and Hp in the development of gastric cancer.Methods A modified Warthin-Starry (W-S) silver staining method was used to detect Hp infection in 27 Mongolian patients with gastric cancer and 30 Han patients with gastric cancer.Immunohistochemical (S-P) method and reverse transcription polymerase chain reaction (RT-PCR) method were used to detect protein and mRNA expressions of HIF-1α in cancer tissue,paracancerous tissue and normal tissue samples.The relationship of HIF-1α protein and Hp expression was analyzed between gastric carcinoma tissue samples of Mongolian patient group and Han patient group.Results Hp infection rates were 81.5% (22/27) and 66.7% (20/30) in Mongolian and Han patients with gastric cancer.The difference in infection rate was not statistically significant between two groups of patients (~=1.608,P > 0.05).The positive expression levels of HIF-1α protein were 62.96% (17/27) and 70.00% (21/30) in gastric cancer tissue samples of Mongolian and Han groups.The relative expression levels of HIF-1α mRNA were 0.891 4±0.075 0 and 0.907 4±0.065 5.There was no significant difference in the HIF-1 α mRNA expression between the two nationalities.No positive expressions of HIF-1α protein and mRNA were found in paracancerous tissue and normal tissue samples.There was a positive correlation in the positive expression of HIF-1α protein and Hp infection in gastric cancer tissue samples of Mongolian and Han patient groups (r,=0.424 and 0.617,P < 0.05).Conclusion HIF-1α and Hp are pathogenic factors of gastric cancer.Hp and HIF-1α may promote the occurrence of gastric cancer together.The pathogenic mechanism of HIF-1α and Hp in gastric cancer may be the same for Mongolian and Han patients.
6.Clinical value of time-intensity curve analysis of hysterosalpingo-contrast-sonography in the diagnosis of women infertility due to tube obstruction

Objective To investigate the clinical value of hysterosalpingo-contrast-sonography (HyCoSy) with high-pressure syringe injection and time-intensity curve analysis in the diagnosis of women infertility due to tube obstruction.Methods Fifty-six women with infertility underwent HyCoSy with compressive injection of SonoVue with a balloon catheter and high-pressure syringe.Time-intensity curve based on Autotracking Contrast Quatification (ACQ) software was analyzed in all patients.The arrival time (AT) of microbubbles from uterine cavity to tube fimbria was analyzed between partially obstructed tube group and control group.Results The AT in the group of partially obstructed tubes was (3.49 ± 1.70) s,while the AT in the group of freely patent tubes was (1.14-± 0.37)s,there was statistical difference of AT between the group of partially obstructed tubes and the group of freely patent tubes (P =0.000).According to ROC results,the optimization point of AT was 1.65 s,the sensitivity,specificity,positive predictive value and negative predictive value were 92.6%,93.3%,96.2% and 87.5%,respectively.Conclusions High-pressure syringe injection in HyCoSy is useful for regular administration and high image quality.The AT could be considered as the diagnostic indices in the evaluation of fallopian tube patency for infertile women.
7.Recombinant lentiviral vector transfected sheep bone marrow mesenchymal stem cells and osteogenic gene expression changes

METHODS:Lentiviral vectors carrying bFGF and BMP-2 were constructed to transfect sheep bone marrow mesenchymal stem cells. cells were divided into four groups:bFGF group, BMP-2 group, co-transfection group BACKGROUND:Basic fibroblast growth factor (bFGF) can promote the proliferation of bone marrow stromal cells, and bone morphogenetic protein-2 (BMP-2) has an important significance in the induction of new bone formation.
OBJECTIVE:To analyze the effects of bFGF, BMP-2 and their co-transfection on the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells and to compare the relative expressions of col agen I, osteocalcin and osteopontin before and after celltransfection, thereby providing theoretical implications for seed cells in the construction of tissue-engineered bone.

RESULTS AND CONCLUSION:Significant difference in non-specific osteogenic gene expressions was found among the four groups (P<0.05). bFGF and BMP-2 showed an interaction (P<0.05). Expressions of col agen I and osteocalcin in the co-tranfection group were higher than those in the other three groups (P<0.05), but osteopontin expression exhibited no difference (P>0.05). In vitro experiments showed that the relative expression of col agen I, osteocalcin and osteopontin were higher in the co-transfection group, indicating the cells from the co-transfection group have strongest osteogenic capacity that are suitable for seed cells for bone tissue engineering.

Fifteen patients with spasticity were treated by phenol blocks. The results are satisfactory.Phenol blocks has the advantages of high selectivity,few side-effects and low cost. The anthor recommends that phenol blocks could be used in clinical antispasticity treatment.
9.An experimental study of physical intelligence teaching on sensory integration function of 4-5-year-old children

Objective:
To examine the impact of physical intelligence teaching on the function of children s sensory integration, so as to provide reference for promoting the development of sensory integration system.
Methods:
From February to May 2023, the intervention was implemented for 12 weeks among 136 children aged 4-5 (68 in the intervention group and 68 in the control group). The intervention group received situational and game based physical intelligence teaching, the control group received sports game teaching according to the original curriculum objectives of the kindergarten. Intervention was administered 3 times a week for 40 minutes each time. The sensory integration ability of the intervention group and the control group were evaluated before and after the intervention with Chi square test and t test.
Results:
The vestibular sensation, proprioception and tactile sensation of between boys and girls in the intervention group were significantly improved compared with before intervention (boys:44.14±11.52 vs. 53.34± 9.49 ,44.57±12.76 vs. 50.54±11.86,49.31±12.18 vs. 55.00±10.24,girls:46.00±11.01 vs. 54.58±10.06,48.79±13.17 vs. 53.64±11.97,52.67±11.67 vs. 56.91±10.42, t =-3.24,-2.49,-2.09,-5.24,-12.94,-2.56, P <0.05). There was no significant difference in vestibular sensation between boys and girls in the control group (boys:45.91±11.66 vs. 46.31± 11.20,girls:48.27±13.56 vs. 48.45 ±13.54, t =-0.87,-0.07, P >0.05), but there was a significant improvement in proprioception and tactile sensation in both boys and girls (boys:46.63±11.76 vs. 48.06±11.69,51.63±11.98 vs. 52.40±12.18,girls:50.45±12.16 vs. 51.67± 12.03 ,53.36±12.48 vs. 54.39±12.57, t =-3.36,-2.08,-4.66,-2.86, P <0.05). After the intervention, compared with the control group, the vestibular sensation of both boys and girls significantly improved ( t=2.83, 2.08, P <0.05), with exception of proprioception and tactile sensation ( t =0.88,0.67,0.97,0.88, P >0.05). In the experimental group, the number of normal boys increased from 12 to 24, while the number of dysfunctional boys decreased from 23 to 11, with a statistically significant difference ( χ 2=11.53, P <0.01). There was no statistically significant difference in sensory integration in boys of the control group before and after the experiment ( χ 2= 1.10 , P >0.05). After intervention,the number of normal girls in the experimental group increased from 15 to 27, while the number of dysfunctional girls decreased from 18 to 6, with a statistically significant difference ( χ 2=10.39, P < 0.05 ). There was no statistically significant difference in sensory integration in girls from the control group before and after the experiment ( χ 2=2.08, P > 0.05 ).
Conclusion
Physical intelligence teaching can effectively improve children s sensory integration ability, especially for vestibular function.
10.Neurocognitive Graphs of First-Episode Schizophrenia and Major Depression Based on Cognitive Features.

Neurocognitive deficits are frequently observed in patients with schizophrenia and major depressive disorder (MDD). The relations between cognitive features may be represented by neurocognitive graphs based on cognitive features, modeled as Gaussian Markov random fields. However, it is unclear whether it is possible to differentiate between phenotypic patterns associated with the differential diagnosis of schizophrenia and depression using this neurocognitive graph approach. In this study, we enrolled 215 first-episode patients with schizophrenia (FES), 125 with MDD, and 237 demographically-matched healthy controls (HCs). The cognitive performance of all participants was evaluated using a battery of neurocognitive tests. The graphical LASSO model was trained with a one-vs-one scenario to learn the conditional independent structure of neurocognitive features of each group. Participants in the holdout dataset were classified into different groups with the highest likelihood. A partial correlation matrix was transformed from the graphical model to further explore the neurocognitive graph for each group. The classification approach identified the diagnostic class for individuals with an average accuracy of 73.41% for FES vs HC, 67.07% for MDD vs HC, and 59.48% for FES vs MDD. Both of the neurocognitive graphs for FES and MDD had more connections and higher node centrality than those for HC. The neurocognitive graph for FES was less sparse and had more connections than that for MDD. Thus, neurocognitive graphs based on cognitive features are promising for describing endophenotypes that may discriminate schizophrenia from depression.
