OBJECTIVE:To explore the standardized package of Metformin tablets to meet clinical needs. METHODS:Statis-tics was conducted for the utilization data of Metformin tablets in medical and health institutions from 6 cities of China;question-naires were designed to investigate and analyze the evaluation for the suitability of physicians, pharmacists and patients in the pre-scription,deployment and use links to Metformin tablets with different packaging loaded amount in Beijing and Haikou. RE-SULTS:For 0.5 g/tablet,the daily dose of 1.5 g accounted for the largest proportion (32.23%-69.91%) in 5 cities except for Chengdu. Totally 490 questionnaires about package suitability of Metformin tablets in outpatient department were sent out,includ-ing 478 valid questionnaires with effective rate of 97.5%. Results showed that packaging quantity with 4 weeks was considered as appropriate by physicians, pharmacists and patients in Beijing;however, packaging quantity with 1-2 week(s) was considered as appropriate by physicians, pharmacists and patients in Haikou;300-500 tablets of packaging quantity were preferred to be appropri-ate with the matching degree of automatic dispensing machines in both places. CONCLUSIONS:Considering the results in 2 plac-es,for 0.5 g/tablet,2 weeks is appropriate for the packaging loaded amount in outpatient department, that is 0.5 g×45 tablets/box;and 300-500 tablets/box is appropriate for inpatient pharmacies.

To study the method of anodic oxidation followed by hydrothermal treatment for cp titanium and to know bone response to thin hydroxyapatite layers in vivo, commercially pure titanium plates were anodized at 200 V-400 V with direct electric current density no more than 50 mA/cm2 for 15 minutes in the electrolytic trough. beta-glycerophosphate sodium(0.03-0.04 M) and calcium acetate(0.2-0.3 M) were used as electrolytes. Then, titanium plates were hydrothermal treated in the autoclave for 2 hours at 280 degrees C-300 degrees C. Polishing and grit-blasting surface was used as control to learn bone response to thin layers. Twelve rabbits were evenly divided into 3 groups, each group was implanted with 12 implants into the rabbits femoral bone. After 4, 8 and 16 weeks, implants were taken out and collected respectively and were made grinding slices. The bone-implant interface was observed with light microscope. And the bone-implant interface of polishing and layered implants after 8 weeks implantation was observed with scanning electron microscope. The element contents at the interface of polishing and layered implants before and after 8 weeks implantation were detected with EDAX. Results showed that there was hydroxyapatite(HA) precipitated on the titanium surfaceamellae bone in 8 weeks for thin HA coatings, and no HA debris were found at the interfacial zone. In addition, Ca and P content on the hydrothermal treated implant surface increased much more after implantation than that of polished implants. It was concluded that anodic oxidation followed by hydrothermal treatment could precipitate thin hydroxyapatie layer on the surface of cp titanium, which could improve early formation of woven bone and accelerate woven bone transferring to lamellae bone at the implanted site.

Chinese Journal of Tissue Engineering Research 2014;(37):5929-5934
BACKGROUND:Studies about low-frequency pulsed electromagnetic fields interfering with bone marrow mesenchymal stem cells proliferation and differentiation are many, but the Raman spectra of single stem cells irradiated in electromagnetic fields analyzed by surface Raman spectroscopy analysis are rarely reported. OBJECTIVE:To compare the difference in Raman spectra of bone marrow mesenchymal stem cells with or with no irradiation of 3 000 Hz pulsed electromagnetic fields. METHODS:Bone marrow mesenchymal stem cells isolated from Sprague-Dawley rats were cultured and identified. Passage 3 cells were inoculated into 6-wel plates and divided into two groups:pulsed electromagnetic field irradiation group and blank control group. After cultured for 7 days, cells in the two groups were transferred to physiological saline, and 30 cells were randomly col ected from each group. Four Raman spectra were harvested from each celland the average relative intensity of Raman spectra was calculated and compared between two groups. RESULTS AND CONCLUSION:There were the same Raman peaks in the two groups, and the waveforms were basical y same in the two group based on the curve mapping by origin 7.0 software. The peak value in the irradiation group was decreased compared with the blank control group. Laser optical tweezers Raman spectroscopy can be applied to study the biochemical changes of a single stem cellat the molecular level. The Raman spectra of bone marrow mesenchymal stem cells irradiated by 3 000 Hz pulsed electromagnetic fields differ from those without irradiation, and the peak also lowered after irradiation.

Objective To investigate the clinical efficacy ofHejie Fenxiao Decoction on damp-heat patients with acute kidney injury (AKI).Methods Totally 68 patients were divided into the control group and the treatment group according to the patients’ willings. Both groups were given treatment, such as elimination of the incentives and symptomatic treatment. Besides, patients of the treatment group acceptedHejie Fenxiao Decoction for 30 days. The changes of renal function indexes, such as serum creatinine (SCr), blood urine nitrogen (BUN), uric acid (UA), albumin (ALB) and integral of TCM syndrome, were observed, and recovery ratio and clinical endpoints were compared. Results SCr and eGFR of both group were improved on day 14 (P<0.05). After treatment, the renal function recovery ratio and renal replacement ratio in the two groups showed obvious differences (P<0.05);although survival rate of treatment group was higher than control group, it showed no statistical significance (P>0.05);clinical endpoints analysis of the two groups showed statistical significance (P<0.05). The difference of integrals of TCM syndrome between the two groups showed statistical significance (P<0.05) on day 14.ConclusionHejie Fenxiao Decoction combined with basic regular Western medicine can better improve the clinical symptom, recover the renal function, and decrease the rate of renal replacement and death of AKI damp-heat patients.

AIM:To explore the influences of semaphorin 3A (Sema 3A) on hydrogen peroxide (H2O2)-induced injury in human umbilical vein endothelial cells (HUVECs).METHODS:Sema 3A over-expression vectors were constructed and transfected into the HUVECs by Lipofectamine 2000, and the over-expression effect was verified by qPCR and Western blot. The HUVECs in different groups were treated with or without 200 μmol/L H2O2 for 4 h.The levels of inflammatory cytokines were measured by qPCR.The levels of lactic dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde (MDA) were detected by corresponding colorimetry. The cell viability was measured by MTT assay. The cell apoptosis was analyzed by flow cytometry. The levels of apoptosis-related proteins cleaved caspase-3 and Bcl-2 were determined by Western blot.RESULTS:H2O2 induced inflammatory cytokine secretion, increased the levels of LDH and MDA, decreased SOD activity and cell viability, and increased cell apoptosis in the HUVECs.Over-expression of Sema 3A enhanced the above processes. No injury effect of Sema 3A over-expression on HUVECs without H2O2 treatment was observed, indicating that the injury effects of Sema 3A on HUVECs depended on H2O2.CONCLUSION:Sema 3A markedly enhances H2O2-induced injury in the HUVECs, which depends on H2O2.Sema 3A may promote oxidative stress-caused endothelial cell injury.

Objective To develop an ideal hypertension combined hyperlipidemia(HP/HL)rat model by feeding spontaneously hypertensive rats(SHRs)with high fat diet, and to evaluate the pathological changes in target organs including heart and kidney. Methods Twenty 3-week old male SHRs were randomly divided into two groups: normal fat control group(SHR-NC)and high fat group(SHR-HF). Moreover, ten 3-week old male Wistar-Kyoto rats(WKY)were taken as the model control group(WKY-NC). The rats in SHR-HF group were fed with high-fat diet to induce HP/HL, while rats of WKY-NC and SHR-NC groups were fed with normal diet. The systolic blood pressure(SBP)and body weight were measured every week. At the end of the experiment, the rats were sacrificed to take serum samples for blood lipid analysis including high density lipoprotein(HDL-C), low density lipoprotein(LDL-C), total cholesterol(TC)and triglyceride(TG). Heart and kidney tissue samples were collected to examine the pathological changes using HE and Masson staining. Results Compared with the SHR-NC group, the SHRs fed with high-fat diet for 23 weeks presented significant increase of blood pressure and TC, TG, LDL-C, and decrease of HDL-C. The HP/HL rat model showed pathological changes in the HP/HL target organs, heart and kidney. Renal tissues were severely damaged and showed a large area of fibrosis. Besides, left ventricular hypertrophy and myocardial fibrosis were also observed. Conclusions A HP/HL rat model is successfully constructed by feeding SHRs with high-fat diet for 23 weeks. Most importantly, this model exhibits progressive renal and cardiac alterations, similar to those of patients with HP/HL. This HP/HL rat model may become widely used for evaluation of HP/HL therapeutic drugs.

Journal of International Oncology 2018;45(6):365-367
Lymph node metastasis is the main mode of metastasis of thyroid papillary carcinoma. In the central compartment lymph node dissection, the lymph node posterior to right recurrent laryngeal nerve is hidden because of its deep anatomical position, which is easy to be missed during the operation and becomes one of the factors that affect the recurrence of thyroid papillary carcinoma. The metastasis rate of lymph node posterior to right recurrent laryngeal nerve is 10%-30%,which is related to the tumor size, location and other factors, and its position in the surgical treatment of thyroid cancer is increasingly prominent.

OBJECTIVETo prepare a new synthetic macromolecular resin class of denture adhesive (named Comfort-DA).
METHODSTwelve different formulae for denture adhesive were designed by orthogonal design. Each factor affecting tensile bond strength of adhesive was analyzed by ANNOVA to select the best formula. Then the chemical and physical properties, such as tensile bond strength, pH value, and viscosity, were evaluated and compared with Fittydent in vitro also.
RESULTSThe effect of content of polyvinyl acetate and cellulose ether on tensile bond strength exhibited no statistical significance (P > 0.05), while ethyl alcohol and CMC-Na showed statistical significance and mutual action (P < 0.05). Results of testing viscosity and shearing stress showed that there was no significant difference between Fittydent and Comfort-DA (P > 0.05), while Fittydent exhibited a significantly lower tensile bond strength than the prepared one (P < 0.05). The prepared adhesive demonstrated a neutral or slightly basic pH.
CONCLUSIONSThe generally favorable in vitro performance of the prepared adhesive (Comfort-DA) formulation indicated that it might be efficacious denture adhesive.
