1.Uterine artery embolization for the treatment of uterine adenomyosis:comparison study of different embolic agents

Objective To investigate the clinical effects and complications of uterine artery embolization (UAE) therapy using different embolic agents in treating uterine adenomyosis. Methods During 2004—2011, a total of 45 female patients with uterine adenomyosis were admmitted to authors’ hospital to receive uterine artery embolization therapy. The embolic agents used in the procedures included domestic iodized oil, sodium alginate microspheres and Embosphere microspheres. The patients were randomly divided into group A (n=15, using domestic iodized oil as embolic agent), Group B (n=13, using sodium alginate microspheres as embolic agent) and group C (n = 17, using Embosphere microspheres as embolic agent). After the treatment, all the patients were followed up for over 2 years, focusing on the observation of the clinical symptoms such dysmenorrheal, changes of MRI signs and severe complications. The results were analyzed and compared among the three groups. Results After UAE, the clinical symptoms such as the degree of dysmenorrhea, menstrual quantity, etc. were improved in all patients. Six months after UAE, the obvious remission rate of dysmenorrheal in group A, B and C was 33.3% (5/15), 30.8% (4/13) and 41.1%(7/17) respectively. Twelve months after UAE, MRI scanning showed that the uterine sizes in group A, B and C were reduced to 49.19%, 48.25% and 50.05% respectively. Follow-up examination at 24 months after UAE showed that recurrence of dysmenorrheal in group A, B and C was seen in 2, 4 and 2 cases respectively, and amenorrhea was seen in 2 cases of group A and one case in group C. Conclusion The use of domestic iodized oil, sodium alginate microspheres or Embosphere microspheres as embolic agents in performing super-selective uterine artery embolization for adenomyosis can effectively relieve the degree of dysmenorrheal, decrease the uterine size. In addition to ovarian dysfunction, the uterine cavity adhesion may be also a possible cause of amenorrhea occurring after UAE may. In order to ensure a similar clinical efficacy, the use of granular solid embolic agent is preferred as its safety is possibly higher than the liquid embolic agents.

3.Effects ofHuanshuai Oral Liquid in the Process of Hypoxia-Microvascular Waste-Renal Interstitial Fibrosis in Mice with Atherosclerosis Renal Artery Stenosis

Objective To explore the mechanism ofHuanshuai Oral Liquid in the process of hypoxia-microvascular waste-renal interstitial fibrosis in mice with atherosclerosis renal artery stenosis.Methods The atherosclerosis renal artery stenosis model was established on Apoe-/- mice by high fat feed and uninephrectomy. After 2 weeks of renal ablation, model mice were randomly divided into TCM group (Huanshuai Oral Liquid) and model group. C57BL/6J mice were taken as the blank group. The mice of TCM group receivedHuanshuai Oral Liquid 0.01 mL/(g?d) for gavage, while the blank group and the model group received normal saline 0.01 mL/(g?d) for gavage. Renal function, atherosclerosis and renal interstitial fibrosis were assessed in the 12th week after taking medicine via immunohistochemical method to detect the expressions of the collagen typeⅣ, alpha smooth muscle actin (α-SMA) andⅧ factor related antigen (vWF), and via RT-PCR to detect the expressions of HIF-1α and TGF-β.ResultsCompared with the blank group, the model group suffered severe renal artery stenosis and renal fibrosis, and the expressions of HIF-1α, TGF-β, collagen typeⅣ andα-SMA significantly increased (P<0.05), while the expression of vWF decreased (P<0.05). Compared with the model group, the expressions of HIF-1α, TGF-β, collagen typeⅣ andα-SMA group, the expressions of HIF-1α, TGF-β, collagen typeⅣ andα-SMA significantly decreased in the TCM group (P<0.05), while the expression of vWF significantly increased (P<0.05).ConclusionHuanshuai Oral Liquid could significantly improve the renal function and the renal tubular-interstitial fibrosis. The mechanism might be associated with regulating the expressions of HIF-1α and TGF-β.
4.Percutaneous interventional techniques for the treatment of complications of AIDS

Objective To observe clinical application value of percutaneous interventional techniques for the treatment of complications of AIDS.Methods Twenty-one AIDS patients with complications were enrolled.CT guided percutaneous needle biopsy and drainage were conducted,and the correct rate of diagnosis,clinical curative effect and complications post-interventional therapy were observed.Results The operations of all patients executed successfully.Fourteen patients underwent 16 times biopsy,11 were correctly diagnosed,the diagnostic accordance rate was 81.25%.Drainage was performed in 6 patients with abscess or cystis,then the lesions completely disappeared in 3,deflated in 2,while the rest one needed continously draining.Partial splenic embolization (50%) was performed in 1 patient,and after operation platelet content increased,while hepatic function decreased temporality.No complicated infection and other severe complications occurred.Conclusion Percutaneous interventional techniques for the treatment of complications of AIDS are safe and feasible.Medical staffs should be careful to exposition of HIV infection.

6.Effect of trimethyltin chloride on brain metabolism detected by PET-CT

{L-End}Objective To explore the feasibility of using positron emission tomography (PET) -computed tomography (CT) to detect brain metabolic abnormalities caused by trimethyltin chloride (TMT) poisoning. {L-End}Methods Specific pathogen free healthy SD rats were randomly divided into model group and control group with six rats in each group. Rats in the model group were intraperitoneally injected with a single dose of 10 mg/kg body mass of TMT solution, and rats in the control group were intraperitoneally injected with a single dose of an equal volume of 0.9% sodium chloride solution. Rats were anaesthetized after three days of modeling and underwent PET-CT brain scanning to detect the standardized uptake value (SUV) of 18F-2-fluro-D-deoxy-glucose (18F-FDG). After scanning, rats were sacrificed and brain tissues were collected for brain organ coefficients calculation and brain histopathological analysis. {L-End}Results The rats in the model group showed symptoms of head tremor, limb twitching, irritability and others after TMT modeling. There was no significant difference in the body mass between the two groups of rats on the third day of modeling (P>0.05). The 18F-FDG uptake in the cerebral cortex, cerebellum and brainstem of the rats in the model group was significantly weakened compared with the control group, with deceased SUV values (all P<0.05). No obvious abnormalities were found in CT images and freshly collected brain tissues of rats of the control and model groups. The brain organ coefficients of rats in the two groups showed no significant difference (P>0.05). The results of hematoxylin-eosin staining of brain tissue showed that the cerebral cortex of rats in the model group had more tiny cavities than that of the control group, and some neuronal cells and a small number of hippocampal vertebral cells were tightly and deeply stained, with the cytoplasm and nucleus poorly demarcated, and pericellular space enlarged. The results of Nissen staining showed that the arrangement of neuronal cells in the model group was slightly disordered, and the interstitial space was slightly enlarged, but no other significant abnormal changes were observed. {L-End}Conclusion PET-CT can be used in detecting the metabolic abnormalities of brain in TMT poisoning rat model, making it a sensitive detection method for TMT poisoning.
7.Lentivirus-mediated stable Fas gene silencing in human umbilical cord-derived mesenchymal stem cells.

OBJECTIVETo construct a lentivirus-mediated vector for RNA interference (RNAi) of Fas and establish a human umbilical cord-derived mesenchymal stem cells (UC-MSCs) line with stable Fas gene silencing.
METHODSFour short hairpin RNA sequences targeting the coding region of human Fas mRNA were designed. The synthesized oligonucleotides were ligated with the lentivirus vectors harvested from BamHI and EcoRI double digestion of LV3 recombinant vector. The recombinant lentivirus vectors were transfected into the packaging cells 293T, and the lentivirus titers were determined. Cultured UC-MSCs were infected with the lentivirus, and real-time PCR and Western blotting were used to detect the expressions of Fas mRNA and protein in the transfected cells.
RESULTSRestriction digestion and DNA sequencing showed that the lentiviral vectors were successfully constructed, and the titer of lentivirus reached 3 × 10⁸ TU/ml in the packaging cells. Real-time PCR and Western blot demonstrated significantly suppressed Fas gene expression in UC-MSCs after infection with the recombinant lentivirus.
CONCLUSIONLentivirus-mediated RNAi can effectively inhibit Fas gene expression in cultured UC-MSCs.

8.Relationship between the neutrophil to lymphocyte ratio and prehypertension

Objective To investigate the relationship between the ratio of neutrophils to lymphocyte and prehypertension and its clinical significance. Methods In May 2015,three thousand one hundred and twenty-six subjects who underwent physical examination in Tangshan People's Hospital Health Examination Center were enrolled in the study. 120 cases of pre hyperton were selected as the observation group,and 120 with normal blood pressure were used as the control group. The neutrophils and lymphocyte counts of the subjects were detected. The relationship among the two groups of test indicators,the ratio of neutrophils to lymphocyte counts and blood pressure was analyzed. Results The ratio of neutrophil and lymphocytes in the observation group was (2. 28± 0. 39), significantly higher than that in the control group ( 1. 41 ± 0. 32) ( t= 1. 196 P =0. 028). According to the different ratio of neutrophils / lymphocytes,the patients were divided into 3 groups,group A (1. 12 ± 0. 23), group B ( 1. 73 ± 0. 08), group C ( 2. 57 ± 0. 39), aling with the ratio increasing, the blood pressure increased,the systolic blood pressure of group A,B,C was (115±9) mmHg,(117±11) mmHg,(128 ±6) mmHg,respectively. The difference was statistically significant (F=4. 404,P=0. 013),the diastolic blood pressure of group A,B,C was (73±5) mmHg,(78±7) mmHg,(86±4) mmHg,respectively. The difference was statistically significant (F=4. 553,P=0. 027). Multivariate Logistic regression analysis showed that the ratio of neutrophils to lymphocytes was affected by systolic blood pressure,blood glucose,high density lipoprotein,BMI and triglyceride. Conclusion There is correlation between the ratio of neutrophil to lymphocyte and prehypertension, suggesting that inflammation may be involved in the occurrence and development of hypertension.
9. A qualitative research on head nurses′ management experience about standardized training nurses

Objective:
To understand the real experience of the head nurses in the management process of standardized training nurses and to provide a reference for perfecting the standardized training project and improving the training quality.
Methods:
Semi-structured interviews were conducted among eleven head nurses and data were analyzed by Colaizzi method.
Results:
Three themes were extracted as follows: positive management methods; difficulties and challenges; need of support from superior nursing managers.
Conclusion
Understanding the experiences and feelings of head nurses in the management process of standardized training nurses can help sum up experience and find out the defect,then pay attention to seek effective solution and enhance training result.
10.Association between DNA methylation of MMP-9 gene and cognitive impairment in patients with chronic schizophrenia

Objective To investigate the association between DNA methylation of matrix metallo-proteinase 9(MMP-9) gene and cognitive function including continuous attention,cognitive flexibility and visuospatial memory in patients with chronic schizophrenia.Methods 71 male schizophrenia patients and 69 healthy controls were recruited and detected their cognitive function.Pyrosequencing was used to determine DNA methylation at CpG site in exon 4 and exon 5 of MMP-9 gene.RT-qPCR was used to detect MMP-9 gene expression.Clinical symptoms were assessed by BPRS.Results (1) Cognitive function including Trail Making Test A,Trail Making Test B,Stroop words only,Stroop colors only,Stroop interference,Spatial Processing Block design and Block Design all scores were significantly lower in schizophrenia patients with schizophrenia than those in health controls (P<0.01);(2) The MMP-9 gene expression in PBMCs was significantly higher in schizophrenia patients (1.31± 0.48) than that in health control (1.11 ± 0.45,P< 0.05).The DNA methylation of CpG4-1,CpG4-2,CpG4-3,CpG4-4,CpG4-5,CpG5-1,CpG5-2,CpG5-4 of MMP-9 gene in schizophrenia patients were lower than those in health controls (P<0.01).(3)DNA methylation of CpG4-3 was positive correlated with Stroop interference (r=0.324,P<0.01) in health controls.Correlation analysis showed that DNA methylation of CpG4-1 was negatively correlated with sustained attention function (r =-0.329,P<0.01),cognitive flexibility (r =-0.337,P< 0.01) and visuospatial memory (r =-0.308,P< 0.05) in schizophrenia patients.There were positive correlations between DNA methylation of CpG4-5 (r=0.305,P<0.05),CpG5-1 (r=0.262,P<0.05),CpG5-3 (r=0.260,P<0.05),average DNA methylation at exon 5 (r=0.288,P<0.05) and visuospatial memory.DNA methylation of CpG5-4 was positive correlated with sustained attention function (r=0.251,P<0.05) and visuospatial memory (r=0.350,P< 0.01) respectively.Conclusion Male patients with schizophrenia suffer from extensive cognitive impairment,and methylation of MMP-9 gene may be associated with cognitive impairment of schizophrenia.
