1.Clinical observation on rehabilitation of extremity motor function in the hemiplegic patients with stroke by acupuncture

ObjectiveTo explore an effective rehabilitation method to treat hemiplegic patients with stroke by acupuncture.Methods62 hemiplegic patients were treated by Opposing needling and Non-opposing needling twice a day in alternate way.The motor function of patients was assessed with Brunnstrom Assessment and activities daily living(ADLs) were assessed with Barthel Index revised.ResultsAt 3 months after the treatment,The rate of motor function improved above Ⅳ-Ⅳ level accounted for 62.90%,that of ADL improved 60 points accounted for 56.45%.There were significant difference in Brunnstrom level among pre-treatment and 1,2 months after the treatment(P<0.01),but no significant difference between 2 and 3 months (P>0.05).ConclusionsOpposing needling and Non-opposing needling used twice a day in alternate way can promote the recovery of motor function and ADL of hemiplegic patients with stroke,especially early (less than 2 months) recovery.
2.Preparation of immunotoxin BI7D12-PE38KDEL directed to EGFR and determination of its activity in vitro

Objective:To prepare nanobody-based immunotoxin BI7D12-PE38KDEL targeting EGFR and to examine its cytotoxicity against EGFR positive tumor cells.Methods:By using molecular cloning strategy,prokaryotic expression construct of pET28a-BI7D12-PE38KDEL was generated which consisted of nanobody 7D12 targeting EGFR in the form of a divalent fused with PE38KDEL,a truncated form of pseudomonas exotoxin A via a flexible peptide(G4S)4,and then transformed into E.coli BL21(DE3).Protein expression was induced by adding IPTG,purified by Ni-affinity column chromatography,and verified by Western blot.The binding capacity of the resulted immunotoxin to EGFR-positive cells A549,HT29,MCF-7 and EGFR-negative cells CEM,Jurkat were determined by flow cytometry assay,and its cytotoxicity against the target cells was examined.Briefly,tumor cells were treated with different dosage of the immunotoxin,and the killing efficacy of BI7D12-PE38KDEL on these cells were assessed by WST-1 assay after 72 hours.Results:The SDS-PAGE and Western blot results showed the recombinant immunotoxin BI7D12-PE38KDEL was successfully prepared,and majority of them was expressed in soluble form.BI7D12-PE38KDEL could selectively bind to EGFR-positive cells of A549,HT29,and MCF-7.More importantly,the immunotoxin exhibited much more significant killing effect on these EGFR positive cells compared to the negative control group of CEM and Jurkat cells(P<0.01).Conclusion:In the current study,the nanobody-based immunotoxin BI7D12-PE38KDEL targeting EGFR was successfully prepared and exhibited a superior inhibition effect for the growth of EGFR-positive cells.
3.Correlation analysis between interleukin-18,interleukin-1β,copeptin and acute cerebral infarction

Objective To investigate the dynamic changes of serum interleukin(IL)-18,IL-1β and copeptin in acute cerebral infarction(ACI),and analyze the relationship with the serious degree of ACI.Methods The levels of serum IL-18,IL-1β and copeptin were measured by Double-antibody sanduicb enzyme-linked immunosorbent assay(ELISA)in 83 patients with ACI who were admitted to the hospital within 24 h,and neurological impairment were evaluated by European stroke scale(ESS)at the 1st,3rd and 7th day respectively after hospitalization.At the same time,the patients with ACI were compared with 32 normal adults.Results The levels of serum IL-18,IL-1β and copeptin at the 1st,3rd and 7th day after hospitalization were(131.30±31.62),(168.30±28.12),(141.26±24.23)ng/L,(0.35±0.04),(0.82±0.10),(0.52±0.21)μ g/L,and(3.64±0.26),(4.18±0.53),(3.26±2.41)μ g/L respectively.There were significant differences among different times respectively(P<0.05),furthermore they were higher than those in normal adults[(119.12±27.42)ng/L,(0.21±0.08)μ g/L,(2.63±0.23)μ g/L](P<0.05).Univariate Logistic regression analysis showed that age,copeptin,IL-18,IL-1β and ESS score was the influencing factor in the serious degree of ACI.There was no significant difference in assessing the serious degree of ACI by receiver operating characteristic(ROC)curve between IL-18 and age,copeptin,IL-1 β,ESS score(P>0.05).Conclusions The serum levels of IL-18,IL-1β and copeptin are increasing after ACI,indicating that the inflammatory and immune factors may be involved in the development process of ACI.The serum levels of IL-18,IL-1β and copeptin can reflect the serious degree of ACI.
4.Clinical comparison of four treatment methods for cesarean scar pregnancy.

OBJECTIVETo explore the best approach to treatment of cesarean scar pregnancy (CSP).
METHODSA total of 138 patients with CSP treated between January and December, 2013 were retrospectively analyzed. The patients were treated with conservative drug therapy, direct curettage, uterine curettage after embolization, or open or transvaginal surgery. The amount of blood loss, proportion of patients with blooding loss greater than 50 mL, hospitalization days, and hospitalization expenses were compared among the groups.
RESULTSThe median volume of blood loss was 370 mL in the conservative treatment group, 59 mL in direct curettage group, 67 mL in interventional therapy group, and 1425 mL in the surgical group, and the proportion of patients with blood loss over 50 mL was 76.9%, 38.8%, 27.5%, and 100% in the 4 groups, respectively. The midian hospital stay of the 4 groups was 9.0, 4.0, 6.0 and 10.0 days, with median hospitalization expenses of 12281.0, 3843.5, 14805.0, and 17202.2 RMB Yuan, respectively. All these data were significantly different among the 4 groups (P<0.05).
CONCLUSIONDirect curettage surgery should be encouraged for treatment of CSP. Embolization therapy can reduce the risk of bleeding but is associated with potential complications and more costly, and should be performed with caution. Open or trasnvaginal surgery can be considered in difficult cases of CSP, and its combination with interventional therapy is an option to better preserving the uterus.

5.Osteogenesis of dextran sulfate/recombinant human bone morphogenetic protein-2/chitosan nano microspheres combined with coralline hydroxyapatite in repair of large segmental bone defects

Objective To investigate the osteogenic ability of dextran sulfate/recombinant human bone morphogenetic protein-2/chitosan (DS/rhBMP2/CS) nano microspheres combined with coralline hydroxyapatite (CHA) in repair of segmental bone defects.Methods DS/rhBMP2/CS microspheres prepared by ionic crosslinking method were adsorbed into CHA by lyophilization.Seventy-two New Zealand rabbits were randomly divided into 3 equal groups after they had been made into models of bone defect at the right radius.The defects in the 3 groups were implanted respectively with CHA,rhBMP2/CHA and DS/rhBMP2/ CS/CHA.Another 18 animals served as a blank control group.Blood and bone samples were obtained at 4,8 and 12 weeks after implantation.The serum BGP was detected,and the bone grafts were scanned by micro-CT for calculation of volume ratio of the new bone.Hematoxylin and eosin (HE) staining was performed after bone decalcification.Results All the 72 animals recovered well without any infection or graft exposure.Gross observation at postoperative 12 weeks showed that the DS/rhBMP2/CS/CHA group was the best in the quality,quantity and strength of the new bone,as well as in the healing of bone defects.The serum levels of bone gamma-carboxyglutamic-acid-containing protein at all time points in the DS/rhBMP2/CS/CHA group were significantly higher than those in the CHA and rhBMP2/CHA groups (P < 0.05).Micro-CT scanning demonstrated obvious progress in bone formation,cortical bone and marrow cavity at all time points in the DS/rhBMP2/CS/CHA group which showed significantly faster bone reconstruction synchronized with material degradation and significantly higher volume ratio of the new bone than the other 2 groups (P < 0.05).Histological examinations showed better morphology of mature cortical bone and new marrow cavity at all time points in the DS/rhBMP2/CS/CHA group than in the other 2 groups.Conclusion Since DS/rhBMP2/CS/CHA possesses a better mechanism of sustained-releasing rhBMP2 to induce bone formation because of its reticular and hole-hole-connected structure,it may perform better in repairing segmental bone defects than simple CHA or rhBMP2/CHA.
6. Proteomic analysis and verification of protein expression after upregulation of human CD99 in Hodgkin lymphoma cell line L428

Objective:
To investigate the proteins expression difference after upregulation of human CD99 in Hodgkin Lymphoma cell line, L428 cell, and verify the function of differential proteins.
Methods:
The differential proteins were detected by two-dimensional fluorescence difference gel electrophoresis and mass spectrometry analysis, cluster analysis was done by GOfact.
Results:
There were 38 proteins screened out, of which 21 proteins were positively associated with CD99, while 17 proteins were negative. Among the 38 proteins, 32 proteins participated in biological process, and 35 proteins were involved in the composition and construction. And 28 proteins participated in multifaceted biological activities including antioxidation, protein binding, catalytic activity, regulation of enzyme, signal transduction, molecular structure, regulation of translation and ion transport.
Conclusions
The changes of the differential proteins, correlated with cytoskeleton, cell differentiation, signal pathway and regulating gene expression, are closely relevant to the translation between Hodgkin/Reed-Sternberg and B lymphocyte cell.
7. Influence of psychological capital on occupational stress of employees in labor-intensive electronic companies

OBJECTIVE: To explore the effects of psychological capital on occupational stress of employees in labor-intensive electronic enterprises. METHODS: A total of 1 723 employees in 5 labor-intensive electronic enterprises were selected as study subjects by cluster sampling method. The Psychological Capital Questionnaire and Simple Job Stress Questionnaire were used to investigate their psychological capital and job demand-control( JDC) occupational stress. The effect of psychological capital on occupational stress was analyzed. RESULTS: Total average score of psychological capital of the study subjects was( 4. 4 ± 0. 7). The average scores of self-efficacy,hope,resilience,optimism dimensions were( 4. 3 ± 0. 9),( 4. 5 ± 0. 8),( 4. 5 ± 0. 8) and( 4. 4 ± 0. 7),respectively. The proportions of active,relaxed,nervous and passive occupational stress accounted for 24. 2%,24. 7%,23. 5% and 27. 6% respectively. The detection rate of JDC mode high occupational stress was 68. 7%( 1 184/1 723). The proportion of active occupational stress of the low-psychologicalcapital group was lower( 28. 3% vs 20. 2%,P < 0. 001),the proportion of passive occupational stress was higher( 23. 4% vs 31. 7%,P < 0. 001),the positive rate of JDC high occupational stress was higher( 63. 3% vs 74. 1%,P <0. 01),compared with the high-psychological-capital group. The multivariate logistic analysis showed that the risk of JDC high occupational stress in the low-psychological-capital group was higher than that in the high-psychological-capital group( P < 0. 01),after eliminating the confounding factors such as length of service,education level and personal monthly income. CONCLUSION: The psychological capital of employees in labor-intensive electronics enterprises was associated with the occupational stress of the JDC model. The occupational stress can be reduced by improving the psychological capital of workers.
