1.Application of the continuous language rehabilitation nursing in the motor aphasia patients with abrain stroke

Objective To investigate the clinical effect of continuous language rehabilitation nursing on the language function in patients with stroke motor aphasia. Methods A total of 60 motor aphasia patients with a brain stroke were recruited and divided into experimental group and control group with 30 cases each by random digits table method. The control group was treated with the routine stroke care in traditional Chinese medicine, while the experimental group with the continuing language rehabilitation nursing base on the routine stroke care. The language function of both groups were detected by the Chinese Rehabilitation Research Center Standard Aphasia Examination before and after 3 months treatment, and a survey of nursing satisfaction of patients when discharged was conducted. Results The right rate of hear and understand, retelling, say, read aloud, reading comprehension, transcription, description, dictation, calculation in the Chinese Rehabilitation Research Center Standard Aphasia Examination was (44.35 ± 18.69), (32.37 ± 22.25), (21.49 ± 14.91), (27.63 ± 12.54), (46.87 ± 15.30), (14.25 ± 6.11), (7.33±3.23), (11.63±4.82), (18.93±6.82)%before the treatment and (62.14±10.81), (55.36±19.53), (41.42 ± 13.75), (59.16 ± 11.05), (55.57 ± 13.90), (32.39 ± 7.12), (9.73 ± 3.92), (27.45 ± 4.71), (37.46 ± 6.91)%after the treatment in the experimental group, except for description, the differences were significant ( t=2.096-14.540, P<0.01 or 0.05). The right rate of hear and understand, retelling, say, read aloud, reading comprehension, transcription, description, dictation, calculation in the Chinese Rehabilitation Research Center Standard Aphasia Examination was (43.86±15.89), (34.28±20.27), (22.84±15.40), (28.63±12.45), (47.23±11.30), (13.40±6.24), (7.27±3.70), (11.10±4.73), (19.50±6.80)%before the treatment and (49.46± 14.27), (42.36±20.58), (30.33±13.36), (40.31±11.88), (49.97±10.13), (15.37±5.59), (8.50±3.74), (12.83± 4.36), (21.47 ± 7.74)% after the treatment in the control group, the differences of hear and understand, retelling, say, read aloud were significant (t=2.263-3.991, P<0.05 or 0.01). There were significant differences in hear and understand, retelling, say, read aloud, transcription, dictation, calculation after the treatment between two groups (t=2.510-10.298, all P<0.05). The score of satisfaction was (94.36 ± 5.55) in the experimental group and (88.25 ± 5.46) points in the control group, and there was significant difference (t=4.299, P<0.01). Conclusions Continuing language rehabilitation nursing can improve the recovery of the language function of the motor aphasia patients with a brain stroke.
2.Imaging follow-up study of disc degeneration and Modic changes caused by endplate fracture

Objective:To investigate the imaging follow-up value of discectomy and Modic changes caused by endolate fracture.Methods:Forty-six patients with end-plate fractures admitted to Changxing People′s Hospital during January to December 2016 were selected as subjects and followed up for 1 year. All patients underwent MRI examination to analyze the changes of Modic, disc degeneration and lumbar function recovery before and after follow-up.Results:A total of 230 lumbar intervertebral discs in 46 patients in this group had Modic changes. After 1 year′s follow-up, Modic changes occurred in 185 lumbar intervertebral discs in 46 patients. Modic changes occurred in L 5-S 1 [38.92%(72/185)] and L 4 - L 5 [40.0%(74/185)] segments. After 1 year′s follow-up, the rate of intervertebral disc 3-4 grade was higher than that before discharge [70.43%(162/230) vs. 60.43%(139/230)], the rate of intervertebral disc 6-8 grade was lower than that before discharge[8.70%(20/230) vs. 15.22%(35/230)], and the differences were statistically significant ( χ2 = 5.085, 4.646, P<0.05). After 1 year′s follow-up, the lumbar spine function score was significantly higher than that before discharge [(23.47 ± 2.86) scores vs. (9.15 ± 1.32) scores], and the difference was statistically significant ( P<0.05). Conclusions:Diffusion-weighted MR imaging has the characteristics of convenient operation, good repeatability and high accuracy. It is suitable for diagnosis and follow-up of disc degeneration and Modic change caused by endplate fracture. It is worthy of clinical promotion.
3.Isolation, identification and anticancer activity of an endophytic fungi from Juglans mandshurica.

The endophytic fungus named FSN006 was isolated from the inner bark of Juglans mandshurica. It grew quickly and formed circular colony on PDA plate. The upper side of the colony was white, while the lower side of the colony and the conditioned medium were light yellow as a result of significant yellow pigment substances were produced and secreted by the fungi. Green elliptic conidia appeared when cultured on CMX plate. Based on the morphology identification and ITS sequence, it was clear that this fungus belonged to the Deuteromycotina, HyPhomycetes, Moniliales, Trichoderma longibrachiatum. The conditioned medium of FSN006 showed a high anti-tumor ability against liver cancer cell-HepG2, and reached its IC50 concentration after being diluted 20 times, while the IC50 concentration of curcumine was(11.49 +/- 0.12) mg x L(-1). In addition, there was preeminent selective inhibiting effect against the normal liver cell strain HL-7702 and its caner counter strain HepG2. The inhibiting effect against strain HL-7702 was only one quarter of that against HepG2 at the concentration of IC50. Therefore, the fermentation of FSN006 may provide a possible way to produce anticancer drug with higher efficiency and lower toxicity.

4.ROBO3 deficiency promotes chemotherapy-induced transition of macrophage to foam cell

Objective To explore the effect of chemotherapeutic drugs doxorubicin or cisplatin on lipid metabolism of macrophages and its regulatory mechanism.Methods Macrophage RAW264.7 was treated with doxorubicin or cisplatin,and intracellular lipid level was detected by oil red O and ELISA;RNA sequence screen-ing and Western blot were used to confirm the changes of gene expression after chemotherapeutic drug treatment;The effects of silencing ROBO3 on cellular lipid metabolism were explored,and changes in key target genes of lipid metabolism were detected by Q-PCR and western blot.Results Adriamycin or cisplatin induced disturbances in macrophage cholesterol metabolism and exacerbated macrophage foaminess.Further studies showed that the expression of the axon guidance factor receptor,ROBO3,increased and then decreased during the chemotherapeutic drug-induced macrophage foaming process.Further intervention with ROBO3 exacerbates oxldl-induced cholesterol accumulation and foam formation in macrophages.Mechanistically,ROBO3 deficiency promotes the expression of cholesterol synthesis-related gene DHCR24 and inhibits the expression of cholesterol elimination-related gene ABCG1,resulting in cholesterol accumulation in macrophages.Conclusion This study found that ROBO3 plays an important regulatory role in the disruption of cholesterol metabolism and its foaming process in macrophages induced by chemotherapeutic drugs,which may provide new targets and ideas for the prevention and treatment of chemotherapy-related atherosclerosis.
5.Effects of indium exposure on relative content of mitochondrial ND1 gene in human peripheral blood lymphocytes in vitro.

OBJECTIVETo study the effects of indium exposure on the relative content of mitochondrial ND1 gene in lymphocytes.
METHODSVenous blood was obtained from 14 healthy workers and anticoagulated with heparin. Blood lymphocytes were separated and divided into three tube cultures. For two tubes in the exposed group, indium chloride was added to final concentrations of 0.2 mmol/L and 0.8 mmol/L, respectively. For one tube in the control group, an equal volume of normal saline solution was added. After incubation for 72 h, the relative content of mitochondrial gene in each group was determined using quantitative real-time PCR.
RESULTSLymphocytes exposed to 0.8 mmol/L indium chloride had a significantly higher relative content of mitochondrial gene than those exposed to 0.2 mmol/L indium chloride and those in the control group (P < 0.05, P < 0.05).
CONCLUSIONLymphocytes exposed to a high concentration of indium and its compounds have an elevated relative content of mitochondrial ND1 gene, indicating increased oxidative DNA damage induced by exposure to a high concentration of indium and its compounds.
