Objective To investigate the role of large conductance calcium-activated potassium channels (BK) in neuronal Ca2+ overload following traumatic brain injury (TBI). Methods Neuronal cells of C57BL/6 mouse cortex were collected and cultured. Patch-clamp technique was applied to investigate the changes of intracellular free calcium [Ca2+] i and firing frequency of neuronal action potentials in rest condition or evoked by 100 pA electric current lasting 500 ms after perfusion of Iberiotoxin ( 100 nmmol/L), a BK specific blocker. The cells were divided randomly into experimental group ( plus Iberiotoxin) and control group. Extracellular solution of cultured neurons was further perfused with KCl (20 mmol/L) to induce elevation of [Ca2 +]i and influence of Iberiotoxin ( 100 nmol/L) on amplitude of [Ca2 +] i elevation was determined. Results No significant changes of neuronal spontaneous action potential frequency and [Ca2 +]i were observed in rest condition after perfusion of Iberiotoxin (P>0.05).However, when evoked by electric current, the frequency of action potential was (10.4 ± 3.0) Hz,which was increased to ( 13.8 ± 3.7 ) Hz after perfusion of Iberiotoxin, with statistical difference (P<0.05 ). The [Ca2 +] i level was ( 14.21 ± 16.98 ) nmol/Lbefore perfusion with Iberiotoxin but was increased to (44.07 ± 34. 4) nmol/L after perfusion of Iberiotoxin (P < 0.05 ). Extracellular high concentration of KCl increased [Ca2 +] i of neurons, while perfudion of Iberiotoxin further elevated [Ca2 +]i (P < 0.05).Conclusion BK may play an important role in the regulation of neuronal [Ca2+] i and in neuronal Ca2+overload following TBI.

Objective To explore the application of auto-regressive integrated moving average (ARIMA) product season model in predicting number of inpatient and hospitalized expense of malignant tumor, and to provide scientific basis for hospital business management. Methods We collected inpatient data of malignant tumor from January 2007 to December 2015 in one hospital for model fitting, and used monthly data 2016 to verify the effect of model prediction. We predicted the number of inpatient and hospitalized expense of malignant tumor in 2017.Results ARIMA (0,1,1) (1,1,0) 12 was the best model for number of inpatient and hospitalized expense of malignant tumor, with prediction fitting errors of 1.1% and 1.47 %,respectively. The number of inpatient and hospitalized expense of malignant tumor in 2017 were predicted to be 7631 and 0.336 billion. Conclusion ARIMA product season model can better applied in the predicting of hospital business management.

Objective To investigate the effect of benazepril on expression of connective tissue growth factor(CTGF)gene in kidney of diabetic rats and renal lesion. Methods Forty Wistar rats were randomly divided into control group(CON),diabetic group(DM) and diabetic rats treated with benazepril(DM+B) group. After benazepril was orally administered(15 mg?kg~(-1)?d~(-1))to the diabetic rats for 12 weeks, serum creatinine(Scr) and blood urea nitrogen(BUN) levels, urinary protein excretion(UAER)and angiotensin Ⅱ(AngⅡ) contents in renal cortex were determined.CTGF mRNA expression in the renal cortex was detected by RT-PCR.Immunohistochemstry was used to examine the expressions of fibronectin(FN) and type Ⅳ collagen(Col Ⅳ) in glomeruli. Results At the end of the experiment,UAER in DM group(12.92?3.56 mg?24 h~(-1)) was significantly higher than that in CON group(2.74?1.09 mg?24 h~(-1))(P

Objective To evaluate the teaching activity on public health courses from clinical medical students in our university in order to provide a scientific basis for improving the curriculum design and teaching reform. Methods The “Questionnaire on Teaching Evaluation in Public Health Courses”, including teaching attitude, teaching content, teaching methods and teaching effectiveness was designed, and a general investigation was conducted among the clinical medical students of five-year program (840 students) and eight-year program (278 students) in these three aspects to under-stand students' evaluation to the course, who had finished the public health courses, including Preven-tive Medicine, Medical Statistics and Epidemiology (hereinafter referred to as: statistics, epidemiology, prevention) in Sun Yat-sen University. Statistical analysis was made using SPSS 13.0 software. Data analysis methods contain descriptive analysis, T-test, ANOVA, LSD, SNK, hierarchical logistic regres-sion analysis, etc. Results The overall score of teaching evaluation is (4.04±0.60). Differences exist between the evaluation in the five-year medical students and the eight-year medical students. The P values were 0.000 (Medical Statistics), 0.269 (Epidemiology), 0.047 (Preventive Medicine). The com-parison of scores among the four dimensions shows: Teaching effectiveness < Teaching methods

Objective To observe the changes in cytokines, oxygen free radicals, acidulous production in serum and to investigate the effectiveness of monitoring metabolism of cells and the method for evaluating cell injury in patients with severe trauma. Method The detailed data of 117 patients rescued and managed carefully from May 2005 to February 2007 were assessed and stratified with ISS and APACHE Ⅱ , and the serum levels of arterial blood lactate(ABL) ,base deficit(BD) ,superoxide(SOD) ,lipid peroxide(LPO) ,TNF-α and IL-6 were measured in real-time according to the condition of the patient. Results The monitoring biomarkers obviously changed with injury severity which endangered the situation of patients after trauma (P < 0.05) , especially in the patients with ischemia , hypoxia, shock, iniection, SIRS, and MODS (P < 0.01). The persistence of extremely elevated levels of biomarkers meant the organ failure and fatality of patients after trauma, and there was a obvious differece between those cases and cases without elevation of biomarkers ( P < 0.01). Conclusions The outcome of patients after trauma is closely correlated with injury severity, infection and MODS,and the levels of biomarkers including ABL, SOD, IPO, TNF-a and IL-6 are useful indicators of outcome measure.

Objective To investigate the effect of atorvastatin on the expression and activity of matrix metalloproteinases (MMPs) in rabbit abdominal arterial atherosclerotic plaque. Methods Eighteen male New Zealand rabbits weighing 2 kg were randomized to normal control group (n=6) and hypercholesterolemia group (n=12). The latter was given hypercholesterol diet for 2 weeks, and then catheter-induced abdominal aortic wall injury was performed. Rabbits in hypercholesterolemia and aortic injury group were randomized to model group (n=6,4 weeks of hypercholesterol diet) and atorvastatin(5 mg?kg-1?d-1 for 4 weeks)group (n=6). Finally, the levels of MMP2 and MMP9 protein and mRNA in the abdominal aortic artery were measured by immunohistochemical analysis, reverse transcription-polymerase chain reaction (RT-PCR), sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE) zymography. Results The intimal-medial thickness[(0. 49?0. 072) mm vs (0.66?0.079) mm, P
