1.The experience of diagnosis and treatment of 121 cases of laparoscopic appendectomy

Objective To explore the operation al skills of laparoscopic appendectomy(LA) and to improve its proficiency.Methods The clinical data of 121 patients under went LA during February 2013 to May 2014 were summarized retrospectively.Results All the 121 cases were completed successfully with no complication.Conclusion With the development of laparoscope and accumulation of experiences LA can be safe and complications can be minimized if the this operation skill being obtained.
2.Relationship between hemoglobin concentration, cytokine and cardiac function in anemic patients with ischemic cardiomyopathy

Objective To study the relationships between hemoglobin concentration,cytokine and cardiac function in patients with ischemic cardiomyopathy.Methods Hemoglobin concentration,sernm TNF-α,IL-6 in 121 patients with ischemic cardiomyopathy were measured,left ventricular ejection fraction(LVEF)by echocardiography,according to hemoglobin concentration,these patients were distributed to the anemic group and the non-anemic group,contrasting to 27 healthy persons.Results The levels of serum TNF-α,IL-6 in the anemic patients group were higher than those in the non-anemic group subjects,but LVEF Wag lower.With the increase of anemia severity,the levels of serum TNF-α,IL-6 significantly increased;there was a positive correlation between LVEF and the levels of serum TNF-α,IL-6(P<0.01),and a negative correlation between LVEF and Hb concentration.Conclusion The levels of cytokine promote the pathophysiological procedure between cardiac failure and anemic in ICM.
3.Cell suspension culture of Gentiana macrophylla(Ⅰ)

Objective To establish cell suspension culture system of Gentiana macrophylla.Methods Good cell line was selected by the methods of small cell aggregate and the effects of different media,inoculation age of cells,initial pH value,culture temperature,and rotation speed on cell growth and gentiopicroside accumulation were investigated.Results The eligible cell suspension culture conditions were:MS media as the basic medium,cells with inoculation age of 15 d,initial pH 7,and rotation speed of 110 r/min at 25 ℃,culturing in Erlenmeyer flasks.Conclusion Optimal cell suspension culture system of G.macrophylla has been established by shake flasks.The different effects of media on cell growth and gentiopicroside accumulation indicate that some inorganic ions have significant effects on regulation.
4.Cell suspension culture of Gentiana macrophylla(Ⅱ)

Objective To investigate the effects of carbohydrate and its concentration,nitrogen source,and metal ions on suspension cell growth of Gentiana macrophylla and gentiopicroside accumulation. Methods The cell dry weight and gentiopicroside content were measured under factors of different carbohydrate,NH4+/NO3-,Ca2+,and Zn2+ at various concentration. Results The optimum culture conditions are as follows:3% sucrose,NH4+/NO3-of 20/40,3 mmol/L Ca2+,0.007 49 mmol/L Zn2+. Conclusion Different carbohydrates and their concentration have significant effects on cell growth and gentiopicroside accumulation while high concentration of carbohydrates does not have such effect. NO3-is better than NH4+ for the cell growth. Ca2+ Remarkably influences both cell growth and gentiopicroside accumulation while Zn2+ contributes only to gentiopicroside content.
5.Influence of elicitors on growth and baicalin biosynthesis in hairy root of Scutellaria baicalensis

Objective To study the influence of elicitors on growth and baicalin biosynthesis in hairy root of Scutellaria baicalensis(HRSB).Methods The elicitors from the solution of Ca2+,Mg2+,Co2+ and fungal elicitors extracted from Aspergillus niger,A.oryzae,and Armillaria mellea were cocultivated with HRSB and the content of baicalin was determined by HPLC.Results Every elicitor has different influences on the growth and baicalin biosynthesis.When the concentration of A.niger was 40 mg/L,A.oryzae was 20 mg/L,and the Co2+ is 1.53?10-4 mmol/L,the content of baicalin increased respectively to 9.18%,8.81%,and 8.62%.Conclusion Elicitors could improve some specific secondary metabolic synthesis.The test implies that the elicitors of A.niger,A.oryzae,and Co2+ could effectively improve the synthesis of baicalin.
6.ENDOSCOPIC DRAINAGE IN ACUTE OR CHRONIC PANCREATITIS

Endoscopic drainage was practised in 23 patients with acute or chronic pancreatitis. Among them 13 cases with acute pancreatits (AP) including 5 serious ones were treated with endoscopic sphincterotomy (EST) and naso biliary drainage (ENBD).Following successful drainage ( n =11) all the patients were cured with an average of 10 4 days (5-18 days). Among 11 patients with chronic pancreatitis, there were 7 cases treated by endoscopic pancreatic sphinterotomy (EPS), 2 cases by extraction of pancreatic duct stones with balloon, 5 cases by dilatation of pancreatic duct with bougienage, 6 cases by endoscopic naso pancreatic drainage(ENPD), and 7 cases by pancreatic duct drainage(PDE). After the operation, abdominal pain was eliminated in 5/7 cases. Pancreatic duct stones were extracted out in 2 patients and pancreatic cyst disappeared in 4 patients. It is concluded that endoscopic drainage is safe and useful in management of acute or chronic pancreatitis.
7.Expression of Nupr1 in Gastric Cancer and its Clinical Significance

Background:The morbidity and mortality of gastric cancer remain high worldwide,and it is urgent to explore new targets for the diagnosis and treatment of gastric cancer. Nuclear protein 1 (Nupr1)has a variety of biological functions, especially in the tumorigenesis and development of the malignancies. Aims:To investigate the expression and clinical significance of Nupr1 in gastric cancer. Methods:Quantitative RT-PCR was used to determine the mRNA expression of Nupr1 in 72 cases of gastric cancerous tissue and the paired paracancerous tissue. Western blotting and immunohisto-chemistry were employed to detect the protein expression and cellular localization of Nupr1. The correlation of Nupr1 with the clinicopathological characteristics of gastric cancer was analyzed. Results:Expressions of Nupr1 mRNA and protein in gastric cancer were both significantly higher than those in paracancerous tissue (P < 0. 05),and their expressions were coincident. Nupr1 protein was expressed mainly in cytoplasm,and the nuclear expression was reduced. Expression of Nupr1 mRNA in gastric cancer was significantly correlated with tumor size,differentiation,pathological type,Bormann's classification,depth of invasion,lymph node and distant metastases and TNM staging (P < 0. 05),and was not correlated with gender,age and site of tumor (P > 0. 05). Conclusions:Nupr1 is overexpressed and correlated with invasion, metastasis and progression of gastric cancer. It can be used as a novel biomarker for early diagnosis and prognosis evaluation,and as a potential target for treatment of gastric cancer.

Objective: To investigate the non-specific and inaccurate amplification in cases of highly similar sequences among family members and the length heterogeneity of mature microRNA ( miRNA) ,and find a condition that discriminates maximally among similar miRNA family members and detects the accurate expression level of miRNAs. Methods: Primers with their mismatches and/or 3' end at different positions were designed. Amplification efficiencies were compared using matched and various mismatched primers by RNA-tailing and primer-extension RT-PCR at different annealing temperatures. Expression levels of several miRNAs in mouse brain were compared using miRNA specific primers with different termini. Results: Raising annealing temperatures 12℃-14℃above the T_m of the primers maximally increased amplification specificity without sacrificing sensitivity. Primers designed with their termini on or near variant positions could efficiently discriminate between miRNA isoforms. Using primers that terminated before the end of the mature miRNA did not miss the detection of shorter mature miRNA and provided accurate expression levels. Conclusion: Careful primer design and higher annealing temperature can increase specificity and accuracy of real time PCR miRNA detection.
10.Apoptin induces G_2-M arrest in cancer cells

Objective: To further study the effect of apoptin in inducing cancer cell specific apoptosis and the possible applications in cancer therapy. Methods: Apoptin gene was amplified by PCR and inserted into pcDNA3.1(+) with a FLAG tag in front of the multi-cloning-site. Apoptin gene with the FLAG tag was sub-cloned into an adenovirus vector. Several cancer cell lines were transfected with pcDNA3. 1/FLAG/apoptin or infected with apoptin containing recombinant adenoviruses to study the morphologic changes. Ad/apoptin infected cells were also analyzed by flowcytometry after staining with PI. Results: Expressed apoptin was localized in the nucleus of cancer cells. Chromatin condensation occurred 2 or 3 days after Ad/apoptin(+) infection. Cell number in G 2-M phase increased dramatically after Ad/apoptin(+) infection. Conclusion: Apoptin can induce cell cycle G 2-M arrest and chromatin condensation in cancer cells.
