1.The influence of dichloroacetate combined with cisplatin on the apoptosis of colorectal carcinoma cells HCT116

Objective To explore the effects of dichloroacetate (DCA) combined with cisplatin on the apoptosis of HCT116 and possible mechanisms.Methods The inhibitory effects of DCA and cisplatin alone or in combination on colorectal carcinoma cell line HCT116 were examined by MTT and Hoechst 33342 staining,the mitochondrial membrane potential changes were measured by Rodanmine123 staining under fluorescent microscope.The expression of bcl-2 was checked by qPCR.The activity of caspase-3 was assayed.Results DCA or cisplatin alone could inhibit the growth of HCT116 in a time and dose dependent manner.Compared with single drug treatment,there was significantly synergistic effect after treatment of DCA combined with cisplatin for 48 hours.Compared with the single drug treatment,the nuclear morphological changes such as chromatin condensation and fragmentation were more severe,and the mitochondrial transmembrane potential declines were markedly apparent for DCA + cisplatin group.The expression of bcl-2 gene in combination group was inhibited (P < 0.05),and the activity of caspase-3 significantly increased (P < 0.01).Conclusions DCA could inhibit the proliferation and induce the apoptosis of HCT116 cells in a time and dose dependent manner.The combination use of DCA and cisplatin has a synergistic effect on the biological action of HCT116.This may be attributed to lowering of mitochondrial transmembrane potential and the suppressed expression of bcl-2 gene.
2.Abnormal glycometabolism in tumor cells

The abnormal glucose metabolism of tumor cells is associated with a variety of mechanisms.Hypoxia inducible factor (HIF) is able to activate the glycolytic enzymes,which is conducive to getting energy through glycolysis.The dysfunction or the depletion in numbers of mitochondria can inhibit the oxidative phosphorylation pathway of glucose to some extent.The activation of oncogenes and the inactivation of tumor suppressor genes are also involved in the regulation of mitochondrial respiratory chain and glycolytic enzymes,thus affecting the process of glucose metabolism.Compared with normal cells,the synthesis of oxidative phosphorylation enzymes is inhibited in cancer cells.In addition,the abnormal glucose metabolism plays an important role in the growth,invasion and metastasis of tumor cells.
3.Glycolysis and tumor

Tumor cells obtain energy mainly by glycolysis to maintain growth and proliferation. It is related to the DNA mutations of mitochondria and anti-oncogene mutations. Inhibition of glycolysis can inhibit proliferation and kill tumor cells. The rate-limiting enzyme of glycolysis and hypoxia inducible factor may be the new targets for cancer therapy.
4.Interferon/poly (lactic-co-glycolic acid) and chitosan/poly (lactic-co-glycolic acid) composite membranes for prevention of epidural scar adhesion following laminectomy

BACKGROUND: Epidural resistance materials are an approach to prevent the epidural scar adhesion after laminectomy. A composite membrane of mechanical separation and fibroblast inhibition is promising to be widely studied.OBJECTIVE: To prepare the interferon (IFN)/poly (lactic-co-glycolic acid) (PLGA) and chitosan/PLGA composite membranes, and to observe their effects on preventing epidural scar adhesion after laminectomy in rabbits.METHODS: L2 laminectomy was performed in 120 rabbits, which were randomly and evenly divided into six groups. The control group received nothing on epidurals. In the autologous free fat group, the exposed dura was covered with subcutaneous fat. In the sodium hyaluronate group, 1 mL sodium hyaluronate was dripped onto the dura. In the PLGA, chitosan/PLGA, and IFN/PLGA groups, the exposed dura was covered with one of the three membranes at a sufficient size. The animals were sacrificed at 2, 4, 6, 8 weeks postoperatively. The scar formation and adhesion to the dura in L2 laminectomy sites were observed and scored by gross observation. At 4 weeks postoperatively, fibroblast ultrastructure was observed through the use of transmission electron microscope.RESULTS AND CONCLUSION: Widespread scars formed in the control group. The scars in the autologous free fat and PLGA groups were less compared with the control group, but there was no significant difference between these two groups. In the early stage, the scars in the sodium hyaluronate group were obviously less compared with the autologous free rat and PLGA groups, while in the later period, the difference between them was not significant, but the results from the sodium hyaluronate group were still better compared with the control group. The scars and slight adhesion to the dura were observed in the chitosan/PLGA and IFN/PLGA groups. The effect of preventing scar adhesion in these two groups was remarkably superior to that in the other groups, but there was no significant difference between these two composite membrane groups. At 4 weeks postoperatively, fibroblast morphology and function in the control, autologous free rat, and PLGA groups were better than those in the chitosan/PLGA and IFN/PLGA groups. Chitosan/PLGA and IFN/PLGA composite membranes are effective materials to prevent epidural scar adhesion after laminectomy.
5.Quality Standard for Zhitong Huazheng Dispersed Tablets

Objective To establish the quality standard for Zhitong Huazheng Dispersed Tablets.Methods Rhizoma corydalis,Radix astragali,Fructus toosendan in Zhitong Huazheng Dispersed Tablets were identified by TLC.The content of Danshensu was determined by HPLC.Results The spots on the TLC could be well separated and the method had strong specificity.The method for determinating the content of Dispersed tablets by HPLC was suitable for the quality standard with sufficient accuracy,stability and reappearance.The average recovery of Danshensu was 99.61%,RSD=1.60%(n =9).Conclusion The qualitative and quantitative analytic methods are stable and reliable.It can be used for quality control of Zhitong Huazheng Dispersed Tablets.
6.Molecular Recognition Properties of p-Phenylenediamine-Imprinted Polymers

Two p-phenylenediamine (p-PD)-imprinted polymers, P (MAA) and P (AA), were synthesized using methacrylic acid (MAA) and acrylamide(AA) as functional monomer, respectively, in order to prepare molecular recognition material with high selectivity for p-PD and explore the feasibility of methods such as molecular spectrometry and computational approach of quantum chemistry for the selection of functional mono mer with high imprinting efficiency.The molecular recognition properties of the imprinted polymers were evaluated by high performance liquid chromatography.The results indicated that P(AA) exhibited no imprint ing effect for p-PD, while P(MAA) can bind p-PD selectively(k' =3.57), which showed remarkable imprint ing effect (IF=2.95), and p-PD and its analogues o-phenylenediamine and p-aminobenzoic acid can almost realize baseline separation on P (MAA) column in the mobile phase of methanol.Furthermore, we made a comparative study on the interaction of p-PD with MAA and AA by spectroscopic techniques such as UV and fluorometry as well as HF/6-31G~* computational approach.The results demonstrated that the complex of p-PD-MAA was more stable than that of p-PD-AA, which can give a good explanation for the molecular recog nition properties of P (MAA) and P (AA).The study indicated that both molecular spectrometry (UV and fluorometry)and computational approach of quantum chemistry can be employed as efficient means for the selection of efficient functional monomer.The results showed that fluorometry is sensitive and convenient for the choice of functional monomer if the template molecule is fluorescent.
7.A comparative study of escitalopram angumentation with psychoanalysis in the treatment of refractory depression

Objective To investigate the efficacy of escitalopram combined with psychoanalysis in the treatment of refractory depression. Methods A total of 63 patients were randomly divided into escitalopram group ( n = 32) and escitalopram combined with psychoanalysis group( n = 31 ). All patients were evaluated with Hamilton depression Rating Scale(HAMD). Results After treatment,the scores of HAMD in two groups were both significantly lower than those before treatment. In the 8th ( ( HAMD ( 17.35 ± 2.98 ) ), 12th ( ( HAMD (9. 26 ±3.46) )weekend of treatment, the scores of HAMD in study group were significanlly lower than those in control group(8 th:21.97 ± 3.26; 12 th: 15.28 ± 3. 18 ). There were no significant differences in side effects between study group and control group. Conclusion Escitalopram angumented with psychoanalysis takes effects better than escitalopram single and doesn't increase side effects in the treatment of refractory depression.
8.The diagnostic value of MR perfusion weighted imaging on complications and tumor recurrence in hypopharyngeal and laryngeal cancer after operation and radiotherapy

Objective To explore diagnostic value of MR perfusion weighted imaging(PWI)on complications and tumor recurrence in hypopharyngeal and laryngeal cancer after operation and radiotherapy. Methods MR PWI was performed in 31 patients with hypopharyngeal and laryngeal cancer who suffer from tumor recurrence, granulation or infection after operation and radiotherapy. Blood flow (BF), blood volume(BV), mean transit time(MTT) and time to peak (TTP) values were measured in the lesions. Analysis of variance and pair-wise comparison of q test were used for statistical analysis. Results Eleven cases with tumor recurrence,11 case with granulation tissues, 9 cases with infection were confirmed by pathological examinations. Average BF, BV, MTT and TTP values of recurrent tumors were (145.1 ± 29.3)ml · 100 g-1 · min-1,(14.2 ± 3.2)ml · 100 g-1,(5.4 ± 0.6)s,(13.7 ± 1.3)s respectively;Average BF, BV MTT and TTP values of granulation tissues were(109.1±27.4)ml·100 g-1·min-1,(11.1±1.7)ml·100 g-1,(7.7±1.0)s, (19.8 ± 2.1)s respectively;Average BF, BV, MTT and TTP values of infectious tissues were(86.9 ± 7.7)ml · 100 g-1·min-1,(8.8±1.0)ml·100 g-1,(8.0±0.9)s,(19.7±1.3)s respectively. Average BF and BV values of the recurrent tumors group were higher than those of the granulation(q=4.89, 4.64 respectively,P<0.01)and infection group(q=7.52,7.71 respectively,P<0.01)respectively. Average BV values of granulation group was higher than that of the infection group(q=3.31,P<0.05), Average MTT and TTP values of recurrent tumor group were lower than those of the granulation (q=9.38, 12.48 respectively,P<0.01)and infection group(q=9.77, 11.53 respectively,P<0.05). There were no significant difference in average BF, MTT and TTP between the granulation and infection group. Conclusion MR PWI can be helpful in the diagnosis of complications and tumor recurrence in hypopharyngeal and laryngeal cancer after operation and radiotherapy.
9.Analyzing servicing and studying on zero spot in H-300 transmission electron microscope

In this paper,we present analysis,servi-ce and study for zero grating in H-300.Transmission electron microscope made in Japan.It is well known that this kind of electron microscope have been used in most university and institute over twenty years, so all most of the electron microscope have problem in its dependability.In order to analyze and service the electron microscope,we make much efforts in the material chosen and the service technics,the experimental results show that our methods are available for servicing those electron microscope. Moreover,we discuss the failure of the electron microscope and the jointing crafts,these results are useful for those who works in this field.
10.Changes in bone mineral density and bone turnover in postmenopausal osteoporotic patients treated with salmon calcitonin nasal spray

Objective To study the changes of bone mineral density(BMD)and bone turnover in postmenopausal osteoporotic patients treated with salmon calcitonin nasal spray. Methods Sixty-seven postmenopausal osteoporotic patients were enrolled in our trial. All of them received calcium and vitamin D; 37patients were treated with salmon calcitonin nasal spray for 12 months and the other 30 patients received calcium and vitamin D only. Dual-energy X-ray absorptiometry(DEXA)and measurements of a series of bone turnover indices were performed before and after medication for 6 and 12 months. Results After treatment with salmon calcitonin nasal spray for6 months, BMD in lumbar spine 2-4 increased but no change occurred in femoral neck. However, after treatment for 12 months, BMD in both lumbar spine 2-4 and femoral neck increased. In the control group, BMD in lumbar spine 2-4 decreased after treatment for 6 and 12 months, but BMD in femoral neck decreased only after 12months. Comparing with the control group, after treatment with salmon calcitonin nasal spray, BMD in lumbar spine 2-4 and femoral neck were increased obviously. The level of TRACP-5b and NTX/Cr decreased after treatment with salmon calcitonin nasal spray for6 months and 12 months, while BALP increased only after treatment for 12 months. In the control group, BALP decreased after treatment for 12 months. The level of 25-(OH)vitamin D increased after treatment for 6 months and 12 months in both groups. Conclusions Long-term treatment with salmon calcitonin nasal spray prevents bone loss and may increase bone mass.
