1.The expression of high mobility group box-1 in patients with acute coronary syndrome and the treatment of atorvastatin

Objective To investigate the expressions of high mobility group box-1(HMGB1) and high sensitivity C-re?active protein (hs-CRP) in patients with acute coronary syndrome (ACS) and the effects of atorvastatin on the two inflamma?tory cytokines. Methods A total of 90 patients with ACS and 90 cases of normal control subjects were selected in this study. The serum concentrations of HMGB1 and hs-CRP were measured before treatment in patients of ACS. Patients were randomly divided into two groups:control group (n=45) and atorvastatin group (n=45). Atorvastatin was given 20 mg/24 h and 40 mg/24 h. Blood samples were obtained from the patients for detection of HMGB1 and hs-CRP one week after treatment with atorvastatin. Results There were significantly higher serum levels of HMGB1 and hs-CRP in patients with ACS than those of control subjects (P<0.01). The level of HMGB1 was positively correlated with the level of hs-CRP in patients of ACS (r=0.389, P<0.01). Before treatment, there were no significant diffferences in level of HMGB1 and hs-CRP in patients with ACS between the two groups. After treatment with atorvastatin, the levels of HMGB1 and hs-CRP were decreased in the two groups of ACS, and those were significantly lower in the intensive group than the standard group (P<0.05). Conclu?sion HMGB1 could stimulate the secretion of hs-CRP and other inflammatory cytokines, playing an important role in the process of occurrence and development of atherosclerosis. High loading dose of atorvastatin may reduce the expression of HMGB1 and decrease the inflammation, and stabilize the plaques in patients with acute coronary syndrome.
2.Liraglutide attenuates myocardial ischemia/reperfusion injury possibly through reducing cardiomyocytes apoptosis and oxidation in rats

Objective To explore the effect of liraglutide on myocardial ischemia/reperfusion (I/R) injury in rats and related mechanisms.Methods Twenty-four male SD rats were divided into sham group,I/R injury group,and liraglutide group by table of random number (n =8 each).Myocardial I/R injury model was established by occlusion of the left anterior descending artery for 30 min followed by 2 h of reperfusion.HE stain method was used to observe cardiomyocyte status under light microscope,myocardial tissue samples were stained with TTC to measure the myocardial infarction size,protein expression of p53 and caspase-3 was analyzed by immunohistochemical technique and Western blot respectively,xanthine oxidase method was used to detect SOD activity,Thiobarbituric acid method was used to measure the concentration of MDA.Results Compared with the I/R group,the degree of myocardial damage of liraglutide group was significantly reduced and the myocardial infarct area was significantly lower ((44 ± 8) % vs.(62 ± 8) %,P < 0.05) while protein expression of caspase-3 and p53 in liraglutide group was significantly downregulated (0.19 ±0.03 vs.0.24 ± 0.02 and 0.27 ± 0.03 vs.0.39 ± 0.04,P < 0.05).SOD activity was significantly increased and MDA concentration was significantly decreased (74.20 ± 11.10 vs.44.04 ± 14.30 and 4.41 ± 1.07 vs.8.72 ± 2.20,P < 0.05) in liraglutide group compared to I/R group.Conclusion Liraglutide protects myocardium against I/R injury possibly through reducing cardiomyocytes apoptosis and oxidation.
3.Study on 27 Serum Cytokines in EV71-induced Hand Foot and Mouth Disease in Jinan

Objective To detect and analyze the levels of 27 serum cytokines expression in EV71-induced Hand Foot and Mouth Disease(HFMD) in Jinan and revealed their correlations with disease severity based on the platform of liquid chip.Methods 43 serum samples were collected from EV71-infected HFMD patients in Jinan in 2009-2014,including 16 mild cases and 27 severe cases.10 serum specimens from healthy people were also collected as controls.27 serum cytokines were analyzed on Bio-Plex liquid chip platform.Results Compared to healthy controls,22 cytokines expression levels increased in severe groups,including IL-1 ra,IL-2,IL-4,IL-5,IL-6,IL-7,IL-8,IL-9,IL-10,IL-12 (p70),IL-13,IL-15,IL-17,etotaxin,G-CSF,IFN-γ,IP-10,MCP-1,PDGF-BB,MIP-1β,RANTES and TNF-α(P < 0.02).21 cytokines expression levels elevated than healthy controls (P < 0.02).GM-CSF was found decreased levels in both mild and severe groups than in the healthy controls(P < 0.02).The correlation analysis showed that 12 cytokines (IL-1 ra,IL-7,IL-9,IL-10,IL-13,IL-17,etotaxin,IP-10,PDGF-BB,MIP-1β,RANTES and TNF-α) were moderately correlated with the severity of EV71-infected HFMD.GM-CSF was negatively correlated with HFMD.Conclusions Many kinds of serum cytokine changed obviously in the EV71-infected HFMD patients and cytokine levels were closely associated with the severity of HFMD.The analysis of liquid chip for serum cytokines provides an efficiently method.
