1.Protective Effect of 3-hydroxybutyrate Derivate on the Apoptosis of PC12

Objective To investigate the protective effect of 3-hydroxybutyrate derivate( methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate)on the apoptosis of PC12 induced by FBS deficiency. Methods Methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate were prepared by chemical degradation. PC12 cells were divided into different groups:medium with FBS as negative control,medium without FBS as positive control,medium with different concentrations of methyl-3-hydroxybutyrate or ethyl-3-hydroxybutyrate without FBS as sample groups. The shape and viability of cells in each group were analyzed by optical microscope and MTT. Results Compared with the negative control,cells in the positive control group demonstrated a large number of apoptosis,smaller and thinner morphology,and lower activity. However,the activity of cells was improved in the sample groups.Low concentration(0. 01 and 0. 001 mg·mL-1)of methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate showed a protective effect on cell apoptosis,and 1 mg · mL-1 had the best protection effect. Conclusion High purity methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate can be produced by chemical method,and both chemicals present good effect on protecting the PC12 from apoptosis.
2.Plasma leakage monitoring for the blood circle treatment system

This paper introduces the working principle of the blood circle treatment system outside the body. A monitoring instrument of plasma leakage suited for the blood circle treatment system outside the body is developed based on the absorption spectrum experiments of plasma leakage. Photoelectric detection technology and virtual instrumentation are utilized in the development. A series of detecting experiments of waste solution containing plasma with continuously changing concentration show the monitoring system possesses a relatively high sensitivity. Moreover, the experiments of continual detection with plasma concentration at one certain point indicate the monitoring system has a quite good stability. The monitoring instrument is adapted to dynamically detecting the plasma leakage when the blood circle treatment system outside the body is working.

4.Correlation between domestic violence in pregnancy and the levels of plasma amino acids and cortisol in the neonates

Objective To explore the correlation between domestic violence in pregnancy and plasma glutamate(Glu),γ-aminobutyric acid(GABA)and coaisol levels in neonatels.Methods By crosssectional survey and domestic violence(DV)interview with Abuse Assessment Survey(AAS),56 abused pregnant women were screened and 107 non-abused pregnant women served as controls.Blood from umbilical vein of neonatal were collected immediately after delivery.Levels of Glu,GABA in plasma were assayed by using high performance liquid chromatography with electrochemical detection on samples.Levels of plasma cortisol were detecteded by with immunoradiometric assay.All variables of the two groups were compared.Chi-square,t/z-test,Spearman correlation,liner regression analysis were adopted.Results In the DV group,there were 49 women(87.5%)experiencing psychological abuse and 2(3.6%)physical abuse during pregnancy.Moreover,29 women(51.8%)experienced sexual abuse in the period of prognancy.By comparison,our study showed higher levels of plasma Glu,GABA and cortisol in neonates of the DV group than those of the controls[(1509±339)pmol/L vs(811±270)pmol/L,(1460±369)pmol/L vs (707±296)pmol/L,(491±87)μg/L vs(392±108)μg/L,t=14.326,t=14.138,t=5.916,P<0.001]. Spearman correlation analysis showed significantly positive correlation between severity of psychological abuse and levels of Glu,GABA and cortisol(r=0.705,0.696,0.425,P<0.01)and positive correlation between the severity of sexual abuse and levels of Glu,GABA and cortisol(r=0.471,0.424,0.274,P<0.01).Conclusion Domestic violence in pregnancy may alter the levels of some excitatory and inhibitory amino acids and change the endocrinic function in neonatels.

6.Expressions of Protease Activated Receptors on Human monocyte.

Objective To explore the expression of protease activated receptors (PARs) on human monocytes. Methods Flow cytometry and RT - PCR was used to detect the expressions of PARs on human monocytes that purified by MACS. Results FACS analysis showed that monocytes expressed PAR - 1 , PAR - 3 , PAR - 4, but not PAR - 2.RT - PCR revealed that monocytes expressed PAR - 1 and PAR -3, but not PAR -2 and PAR -4 mRNA. Conclusion PARs is expressed on human monocyte cells,which may facilitate further investigation of the potential functions of PARs on monocytes.
