1.Important trials of targeted therapy in patients with advanced gastric carcinoma

The conventional treatment options for advanced gastric carcinoma patients remain unsatisfactory in terms of response rate and overall survival benefit. Targeted therapy plus systemic chemotherapy now becomes a new promising therapeutic option. In this review we profiled important trials of international multicenter phase Ⅲ clinical studies in patients with advanced gastric carcinoma, including ToCA trial, EXPAND trial, LOGIC trial, AVAGAST trial, REAL3 trial, and granite-1 trial. These trials warrant the role of molecular targeted therapy in patients with advanced gastric carcinoma.
2.Sompatible Stability of Cefoperazone Sodium/Sulbactam Sodium for Injection Mixing with Doxofylline for Injection

OBJECTIVE:To study the compatible stability of doxofylline for injection with cefoperazone sodium/sulbactam sodium for Injection. METHODS:The contents of 2 constituents in the mixed solution stored at (25?1)℃ within 6 hours were determined by dual ultraviolet wavelength spectrophotometry,and the appearance and pH value of the mixture were compared with those before mixing. RESULTS:There was no significant change in the content of doxofylline after mixing,while the contents of cefoperazone sodium and sulbactam sodium showed a slight reduction. No significant change was noted for other indexes. CONCLUSION:Doxofylline injection is compatible with cefoperazone sodium and sulbactam sodium for injection in 0.9% sodium chloride injection at 25℃ within 4 hours' mixing.

4.Expression of surface antigen of hepatitis B virus in tobacco

Objective Plant vaccine is a new kind of vaccine with good safety profiles, low cost and convenience. Our purpose was to establish a good plant system to express HBsAg and pave the way for further research on plant vaccine. Methods Transfer HBV s gene into binary vector pCAMBIA1301 which had plant promoter and hygromycin resistant gene. The recombinant vector was transferred into agrobacterium LBA4404. Then we used agrobacterium to infect tobacco. Results The GUS staining result of transformant leaves showed blue, which meant the target gene had been transferred into tobacco and expressed HBsAg seccessfully. We confirmed the insertion of S gene by PCR and its expression further by ELISA and Western blot. Conclusions HBsAg can be expressed successfully in tobacco.

6.Analysis of indicators in 2 521 patients before blood transfusion

Objective To investigate the infection status of infectious diseases for 2 521 patients before blood transfusion in a Hospital in Changsha.Methods A total of 2 521 patients who would be transfused were selected,and six kind of serum hepatitis B virus indicators,hepatitis C virus antibody (antiHCV),human immunodeficiency virus antibody (anti-HIV/1 + 2),and treponema pallidum antibody (antiTP) of nine common infectious disease targets were detected with enzyme-linked immunosorbent assay (ELISA).Results Among 2 521 patients,HBsAg-positive cases were 8.33%,anti-HCV positive were 0.59%,anti-HIV positive [confirmed by the Provincial Center for Disease Control (CDC)] was 6 cases,and TP-positive were 2.301%.A total of 289 patients were tested positively,with a total positive rate of 11.46%.Conclusions Detection before transfusion may reduce infection risk and decrease the risk of occupational exposure,strengthen medical staff self-protection,and reduce medical malpractice caused by blood transfusion.
7.Drug Release Characteristics of Mu'an-Eye-Gel in Vitro

OBJECTIVE:To compare home-made mu'an-eye-gel(acyclovir plus honey) with commercial aciclovir(ACV)-eye-gel in releasing drug characteristics in vitro.METHODS:The in vitro drug release test was conducted by the third method of dissolution determination stated in Chinese Pharmacopeia together with bag filler method.The cumulative drug-releasing percentage and the acyclovir amount in mu'an-eye-gel versus ACV-eye-gel were determined by UV spec-trophotometry,and the accumulative releasing drug percentages of the two preparations were computed and their drug release behaviors w ere compared.RESULTS:The in vitro releasing behaviors of mu'an-eye-gel followed the Weibull kinetic equa-tion,however the vitro releasing behavior of commercial ACV-eye-gel followed the zero order kinetic equation,and the T80%and Q8 h had statistical significances between(mu'an-eye-gel:T80%=3.156?0.013(h),Q8 h=93.28?0.010(%);ACV-eye-gel:T80%=10.16?0.009(h),Q8 h=67.85?0.025(%)) 2 kinds of preparation.CONCLUSION:Mu'an-eye-gel is superior to the commercial ACV ophthalmic gel in both releasing velocity and accumulative drug release percentage.
8.Activated Notch1 signaling inhibits growth of EC109 cell line and its mechanism

Background and purpose: It has been reported that activation of Notch1 could strongly inhibit proliferation of HPV (human papilloma virus)-positive HeLa cells by down-regulation of the E6 and E7 genes. The aim of this paper was to investigate the role of the Notch signaling pathway in growth arrest of EC109 cells in vitro and the molecular mechanism. Methods: EC109 cell lines, a well differentiated human ESCC (esophageal squamous cell carcinoma) cell line with HPV18-positive, was used in the study. Exogenous intracellular domain of Notch1(ICN) was transfected into cultured EC109 cells by lipofectamine transfection, the proliferation of the transfected cells was measured by an MTT assay. Cell cycle distribution was analyzed by flow cytometry. Human papilloma virus type 18 (HPV18) E6/E7 mRNA expression was detected by RT-PCR, and p53 protein expression was detected by Western blot.Results: Activation of Notchl signaling resulted in inhibition of EC109 cell proliferation with the induction of G_2/ M arrest. There was a significant difference in terms of the percentage of G_2/M phase cells among the ICN-transfected group (42.57±1.57)% and the non-transfected group (1.88±0.66)% or the empty plasmid transfected group (1.99±1.02)% (P<0.01). Down modulation of HPV18 E6/E7 gene expression and upregulation of p53 expression was (2.15±0.23) in ICN-transfected group higher than non- transfected group (0.45±0.07) and empty plasmid transfected group (0.46±0.02) (P<0.01). Conclusion: Repression of HPV18 E6/E7 expression by Notch1 signaling results in growth suppression of HPV18-positive EC109 cells with concomitant activation of p53-mediated pathways.
9.The protective effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats

Objective To evaluate the effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats, and to investigate the possible mechanism. Methods Thirty male Sprague-Dawley ratswere randomly divided into 5 groups (n = 6 each): sham group (group S), I/R group (group I/R), low dose of sufentanil postconditioning group (group SL), high dose of sufentanil postconditioning group (group SH) andsufentanil postconditioning plus wortmannin group (group WM). Different drugs were injected before reperfusion: normal saline (NS) and sufentanil 1 μg/kg in group SL, NS and sufentanil 3 μg /kg in group SH, wortmannin 15 μg/kg andsufentanil 1 μg/kgin group WM, and NS in group S and I/R. At the end of reperfusion, artery blood was collected for assessment of plasma cTnI concentration; And the heart was harvested for determination of infarct size (IS) and area at risk (AAR). Results Compared with group S, cTnI concentration was increasedin the rest groups (P< 0.01); Compared with group I/R, cTnI concentration was decreased in group SL, SH and WM, while IS/AAR reduced in group SL and SH (P < 0.01); Compared with group SL, cTnI concentration and IS/AAR were decreased in group SH while increased in group WM (P <0.01). Conclusion Sufentanil postconditioning could attenuate myocardial ischemia-reperfusion injury in rats, and the mechanism involved PI3K related pathway.

Bone is the most common metastatic site of breast cancer . And breast cancer metastasis to bone can lead to osteoporosis , hypercalcemia , spinal cord compression and pathological fracture , which seriously impact the patientsˊ quality of life and survival . Chinese herbs have a potential advantage in the treatment of breast cancer metastasis to bone. In recent ten years, a number of studies showed that Chinese medicine monomers, traditional Chinese medicine (TCM) extracts, and Chinese herbal compound prescriptions played a role in in-hibiting breast cancer cell proliferation , invading and relieving the bone destruction and bone pain by different molecular mechanisms .
