Objective To study the relationship among leptin, neuropeptide Y and plasma lipid in the patients with essential hypertension. Methods In 116 patients with essential hypertension (EH) and 26 healthy subjects, the plasma concentrations of leptin(LE), neuropeptide Y(NPY), total cholesterol(TC), triglyceride(TG) and high density lipoprotein cholesterol(HDL) were determined using RIA. 3.Influence of different root exposure in long-term effect of crown lengthening

Objective To study the influence of different root exposure in long-term effect of crown lengthening .Methods To-tally 142 teeths(66 of the front teeths ,46 of premolars teeths ,30 of molars teeths ) of 110 patients collected from August 2010 to October 2012 in stomatological hospital to affiliated zunyi medical college were chosen as research subjects ,then they were randomly divided into the experimental group and the control group .There were 33 anterior teeths ,23 premolar teeths and 15 molar teeths in each group .The experimental group accepted 3 mm root exposure while the control group accepted 4 mm root exposure .The post-operative effect of the two groups was compared .Results The tooth mobility of the anterior cases of control group after 3 months was statistically higher than that of the experimental group (P<0 .05) and the difference of tooth mobility between the two groups in other teeths and times was not statistically significant (P>0 .05) .The bleeding index after 3 months ,the anterior teeths ,premo-lar teeths and molar teeths of the experimental group was statistically higher than that of the control group (P<0 .05) ,while the difference of tooth mobility between the two groups in other times was not statistically significant (P>0 .05) .The postoperative probing difference of depth and attachment loss between the two groups showed no statistically significant (P>0 .05) .Conclusion The root exposure of 3 mm and 4 mm both have a significant effect in crown lengthening .But for the aesthetic point of view ,3 mm should be taken as the root exposure of the anterior cases .
4.Construction and identification of recombinant human cytomegalovirus with ganciclovir-resistance associated mutations in UL97 gene

Objective To construct drug-resistant variant recombinant human cytomegalovirus (rHCMV)and identify drug susceptibility by phenotypic and genotypic assays.Methods The UL97 fragments containing Pine I recongnition site and resistant mutation were introduced by site-directed mutagenesis using gene splicing by overlap extension polymerase chain reaction(PCR),and blended with human cytomegalo-virus(HCMV)standard strain ADl 69 genomic DNA proportionally,then the DNA mixture were transfected into MRC-5 fibroblasts by the vector of liposomes.HCMV-PP65 antigen was detected by indirect-immunofluorescent assay to verify rHCMV infection of MRC-5 fibroblasts.When the eytopathic effect(CPE)of homologous recombinant virus reached 100%,the virus was harvested.The purified target virus was screened by plague with different concentrations of ganciclovir(GCV)and the recombinant virus was identified by plague reduction test and DNA sequencing of drug-resistant genes(UL97 and UL54).Results The UL97 fragments containing intended mutations for transfection were constructed successfully.After cotransfected with AD169DNA mixture for 7 days,rHCMV formed cytopathology was obviously visible,which was verified as rHCMV infected focus by HCMV-PP65 antigen test.The UL97 genotypic analysis of recombinant virus obtained by cloning was as expected.No mutation was found by UL54 gene sequencing.The GCV susceptibility of rHCMV positive clone was 15 μmol/L (50% inhibiting concentration),which was 12-fold of standard AD169 strain and was drug-resistant phenotype.Conclusions The rHCMV containing intended mutations is constructed successfully by cotransfeetion into MRC-5 cells and the recombinant virus strain is obtained by GCV screening and plaque purifying.The establishment of this method provides technique platform for identifications of new drug-resistant mutations of HCMV during anti-viral therapy.
5.Seroreversion of serological tests for syphilis in neonates born to mothers with syphilis: a follow-up study

Objective To observe the seroreversion of serological tests for syphilis in neonates born to mothers with syphilis and to evaluate the influence of antisyphilitic treatment on the time of seroreversion in uninfected neonates.Methods A total of 115 neonates born to mothers with syphilis were recruited in this study,and underwent toluidine red unheated serum test (TRUST),Treponema pallidum particle assay (TPPA)and 19S IgM-TPPA at birth.Follow up was scheduled at 1,3,6,9,12,18 and 24 months of age,and cancelled as soon as both TRUST and TPPA became negative during the 24 months.Both TRUST and TPPA were carried out at each follow-up visit.Kaplan-Meiter survival analysis and Wilcoxon rank-sum test were conducted to assess these data by using SPSS 13.0 software.Results At birth,TPPA was positive in all (100%) of the neonates,TRUST in 97 neonates (84.3%) with the titer ranging from 1 ∶ 1 to 1 ∶ 256,19S IgM-TPPA in 3 neonates who were diagnosed with congenital syphilis (CS).CS was ruled out in the other 112 neonates with negative 19S IgM-TPPA after follow up.Of the 3 neonates with CS,all became negative for TRUST within 12-18 months after birth,2 remained positive for TPPA at 24 months of age,1 remained positive for TPPA at 20 months of age,and all were still followed at the time of this writing.The cumulative seroreversion rate was 82.2%,98.9% and 100%for TRUST at 0-3,4-6 and 7-9 months of age,respectively in 90 uninfected neonates,3.1%,30.2%,85.4% and 100% for TPPA at 0-3,4-6,7-9 and 10-12 months of age,respectively in 96 uninfected neonates.Of 16 neonates who missed some follow-up visits,all turned negative for TRUST and TPPA within 15 months after birth.As far as the TP-seropositive uninfected neonates were concerned,no statistical differences were observed in the seroreversion time of TRUST or TPPA between neonates receiving antisyphilitic treatment and those without antisyphilitic treatment (x2 =0.54,2.41,respectively,both P > 0.05).The seroreversion of TRUST occurred earlier than that of TPPA (Z =10.45,P < 0.01).Conclusions Most uninfected neonates born to syphilitic mothers turn negative for TRUST within 6 months after birth,and for TPPA within 12 months.And the seroreversion of TRUST usually occurs earlier than that of TPPA.Antisyphilitic treatment shows no significant influence on the seroreversion time of TRUST or TPPA in these seropositive uninfected infants.
6.The changes of expression of neuropeptide Y and oxytocin in the plasma of first-episode schizophrenia patients

Objective To investigate the clinical significance of plasma neuropeptide Y (NPY)and oxytocin (OXT) and the mRNA level of OXT gene in first-episode schizophrenia (FES) patients and healthy controls (HC).Methods The plasma concentration of NPY and OXT in 55 FES patients and 33 HC were measured by enzyme linked immunosorbent assay (ELISA) and the mRNA level of OXT gene were performed by real-time quantitative polymerase chain reaction (RT-qPCR).Results The NPY levels in FES patients ((1386.88±254.57) pg/ml) were significantly higher than that in HC ((1 140.62±266.63)pg/ml)(P<0.01),and both the males and females in FES patients ((1 489.97±231.06)pg/ml and (1 279.97±236.30) pg/ml) were higher than their counterparts in HC ((1 305.40 ± 238.80) pg/ml and (965.55 ±165.45) pg/ml) (P<0.05,P<0.01).The OXT level in FES patients ((553.26± 180.49) pg/ml) was lower than that in HC ((696.27±280.77) pg/ml) with significant difference (P<0.05),especially the females FES patients ((597.49±178.63)pg/ml vs (785.51±329.60)pg/ml);but the mRNA expression of OXT gene in FES patients (0.2075 (0.1653,0.3388)) were significantly higher than that in HC (0.1615 (0.1321,0.2200)) (P<0.05),and there were no differences between the females of the two groups (P>0.05).Conclusion The FES patients presented abnormal expression of NPY and OXT,and the concentration changes of plasma NPY and OXT will provide referential significance for the diagnosis of schizophrenia(SZ).
7.Clinical application and prospect of SBRT in treatment of oligo-metastatic NSCLC

The rapid progress on immunotherapy and targeted therapy has brought long-term survival benefits for locally advanced non-small cell lung cancer (NSCLC). The oncology community has also paid more attention to the local treatment for advanced NSCLC, especially for patients with limited metastatic lesions, also known as oligo-metastasis. Many studies have reported that oligo-metastatic NSCLC patients could benefit from the combination of local and systematic treatment, and even to be cured. In recent years, with the advances in radiation technology, stereotactic body radiation therapy (SBRT) has achieved precise high-dose radiotherapy for small target tumors. Currently, SBRT has been widely applied in the treatment of inoperable early lung cancer, and its application value and safety in patients with advanced lung cancer are also being actively explored. In this article, the research status, progress and future development direction of SBRT in the treatment of oligo-metastatic NSCLC were discussed.
