Objective To investigate the effects of ulinastatin on the expression of NF-κB in lung tissues during limb ischemia-reperfusion(I/R) in rats. Methods Forty-eight male SD rats weighing 230-260 g were randomly divided into 3 groups (n=16 each):sham operation group (group S);I/R group; ulinastatin group (group U).A rat model of lung injury induced by I/R of hind limbs which was produced by occlusion of the bilateral femoral arteries for 2 h followed by reperfusion was established. The rats were sacrificed at 2 and 4 h of reperfusion(8 rats at each time point) and the lung tissues removed for determination of NF-κB expression (by immuno-histochemistry) and microscopic examination.W/D lung weight ratio was calculated. Results W/D lung weight ratio was significantly increased and NF-κB expression was up-regulated in group I/R compared with group S(P< 0.05). W/D lung weight ratio was significantly decreased and NF-κB expression was down-regulated in group U compared with group I/R (P<0.05). The lung injury induced by I/R was reduced in group U compared with group I/R. Conclusion Ulinastatin can reduce the lung injury induced by limb I/R by down-regulating the expression of NF-κB in rat lung tissues.

Nitrogen source is one of the important factors that affect the microalgae growth and lipid accumulation. We studied the effects of various nitrogen sources (i.e. NaNO3, CO(NH2)2, NH4Cl and CH3COONH4) and amount on the growth density, lipid yield, and eicosapentaenoic acid (EPA) content of Nannochloropsis oculata by single factor experimental method. The results show that N. oculata preferred NH4+ as nitrogen source rather than NO3- and CO(NH2)2. NH4+ could promote the growth and lipid accumulation of N. oculata. With the increase of nitrogen concentration, the biomass and the content of polyunsaturated fatty acid (PUFA) increased, but the content of lipid decreased. CH3COONH4 was the most suitable for growth, accumulation of lipid and EPA of N. oculata among the four investigated nitrogen sources. The optimal concentration was 5.29 mmol/L.

A TurboFlow ( TF ) online purification-ultra performance liquid chromatography-tandem mass spectrometric method was developed for the determination of nitroimidazoles and their metabolites ( ornidazole, IPZ, MNZ, MNZOH, DMZ, RNZ, IPZOH and HMMNI) in honey. The honey sample was extracted by H2 O containing 0. 1% formic acid solution. Then, the extraction solution was analyzed by TF-UPLC-MS/MS. The main factors influencing the purification efficiency including TF column, mobile phases and elution solutions were optimized. The compounds were detected by selective reaction monitoring ( SRM) in positive ion mode electrospray ionization ( ESI+) . The linear range of the method ranged from 0 . 1 to 50μg/L for nitroimidazoles and their metabolites, with the correlation coefficient ( R2 ) of over 0. 997. The limits of quantification were 0. 1 μg/kg for ornidazole, IPZ, 0. 2 μg/kg for MNZ, MNZOH, DMZ, RNZ and IPZOH, and 1. 0 μg/kg for HMMNI, respectively. The recoveries were in the range of 73. 7% to 116. 4% at four spiked levels with the relative standard deviations ranged from 1. 1% to 9. 1% in honey samples. The results indicate that the developed method is simple, efficient and precise, and can be used for the determination of nitroimidazoles and their metabolites in the actual honey samples.
