1.Influence of esculentoside A on proliferation and CDK2 of IL-1βinduced glomerular mesangial cell *

Objective To observe the effect of esculemoside A (EsA)on the proliferation and expression of CDK2 and P27 of rats glomerular mesangial cell .Methods rGMC was cultured in vitro ,The cell growth and cell toxicity were detected by MTT assay ;rGMC proliferation was observed by Rnase/PI-Flous ;The expression of CDK2 and P27 were measured by Western blotting .Results EsA at observed dose has not apparent cytotoxicity effect on rGMC .EsA(2 .5-5 .0 mg/L) inhibited the proliferation of rGMC after 48h .EsA increased the number of the G1 phase and reduced the number of the S phase of IL-1βinduced rGMC .At the same time ,EsA inhibited the expression of CDK2 and promoted the expression of P27 of IL-1βinduced rGMC .Conclusion The GMC is one of the mainly target cell which EsA bing therapeu tical action .EsA inhibited proliferation of IL-1βinduced the GMC ,inhibited the expression of CDK2 and activated the expression of P27 may be its mechanism .
2.Effects of panax notoginseng saponins pretreatment on the expression of wnt4 protein in renal ischemia reperfusion injury in rats

Objective To observe the effect of panax notoginseng saponins (PNS) pretreatment on the expression of wnt4 protein in renal ischemia reperfusion injury in rats. Methods Seventy-two male SD rats were randomly averaged into control group (CON group), renal ischemia-reperfusion group (IR group) and PNS group. Rats in CON group were removed the right kidneys, and the left kidneys were exposed for 30 minutes, and then the incisions were closed. Rats of IR group were removed the right kidneys, and the left renal artery were clamped for 30 minutes, and then the incisions were closed. Rats in PNS group were injected PNS (150 mg/kg) via tail vein for 3 days before experiment, and the rest steps were the same with IR group. After 24 h, 48 h and 72 h reperfusion, 8 rats in each group were killed, and blood and kidney samples were collected to examine the serum creatinine (Scr) and blood urea nitrogen (BUN). The left renal tissues were stained with HE. Pathological changes were observed under light microscopy. The score of renal tubular injury was evaluated. Immunohistochemistry was performed to detecte the expression of wnt4 in kidney tissue. The expression of wnt4 was detected by Western blot assay. Results There was significant renal injury in IR group. The renal tubular injury score, Scr and BUN reached the peak levels of injury at 24 h, which were then decreased gradually, but at 72 h still significantly higher than those of CON group (P<0.05). The renal tubular injury score, Scr and BUN values at different time points were significantly lower in PNS group than those of IR group (P<0.05). The results of wnt4 immunohistochemistry and Western blot assay were consistent, which showed that the expression of wnt4 peaked at 24 h in IR group and PNS group, but the expression levels were significantly higher in PNS group than those of IR group at each time point (P<0.05). Conclusion PNS could protect kidney from ischemia-reperfusion injury, which may be related to the enhanced expression of wnt4 protein.
3.Therapeutic effect of traditional Chinese herb and selective medial vastus muscle electric stimulation in treatment of chondromalacia of patella

[Objective]To investigate the therapeutic effect of traditional Chinese herb and selective medial vastus muscle electric stimulation in treatment of chondromalacia of patella(CMP).[Method]Ninety CMP cases were selected and divided into 3 treatment groups,with 30 cases in each one.Traditional Chinese herb was applied in group A.Selected medial vastus muscle electric stimulation was applied in group B.Combination of both treatments was applied in group C.All cases were followed up for 9-12 months.Congruence angle(CA) and lateral patello-femoral angle(LPA) were measured and statistical analysis was used to investigate the therapeutic effect.[Result]CA and LPA showed significant differences between pre-and postoperative results(P0.05).[Conclusion]Traditional Chinese herb combined with selective medial vastus muscle electric stimulation is a effective therapy in treatment of chondromalacia of patella.
4.Comparison of efficacy between emergency ESWL and elective ESWL in management of ureteral stones

Objective To compare the efficacy of the emergency ESWL within 24 hours after the onset of renal colic and elective ESWL at 5 - 7 days after the onset of renal colic in the treatment for ureteral stones.Method From January 2008 to October 2009, 365 patients in the Second Hospital of Nanchang, with ureteral stones were admitted to our department for renal colic. They underwent emergency ESWL or elective ESWL with HK. ESWL- 108 energy electromagnetic shock wave lithotripter. Success rate of fragmentation, stone-free rate after 2 weeks of operation, pain recurrence rate, and complications were evaluated. The data were analyzed using SPSS 15.0. Results The success rate of fragmentation and the stone-free rate after 2 weeks of emergency ESWL were 96.76% and96.21%, respectively, which were higher than those (88.33% and 87.22%, respectively) of elective ESWL ( P < 0. 05). The postoperative pain recurrence rate of emergency ESWL ( 4.32 % ) was lower than that (9.44%) of elective ESWL ( P <0.05). There was no difference of total complications between emergency ESWL (3.24%) and elective ESWL (2.78%) group (P >0.05). Conclusions Emergency ESWL is superior to selective ESWL and is the safe and effective method for the management of ureteral stones.
5.Comparative analysis of chemical constituents of Injectio Erigerontis with breviscapine by HPLC-DAD

AIM: To compare the chemical constituents of Injectio erigerontis and breviscapine. METHODS: High performance liquid chromatography and diotron array detection (HPLC-DAD) was adopled with Hypersil ODS2 Column (250 mm?4.6 mm,5 ?m), and mobile phase was acetonitrile-H 2O-H 3PO 4(18∶82∶0.2),detection wavelength was at 286 nm, temperature was at 40 ?C. RESULTS: Weak peaks of Seutellarin and caffeic acid were resembling between injetio erigerontis and breviscapine in UV spectra. but Injectio eriperontis possibly contained 10 catteic acid derivatives. CONCLUSION: Injectio erigerontis and breviscapine are completely different in chemical constituents,though their clinic application and curative effect are equal.

Objective To observe the influence of serum containing esculentoside A(EsA) on the proliferation of glomerular mesangial cell (GMC) and the activation of ERK1/2-AP-1 pathway of glomerular mesangial cell induced by IL-1β.Methods SD rats were gavaged by different doses of EsA(5,10,20,40 mg/kg) for getting medicated sera.The control group was set (gavage by 0.5sodium carboxymethylcellulose);the EsA medicated serum was used to treat rGMC.The control serum group was set.The influence of EsA medicated serum in each group on rGMC proliferation was detected by MTT;the rGMC was divided into the blank control group,IL-1β single action group,IL-1β+ EsA double action group,IL-1β+ U016 double action group and IL-1β+ U0126 + EsA combined action group,which were synchronized and then cultured for 48 h.Western blot was used to detectthe expression of p-ERK1/2 and AP-1 an the imaging analysis was performed.Results The EsA medicated serum(5-10 mg/kg gavage) inhibited the cellular proliferation(P<0.05 or P<0.01);the IL-1β group promoted the expression of p-ERK1/2 and AP-1 in rGMC(P< 0.05),after acting on rGMC in the IL-1β+EsA double action group,IL-1β+U0126 double action group and IL-1β+U0126+EsA combined action group,the expression of p-ERK1/2 and AP-1 was decreased(P<0.05).Conclusion Serum containing EsA (5~10 mg/kg gavage) significantly inhibits the rGMC proliferation;EsA inhibits IL-1β induced rGMC proliferation,its action pathway on ERK1/2-AP-1 is one of mechanisms for inhibiting rGMC proliferation.

Objective:To investigate associations between glycosylated hemoglobin (HbA1c) levels and nonalcoholic fatty liver disease (NAFLD) in population who underwent periodic heath examination.Methods:A total of 912 subjects were selected from the health examination adults in University of Hong Kong Shenzhen Hospital from June to November 2013,and they were assigned into a NAFLD group,an else hepatic disease group,and a control group.The physical examination results of these subjects were analyzed retrospectively.Results:The prevalence of NAFLD was 29.7%.The related factors such as age,body mass index,and the levels of systolic blood pressure,diastolic blood pressure,total cholesterol,triglyceride,low density lipoprotein cholesterol,fasting glucose,HbA1c and alanine aminotransferase in the NAFLD group were higher than those in the else hepatic disease group and the control group (P<0.05).Aspartate aminotransferase levels in the NAFLD group were higher than those in the control group (P<0.05).High density lipoprotein cholesterol levels in the NAFLD group were lower than those in the else hepatic disease group and the control group (P<0.05).According to the quartile of HbAlc level,these subjects were divided into 4 groups,Q1,Q2,Q3,and Q4,and the prevalence of NAFLD in the Q1,Q2,Q3,and Q4 were 17.5%,21.5%,31%,and 59.4%,respectively.The prevalence of NAFLD increased along with the increase in the level of HbAlc (x2=100.092,P<0.05).Logistic regression analysis showed that the risk factor for NAFLD was increased in the subjects of Q4 as compared with the subjects of Q1 after adjusting for the related factors including age,gender,body mass index,etc.Conclusion:There is a positive correlation between the prevalence of NAFLD and HbAlc level,and the risk of NAFLD increases with the elevating level of HbA1c.High HbA1c level is an independent risk factor for NAFLD.Thus it should be listed as a routine medical test in the health examination.
9.Effect of dual antiplatelet drugs on Fibulin-5, vWF and P-selection in serum of patients with acute cerebral infarction

Objective To analyse the effect of dual antiplatelet drugs on fibulin-5, vWF and P-selection in serum of patients with acute cerebral infarction.Methods Diagnosed 60 patients with acute cerebral infarction in our hospital and collected. All patients were randomly divided into experimental group and control group, 30 cases in each group.The control group was treated with conventional treatment and aspirin, and the experimental group was treated with clopidogrel on the basis of control group.After treatment, the serum levels of Fibulin-5, vWF, P-selection and adverse reactions were detected in all patients.ResuIts After treatment, compared with control group, the serum Fibulin-5 level was significantly lower in experimental group ( P<0.05 );the serum vWF level in experimental group was significantly lower ( P<0.05 );the serum P-selectin level in experimental group was significantly lower (P<0.05);there was no significant difference in the incidence of adverse reactions between two groups. ConcIusion Dual antiplatelet drugs can reduce serum vWF, P-selectin and fibulin-5 in serum of patients with acute cerebral infarction, adverse reactions do not significantly increase, have guiding significance to clinical application.
10.Identification of two peaks in HPLC chromatogram of Erigeron Injection

AIM:To study the chemical constituents of Erigeron Injection by HPLC. METHODS: Under the guidance of HPLC analysis,two constituents were separated and purified by the aid of column chromatography with silica gel,and identified by UV、 IR、 MS、 NMR respectively and documentary data. RESULTS: Two compounds from semi-finished products of Erigeron Injection were identified as caffeic acid(Ⅰ) and chlorogenic acid(Ⅱ). CONCLUSION: Two main peaks in HPLC chromatogram of Erigeron Injection are Ⅰ and Ⅱ.
