1.Molecular cloning, expression and identification of recombinant hyaluronan synthase from Pasteurella multocida

Aim: To set a suitable route for efficient expression and purification of recombinant hyaluronan syn-thase from Pasteurella multocida (rPmH AS). Methods: Coding sequences were cloned and expressed in Esche-richia coli strain of BL-21( DE3). Then rPmHAS was purified through a simple Ni-affinity chromatography and identified by hyaluronic acid binding protein ( HABP) based ELISA assay. Results: High yield expression of functional rPmHAS exceeding the highest production reported hitherto was achieved, and the purity of rPmHAS was as high as 90%. Conclusion: In this study, an optimized system of the expression, purification and identifica-tion route for large scale preparation of rPmHAS was established, which will be helpful for accelerating further study of PmHAS and facilitating fine researches on peculiar hyaluronic acid with special properties.
2.Study on focal distance of II type uterine fibroids under mucous membrane treated by high intensity focused ultrasound ablation

Objective To assess the relationship between endometrium damage and focal distance of high intensity focused ultrasound ablation(HIFU).Methods 150 patients with requirement of atoke from Ⅱ type uterine fibroids under HIFU were collected and divided into two groups by the location of fibroids,one was antetheca group, and the other was posterior group,which were further divided into three groups,A group(25 cases),B group(25 cases), and C group(25 cases),in which their distance were 15mm,18mm,and 20mm from endometrium respectively.Those patients were treated with HIFU,and improvement of symptoms was assessed by uterine fibroids symptom (UFS) score,energy efficiency factor(EEF)was served as total energy required by treatment,the closest distance from mar-gin of necrosis of fibroids to endometrium and the rate of ablation were detected by MRI,as well as integrity of endo-metrium and vaginal bleeding were observed on postoperative 2 days and 3 months.Results After treatment by HIFU,significant improvement of symptoms was found,especially for the fibroids in antetheca(t =3.868,P =0.01), moreover,as for the fibroids in antetheca,the obvious efficacy was found in B group and C group(F =4.711,P <0.05),while for posterior fibroids,the efficacy was not associated with the location of HIFU(F =3.898,P >0.05).In addition,high rate of ablation and low EEF for the fibroids in antetheca was found(t =9.818,11.224,respectively,all P <0.01),and the highest rate of ablation in A group was revealed(F =105.673,P <0.01),followed by that of B group,the worse was C group.While in posterior fibroids,the distance was not associated with the rate of ablation(F =0.485,P >0.05).There was significant difference of integrity rate for endometrium between antetheca and posterior fibroids on postoperative 2 days(93.33% vs.77.33%,χ2 =7.67,P <0.05),and the rate in antetheca fibroids was higher than that of posterior fibroids(94.67% vs.82.67%)on postoperative 3 months.After postoperative 2 days,as for the fibroids in antetheca,the rate of integrity in B group(100.00%)and C group(100.00%)was higher than that in A group(80.00%),while in posterior fibroids,the highest rate was in C group(100.00%),followed by B group, which of A group was the worst(60.00%);After postoperative 3 months,in the fibroids of antetheca,there was no sig-nificant difference of rate between B group and C group,which were higher than that in A group(84.00%),while in posterior fibroids,the highest rate was in C group(100.00%),followed by B group(84.00%).In addition,significant difference between varied focal distance and improvement of vaginal bleeding existed in antetheca and posterior fibroids(P <0.05),for example the score of vaginal bleeding in B group and C group at postoperative 3 months was lower than that in A group(F =7.292,P <0.01),while for posterior fibroids,the efficacy of C group was higher than that of B group(F =14.559,P <0.05 ).Conclusion Although improved efficacy of Ⅱ type uterine fibroids is offered by HIFU,for the minimum damage of endometrium,its focal distance is varied with the locations of fibroids, namely,the safe distance in antetheca is more than 18mm,while that in posterior fibroids is more than 20mm.
3.Comparison of Etomidate-Dexmedetomidine and Propofol-Dexmedetomidine sedation in patients with gastroesophageal relfux disease treated by endoluminal radiofrequency ablation

Objective To compare the efficacy and safety of Etomidate-Dexmedetomidine and Propofol-Dexmedetomidine sedation in patients undergoing endoscopic radiofrequency ablation of gastroesophageal relfux disease (GERD).Methods Sixty adult patients, scheduled for elective endoluminal radiofrequency ablation for GERD under sedation were prospectively randomized into Etomidate-Dexmedetomidine (Group-E,n = 30) or Propofol-Dexmedetomidine (Group-P,n = 30) group. A bolus of 0.2 μg/kg of intravenous Dexmedetomidine was followed by intermittent Etomidate or Propofol injection during the procedure in order to maintain a proper depth of sedation with a Ramsay sedation scores of 5~6. Heart rates, mean blood pressure, oxygen saturation, respiratory rates and Ramsay sedation scores were recorded before sedation (T0), at the beginning of the examination (T1), during radiofrequency energy delivery (T2), at the time of gastroscopy (T3) and at the end of therapy (T4). Inter-group differences in sedation proifles (duration, time to recovery, incidence of body movement, Ramsay sedation scores and satisfaction of patient and endoscopist) and cardio-respiratory responses (heart rate, mean arterial pressure, oxygen saturation) were determined during and after radiofrequency ablation.Results No difference was found for therapy duration, anesthesia time or the time to recovery between Group-P and Group-E. Patients receiving Propofol experienced more bradycardia (Ρ = 0.032) and had higher incidences of vasoactives used (Ρ = 0.002) compared with that receiving Etomidate. Oxygen saturation in T1 (Ρ = 0.023) and T2 (Ρ = 0.009) was lower in the Group-P. No significant difference was found for other indicators.Conclusion Etomidate-Dexmedetomidine sedation was superior to Propofol-Dexmedetomidine sedation for GERD radiofrequency therapy with more stable cardio-respiratory responses.
4.Application of “double low”CTU method in children with urinary system malformation

Objective To probe the application value of “double low”CTU method in diagnosis of urinary system malformation in children.Methods Patients with urinary system malformation were screened by ultrasound,and renal calculi,tumor and tumor-like lesions were excluded.Double phase contrast media injection (iodixanol contrast media 270 mg I/mL)was used in one phase scan-ning,and the cortex-secretory phase images were obtained.The images were reviewed and evaluated independently by two readers. Results The coincidence rate of diagnosis was 100% with well images quality.Conclusion CTU is an important tool for the diagno-sis of urinary tract malformations in children.The “double low”method with lower radiation dose and contrast media quantity may ensure the quality of diagnosis.
5.The Effect of Intercostal Bronchial Artery Embolization in Treating Massive Hemoptysis

Objective To study the effect of intercostal bronchial artery embolization(BAE) with gelatinum sponge(GS) or polyvinyl alcohol(PVA) in treating acute massive hemoptysis.Methods Fifty-two patients with acute massive hemoptysis underwent selective bronchial arteries angiogaphy first,then superselective bronchial arteries embolization using 3F SP catheter with GS in group A(n=24) and with PVA in group B(n=28).Results In group A,immediate control of hemoptysis was achieved in 16 patients(66.67%),marked reduce in bleeding was seen in 7 patients(29.17%),the effective rate was 95.83%.In group B,immediate control of hemoptysis was achieved in 24 patients(85.71%),marked reduce in bleeding was seen in one patients(14.29%),the effective rate was 100%(P﹥0.05).All patients were followed-up for one year.The recurrent rate was 25%(6 cases) in group A and 3.57%(1 case) in group B(P﹤0.05).Conclusion BAE with PVA is effective and safe method in treating serious hemoptysis.
6.Effect of Polypeptide Extract from Scorpion Venom on MMP2 and MMP9 Expression in Leukemia -NOD/SCID Mice

Objective: To investigate the effect of polypeptide extract from scorpion venom (PESV) on the matrix metalloproteinase2(MMP2)and matrix metalloproteinase9(MMP9)expression in leukemia-NOD/SCID mice, and the intervention mechanism of PESV in the multiplication and infiltration of leukemic cells thereof. Methods: In order to establish the animal model of outer marrow infiltration of human leukemia,bone marrow mononuclear cells of leukemia patients, irradiated 270 cGy on body by ~(137)Cs, were injected into NOD/SCID mice. The mice were randomly divided into five groups. The groups I, II and III were treated with different concentrations of PESV. Group Ⅳ was the model group injected by the normal saline solution. GroupⅤ was taken as control. The peripheral white blood cell count and blood smear were observed in groups. All of the mice were killed after four-week observation and MMP2 and MMP9 expressions were examined using Real timePCRmethod. Results: The expression levels of MMP2 and MMP9 were significantly lower in group I, group II and group III than that of the model group(P < 0.05). The expression levels of MMP2 and MMP9 were related to the concentration of PESV. Moreover, the peripheral white blood cell count and blood smear were more normal in mice treated with PESV than those of mice of model group. Conclusion: PESV inhibited the overexpression of MMP2 and MMP9 in leukemia-NOD/SCID mice, which significantly inhibited the multiplication and infiltration of leukemic cells.

8.Mechanism of L-Ca2+/calpain signal transduction in verapamil inversing resistance of papillary thyroid carcinoma to doxorubicin

[ ABSTRACT] AIM:To investigate the mechanism of L-type calcium channel ( L-Ca2+)/calpain signal transduc-tion pathway in verapamil inversing resistance of papillary thyroid carcinoma to doxorubicin .METHODS:Human papillary thyroid carcinoma TPC-1 cells were cultured for 2 d.For determining the appropriate concentrations and treatment time of verapamil and doxorubicin , a compatibility test was conducted to detect the cell viability by CCK-8 assay.The cells were divided into control group , doxorubicin group , verapamil group and doxorubicin +verapamil group .The techniques of whole-cell patch-clamp was used to record L-Ca2+currents.The protein expression levels of calpain 1 and LC3 were detec-ted by Western blot .RESULTS: Compared with control group , the density of L-Ca2+current decreased in doxorubicin group and verapamil group (P<0.05).Compared with verapamil group , the density of L-Ca2+current decreased in doxo-rubicin+verapamil group (P<0.01).Compared with control group, the expression of calpain 1 decreased in doxorubicin group and verapamil group (P<0.05).Compared with doxorubicin group , the expression of calpain 1 decreased in doxo-rubicin+verapamil group (P<0.05).Compared with control group , the expression of LC3 increased in doxorubicin group and verapamil group (P<0.05).Compared with doxorubicin group , the expression of LC3 increased in doxorubicin +ver-apamil group ( P<0.01) .CONCLUSION:The drug resistance of TPC-1 cells to doxorubicin may be related to the in-crease in autophagic activity .Verapamil further increases autophagic activity of TPC-1 cells, resulting in autophagic death and inversing the resistance of TPC-1 cells to doxorubicin .The mechanism may be involved in L-Ca2+/calpain 1 signal transduction pathway of autophagy .
9.Comparison of the effects of different fixation methods on open-door side in posterior expansive open-door laminoplasty

Objective To investigate the clinical outcome and complications of different types of expansive open door laminoplasty in cervical spondylotic myelopathy.Methods From December 2008 to June 2012,221 patients with cervical spondylotic myelopathy treated with expansive open-door laminoplasty were divided into micro-titanium plate fixed group (96 cases),wire hanging fixed group(66 cases),anchor suspension fixed group (59 cases).The clinical outcome was evaluated including JOA score,operation time,bleeding volume,spinal cord pressure (a/M),cervical range of motion (ROM),open door angle,posterior shift distance of spinal cord and postoperative complications.Results All patients were followed up for more than 2 years.There were no significant differences in general condition,operation time,intraoperative blood loss,preoperative and postoperative 2 years JOA score,JOA score improvement rate,spinal cord shift distance,spinal cord compression among three groups;In the incidence of C5 nerve root paralysis,there was no significant difference between the silk suspension fixation group (12.1%) and the anchored suspension group (11.9%).The incidence of C5 nerve root paralysis for microplate fixation group (2.1%) was significantly lower than that of the silk suspension fixation group and anchored suspension fixation group,and the difference was statistically significant.There was no significant difference in the fusion rate between the 3-month and 6-month portal arteries in the group of silk suspension fixation group(65%,82%) and anchor fixation group(61%,82%).The fusion rate of the microtiter plate fixation group(90%,98%) was significantly higher than that of the silk suspension group and the anchorage group at 3 months and 6 months after operation.There were no significant differences in the incidence of recurrence,the total amount of cervical spine loss and the incidence of axial symptoms after 6 months of operation in the suspension group and the anchor suspension group.The incidence of occlusion,the total amount of cervical spine loss and the incidence of axial symptoms were significantly lower in the micro titanium plate fixation group than those in the silk suspension group and the anchorage fixation group,the difference was statistically significant.Conclusion Using micro-titanium plate to fix the side of open door could achieve full spinal cord decompression and access to satisfactory neurological improvement rate.At the same time,compared with the traditional silk or anchor suspension,the use of micro-titanium plate is more conducive to the door axis bone healing,prevent the occurrence of reclosing,significantly reduce the loss of postoperative cervical range of motion,and reduce the incidence of axial symptoms with the satisfaction of the clinical outcome.
10.Effect of light quality on growth and polysaccharides content of Ganoderma lucidum.

OBJECTIVETo study the effect of light quality on growth, polysaccharides content of Ganoderma lucidum.
METHODG. lucidum was planted under different light qualities. The growth of G. lucidum was observed and polysaccharides content was determined in different growth periods.
RESULTThere were significant differences in the form of the thickness and the numbers of ringed of the fruit before the ejection. The maximum content of G. lucidum polysaccharides under different light qualities were appeared in different periods. The content of polysaccharides was higher than that of CK under blue light in the period of budding, parachute phase, after spore ejection. The content of polysaccharides was increased under green light in the period of growth stage. The yield of fruits and spores were different in different light qualities.
CONCLUSIONThe light quality could increase the content of polysaccharides in G. lucidum.
