3. Analysis of the chemical stability of Morinda officinalis oligosaccharides during extraction
Chinese Pharmaceutical Journal 2014;49(2):102-105
OBJECTIVE: To study the chemical stability of oligosaccharides in Morinda officinalis (M. officinalis) extracted with different solvents. METHODS: The change of M. officinalis oligosaccharides extracted with water, ethanol, and aqueous solutions at different pHs was detected by HPTLC. RESULTS: The oligosaccharides began to constantly hydrolyze after 0.5 h when being extracted with water for different time. There was a fall of oligosaccharides concentration while the contents of fructose and sucrose were on the rise. New unknown compositions were generated after 0.5 h. When M. officinalis oligosaccharides were extracted with different percentages of ethanol, it only hydrolyzed when ethanol was in the range of 10%-30%, with little unknown compositions. Oligose was almost completely hydrolyzed at pH 2-3 but the hydrolyzation was alleviated at pH 3-4, with many kinds of new unknown composition generated. The oligosaccharides became stable when the pH was between 6 and 10. CONCLUSION: M. officinalis oligosaccharides is unstable in water, but rather stable in ethanol. However, there is significant difference in aqueous solutions at different pHs.

Aim To apply stochastic resonance algorithm (SRA) to quantitative analysis of weak chromatographic signal, which was embedded in the noise. Methods Based on the theory of stochastic resonance (SR), a simple and effective SRA has been established to improve analytical detection limits of chromatographic analysis, which apply to enhance the signal to noise ratio by the optimization of the parameters and Runge-Kutta method, was established. The method was used to quantitative analysis of phenazopyridine in human plasma by HPLC/UV. Meanwhile this method is compared with HPLC/MS.Results By experimental chromatographic data sets, an excellent quantitative relationship between concentrations of phenazopyridine and their responses had been obtained. The concentration of phenazopyridine in plasma determined by HPLC/UV with SRA and HPLC/MS showed that there was no significant difference (P > 0.05) between the two methods. Conclusion The new method was feasible.

In order to search for a new pathway to improve the yield of ginseng through growing at the full sun shine accompanied by salicylic acid (SA), the net photosynthetic rate (P(n)), superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), malondialdehyde (MDA) in Panax ginseng leaves, and the content of ginsenosides in roots were compared under various concentrations of SA and full sun shine with the traditional shade shed. Under the full sun shine, 0.05, 0.2 mmol x L(-1) SA increased net photosynthetic rate to a great extent. Under the cloudy day, the average net photosynthetic rate increased by 127.8% and 155.0% over the traditional shade shed, 13.9% and 27.5% over the treatment without SA respectively; under the clear day, 23.5% and 30.4% over the traditional shade shed, 8.6% and 14.6% over the treatment without SA, particularly obvious in the morning and late afternoon. With such concentration, SA increased activities of SOD, CAT, POD, and decreased the contents of the MDA. This difference resulted from different light intensity, rise of light saturation point, and fall of compensation point. Full sun shine decreased ginsenosides contents, but with SA, the ginsenosides regained, the content of Rg1 and Re, Rb1, total six types of ginsenosides in SA 0.2 mmol x L(-1) group were higher than those in the control group (P < 0.05) and other groups. The application of 0.2 mmol x L(-1) SA under full sun shine during a short time has little threat to the P. ginseng in spring, and could enhance the resistance to the adversity, which would improve the yield of ginseng heavily.

OBJECTIVE:To explore the teaching model and method of pharmacy case analysis writing in standardized clinical pharmacist training. METHODS:Based on the training experience in our hospital, the purpose, contents, common problems and summary of pharmacy case analysis writing were analyzed. RESULTS:The aim of pharmacy case analysis writing was to cultivate students’pharmaceutical thinking and the ability to solve real-world pharmaceutical problems. The key was to reflect the clinical pharmacist’s role in the event. The common problems included inappropriate selection of topics, insufficient evidence in discus-sion,and unclear organization of contents, where teachers should pay more attention to. CONCLUSIONS:Improving the teaching of pharmacy case analysis writing can improve the quality of clinical pharmacy student’s case analysis and cultivate their practical skills.

OBJECTIVE: To compare the effects of de-addiction with the therapy of acupuncture, acupuncture plus opium, opium plus buprenorphine and opium plus Han's instrument for de-addiction and to study the effects of the four therapeutic methods on the protracted withdrawal syndrome and craving. METHODS: The effects of de-addiction were assessed with the opiate withdrawal scale and the craving degree with visual analogue scale (VAS). RESULTS: The dominance of acupuncture treatment for withdrawal syndrome appeared to be after the 6th day, and the dominance for controlling craving showed after the 8th day, moreover, there were little side effects. CONCLUSION: Acupuncture treatment had the potentiality of preventing relapse and could be used for treating the protracted withdrawal syndrome and psychic dependence during the period between the stages of abstinence and rehabilitation.

BACKGROUND: Microglia play an important role in immune surveillance in their quiescent state, but the role of the activated microglia is under discussion. OBJECTIVE: To analyze the mechanism of activated microglia in acute cerebral infarction. METHODS: Totally 96 male Kunming mice were selected and randomly divided into four groups, including transplantation, placebo, blank control and sham operation groups. Permanent occlusion of the middle cerebral artery was performed using suture method in the mice of the transplantation, placebo and blank control groups, followed by injection of microglia suspension via subclavian vein, medium containing the same volume of microglia, and nothing, respectively, at 12 hours after modeling. In the meanwhile, the same amount of microglia suspension was injected into the mice of the sham operation group. The Zea-longa scale and brain-derived neurotrophic factor expression at 12, 24 and 72 hours after modeling, the volume of cerebral infarction and the number of nerve cells positive for microtubule-associated protein-2 at 72 hours after modeling were detected. RESULTS AND CONCLUSION: The Zea-longa scale score was 0 point in the sham operation group, which was significantly lower than that in the other three groups at each time point after modeling (P < 0.01). The Zea-longa scores in the transplantation group were significantly lower than those in the placebo and blank control groups at 24 and 72 hours after transplantation (P < 0.01). The positive expression rate of brain-derived neurotrophic factor in the transplantation group was significantly higher than that in the other three groups after transplantation (P < 0.01). The sham group showed no infarction, while the size of cerebral infarction in the transplantation group was significantly lower than that in the placebo and blank control groups (P < 0.01), and the microtubule-associated protein-2 positive rate was significantly higher than that in the placebo and blank control groups (P < 0.01). These results manifest that the activated microglia can improve the survival rate of nerve cells, promote the recovery of cerebral nerve function and reduce the size of cerebral infarction.
