1.Effect of tea polyphenols on oxidative damage and apoptosis in human bronchial epithelial cells induced by low-dose cigarette smoke condensate

Objective To determine the effect of tea polyphenols on oxidative damage and (apoptosis) in human bronchial epithelial cells induced by low-dose cigarette smoke condensate (CSC).Methods We prepared CSC. 3-(4,5-dimethyl thiazoly) 2,5-diphenyl-tetrazoliun bromide (MTT) assay was used to determine the growth of cultured human bronchial epithelial cells (HBE135-E6E7). Fluorescent-chemiluminescent analyzer was used to measure cell reactive oxygen species (ROS) level. DNA ladder method was used to detect HBE135-E6E7 apoptosis. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect Bcl-2 and Bax mRNA expression.Results Concentration of intracellular ROS in the CSC group and CSC + TP group was significantly higher than that in the control group (P<0.01); concentration of intracellular ROS in the CSC + TP group was significantly lower than that in the CSC group (P<0.01). Apparent DNA breakage of the tail belt appeared in the CSC Group,while only a small amount of DNA breakage of the tail belt appeared in the CSC + TP group. Compared with the control group, Bcl-2 mRNA expression was reduced and Bax mRNA expression was increased in the CSC group (all P<0.01). Compared with the CSC group, Bcl-2 mRNA expression was increased and Bax mRNA expression was reduced in the CSC+TP group (all P<0.01). Ratio of Bcl-2 mRNA/ Bax mRNA in the CSC group and CSC+TP group was significantly lower than that in the control group (all P<0.01).Conclusion TP can antagonize CSC-induced airway epithelial cell apoptosis through the effective removal of ROS, promoting Bcl-2 mRNA expression and inhibiting the expression of Bax mRNA.

Objective To investigate the role of PI3K/AKT and ERK1/2 signaling pathways in the process of cardiomyopathy induced by doxorubicin(DOX).Methods Twenty SD rats were divided into two groups:DOX group(10 rats that were injected through caudal vein with 2.5 mg/kg DOX,once a week,for 6 weeks)and control group(10 rats).At 5th week after the end of DOX injection,the structural changes and functional damage were examined,and the apoptosis rate of cardiomyocytes and the expression levels of p-AKT and p-ERK1/2 were determined.Results Eleven weeks after the DOX injection,the myocardial pathological changes were mainly myofibrillar loss and cytoplasmic vacuolization.DOX could induce compensated heart function(the value of FS% was 34,P0.05)and Bax to 338%(P

T he concentrations of clonazepam and 7-am in-oclonazepam w ere detected in each hair section. Results C lonazepam and its m etabolite 7-am inoclon-azepam w ere detected in parts of hair sections from the 6 victim s. T he occurrence tim e of drug peak concentration w as consistent w ith the intake tim ing provided by victim s. Conclusion Segm ental analysis of hair can provide the inform ation of frequency of dosage and intake tim ing, w hich show s an unique evidential value in drug facilitated crim es.
4.A preliminary study on visual cortex and optic radiation with diabetic retinopathy by 1H-MR spectroscopy

Objective To study the metabolic change of proton magnetic resonance spectroscopy (1 H-MRS)in the visual cortex and optic radiation region of patients with diabetic retinopathy (DR).Methods 1H-MRS was performed in 20 patients with DR and 20 healthy volunteers on GE 1.5 T MR system respectively.Metabolic peaks of N-acetylasparte(NAA),creatine(Cr,in 3.02 and 3.94 ppm),choline-containing compounds(Cho)and myo-inositol(mI)were observed,and the ratios were analyzed by each other.Independent-samples t test was performed between two sets of data.Results In both visual cortex and optic radiation,the ratios of ml/Cr and mI/CrSee in DR group(0.664 ± 0.052 and 1.453±0.068 in visual cortex,0.717±0.074 and 1.484±0.114 in optic radiation)were significant higher than those in normal group(0.602±0.047 and 1.249±0.044 in visual cortex,0.679 ± 0.075 and 1.334±0.089 in optic radiation,P<0.05).In DR group,the ratios of CrSec/Cr,Cho/Cr and NAA/Cr in visual cortex and optic radiation were 0.458±0.043 and 0.488±0.052,0.481±0.057 and 0.807±0.110,1.633±0.105 and 1.709±0.140 respectively.In control group.the ratios of those were 0.484±0.041 and0.502±0.056,0.471±0.065 and 0.786±0.109,1.625±0.098 and 1.716±0.135 respectively.The ratios of CrSec/Cr,Cho/Cr and NAA/Cr had no statistic difference between two groups(P<0.05).Conclusion mI/CrSec is a typical change in the visual cortex and optic radiation region,1H-MRS as a noninvasive examination could provide biochemical and metabolic informations for diabetic patients.
5.Effect of of propofol repeated anesthesia on expression of CaMK Ⅱα in hippocampus in neonatal rats

ObjectiveTo investigate the effect of propofol repeated anesthesia on the expression of CaMK Ⅱ α in the hippocampus in neonatal rats.MethodsThirty-two SD rats aged 7 days weighing 12-16 g were randomly divided into 2 groups (n =16 each): group C received intraperitoneal 0.9% normal saline 7.5 ml/kg once a day for 7 days and group P received propofol 75 mg/kg once a day for 7 days.Learning and memory function were assessed using Morris warier maze at 28 days old of rats.The animals were sacrificed at 24 h after the tests and brain tissues were removed.The expression of CaMK Ⅱ α and phosphorylated CaMK Ⅱ α (pCaMK Ⅱ α) in hippocampal CAI region were determined by immunochemistry and Western bolt.ResultsCompared with group C,the escape latency was significantly prolonged,space exploration time shortened and expression of CaMK Ⅱ α and pCaMK Ⅱ α down-rugulated in group P than in group C( P < 0.01 ).ConclusionPropofol repeated anesthesia decreases congnitive function through down-regulating the expression and inhibiting the activity of CaMK Ⅱ α in hippocampus in neonatal rats.
6.Simultaneous Determination of Five Compounds in Isatidis Radix by HPLC

Objective To establish a HPLC method for simultaneous determination of uracil,hypoxanthine,guanosine, thymine,and( R,S)-goitrin in Isatidis Radix. Methods The determination was performed on a Hanbon Hedera C18 column (4. 6 mm×250 mm,5 μm). The mobile phase consisted of acetonitrile( A)and water( B)with linear gradient elution at the flow rate of 1.0 mL·min-1. The column temperature was 30 ℃. Detection wavelength was 254 nm. Results The linear range of uracil,hypoxanthine,guanosine,thymine,and(R,S)-goitrin was 1.97-39.40 mg·L-1(r=0.999 9),1.25-50.00 mg·L-1(r=0.999 9),0.25-10.40 mg·L-1(r=0. 999 6),2. 84-56. 70 mg·L-1(r=0. 999 9),and 0. 72-28.80 mg·L-1(r=0.999 8), respectively. The average recovery was 99. 36%(RSD=0. 91%),99. 79%(RSD=1. 12%),100. 90%(RSD=1. 71%),98. 67%(RSD=1. 50%),and 99. 33%(RSD=0. 94%),respectively. Conclusion The method is simple,accurate,reliable,reproducible and sensitive,which can be used as an effective method for quality control of Isatidis Radix.

Background and Objective:Lung adenocarcinoma is,at present,the most common malignancy in the world and its overall 5-year survival rate is disappointing.Because most of the patients are diagnosed at advanced stage,early diagnosis may improve the prognosis of patients with lung adenocarcinoma.Surface-Enhanced Laser Desorption and Ionization Mass Spectrometry(SELDI) is one of the currently used techniques to identify biomarkers for cancers.This study explored the application of serum SELDI proteomic patterns to distinguish lung adenocarcinoma patients from healthy individuals.Methods:Serum samples from 71 lung adenocarcinoma patients,71 healthy volunteers with matched gender,age and history of smoking were analyzed using WCX2 ProteinChip to select potential biomarkers.Proteomic spectra were generated by mass spectrometry.Results:Five highly expressed potential biomarkers were identified with the relative molecular weights of 4047.79?1.60,4203.99?1.91,4959.81?2.13,5329.30?2.55 and 7760.12?4.11.The sensitivity for diagnosing lung adenocarcinoma was 90.14%,78.87%,50.70%,57.75%,73.24%;and specificity was 97.18%,92.96%,70.42%,76.06%,94.37%,when the critical point was made 1.5.Conclusions:SELDI-TOF-MS ProteinChip technology is a quick,easy,convenient,and highoutput analyzing method that it is capable of selecting several relatively specific,potential biomarkers from the serum of lung adenocarcinoma patients and may have clinic value.
8.Combination of chest physiotherapy and intermittent non-invasive mechanical ventilation for chronic obstructive pulmonary disease patients with respiratory failure

Objective To determine the effect of combination of chest physiotherapy (CPT) and intermittent non-invasive ventilation for chronic obstructive pulmonary disease (COPD) patients with respiratory failure. Methods Ninety COPD patients with intermittent bi-level positive airway pressure (BiPAP) ventilation were randomly divided into 2 groups : control group (n=45) received BiPAP treatment after conventional anti-infection, phlegm treatment and support treatment; CPT group (n=45) received CPT before BiPAP treatment. Clinical symptoms, chest signs, chest X-ray, time of BiPAP therapy, PaO2, and PaCO2 after the treatment were evaluated. Results Compared with with the control group, patients in the CPT group significantly improved in clinical symptoms, chest signs, chest X-ray absorption as well as shorter BiPAP therapy time, PaO2 increase and PaCO2 decrease(P<0.05). Conclusion For COPD patients who need intermittent non-invasive BiPAP ventilation, receiving CPT can effectively improve the therapeutic effect. CPT and intermittent non in-vasive BiPAP ventilation cooperate in the treatment of COPD patients with respirstory failure.
9.Association of interleukin-6 receptor gene polymorphism with obesity

To examine the relationship between genetic variants in the interleukin-6 receptor gene and obesity. The result showed that the carriers of Asp358 homozygotes had higher risk in developing obesity when compared with Ala358 homozygotes (OR = 1.32,95% CI 1.07-1.68, P = 0. 041). Interleukin-6 receptor gene pulymorphism was significantly associated with obesity.

This study is to investigate the influence and mechanism of action of asymmetrical dimethylarginine (ADMA) and the induced oxidative stress level on Alzheimer's disease (AD) incidence. ADMA concentration, nitric oxide, Abeta(40)/Abeta(42) ratio, inducible NO synthase (iNOS) activity and the concentrations of the induced free radicals including malondialdehyde (MDA), 3-nitrotyrosine (3-NT) and peroxynitrite (ONOO-) in the cerebrospinal fluid (CSF) from 34 neurologically normal controls and 37 AD patients were quantitatively determined and statistically compared. The results showed that the ADMA concentration significantly decreased in AD patients, and it showed negative correlation with the NO, iNOS activity, and showed positive correlation with MMSE score. ADMA concentration was negatively correlated with Abeta(40)/Abeta(42) ratio (P<0.01) with the observation that Abeta(40)/Abeta(42) ratio increased while ADMA level decreased in CSF in AD patients. The concentration levels of MDA, 3-NT and ROS significantly increased compared with the control with all the P values less than 0.05. These findings suggested that the ADMA disorder and the oxidative damage effect of the induced free radicals in CSF of AD patients are an important mechanism of AD incidence, and their joint regulation may provide new idea for the prevention and clinical treatment of AD.
