3. Screening and determination of estrogen-like quality markers of Cuscuta chinensis
Chinese Traditional and Herbal Drugs 2020;51(10):2671-2679
Objective: To preliminarily screen out the estrogen-like quality markers of small grain Cuscuta chinensis from Heilongjiang Province, so as to provide reference for its subsequent experimental research and quality control. Methods: UPLC- Q-TOFMS/MS was used to qualitatively analyze the extract of C. chinensis and the fingerprints of different polar fractions were established. The estrogenic activity of different polar fractions was evaluated with uterine coefficient, endometrial thickness and serum estrogen level of mice. The bivariate correlation analysis and gray relational analysis were used to construct the composition-activity relationship between chemicals and the effects of estrogen for screening quality markers. And the content determination methods of the five quality markers were established. Results: A total of 10 quality markers related to the estrogenic effect from C. chinensis were found in the positive and negative ion scanning modes. They were hyperoside, astragalin, stigmasterol, neocuscutoside C, apigenin, kaempferol, 6-O-(E)-p-coumaroyl-β-D-fructofuranosyl-(2→1)-α-D-glucopyranoside, quercetin, isorhamnetin, and 2,6-octadecadiynoic acid. The contents of the five quality markers were determined as follows: hyperoside (2.753 ± 0.097) mg/g, quercetin (1.139 ± 0.107) mg/g, apigenin (1.104 ± 0.047) mg/g, kaempferol (1.144 ± 0.079) mg/g and isorhamnetin (0.697 ± 0.074) mg/g. Conclusion: The quality markers of small grain C. chinensis from Heilongjiang Province can be screened out according to the composition-activity relationship, and the method for the detect the concentrations of the quality markers is accurate and stable.

Objective To investigate the role of PI3K/Akt signaling in the regulation of epithelial-mesenchymal transition (EMT) of peritoneal mesothelial cells (PMCs) in peritoneal dialysis in vitro and in vivo. Methods The level of Phosphorylated serine/threonine kinase Akt and the expression of EMT associated gene and protein, including ZO-1,Vimentin and FN,were measured in mice EMT model. In vitro study, phosphorylation level and nuclear translocation of Akt,ZO-1 and Vimentin expression induced by TGF-β1 in human peritoneal mesothelial cells (HPMCs) were also observed. Moreover,HPMCs were pre-treated by one of PI3K/Akt inhibitor,LY294002,or transfected with dominant-negative Akt plasmid to inhibit PI3K/Akt signaling, then analyzed its effect on Zo-1 and Vimentin expression induced by TGF-β1.Results Compared with the control, thickened parietal peritoneum and remarkable decrease in mRNA and protein of the epithelial marker ZO-1,and notable increased in the expression of mesenchymal markers Vimentin and FN were observed in PD mice (all P < 0.01).Moreover,the phosphorylation of Akt also significantly increased under above condition (P < 0.01).In vitro study, with the stimulation of TGF-β1,the expression of Zo-l was down-regulated,while the expression of Vimentin increased (all P < 0.01).In addition,TGF-β1 remarkably increased pAkt in HPMCs (all P < 0.01) in dose-dependent.However,LY294002 and DN-Akt dramatically inhibited the vimentin expression in HPMCs induced by TGF-β1 after inhibition of pAkt.On the other hand, the expression of ZO-1 also was restored (P < 0.01).Conclusion PI3K/Akt signaling is involved in EMT of peritoneal mesothelial cells in peritoneal dialysis, and may be a new target for the prevention and treatment of peritoneal fibrosis during PD.

The effect of insulin treatment on fatty liver was investigated in diabetic mice fed with high-fat diet.C57 BL/6J mice were fed with high fat diet for 12 weeks, and then treated with Glargine for 4 weeks. The results showed that during intraperitoneal glucose tolerance test, blood glucose, serum total cholesterol and triglyceride(TG) were significantly lower in insulin-treated high fat group than those in untreated ones(P<0.05).The hepatic histology showed minimal or barely visible fat in hepmic cells along with TG content in liver decreasing by 20.71%. The results suggest that insulin ameliorates hepatic intracellular lipid accumulation induced by high fat diet.

To learn about the economic studies on traditional Chinese medicines in domestic literatures, in order to analyze the current economic assessment of traditional Chinese medicines and explore the existing problems. Efforts were made to search CNKI, VIP, Wanfang database and CBM by computer and include all literatures about economic assessment of traditional Chinese medicines published on professional domestic journals in the systematic assessment and quality evaluation. Finally, 50 articles were included in the study, and the systematic assessment and quality evaluation were made for them in terms of titles, year, authors' identity, expense source, disease type, study perspective, study design type, study target, study target source, time limit, cost calculation, effect indicator, analytical technique and sensitivity analysis. The finally quality score was 0.74, which is very low. The results of the study showed insufficient studies on economics of traditional Chinese medicines, short study duration and simple evaluation methods, which will be solved through unremitting efforts in the future.

China Journal of Chinese Materia Medica 2014;39(23):4484-4488
Evaluation of the permeability mainly focuses on intestinal absorption in biopharmaceutics classification system (BCS). It is more complicated that the absorption and metabolism under multicomponent environment in biopharmaceutics classification system of Chinese materia medica (CMMBCS) compared with single component environment, which needs suitable mathematical models to be described. Therefore, with full consideration of existing single component mathematical algorithm combining with the characteristics of intestinal absorption and metabolism, we explored and designed a new mathematical algorithm of intestinal absorption and metabolism of multicomponent drug. Then we put forward a new coefficient, P (influence), the relative change rate of the single component's intestinal absorption and metabolism under multicomponent environment compared with single component environment, which described the influences of intestinal absorption and metabolism of the component under multicomponent environment. Moreover, P (influence) highlights the distinctive characteristics of multicomponent drug's intestinal absorption and metabolism, and lays the foundation for the construction of CMMBCS.

Chinese Journal of Applied Physiology 2005;21(3):248-251
AIMIn order to explore the mechanism of central motilin-induced feeding behavior, the effects of erythromycin, a motilin receptor agonist, on glucose responsive neurons in hypothalamus were observed.
METHODSExtracellular recordings were made from single neurons in region of lateral hypothalamic area (LHA) and ventromedial hypothalamic nucleus (VMH) in anesthetized rats. On the basis of their responsiveness to intracarotid injection of 0.58 mol/L glucose solution 0.2 ml, glucose-sensitive neurons (GSNs) in LHA and glucoreceptor neurons (GRNs) in VMH were recognized. Effects of intracerebroventricularly (i. c. v.) administration of 4 microg erythromycin on neural activities of glucose responsive neurons and non-glucose responsive neurons were examined. The mixture of EM and GM-109 1 microl were used to GSNs and GRNs which were sensitive to i. c. v. administration of EM.
RESULTSIn LHA, EM increased activity of GSNs significantly (P < 0.05 vs non-glucose-sensitive neurons group). Whereas in VMH, EM significantly decreased the activities of GRNs (P < 0.01 vs non-glucoreceptor neurons group). The mixture of EM and GM-109 had no effect on GSNs and GRNs.
CONCLUSIONEM, a motilin receptor agonist, can stimulate GSNs in LHA and suppress GRNs in VMH and this may contribute to central motilin's effect on feeding behavior.
