1.Effect of PP60c-Src on Ang Ⅱ- induced signal transduction in rat vascular smooth muscle cells
Chinese Journal of Pathophysiology 2005;21(4):685-689
AIM: The aim of the present study was to clarify the mechanism of intracellular signal transduction in Ang Ⅱ- induced proliferation of vascular smooth muscular cells (VSMC) by observing the effect of c- Src on Ang Ⅱ- mediated mitogen- activated protein kinase (MAPK) activation and c- Fos protein expression in cultured VSMC of rats. METHODS: Cultured aortic VSMCs from SD rats were transfected with anti-sense c-Src oligodeoxynucleotides (ODNs) wrapt with lipofectin to inhibit c- Src activity and protein production. Untransfected VSMCs were used as control. We observed the role of Ang Ⅱ stimulation in MAPK activation and c- Fos protein expression. c- Src kinase activity was measured by protein immunoprecipitation and kinase autophosphorylation. The phosphorylation rate of the substrate myelin basic protein (MBP) was employed to assess MAPK activity.Western immunoblot was used to detect protein expression of c- Src and c-Fos. RESULTS: c-Src protein expression in VSMC transfected with different concentrations of anti- sense ODNs significantly decreased in a negative dose- effect manner. c- Src kinase activity was also markedly inhibited. Following the stimulation of Ang Ⅱ on transfected VSMCs with anti-sense ODNs, the increase rate of c- Src activity was 8.7% of that in control, the activity of MAPK was 1.6% compared with control and c- Fos protein expression was as 30.0% as that of control. CONCLUSION: Ang Ⅱ induces c- Src activation. MAPK activation and c - Fos protein expression by Ang Ⅱ is dependent on c- Src activation. These findings indicate that c- Src is an important signal factor in Ang Ⅱ-induced VSMC proliferation.

Calculus Bovis is a valuable traditional Chinese medicine and has been used for more than two thousand years in clinic with the effects of puring heart, sweeping phlegm, resuscitation, extinguishing wind and detoxification. Since the founding of the People's Republic of China, modern methods have been utilized by traditional Chinese medicine researchers in the resource identification, chemical components, pharmacodynamics, pharmacokinetics, pharmacy, clinical application, etc. It is their continually exploration that makes significant achievements for the modern research of Calculus Bovis. This article statistically analysed the literatures from 1949 to December 2015 in Pubmed, CNKI, Wanfang, Vip database etc. to review Calculus Bovis and its compound formulas, as well as its substitutes, quality control, formulation study, compound prescription, pharmacological mechanism and clinical research. The aim of this article is to provide a valuable reference for future developments and studies of Calculus Bovis.

Four-week-old male Sprague-Dawley rats were rendered diabetic by intraperitoneal injection of streptozotocin (STZ) after feeding high-fat-diet for 8 weeks, and divided into diabetes group and tumor necrosis factorrelated apoptosis ligand(TRAIL) group.Normal rats severed as a control group.Treatment with TRAIL lasted for 3 months.Total cholesterol,triglycerides,low-density lipoprotein-cholesterol,blood glucose, and insulin levels were decreased in TRAIL group,as compared with diabetes group.Area of atherosclerotic lesion in TRAIL group [(23.8 ± 5.7) %] was significantly smaller than that in diabetes group [(47.6 ± 7.8) %].It suggested that TRAIL may reduce the area of atherosclerotic lesion in diabetic rats.

Objective To analyze the impact and quality of life of minimally invasive surgery combined with catheter attract surgery on patients with cerebral hemorrhage. Methods A total of 90 patients with cerebral hemorrhage in our department from February 2013 to De-cember 2014 were randomly divided into invasive group and the control group.Minimally invasive surgery group were given to cerebral hemor-rhage minimally invasive surgery combined with catheter attract surgery, the control group received traditional craniotomy. The postoperative recovery, nerve function scores, quality of life and morbidity between two groups were compared. Results The operation time and edema vol-ume of tissue postoperative 7 days in minimally invasive surgery group was significantly lower than those of the control group. The hematoma clearance rate 1 day after surgery was higher than that of control group, with statistically significant difference(P <0.05).NIHSS scores of two groups were gradually decreased 7 d,15 d,30 d after surgery(P <0.05).NIHSS scores of the minimally invasive surgery group 7 d,15 d, 30 d after surgery were lower than that of control group, with statistically significant difference(P <0.05).The scores of vitality, physiological function, physical function, general health, bodily pain, emotional function, social function, mental health after 6 months of the surgery in the min-imally invasive group was higher than those of control group, with statistically significant difference(P <0.05).The rate of postoperative pulmo-nary infection, electrolyte imbalance, gastrointestinal bleeding in MIS group were lower than those in the control group, with statistically signifi-cant difference(χ2 =5.400 0,4.405 7,4.615 4,P <0.05).Conclusion Minimally invasive surgery combined with catheter attract surgery can improve neurological function and quality of life for patients with intracerebral hemorrhage, and reduce the incidence of complications.
