2.Manumycin inhibits the activity of breast cancer cell line SK-BR-3 via inducing apoptosis

AIM: To investigate the anticancer effect of manumycin on abdominal metastatic breast cancer cell line-SK-BR-3 and its relationship with p38 MAPK . METHODS: The test of anticancer effect was performed by the method of MTT, apoptosis induced by manumycin and affected by SB203580, a specific p38 MAPK inhibitor, were examined by caspase-3 activity assay kit, and the protein expression was detected by immunoblotting assay. RESULTS: The inhibition rates at 24 h after treatment with manumycin of 6 ?mol/L, 18 ?mol/L, 54 ?mol/L were (7.4?3.9)%, (21.0?4.4)% and (64.7?4.1)%, respectively and showed dosage-effect relationship. Background Choroidal melanoma(CM)is a common form of primary ocular cancer in adults.It is reported that indomethacin has inhibitory effect on many tumor in vitro and in vivo,but whether it can inhibit the growth of CM has not been published. Objective This study was to investigate the anti-tumor activity of indomethacin on the growth of human CM OCM-1 cell xenografts in nude mice. Methods OCM-1 cells were subcutaneously implanted on 24 SPF female BALB/C.nu/nu nude mice to establish ectopic models of human CM.The nude mice with the tumor 5 mm were randomly divided into 4 groups:untreated group,normal saline solution(NS) group,indomethacin 1 ms/kg group,indomethacin 2 ms/kg group.The 1 mS/kg or 2 ms/kg indomethacin was intraperitoneally injected for 14 consecutive days in indomethacin 1 ms/kg group and indomethacin 2 me/kg group respectively.and 0.2 ml of 2%NS-DMSO was used at a same way in the NS group.No any agent was used as the untreated group.The volume and weight of implanted tumor as well as inhibitory rates of indomethaein on tumor were calculated.The expression of ki67 and survivin proteins were measured with immunohistochemistry,and the expression of survivin mRNA in CM was assessed by RT-PCR. ResuIts The tumor of indomethacin treatment group was reduced in volume and weight with a significant difference between treatment group and control group as well as indomethacin 1 ms/ks group and indomethacin 2 ms/kg group(P<0.05).The inhibitory rate of indomethacin 1 ms/kg and 2 ms/kg for tumor was 22.86%,48.00%respectively.The prolifiration index (PI)of ki67 in these 4 groups were (76.73±3.34)%,(73.30±2.95)%,(55.97±2.24)%,(32.87±2.91)%respectively,and significant difference was found in PI between indomethacin 2 mg/kg group and untreated group or NS group(P<0.05),but there was not significant difference between indomethacin 1 mg/kg and 2 ms/kg group(P>0.05).Compared with the control group,the indomethacin treatment groups showed the decreased expression of survivin protein and mRNA,and significant difference was found between indomethaein 2 ms/kg group and untreated group or NS group(P<0.05),however,no significant difference was found between indomethacin 1 mg/kg and 2 mg/kg group(P>0.05). Conclusion Indomethacin inhibits the growth of CM in nude mice through inhibiting the expression of survivin in the tumor and accelerating cell apoptosis and inhibiting tumor cell proliferation.
5.Microsurgical treatment of dumbbell tumors of high cervical spine

Objective To summarize experiences of microsurgical treatment of dumbbell tumors of the high cervical spine. Methods A series of 12 patients with dumbbell tumors of the high cervical spine were treated by using microsurgical techniques through posterior approach or antero-lateral approach. Results Complete resection was achieved in 10 patients. Postoperative neurological symptoms improved greatly in all. Conclusion The key points of treatment in dumbbell tumors of the high cervical spine are to analyze the preoperative image carefully and have knowledge about anatomy of high cervical spine as well as the experience of microsurgical technique.
6.Effects of different surface treatments and binders on the bonding strength of zirconia crowns

BACKGROUND: Zirconia all-ceramic restorations have been widely used in the repair of various complicated dental or dentition defects. Considering its surface chemical inertness, zirconia bonding is extremely crucial for the clinical practice of zirconia all-ceramic restorations. Currently, there are a variety of clinically selectable bonding materials and surface treatments for dental restorations. OBJECTIVE: To study the effects of different surface treatments and binders on the bonding performance of zirconia. METHODS: Zirconia tiles with different diameters were made and randomly divided into 16 groups (n=30 per group). Super-Bond C & B, polycarboxylic acid zinc binder, Panavia F resin cements and Fuji I of glass ionomer cements were selected. The surface of zirconia was treated by four different methods, including grinding, sand blasting, silane treatment and sand blasting plus silane treatment. Zirconia tiles were bonded by different binders, placed into static distilled at 37 ℃ for 24 hours, and then the shear bond strength was detected using a universal testing machine. RESULTS AND CONCLUSION: (1) With the use of the same binder, four surface treatments showed different effects on the bonding strength (P < 0.05), and the strongest bonding strength appeared after sandblasting plus silane treatment on the zirconia surface. (2) With the use of the same surface treatment, different binders showed different effects on the bonding strength (P < 0.05). Super-Bond C & B binder exhibited the best bonding effect, followed by Panavia F resin cements and Fuji I of glass ionomer cements, and polycarboxylic acid zinc binder showed the worst bonding effect. To conclude, these four surface treatments can all enhance the bonding strength to a certain extent, and Super-Bond C & B binder with sandblasting and silane treatment can achieve the highest bonding strength on the zirconia surface.
7.Meta-analysis of Acupuncture Treatment for Type 2 Diabetes

Objective To systematically assess the efficacy and safety of acupuncture in treating type 2 diabetes.Methods Related literature was retrieved with a computer. After the data were sifted out from the literature according to the established criteria, they were collected and crossly checked by two researchers independently. The data were input to RevMan5.2 for statistical analysis. Results A total of ten studies were included with a total of 743 patients. The results of Meta analysis showed 1) Post-treatment total efficacy rate was higher in the acupuncture or combined acupuncture and medicine group than in the control group [OR=3.60, 95%CI (2.11,6.13)] and there was a statistically significant difference (Z=4.71,P<0.00001); 2) Post-treatment range of decrease in fasting blood glucose (FBG) was significantly higher in the experiment group than in the control group [WMD=-1.20,95%CI (-1.38,-1.02)] and there was a statistically significant difference (Z=12.88,P<0.00001); 3) Post-treatment range of decrease in 2-h postprandial blood glucose (2hPBG) was significantly higher in the experiment group than in the control group [WMD=-1.27, 95%CI (-1.47,-1.06)] and there was a statistically significant difference (Z=12.02,P<0.00001).Conclusion Clinically acupuncture or combined acupuncture and medicine is more effective than Chinese herbal medicine or Western medication alone in treating T2DM and has no toxic and side effects.

9.Effects of esculentoside A on production of IL-1 and TNF by rabbit synovial cells

Objective: To investigate the influence of esculentoside A(EsA) on production of IL-1 and TNF by rabbit synovial cells induced by LPS. Methods: levels of IL-1 and TNF in the supernatant of rabbit synovial cell were determined by examining proliferation of thymic cells and by bioassay L929 cells as target cells, respectively. Results: EsA in 5-40 μg/ml could significantly inhibit the production of IL-1 and TNF from rabbit synovial cells induced by LPS. Conclusion: EsA can inhibit the production of IL-1 and TNF from synovial cells. It suggests that EsA may play a role in improving the rheumatoid arthritis.
