1.Uncertainty in the Determination of Hardness in Water

Objective To explore the methods for the evaluation of the uncertainty of the determination of hardness in water. Methods A certain water sample was analyzed for the hardness according to EDTA-Na2 volumetry ruled by the Sanitory Standard for Drinking Water(2001).The mathematical model of the determination of hardness in water was established based on JJF1059-1999. The variance composition of the relative uncertainty was analyzed. The procedure of the evaluation of every uncertainty component was described in detail. Results The hardness in the water sample was 137 mg/L calculated according to mathematical model established in this study. The relative uncertainty and the expand uncertainty of the detection of hardness in the water sample were 0.008, 3 mg/L respectively. The expression of the uncertainty in the determination of hardness was (137?3)mg/L. Conclusion This method presented referable value for the evaluation of the uncertainty of volumetry determination method of hardness in water.
2.A finite element model based on medical image for evaluating biomechanical stability of percutaneous vertebroplasty

BACKGROUND: Researches on vitodynamics present that percutaneous vertebroplasty (PVP) can strengthen hardness and intension of single osteoporosis vertebral body after injection of bone cement; however, the infused volume and site of bone cement for maintaining mechanical stability of vertebral body should be further studied.OBJECTIVE: A numerical calculation method on finite element models (FEM) for biomechanical analysis has been developed, while a boundary condition describing the relative L1 -L2 displacement is imposed on the FEM to account for three-dimensional physiological states.DESIGN: Non-randomized control study.SEITING: Beijing Neurosurgical Institute.MATERIALS: One female patient aged 54 years with compressibility fracture at lumbar vertebrae L1-L2 induced by osteoporosis was diagnosed with CT examination, and the patient was told the fact. Based on CT scanning before and after PVP, 90 serial sections with the thickness of 1 mm were obtained and the size of each pixei was 0.33 mm.METHODS: The experiment was carried out Beijing Neurosurgery Institute from October 2005 to June 2006. ①Establishment of three-dimensional FEM: Integrating the anatomical structure from the spine CT and MRI image of a patient, a novel three-dimensional geometric model of lumbar functional spinal units (FSUs) has been built. Meanwhile, 90 serial sections were obtained to exchange data of CT sections and divide imagings. Based on the geometric model, two kinds of three-dimensional FEM of L1-L2 segments for preoperative and postoperative vertebrae were created. ② Model evaluation: The lcad of (500 N, 1 000 N, 1 500 N, 2 000 N, 2 500 N) axial compression were applied to the superior surface of the model in the form of a uniformly concentrated lcad over all L1 superior surface nodes respectively. We could observe the stress distribution of L1-L2 segment by applying the load and clue on the high stress concentration region as the most likely areas fracture.MAIN OUTCOME MEASURES: ① Changes of displacement, stress and strain distributions of FEM at L1-L2 segment of lumbar vertebrae under various loads; ② Effect of increase of bone cement volume on displacement, stress and strain distribution.RESULTS: ① The increase in displacement, stress and strain of FE model with the increase of loading in the postoperarive cases. The tendency was approximately linear which also illustrates the spine have flexible biomechanical characteristics. The region was a common place for injures due to loading. The magnitude of stress in the intervertebral disc depended on the proportion of load applied to the superior surface of the motion segment. The heavier the lcad was, the stronger the stress was. ② With the increase of stress on intervertebral disc, the increasing volume of bone cement could induce transfusion of load of nearby vertebral body.CONCLUSION: Building three-dimensional FEM of L1-L2 segments for preoperative and postoperative PVP can explain three-dimensional physiological status of vertebral body based on calculating relative shift of L1-L2 segment of lumbar vertebrae. Meanwhile, simulative calculation can clearly express distribution of stain and stress and preoperative and postoperative deformity of vertebral body.
3.Effects of PUVA on Apoptosis and Expression of Fas in Leukemia Cells

Objective To explore the effects of psoralen (PSO) and long wave ultraviolet A (PUVA) on apoptosis and expression of Fas in HL-60,K562 and NB4 leukemia cells.Methods The cells were incubated with PSO in different concentrations irradiated with or without UVA.The changes of ultrastructure of cells were observed under the electron microscope.The expression of Fas gene was detected by fluorescent quantitation PCR.The apoptosis ratio and the expression of Fas protein were detected through the flow cytometry.The factorial design and analysis of variance were used to analyze the interaction among the factors.Results There were obvious ultrastructure changes about apoptosis in leukemia cells after treated with PUVA.PSO,UVA and PUVA all increased the apoptosis ratio and expression of Fas gene and protein,and the effects of PUVA were stronger than the other two (P

Detection of the possible role of ERp46,new endoplasmic reticulum protein,on palmitic acid-inducedendoplasmic reticulum stress-apoptosis pathway in βTC6 cells for the new treatment of type 2 diabetes.Results showed that ERp46 played a protective role in palnutic acid-induced cell apoptosis by decreasing the endoplasmic reticulum stress response through three pathway.
5.Changes of Caspase8 and Caspase3 Protein in HL-60 Cells during Apoptosis Induced by PUVA

Objective To study the changes of Caspase8 and Caspase3 protein in HL-60 cells during apoptosis induced by psoralen (PSO) plus ultraviolet A (UVA). Methods HL-60 cells were treated with PSO, extracted from Chinese medicine psoralea fruits in different concentrations, plus UVA of wave length 360 nm in different irradiation time. The apoptosis ratios, ultrastructure changes and the expression of Caspase8 and Caspase3 protein were detected. The factorial design and analysis of variance were used to analyze the interaction among the factors. Results PSO, UVA and PUVA all increased the apoptosis ratios and up-regulated the expression of Caspase8 and Caspase3 protein, but the effects of PUVA were the strongest. There were obvious changes about apoptosis under the electron microscope after treatment with PUVA. Conclusions PUVA can induce the apoptosis of HL-60 cells and activate Caspase8 and Caspase3 gene.
6.Clinical features and treatment of refractory mycotic otitis media

OBJECTIVE To study the clinical features and the treatment of refractory mycotic otitis media. METHODS From February 2005 to February 2009, 62 cases diagnosed as refractory mycotic otitis media were randomly divided into two groups. One group was 31 cases treated with local daubing Miconazole Nitrate Cream and another group was 31 cases treated with oral Itraconazole. The clinical features and fungi of distribution were retrospectively analyzed. RESULTSAspergillus was the most common fungi(70.6%). After two weeks treatment, 29 cases were cured in the group of Itraconazole, while 21 cases were cured in the group of Miconazole Nitrate Cream. CONCLUSIONRefractory mycotic otitis media is deep-seated mycotic infection and the primary pathomycete was aspergillus. The curative effect with oral Itraconazole was significantly better than that with local daubing Miconazole Nitrate Cream.
7.Role of T?SPOT.TB test in diagnosis of active tuberculosis

Objective To assess the value of T?SPOT.TB test in the diagnosis of active tuberculosis. Methods The clinical data of 975 hospitalized patients receiving T?SPOT.TB test were collected in our hospital. The clinical information and testing results were analyzed. The receiver operating curve (ROC) was used to determine the optimal threshold of T?SPOT.TB test for differentiating active tuberculosis. Results T?SPOT.TB test results showed that the positive rate was 29.26%for the non?active tuberculosis group(n=793),but was 91.21%for active tuberculosis patients group (n = 182),which indicated that the test had a significant value in active tuberculosis detection(P<0.001). The sensitivity of T?SPOT.TB test was 0.912 and the specificity was 0.707. The detection threshold of T?SPOT.TB was optimized. As the spot?forming count(sfc)of ESAT?6 antigen threshold was 11.5 and that of the CEP?10 threshold was 9.5,the efficiency of T?SPOT.TB test for detection of active pulmonary tuberculosis was the highest. Conclusions T?SPOT.TB test has a good diagnostic performance for active tuberculosis, and it can be further optimized to better serve the clinical practice.
8.Genomic level analysis of the molecular resistance mechanism for Pseudomonas aeruginosa PA902

Objective To investigate the resistance mechanism of Pseudomonas aeruginosa PA902 by high -throughput sequencing.Method PA902 was sequenced by the Illumina Miseq platform,and sequencing data were analyzed by bioinformatic techniques.Results PA902 was resistant to all common clinical antibiotics except amikacin.Analysis of data reveal resistance genes to β-1actams (blaOXA-10,blaPER-1 和 blaVIM-2),aminoglycosides (aph (3 ‘)-Ⅱd,aac (6')-Ⅰb-cr,aacA4 和 aadA2),sulfonamides (sul1),tetracyclines (tetG),chloramphenicol (floR).Several putative chromosome-located resistance genes were also identified.The results were fully in accordancc with thc susceptibility results.Sequence analysis of the contigs containing resistance genes revealed that they were always clustered on the same contig and correlated mobilization sequences.MLST identified PA902 as ST389,which was first reported in China.Conclusion High throughput sequencing demonstrated the molecular resistance mechanism of PA902,and the findings were in accordance with the susceptibility results.The technique will provide solid support for the traditional clinical microbiology methods.

10.Influence of different plantar flexion angle on X-ray diagnosis of deltoid ligament injury

Objective To improve the accuracy of X-ray diagnosis of the deltoid ligament injury by evaluating the influence of plantar flexion on the diagnosis.Methods Twenty-four patients with ankle supination-extemal rotation fracture which was distal fibula fracture but no medial malleolus fracture were treated from February 2010 to December 2010.All the patients were taken Mortise radiograph with the ankle in four positions of plantar flexion(0°,15°,30° and 45°)applied with rotational stress and the deltoid ligament injury was identified with MRI.The medical clear space(MCS)and the superior clear space(SCS)on each radiograph were measured.The results were tested by a one-way analysis of variance model(ANOVA),based on the following criteria:(1)the MCS≥ 4 mm and MCS > SCS,or when MCS≥5 mm and MCS >SCS.The MRI examination was taken as the gold standard to compare the influence of X-ray diagnosis of the deltoid ligament injury with different plantar flexions and different standards.Results The average MCS in four positions of plantar flexion(0°,15°,30°,and 45 °)with rotational stress were(4.10 ±0.79)mm,(4.55 ±0.72)mm,(4.99 ±0.56)mm and(5.71 ±0.86)ram,with statistical difference(P <0.05).The average SCS in four positions of plantar flexion(0°,15°,30°,and 45°)were(3.56 ±0.41)mm,(3.50 ±0.43)mm,(3.71 +0.44)mm and(3.93 ±0.51)mm,with no statistical difference in the four groups(P >0.05).With the MSC≥4 mm and MSC > SCS as the diagnostic criteria,the prevalence of false-positive findings of deltoid injury based on the ratio with the four positions of plantar flexion(0°,15°,30° and 45°)were 50.0%,66.7%,88.9% and 94.4% respectively.With the MSC≥5 mm and MSC >SCS as the diagnostic criteria,the prevalence of false-positive findings of deltoid injury based on the ratio with the four positions of plantar flexion(0°,15°,30° and 45°)were 5.6%,11.1%,38.9% and 77.8% respectively.Condusions Different plantar flexion angle is an important factor for X-ray diagnosis of deltoid ligament injury of the ankle joint.The prevalence of false-positive findings of deltoid injury increases with the increase of plantar flexion.
