1.Effects of endotoxin preconditioning on inflammatory response after global brain ischemia in rats

Objective To determine the protective effects of endotoxin preconditioning on the brain ischemic injury and explore its protective mechanism. Methods Global ischemia for 20 min by four-vessel occlusion model was used in 140 Wistar rats, in which 70 rats were pretreated with MPL at dose of 20 ?g/kg 24 h before global ischemia. Another 10 Wistar rats served as normal controls. Ischemic neurons and neuron density of the hippocampal CA1 region, MPO activity, TNF-? activity, IL-1? activity were assayed at 4, 12, 24, 48, 72, 120, 168 h after reperfusion. Results Pretreatment with endotoxin produced significant reductions in ischemic neuron number, MPO activity, TNF-? activity and IL-1? activity. Conclusion Endotoxin preconditioning protects brain from subsequent ischemic injury, in which the suppression of cellular inflammation may be the protective mechanism.

3.Effect of Probiotics on Feeding Intolerance in Infants with Very Low Birth Weight

Objective To explore effect of probiotics on improving the feeding intolerance in very low birth weight infants(VLBWI). Methods Sixty cases of VLBWI were randomly divided into two groups, 30 infants as therapy group,another 30 infants as control group.All infants in two groups were given aggressive intravenous nutrition and enteral feeding with preterm formulars. At the same time, probiotics were administered combined with preterm formulars to infants in therapy group. Milk amount, gastric residual, initial time of enteral feeding, time required for full enteral feeding and body weight on day 21 after birth were all recorded.Results Rate of no gastric residual increased significantly in therapy group compared with that of control group(t=6.24 P

5.Effects of endotoxin preconditioning on NOS activity in rat brain after global brain ischemia/reperfusion

Objective To determine the effects and significance of endotoxin preconditioning on iNOS activity,nNOS activity in rat cerebral cortex,hippocampus and cerebella region after global brain ischemia/reperfusion.Methods Global ischemia for 20 min was made by four-vessel occlusion model(4-vo) in 60 Wistar rats,among which 30 were injected of 20 ?g/kg MPL through caudal vein 24 h before model establishment.Another 8 rats undergoing sham operation served as controls.The dynamic change of iNOS activity,nNOS activity and neuron density of the cerebral cortex,hippocampus and cerebella region were observed at 1,4,8 h after reperfusion. Results The activity of iNOS and nNOS decreased significantly after endotoxin preconditioning in the regions mentioned above,as compared with that of rats only undergoing the ischemia/reperfusion.The neuron number in rat hippocampus decreased after ischemia/reperfusion,but no significant difference was found between control and endotoxin preconditioning groups.Conclusion The activity of iNOS and nNOS changed significantly after global brain ischemia/reperfusion.That endotoxin preconditioning decreased iNOS and nNOS activities may be the protective mechanism.
6.Experimental study of the protective effect of endotoxin pretreatment on hippocampal neurons in rat forebrain following ischemia-reperfusion

Objective To investigate the protective effect of endotoxin pretreatment on hippocampal neurons in rat forebrain following ischemia reperfusion and its possible mechanism. Methods Rat forebrain ischemia reperfusion model was used. The effects of endotoxin pretreatment on the changes of superoxide dismutase (SOD), glutathione peroxidase (GSH PX) and malondialdehyde (MDA) activities and the neuron count in CA1 region were observed. Results Pretreament with endotoxin before cerebral ischemia enhanced the activities of SOD and GSH PX but decreased MDA level and the number of ischemic neurons in CA1 region. Conclusion Endotoxin pretreatment can protect the neurons in rat forebrain against ischemia reperfusion injury. The mechanism may be associated with the enhancement of endogenous antioxidant activity in central nervous system.
7.Dynamic changes of inflammatory cytokines in hematoma fluid of intracerebral hemorrhage

30 ml) undergoing minimally invasive evacuation of intracranial hematoma from January 1999 to October 2002 in our department were analyzed retrospectively. The changes of IL 1, IL 6, and IL 8 in hematoma fluid were observed continuously. The content of IL 1 was determined by radioimmunassay and IL 6, IL 8 by ELISA methed. Results IL 1, IL 6, and IL 8 were observed at 6 12 h after hypertensive cerebral hemorrhage, and showed different changes in acute stage. Conclusion Inflammatory cytokines are involved in the pathological process of cerebral hemorrhage.
8.Study on separation and purification of total flavones from Smilax china by macroporous absorption resin.

OBJECTIVETo investigate the technological parameters of the purification process of total flavones from Smilax china with macroporous absorption resin.
METHODThe technical process for purification of total flavones with the optimum macroporous absorption resin was screened by yield of total flavones product.
RESULTThe D140 macroporous absorption resin had the best separating efficiency when the flavones content in the liquid was 0.5 g x mL(-1) equivalent to raw material, the volume of drug 18 BV (resin bad volume) with the adsorption-power 2 BV x h(-1), and the volume of 60% (mL x mL(-1)) ethanol as eluant 5-10 BV (resin bad volume) with desorption-power 1 BV x h(-1). The obtained flavones product has total flavones recovery rate of 84.72%.
CONCLUSIONThe treatment of regenerated resin is easy, this method is advisable.

9.Clinical Analysis of Infantile Cytomegalovirus Hepatitis with Cholestasis

Objective To investigate the clinical features of infantile cytomegalovirus(CMV) hepatitis with cholestasis.Methods The clinical features of infantile CMV hepatitis with cholestasis were retrospected in 48 cases and the liver function were detected before and after therapy in 23 cases.Results Forty-eight cases of infantile cytomegalovirus hepatitis with cholestasis had different degrees of jaundice, hepatosplenomegaly and abnormal liver functions. After therapy of 23 cases, the clinical symptoms and serum biochemical parameters such as bilirubin, alanine aminotransferase were improved , but serum parameters of aspartate aminotransferase ,alkaline phospholipids and ?-glutamyltransferase had no improvement.Conclusion The theraphy of CMV hepatitis with cholestasis shall be individual.
10.Effects of sevoflurane on expression of heparanase and fascin in lung carcinoma cells of mice

Objective To evaluate the effects of sevoflurane on the expression of heparanase ( HPA) and fascin in lung carcinoma cells of mice. Methods Mouse LLC cells were inoculated in the culture plate. After being cultured for 24 h, the cells were equally and randomly divided into 4 groups using a random number table: control group ( group CC) , 1% sevoflurane group ( group Sev1 ) , 2% sevoflurane group ( group Sev2 ) , and 3% sevoflurane group ( group Sev3 ) . Cells in Sev1-3 groups were exposed to 1%, 2% and 3% sevoflurane, respectively, for 4 h, while cells in group CC were not exposed to sevoflurane, and all the cells were then cultured for another 24 h in an incubator. The invasion of cells was determined by Transwell invasion assay, and the invaded cells were counted. The migration of cells was determined by wound healing assay, and cell migration rates were calculated. The expression of HPA and fascin in cells was detected by Western blot. Results Compared with group CC, the number of invaded cells and cell migration rates were gradually decreased, and the expression of HPA and fascin was gradually down?regulated with increasing concentrations of sevoflurane in Sev1-3 groups. Conclusion The mechanism through which sevoflurane inhibits the metastasis of mouse lung carcinoma cells is associated with down?regulated expression of HPA and fascin.
