1.Changes of interleukin-2 and interferon-γ in serum and synovial fluid of patients with joint injure with Kashin-Beck disease

Objective To investigate the changes of interleukin(IL)-2 and interferon(IFN)-γin serum and synovial fluid of patients with joint injure with Kashin-Beck disease(KBD), and study the roles of IL-2 and IFN-γ in KBD joint injure. Methods In accordance with the "Diagnostic Criteria of Kashin-Beck disease"(GB16003-1995),48 cases of KBD patients and 26 healthy people(control group) from KBD endemic area in Long county Shaanxi province were enrolled in the study. KBD patient were 24 males and 24 females, respectively, aged 40 to 65 years (mean age 51 years). Forty-eight serum specimens and 28 synovial fluid specimens of patients(14 males and 14 females,respectively) were collected. Healthy control group were 13 males and 13 females, respectively. Twenty-six serum specimens of healthy controls were collected. Serum and synovial fluid IL-2 and IFN-γ levels were determined by enzyme-linked immunosorbent assay(ELISA). Results In healthy controls and KBD patients, the midian of serum IL-2 were 46.8 ng/L and 55.7 ng/L, respectively, and IFN-γ were 52.3 ng/L and 48.8 ng/L, respectively. The difference was not statistically significant between healthy controls and KBD patients(t = 0.62, 0.70, all P > 0.05).In synovial fluid of KBD patient, the midian of IL-2 and IFN-γwere 48.3 ng/L and 44.1 ng/L, respectively. The difference was not statistically significant between serum and synovial fluid in KBD patients(t = 0.69, 1.72, all P >0.05). Conclusion Serum and synovial fluid IL-2 and IFN-γare not significantly increased in KBD patients with articular damage, indicating that IL-2 and IFN-γare not involved in KBD joint injury.
2.Induction of apoptosis and inhibition of HL-60 cell proliferation by survivin antisense oligonucleotide.

OBJECTIVETo investigate the effects of survivin antisense oligonucleotide (ODN) on cell proliferation and apoptosis of HL-60 cells.
METHODSSynthetic ODN was completely phosphorothioate-modified. Cationic lipid-mediated antisense ODN was transferred into HL-60 cells. The expression of survivin mRNA and protein was detected by RT-PCR and Western Blot. The incorporation of MTT was used as the measurement of HL-60 proliferation. The cell-cycle and apoptosis were analyzed by flow cytometry.
RESULTSHL-60 cells spontaneously expressed survivin mRNA and protein. Both mRNA and protein expression of survivin decreased significantly in the antisense ODN transfected cells in comparison to that in the original cells and cells transfected with sense ODN. Survivin antisense ODN significantly inhibited cell proliferation and induced apoptosis in a dose-dependent manner. The cell-cycle in the antisense ODN-transfected cells stopped at the G2/M phase.
CONCLUSIONSAntisense ODN targeting at survivin mRNA can inhibit HL-60 cell proliferation and induce G2/M stop and apoptosis.

3.Influence of Minkeqing oral liquid on IL-6, IL-8, ET-1, TX-B2 in blood and to observe bronchoalveolar lavage fluid of the rat with inhalating ovalbumin.

OBJECTIVETo observe the effect on infantile allergic cough with Minkeqing oral liquid (Minkeqing) and to study its cell molecular biologic mechanism.
METHODThe rat model was induced by inhalating ovalbumin; then the effects of Minkeqing on IL-6, IL-8, ET-1, TX-B2 in the blood and the bronchoalveolar lavage fluid (BALF) of the animal model were observed.
RESULTMinkeqing could reduce the levels of IL-6,IL-8,ET-1,Tx-B2 in the blood and BALF of the animal model.
CONCLUSIONMinkeqing has the significant function of inhibiting the release of inflammatory mediums.

4.Analysis of coronary arteriographic and fluorescence fundus angiographic characteristics in type 2 diabetic patients with coronary heart disease

Objective To study the relationship between coronary arteriographic and fluorescence fundus angiographic characteristics in type 2 diabetic patients with coronary heart disease.Methods The study was carried out by the analysis of the data from coronary arteriography and fluorescence fundus angiography in 203 type 2 diabetic patients with coronary heart disease in different groups divided according to age or total cholesterol level. Logisitic regression analysis was applied to explore various risk factors to angiographic characteristics.Results With advancing age,there were more involvement of 3 coronary vessels or the left main branch along with stageⅢretinopathy,but less single vessel diseases in the coronary arteries and less stageⅠretinopathy.The difference in coronary angiographic and fluorescence fundus angiographic characteristics between groups with different total cholesterol levels was not significant.Logistic regression analysis suggested that coronary artery diaease was related to age,sex and blood glucose and triglyceride levels while diabetic retinopathy was related to blood glucose level and age.Conclusion There is great difference in coronary arteriography and fluorescence fundus angiography among different age groups.Aging may aggravate the lesions both in the coronary arteries and fundal vessels in type 2 diabetic patients with coronary heart diseease.
5.Study on pelvic injury mechanisms in road traffic fatalities according to Young-Burgess classification.

OBJECTIVE:
To explore the pelvic injury mechanisms in road traffic fatalities (RTFs) according to Young-Burgess classification and its practical value in forensic medicine.
METHODS:
Retrospective study was performed on pelvic X-ray radiographs of adult RTFs outside the automobiles in 128 cases. Pelvic injury mechanisms were investigated according to Young-Burgess classification and then were analyzed statistically combined with the real circumstance.
RESULTS:
The accuracy of pelvic injury mechanism identification using APC subtype (94.1%) and LC subtype (92.9%) were significantly higher than that without using subtypes (63.6% and 70.7%) (P<0.05). LC subtype was helpful to discriminate the direction of force, for example the rear lateral force, anterior lateral force or continuous anterior lateral force.
CONCLUSION
Young-Burgess classification discriminated by various methods of medical imaging may be helpful to study the pelvic injury mechanisms and provide reliable reference for road traffic accidents reconstruction.

6.Effects of different factors on direct induction of microtubers from explants in Pinellia ternate.

OBJECTIVETo study the effects of different factors on direct induction of microtubers. These factors included plant growth substances, casein hydrolysate (CH), active carbon (AC), polyethylene glycol (PEG 4000), sucrose and glucose.
METHODUsing the orthogonal design method.
RESULT AND CONCLUSIONThe optimal media to directly induce microtubers from leaves were MS + MET 0.5 mg x L(-1) + 6-BA 0.5 mg x L(-1) + sucrose 3% and MS+ 6-BA 0.5 mg x L(-1) + IAA 0.5 mg x L(-1) + sucrose 3%. Optimal media to directly induce microtubers from tubers were MS + 6-BA 1.0 mg x L(-1) + PEG 5% + sucrose 5% and MS+ CH 500 mg x L(-1) + MET 0.5 mg x L(-1) + 6-BA 0.5 mg x L(-1) + AC 0.5% + sucrose 5%. Media suitable for plantlet growth were MS + 6-BA 0.5 mg x L(-1) + IAA 0.5 mg x L(-1) + AC 0.5% + sucrose 5% and MS + MET 0.5 mg x L(-1) + 6-BA 0.5 mg x L(-1) + sucrose 3%.

7.Surveillance of soil-borne nematodiasis in population in Hefei City,2012-2015

Objective To analyze the surveillance results of soil-borne nematodiasis in Hefei City from 2012 to 2015,so as to provide the evidence formulating the Hefei"Thirteen-five"parasitic disease prevention strategy.Methods The population in monitoring points in Changfeng County,Lujiang City and Chaohu County were investigated for Ascaris lumbricoides,Trichuris trichiura and hookworm infections with Kato-Katz technique every year from 2012 to 2015,and the children aged less than 6 years were checked for Enterobius vermicularis infection by using the cellophane tape method.Results A total of 12 496 people were investigated from 2012 to 2015,the number of infections was 84,and among them,the number of hookworm infection was the most(70 cases)and the number of A.lumbricoides was the least(3 cases).The infection rate in 2014 was higher than that in other years,and there was a statistically significant difference(P<0.008).There was a statistically significant difference among all age groups over 6 years of age in the infection rates(Χ2=27.229,P<0.05),and the infection rate of the age group over 66 years was higher than that of the other age groups.The infection rate in the female was higher than that in the male(Χ2=5.378, P<0.05).The infection rate in the monitoring point in Changfeng County was higher than that in the other monitoring points (P<0.017).Conclusions From 2012 to 2015,the infection rate of soil-borne nematode infections is lower than 5% in Hefei City.The number of hookworm infection is the most.Therefore,during the〝Thirteen five〝period,the key point of prevention of parasitic infections is hookworm infection.
8.Anti-inflammation and anti-oxidation effects of recombinant human superoxide dismutase on acute lung injury induced by meconium aspiration in infant rats.

OBJECTIVETo investigate the anti-inflammation and anti-oxidation effects of recombinant human CuZn superoxide dismutase(rhSOD) on acute lung injury (ALI) induced by meconium aspiration in rats.
METHODS1 ml/kg of 20% human newborn meconium suspension was intratracheally (IT) administrated to induce the model of ALI in 32 male Sprage-Dawley rats, and the animals were then randomized to 4 groups: 3 treatment groups with IT administration of 5, 10 and 20 mg/kg rhSOD dissolved in 1 ml/kg saline and the control group with IT administration of 1 ml/kg saline. The animals were killed after 24 h of treatments. The measurements included lung tissue wet/dry ratio, broncho-alveolar lavage fluid (BALF) protein, BALF protein/plasma protein (pulmonary permeability index, PPI),lung myeloperoxidase (MPO) and superoxide dismutase (SOD) activity, nitric oxide (NO) and 8-isoprostane levels. Lung injury score was also evaluated.
RESULTSCompared with the control group, pulmonary MPO activity, NO and 8-isoprostane levels were significantly decreased and SOD activity was markedly increased in all rhSOD treatment groups (P<0.05 or 0.01). Compared with the rhSOD 5 mg/kg group, pulmonary 8-isoprostane level was further low in the rhSOD 20 mg/kg group(P=0.01). Lung injury score was decreased in rhSOD 20 mg/kg group (P<0.05). But there were no statistically differences in lung wet/dry, BALF protein and PPI among all groups.
CONCLUSIONThe results suggest that a single IT dose of 5,10 or 20 mg/kg rhSOD can prevent lung damages in rats with ALI following meconium aspiration.

9.Effects of basic fibroblast growth factor on the proliferation of human salivary adenoid cystic carcinoma cell line ACC-2 and extracellular signal-regulated kinase, Cyclin D1, p2waf/cip1 signaling pathway.

OBJECTIVETo investigate the effects of basic fibroblast growth factor (bFGF) on the proliferation of human salivary adenoid cystic carcinoma (ACC) cell line ACC-2 in vitro.
RESULTSThe effect of ectogenic bFGF on proliferation of ACC-2 was observed by MTT assay. Extracellular signal-regulated kinase (ERK) activity was measured by immuno-precipitation. p-ERK1/2, Cyclin D1 and p21waf/cip1 expression were assessed by Western blot.
RESULTSbFGF could enhance the proliferation of ACC-2. Stimulated by bFGF, the proliferation ratio increased significantly. The intracellular ERK activity, p-ERK1/2 and Cyclin D1 expression were increased, while p21waf/cip1 expression was inhibited by different concentrations of bFGF. The above effects of bFGF could be attenuated by MEK inhibitor U0126.
CONCLUSIONbFGF stimulates the proliferation of ACC-2 in a dose dependent manner. The proliferation effect of bFGF may be due to up-regulating ERK, Cyclin D1 and p21waf/cip1 signaling pathway. This research can help us to explore a new pathogenesis and therapy of the ACC.

10.Pulmonary fungal infections after bone marrow transplantation: the value of high-resolution computed tomography in predicting their etiology.

BACKGROUNDThe correct diagnosis of etiology of fungal infection after bone marrow transplantation is very important to the choice of antifungal drugs and a premise for improvement of therapeutic efficacy. This study aimed to compare high-resolution computed tomography (HRCT) findings of the pulmonary fungal infections to determine whether the etiology of various fungal infections could be diagnosed with HRCT.
METHODSEighty-five cases were enrolled. According to the pathogens responsible for fungal infections, the patients were classified into three groups including invasive aspergillosis (n = 52), candidiasis (n = 19) and cryptococcosis (n = 14) groups. All the patients underwent HRCT scans. Two independent radiologists retrospectively analyzed the HRCT scans regarding CT patterns and distribution of lung abnormality.
RESULTSMost fungal infections in the three groups occurred in the neutropenic phase. There was no significant difference in the constituent ratio of fungal infections at different phases after bone marrow transplantation among the three groups. Agreement between the two observers for all the CT characteristics of fungal infections was excellent (k > 0.75). There was a significant difference in occurrence ratio of mass among the three groups (P = 0.02). Occurrence ratio of mass (43.3%, 13/30) in the group with invasive aspergillosis was higher than in each of other two groups (20.0%, 2/10; 14.3%, 1/7). There was no significant difference in other CT characteristics of nodules or masses; including number, margin, halo sign, cavitation and air-crescent sign. There was no significant difference in number, margin, air bronchogram and distribution of air-space consolidation.
CONCLUSIONSThe HRCT appearance of various pulmonary fungal infections has a great deal of overlap and is nonspecific. Mass is more common in invasive aspergillosis, which is helpful to the diagnosis of invasive aspergillosis after bone marrow transplantation.
