Chinese Journal of Biologicals 2025;38(04):436-441+449
Objective To investigate the genetic characteristics of viruses associated with viral diarrhea in Jilin Province, in order to provide a scientific basis for making more effective prevention and control strategies of viral diarrhea. Methods A total of 211 fecal specimens from hospitalized children under 5 years of age with diarrhea and 7 sewage samples were collected in Jilin Province from January to July 2023. Initial screening was performed using qPCR, and positive specimens were further analyzed by RT-PCR to amplify target genes. Sequencing was conducted, and phylogenetic trees were constructed to analyze genetic relationships. Results In fecal specimens, the positive rates for rotavirus(RV), Norovirus(NoV),Sapovirus(SaV), astrovirus(AstV), and adenovirus(AdV) were 10. 9%, 48. 3%, 4. 3%, 1. 4%, and 5. 7%, respectively. In sewage samples, 5 AstV and 4 NoV were detected, along with 3 samples each of RV, SaV, and AdV. Sequencing and genotyping results revealed that RV G8P [8] was the predominant circulating strain. Among NoV strains, GⅡ.4 [P16] was the most prevalent, followed by GⅡ.4 [P31]. In fecal samples, GⅠ group strains included GⅠ.3 [P13] and GⅠ.1 [P1]. One SaV strain of GⅡ.5 was identified. For AstV, three strains of HAstV-1a and one strain of HAstV-5 were detected. Among AdV strains, three were identified as HAdV-41, three as HAdV-2, one as HAdV-1, and one as HAdV-5.Conclusion The pathogenic genes of viral diarrhea in Jilin Province are diverse, the dominant strains of RV have changed, and domestic sewage contains a variety of diarrhea viruses.

Academic Journal of Second Military Medical University 2006;27(10):1072-1075
Objective: To screen for the differentially expressed genes in peripheral blood mononuclear cells (PBMCs) from type 2 diabetes patients with distal symmetric polyneuropathy (DSPN) by gene microarray. Methods: A 5075 gene cDNA microarray was used to examine gene expression in PBMCs of 2 type 2 diabetes patients with DSPN (group 1), 2 type 2 diabetes patients without DSPN (group 2),and 2 age- and sex-matched controls (control group). The differentially expressed genes were identified and subjected to real-time RT-PCR analysis. Results: Twenty-two genes were found to be differentially expressed, including cell metabolism and signal transduction genes, oncogenes and anti-oncogenes, DNA synthesis and repair genes, ion channel and transport protein genes, DNA-binding, transcription and transcription factor genes, and cytoskeleton genes. There were 4 genes up-regulated and 18 down-regulated. Real-time RT-PCR results of AK1 and FBX07 were consistent with that of microarray analysis. Conclusion: Microarray expression profile of PBMC in type 2 diabetes patients with DSPN is differential. DSPN may be involved in cell metabolism, signal transduction, DNA synthesis, etc.

10.Expression of pro-inflammatory cytokine mRNA after spinal cord injury and effect of the methylprednisolone in rats
Chinese Journal of Rehabilitation Theory and Practice 2005;11(2):91-93
ObjectiveTo investigate the expression of pro inflammatory cytokine mRNA including interleukin 1β (IL-1β), interleukin 6 (IL-6) and tumor necrosis factor-α(TNF-α) after spinal cord injury (SCI), and the effect of methylprednisolone (MP) on the expression of the cytokine and neurological outcome after SCI in rats.MethodsA moderate SCI model was made with SD rats according to modified Allen's method. 56 SD rats were divided randomly into the sham group (n=4), SCI group (n=24), saline group (n=14) and MP group (n=14, MP 30 mg/kg injected throuh tail vein 30 min after injury). Samples of injured spinal cord were taken at 1 h, 4 h, 12 h, 24 h, 72 h and 7 day after SCI. The mRNA expressions of IL-1β,IL-6 and TNF-α in the spinal cord of rats were measured by semi-quantitative RT-PCR at 1 h, 4 h, 12 h, 24 h, 72 h and 7 d post-injury, and 4 h post-operation in the control group. The effect of MP (30 mg/kg, iv, within 30 minutes post-injury) on the expression of cytokine at 4 h post-injury was tested with RT-PCR. The neurological outcome of rats was evaluated with BBB scores.ResultsThe expression of the cytokine mRNA was little in the sham group and significantly increased in injured groups with IL-1β at 12 h, TNF-αat 4 h reaching peak after injury, and higher than that in sham group until 72 h (P<0.05). While, the expressions of IL-1β and TNF -α mRNA were inhibited obviously in MP group at 4 h after injury, but MP had no effect on the neurological outcomes.ConclusionMP can inhibit inflammatory reaction obviously.
