2.Investigation of H antigen on AB subgroup RBCs

Objective To find the rule of the distribution of H antigens on AB subgrouperythrocytes.Methods ABO subgroups were confirmed by using serological and molecular biology (PCR-RFLP) methods. AB subgroup with strong H was defined as red cell agglutination by anti-H of 2 scores or more higher than that of B cells.Results Strong H was only found in certain AB subgroups ,CisAB(100%),B(A)(100%), AxB(46.2%) and A2B(43.6%), but seldom in others among Chinese population.Conclusion The fact that H type-3, which comes from A type-2, can hardly be transferred by B and weak A glycosyltransferase can help to explain why some ‘strong’ H combines with ‘weak’ A in AB erythrocytes. Why only little H can be found in 53.8% AxB, 56.4% A2B and all A3B, AmB subgroup samples still cannot been explained.
3.Caveolin-1 expression is downregulated by shear stress and TNF-? in human aortic endothelial cells

AIM: To observe effects of shear stress and TNF-? on caveolin-1 expression. METHODS: Cultured human aortic endothelial cells (HAECs) of passage 3-5 were used in the experiment. Cells were exposed to a laminar flow (shear stress 1.0 Pa) by using a parallel rectangular flow chamber for different time. Caveolin-1 mRNA and protein expression were measured by RT-PCR and Western blot, respectively. Caveolin-1 expression of the cells stimulated by TNF-? were also studied to elucidate the influence of this inflammatory factor. 4.The Role and Mechanism of MiR-130b-5p in Down-regulating IGF-1 and Inhibiting the Migration and Invasion of Human Chorionic Trophoblast Cells

ObjectiveTo explore the effects of microRNA 130b-5p (miR-130b-5p) on the migration and invasion of human chorionic trophoblast cells (HTR8/SVneo) by targeting insulin-like growth factor-1 (IGF-1). MethodsHTR8/SVneo cells were divided into control group, miR-NC group, miR-130b-5p mimics group, miR-130b-5p mimics+pcDNA3.1 group and miR-130b-5p mimics+pcDNA3.1-IGF-1 group. The dual luciferase reporter gene was used to detect the targeting relationship between miR-130b-5p and IGF-1; real-time fluorescent quantitative PCR was used to detect the expression level of miR-130b-5p and IGF-1in HTR8/SVneo cell samples; the CCK-8 method was used to detect the proliferation of HTR8/SVneo cells; holographic cytometer and Transwell method was used to analyze the migration and invasion abilities of HTR8/SVneo cells; Western blot method was used to detect the expression of IGF-1, invasion, and epithelial-mesenchymal transition (EMT) proteins in HTR8/SVneo cell samples. ResultsThe results of the dual luciferase reporter gene showed that IGF-1 was a potential target gene of miR-130b-5p; compared with the miR-NC group, the expression level of miR-130b-5p, the cell proliferation inhibition rate, and the expression level of E-cadherin increased significantly in the miR-130b-5p mimics group. The expression levels of IGF-1, c-Myc, Cyclin D1, migration distance, number of invaded cells, the expression levels of matrix metalloproteinase 9 (MMP9), matrix metalloproteinase 2 (MMP2), neural cadherin (N-cadherin) and Vimentin were significantly reduced (P<0.05); compared with the miR-130b-5pmimics+pcDNA3.1 group, the cell proliferation inhibition rate and the expression level of E-cadherin in the miR-130b-5p mimics+pcDNA3.1-IGF-1 group were significantly reduced, while the expression levels of c-Myc and CyclinD1, migration distance, number of invasive cells, the expression levels of MMP9, MMP2, Vimentin and N-cadherin were significantly increased (P<0.05). ConclusionThe overexpression of miR-130b-5p may inhibit the migration and invasion of HTR8/SVneo cells by inhibiting the expression of IGF-1.

6.Effect of saikosaponin-d on rat's glomerular mesangial cell proliferation and extracellular matrix hyperplasia in vitro.

OBJECTIVETo investigate the effects of saikosaponin-d (SSd) on glomerular mesangial cells (MCs) proliferation and hyperplastic extracellular matrix (ECM) induced by lipopolysaccharide (LPS) to provide experimental proof for its use in prevention and treatment of glomerulosclerosis.
METHODSRat's MCs were cultivated and identified. The cultured MCs were stimulated by LPS and incubated with different concentrations of SSd. Cell proliferation was determined by MTT assay, LDH assay and flow cytometry, respectively. Type IV collagen (Col IV), fibronectin (FN) and transforming growth factor beta1 (TGF-beta1) in the conditioned medium were measured by ELISA. The expressions of cyclin-dependent kinase 4 (CDK4), c-Jun and c-Fos were detected by immunohistochemistry.
RESULTSAfter treated by SSd, MC proliferation was inhibited, cells in G0/G1 phase increased, and apoptosis induced. Moreover, secretion of Col IV, FN and TGF-beta1 and the expressions of CDK4, c-Jun and c-Fos in MC were inhibited.
CONCLUSIONThe inhibitory action of SSd on glomerulosclerosis was realized through inhibiting the expressions of CDK4, c-J un and c-Fos.

7.Study on B(A) phenotypes and identification of novel B(A)641 allele in chinese population

C mutation. All 8 samples displayed the B(A) phenotype. Their real genotypes were B(A)/O. Conclusion Three B(A) alleles in the Chinese Han population were detected. Two alleles,B(A)700,B(A)640 were reported previously. One novel allele B(A)641, was first identified in this study.
8.Cause analysis and control measures of insufficient blood collection

Objective To analyze the causes of blood deficiency in the process of voluntary blood donation and to adopt targeted control measures,so as to effectively reduce blood scrapping and to better ensure theclinical blood use of the hospital.Methods The data of blood collection from January 2014 to December 2016 and the various reasons of insufficient blood collection were summarized;and according to these data as the object of study,the targeted measures were taken to observe the effect.Results According to the the reasons for the lack of blood collection,the targeted measures,such as organizing staff training,strengthening communication with blood donors and so on,were taken.After the implementation of these measures,the phenomenon of insufficient blood collection from 2014 to 2016 showed declining trend,withthe proportion decreased from 0.29% to 0.20%.Conclusion To strengthen the staff staining in order to improve them vein collection technology,to publicize further,to communicate with blood donors effectively and improve the blood donation services,to ease the feelings of blood donors and to create a warm,harmonious and orderly blood donation atmosphere;all of these should be helpful for reducing the occurrence of insufficient blood collection.

10.Dorsal root reflex from Adelta and C afferent fibers induced by electrical stimulation of the sural nerve in rats.

The purpose of the study was to investigate whether the small fiber-evoked dorsal root reflex (DRR) can be obtained by electrical stimulation of the peripheral nerve in rats. Fifty-one DRRs were recorded from different kinds of fiber filaments in the proximal ends of the cut L(5) dorsal root following electrical stimulation of the sural nerve. According to the kind of afferent fibers in the sural nerve associated with the DRRs from dorsal root, these DRRs were divided into five different types: A(alphabeta) fiber-evoked A(alphabeta).DRR (A(alphabeta)- A(alphabeta).DRR), A(betadelta) - A(delta ).DRR, A(betadelta)-C.DRR, A(alphabetadelta)-C.DRR and C-C.DRR. The results obtained show that the DRR can be obtained from either A-fibers (including A(delta )-fibers) or C-fibers of dorsal root filaments by stimulation of the sural nerve. It is therefore suggested that either A(delta ).DRR or C.DRR can be used as a validity index of presynaptic inhibition of the thin primary afferent terminals for investigation of the modulation mechanisms of peripheral effectors.
